Abstract

Producción en la levadura Pichia pastoris, y sistema de aislamiento y purificación, del alergeno recombinante de Olea europaea Ole e 9 para uso en diagnosis y tratamiento de alergias. Se ha clonado y expresado el DNA recombinante que codifica el alergeno Ole e 9 de polen de olivo (Olea europaea). Este alergeno tiene actividad b-1,3-glucanasa y presenta una alta homología con estas proteínas. Se han detectado también proteínas homólogas a este alergeno en otros pólenes como el de otras miembros de la familia de las Oleaceas (Syringa vulgaris, Ligustrum vulgare y Fraxinus excelsior). La proteína recombinante producida en la levadura (Pichia pastoris) se aísla con gran rendimiento, está correctamente plegada y exhibe propiedades inmunológicas equivalentes a las del alergeno natural por lo que tanto ella como las proteínas homólogas, las formas modificadas derivadas de ellas, los fragmentos derivados que contengan al menos un epítopo alergénico, pueden ser empleadas en protocolos de diagnosis einmunoterapia.
