Abstract
Description
The invention relates to a process for the enzymatic breakdown of the pollen envelopes of flowers made up of cellulose in order to thereby obtain an increased release of the immediate pollen principles.
It is known that flower pollens have always occupied a solid position in folk medicine, properties beneficial to health being generally attributed to them by oral administration.
However, it is also known that most flower pollens in European flora have an extremely stable cellulose envelope that cannot be dissolved or ruptured in the human gastrointestinal tract.
This is why, we have also tried, by mechanical treatment, for example by micronization, to obtain an improved bioavailability of the immediate principles of pollen.
However, by comparing by thin layer chromatography the amounts of free amino acids in the case of nan pollens treated and micronized, there are no significant differences.
EP-A-0069 995 discloses a process which provides for the enzymatic attack on pollen from flowers. It is proposed there, inter alia, to act on enzymes such as for example cellulase for 1 to 14 days on the walls of the pollen grains.
This process is very long and provides a product which is not satisfactory as regards the sensory properties. In addition, a verification of the example of the aforementioned prior publication in which cellulase is used, leads to a product having an insufficient content of amino acids.
If one can attribute to the constituents of pollen beneficial properties for health, there is a need for the release, in an optimal way, of the constituents of pollen from flowers by an effective rupture.
We have now found that this problem can be resolved by an enzymatic biological attack on the cellulose envelopes of flower pollens using enzymes capable of degrading the cellulose, preferably cellulases, by treating an aqueous suspension of pollen. flowers for less than 24 hours.
With cellulase - MKC 4000 (manufacturer, Miles Kali
Chemie) commercially available, it is already possible, after an incubation time of 2 hours, to demonstrate using various parameters a clear rupture of the pollen.
The mixture of enzymatically broken pollen substance and water can then be transformed, in a manner known per se, into a stable pulverulent product by controlled dehydration processes such as spray drying or lyophilization.
de préférence 10 à 200 g/kg.For the process in accordance with the invention, the following operating parameters, taken individually or in combination, have proven to be suitable - enzyme concentration: 2 to 200 g of enzyme / kg of pollen,
preferably 10 to 200 g / kg.
- température : 10 à 70"C, de préférence 20 à 50"C. - temperature: 10 to 70 "C, preferably 20 to 50" C.
- durée inférieure à 24 heures, de préférence de 0,5 à
20 heures, en particulier de 1 à 16 heures.- duration less than 24 hours, preferably from 0.5 to
8 p.m., especially 1 a.m. to 4 p.m.
The product obtained according to the process according to the invention is characterized by at least one of the following parameters which can be determined analytically - fraction of free amino acids (determined using a
amino acid analyzer) of at least 3.5% by weight,
preferably at least 4% by weight relative to the preparation
pollen from dried flowers.
poids au maximum.- aminogram (related to the overall content of amino acids
free) - proline: 60% by weight maximum, preferably 55% by
maximum weight.
- remaining amino acids: at least 40% by weight, preferably
at least 45% by weight.

For the detection of the enzymatic breakdown of pollen, the following samples were subjected to a comparative examination: sample l: preparation of pollen from Example 1 under attack
enzymatically, in accordance with the invention
tion, sample 2: commercial micronized pollen preparation, sample 3: small grains of natural pollen ground
mortar, not otherwise treated, sample 4: preparation obtained according to the operating mode
of example 3 of EP-A-0069 995
Determination by thin layer chromatography of increased amounts of released amino acids
Ready-to-use "Merck" silica gel plates were used.

The samples of the cited pollen preparations were suspended in water (1: 10) and chromatographed at 37 "C after an incubation time of 15 minutes.
Unlike samples 2 and 3, the pollen having undergone an enzymatic attack (sample 1) revealed, after development and detection, an increase in the number of amino acid bands.
Also, the bands of amino acids present in the natural state are they considerably reinforced as regards their intensity (see FIG. 1 where the thin layer chromatogram has the following Rf values: 0.53, 0.41, 0, 33, 0.25, 0.18, 0.13, 0.03.
On the other hand, the micronized preparation on the market had approximately the same chromatogram poor in amino acid bands as the preparation of natural pollen.
Gravimetric characterization of solvent extraction of pollen constituents
The samples of the pollen preparations considered were suspended in water at 37 "C (1:10) and filtered after 15 minutes. The weight was determined after washing the filtration residue and drying at 70" C until '' at constant weight.
In the case of the preparation of broken pollen, prepared according to the invention, an 80% fraction of water-soluble pollen constituents was determined.
On the other hand, the micronized pollen-based product on the market contained only a water-soluble fraction of approximately 60%, of the order of magnitude of the untreated pollen.
Gravimetric characterization of the increased quantities of lipid bodies released and extractable by chloroform
The samples of the cited pollen preparations were suspended in water at 37 "C (1:10) and extracted with chloroform. After drying the chloroform phase with anhydrous sodium sulfate, the extraction was removed by evaporation and the residue was dried to constant weight.
The pollen preparation attacked according to the invention offered, compared to the commercial micronized preparation and the natural form of pollen, an increase in the quantity of lipids released by approximately 50%.
Amino acid analysis
Sample 1a (corresponding to Example 3) and Sample 4 were treated according to the following operating mode and then analyzed on the LC 2000 automatic amino acid analyzer from the company Biotronik.
