Abstract
La présente invention concerne une méthode pour déterminer le glutamate oxalacétate transaminase ou glutamate pyruvate transaminase dans un fluide biologique, où du L-glutamate est produit à partir d'alpha-cétoglutarate par transamination en présence de la transaminase dans le fluide, et le L-glutamate est désaminé par oxydation en présence de glutamate déhydrogénase avec production simultanée de béta-nicotinamide adénine dinucléotide réduit en une quantité proportionnelle à la teneur de la transaminase dans le fluide, ce fluide contenant également une substance endogène oxydée en présence de glutamate déhydrogénase avec production simultanée de béta-nicotinamide adénine dinucléotide réduit, interférant ainsi avec la détermination. Selon l'invention, on obvie à l'interférence provoquée par la substance endogène en oxydant cette substance en présence de glutamate déhydrogénase avec réduction simultanée de béta-nicotinamide adénine dinucléotide, on oxyde le béta-nicotinamide adénine dinucléotide réduit résultant en présence de lactate déhydrogénase avec réduction simultanée de pyruvate en lactate, et on inhibe ensuite l'activité de lactate déhydrogénase, ensuite on produit une certaine quantité de L-glutamate à partir de l'apha-cétoglutarate par la transamination pour la désamination par oxydation et production simultanée de bétanicotinamide adénine dinucléotide réduit en une quantité proportionnelle à la teneur de la transaminase dans le fluide. L'invention s'applique notamment à l'analyse du sérum du sang.
