Abstract

Reaction scheme is: acid hydrolysis right arrow pH max. 5.0 -R-COOH=aminoacid residue R3 = alkyl (C1-3) R4=H or alkyl (C1-3) or R3+R4=cyclopentyl or cyclohexyl R1 and R2 = alkyl (C1-3) or R1+R2+O=morpholino 6-APA must be reacted with an N-protected amino-acid (III) or its functional deriv. at 0 degree to 30 degrees C in the presence of an anhydrous inert solvent to give (IV) which is hydrolysed at room temp. with an acid at pH of 5.0 or less to give (I). (III) is most preferably prepared by reacting an aminoacid or its salt with an N,N-disubstd. beta-ketoacid: R3COCHR4CONR1R2. The process give 6-aminoacylaminopenicillanicacids in much higher yields of high purity.
