Abstract
(A) Acyl derivs. of antibiotic WS-4545 of formula (I) are new. (in which R1 is acyl and R2 and R3 are H or acyl, the acyl gps. being selected from 2-10C alkanoyl interrupted by an S atom, 2-10C alkenoyl, 3-methoxycarbonyl-propionyl, 3,4,5-trimethoxybenzoyl, 2-(3,4,5-trimethoxy)phenylacetyl, phenylacetyl, p-nitrobenzoyl, phenoxyacetyl, thienylacetyl or tetrahydro-2-thenoyl). (B) (Not claimed). Cpds. of formula (I) in which the acyl gp (S). are derived from aliphatic, aromatic or heterocyclic carboxylic acids. This is an Addition to 773719. (I) are antibiotics partic. effective against Gram negative bacteria, e.g. Escherichia and Salmonella spp. They give higher concns. after oral administration than WS-4545. Urinary excretion of (I) after oral administration to rats is 40.6-73.1% over 24 hrs., whereas for WS-4545 it is 24.1%. The daily dose of (I) is 0.5-5 (pref. 1-2)g. The acyl gps. are 3-vinylpropionyl, 3-methylcrotonyl, methylthioacetyl, ethylthioacetyl, isopropylthioacetyl, 2-methylthiopropionyl; t-butylthioacetyl, 2-t-butylthiopropionyl and the gps. mentioned specifically.
