Abstract

Un método de ensayo de un ligando en una muestra, el cual método comprende las etapas de (a) incubar la muestra en contacto con un compañero de unión específica para el ligando que se desea detectar, estando dicho compañero de unión específica soportado sobre una superficie de una estructura óptica y en presencia de otro reactivo que es o bienun análogo del ligando marcado fluorescentemente o fosforescentemente, específico para el mismo compañero de unión específica, o bienotro compañero de unión específica marcado uorescentemente o fosforescentemente, para el ligando que se desea detectar, comprendiendo dicha estructura óptica un sustrato, una capa de metal, e, interpuesta entre ellas, una capa de material dieléctrico que tiene un índice de refracción inferior al de dicho sustrato, estando dicho compañero de unión específica directa o indirectamente adsorbido sobre o unido a dicha capa de metal; (b) irradiar otra superficie de dicha estructura óptica a un ángulo o margen de ángulos respecto de la normal adecuado, de forma que tenga lugar resonancia de plasmón de superficie de largo alcance y de forma que se genere fluorescencia o fosforescencia; y (c) analizar la fluorescencia o fosforescencia generada para determinar si está alterada por la formación de complejo, y, si se desea, en qué cuantía y a qué velocidad tiene esto lugar.
