Abstract
Quinuclidinyl and quinuclidinylalkyl esters with cholinolytic, antihistaminic and neurosedative activity. G3A. are new cmpds. of formula (I) including their salts and quaternary ammonium salts, (where R1 and R2 are phenyl, halophenyl, 1-4C alkyl, 1-4C alkoxy or CF3, R is H or 1-4C alkyl and x is a number such that there are n carbon atoms between O in the side-chain and N in the quinuclidine ring, n being 2 or 3; provided that when x = 0, n is 2 and R is H). Prepared (a) by condensing (II), or a salt of (II) with a strong acid, with Y-CHR1R2 (where Y is halogen or a reactive ester residue), opt. in the presence of a basic condensation agent, to give (I; R=H); (b) condensing a 2- or 3- quinuclidinol (III) with a diazo cpd. N2=CR1R2 to give (I; R=H, x = 0); or (c) by condensing (IV) (where X is halogen) with the product may be converted into a salt or quaternary ammonium salt by reacting with an acid or an alkylating agent.
