Abstract

Estimation of uric acid in biological liquids, esp. blood, by addin uricase and catalase, aldehyde-free methanol and pH 8 buffer to the liquid to be assayed, then adding 3-methyl-benzothiazolin-2-one hydrazone in pH plus-or-minus 0.05 buffer and FeCl3 in HCl to give a coloured aqueous solution, which is estimated for intensity of colour and compared with the colour produced by a standard reference sample of uric acid. Reagents are stable for long periods and do not require to be freshly made before the test. Uric acid is converted by uricase into allantoin, CO2 and H2O2, the latter oxidizing methanol into formaldehyde in presence of catalase. The HCHO condenses with the hydrazone which is added, and the derivative gives a blue colour in presence of FeCl3.
