Abstract

Un procedimiento de preparación de 6-ciclopropil-1,3-dihidro-5-fenil-2-oxo-2H-tieno {2,3-e} diazapina, caracterizado por: (a) hacer reaccionar ciclopropilmetilcetona con benzoilacetonitrilo en benceno, en presencia de ácido acético, para formar 2-benzoil-3- ciclopropil-crotonitrilo; (b) tratar este último compuesto con azufre en etanol, en presencia de trietilamina, para obtener 2-amino-3-benzoil-4-ciclopropil-tiofeno; (c) hacer reaccionar bromuro del ácido bromoacético con este último compuesto, en éter y en presencia de piridina, a la temperatura ambiente, para formar 3-benzoil-2-alfa- bromoacetamido-4-ciclopropil-tiofeno; (d) aminar este último con amoniaco en tetrahidrofurano para obtener 2-alfa-aminoacetamido-3-benzoiil-4-cicloporpil-tiofeno y (e) ciclar este último mediante calentamiento a reflujo en tolueno, en presencia de ácido acético, para obtener 6-ciclopropil-1,3-dihidro-5-fenil-2-oxo-2H-tieno {2,3-e} diazapina.
