Abstract
Conjugado de oligonucleótido-fase sólida, en el que la fase sólida comprende un metal tiofílico, en el que el oligonucleótido comprende por lo menos una tiooxonucleobase según la fórmula I:**Fórmula** en la que X es CH o N, en la que R1 es H o NH2, en la que --- indica un enlace covalente, y en el que dicho oligonucleótido se une a dicha fase sólida mediante el átomo de azufre de dicho tiooxonucleótido.

6.2 Procedimiento general para la preparación de conjugados de oligonucleótido-nanopartícula de oro que utilizan tiooxo-nucleósidos como anclaje molecular.
5 Se preparó una solución de nanopartículas de oro de 15 nm a partir de una solución de HAuCl4 mediante reducción de citrato según el protocolo informado por Turkevich y posteriormente descrito por Letsinger y Mirkin (Mirkin C.A. et al., Nature 283:607-609, 1996; Elghanian R. et al., Science 277:1078-1081, 1997; Turkevich J. et al., Faraday Soc. 11:55-75, 1951). Las nanopartículas de oro (~3 nM) se funcionalizaron con los oligonucleótidos 8 y 9, y 32 a 37 que contenían uno de los tiooxonucleósidos 1, y 26 a 28 en los extremo 5' (Tabla 2). Se prepararon los conjugados de
10 ADN-NPAu mediante la mezcla de 1 ml de la solución de nanopartículas de oro con soluciones purificadas de oligonucleótidos (concentración final de oligonucleótido: 3 μM). La reacción de acoplamiento se llevó a cabo a temperatura ligeramente elevada (40ºC). Tras dejar en reposo durante 20 h, se añadieron 5 ml de una solución tampón de NaCl 2 M, fosfato 0,2 mM (pH 7,0) bajo agitación constante para incrementar la concentración de NaCl de la solución de nanopartículas a 0,01 M. Las soluciones se incubaron durante 6 a 8 h a 40ºC. Este procedimiento se
15 repitió tres veces para incrementar la concentración de NaCl de la solución de conjugado de nanopartículas escalonadamente de 0,02 M, 0,05 M y finalmente a 0,1 M de NaCl. Entre medio se dejó en todos los casos que las soluciones envejeciesen durante 6 a 8 h a 40ºC. A continuación, se centrifugaron las soluciones de ADNnanopartículas de oro (8.000 rpm) y se separó el sobrenadante transparente para eliminar los oligonucleótidos no unidos. El precipitado se redispersó en 1 ml de una solución tampón de NaCl 0,1 M, fosfato 10 mM (pH 7,0). Tras la
