Abstract
Chimeric gene that uses the insulin gene or CDNA, especially for diabetes gene therapy. The chimeric gene is directed by a promoter or fusion of promoters, which are preferably adjustable and activated by the diabetic process. Preferably, it is obtained by fusion of the human insulin gene to the PEPCK promoter (p-enolpyruvate carboxykinase). Said promoter (fragment - 460 bp at + 73 bp) is fused to the 5 '' free zone of the human insulin gene (-170 bp to +1). The human insulin gene contains two exons encoding E1 and E2 and two introns A and B. It also refers to an expression vector that allows insulin to be expressed in cells other than the callae {be} of the pancreas and a non-human transgenic animal that expresses the said chimeric gene. It is especially used for gene therapy of diabetes.
