Abstract
Chimeric gene using the insulin gene or cDNA, especially for gene therapy in diabetes.The chimeric gene is directed by a promoter or fusion of promoters, which are preferably regulated and activated by the diabetic process.Preferably it is obtained by fusion between the human insulin gene and the PEPCK promoter (P-enolpyruvate carboxykinase). Said promoter (fragment - 460 pb to + 73 pb) is fused with the 5' franking region of the human insulin gene (-170 pb to +1). The human insulin gene contains two exons encoding E1 and E2 and two introns, A and B. It also refers to a vector of expression which enables the expression of insulin in cells other than the [beta] cells of the pancreas and to a non-human transgenic animal which expresses the aforementioned chimeric gene. Notably for use in gene therapy for diabetes.< IMAGE >The gene for gene therapy of diabetes comprises the fusion product of the human insulin gene with PEPCK (p-enolpyruvate cosboxyquinase) promoter. A vector for expression of insulin in different cells from the pancreas is also disclosed.
