Abstract
Ferner betrifft die vorliegenden Erfindung ein Testsystem (10) zum Nachweis einer Infektion mit Legionellen, sowie der Verwendung eines Testsystems (10) in einem Verfahren zum Nachweis von Legionellen. Außerdem betrifft die Erfindung ein Verfahren zum Nachweis einer Infektion mit Legionellen.

Die vorliegende Erfindung betrifft ein Testsystem zur Erkennung von Legionellen. Ferner betrifft die vorliegende Erfindung ein Testsystem zum Nachweis einer Infektion mit Legionellen, sowie der Verwendung eines Testsystems in einem Verfahren zum Nachweis von Legionellen. Außerdem betrifft die Erfindung ein Verfahren zum Nachweis einer Infektion mit Legionellen.
Zum Nachweis der Legionellose muss derzeit eine spezifische Erregerdiagnostik durchgeführt werden, da das Krankheitsbild unspezifisch ist und somit keinen Schluss auf den Erreger zulässt. Nach dem Stand der Technik können Legionellosen durch eine Reihe von Verfahren nachgewiesen werden.
Der Nachweis der Legionellen erfolgt derzeit hauptsächlich (in 73% der Fälle) über den immunologischen Nachweis von Lipopolysaccharid (LPS). Bei diesen Tests ist die Sensitivität mit 75% jedoch nicht zufriedenstellend, sodass bei negativem Ergebnis eine Legionellen- Infektion nicht zuverlässig ausgeschlossen werden kann. Weiterhin können die am Markt erhältlichen Tests nur die Infektion mit Legionella pneumophila der Serogruppe 1 nachweisen. Legionella pneumophila ist zwar bei über 90% der Fälle der Verursacher von schweren Legionellenkrankheiten, jedoch ist die Serogruppe nur bei zwei Dritteln der Fälle ursächlich. Insbesondere bei nosokomialen Infektionen handelt es sich oft um Stämme anderer Erregergruppen, die von diesem Verfahren nicht detektiert werden können.
Alternativ werden PCR-basierte Tests aus Atemwegsproben (meist Bronchiallavage) durchgeführt. Obgleich die Sensitivität und Spezifität hoch sind, sind diese diagnostischen Methoden jedoch aufwändig, teuer und durch die invasive Probennahme mit einem erheblichen Risiko für eine nachfolgend erforderliche Intubation der Patienten nur eingeschränkt einsetzbar. Zudem bedarf es hochqualifiziertes und geschultes Personal zum Nachweis der Legionellen über PCR, sodass diese Verfahren hauptsächlich für den Klinikbereich in Frage kommen.
Serologische Tests weisen Antikörper gegen Legionellen im Serum des Patienten nach. Diese werden allerdings erst nach 3 bis 10 Wochen nach Ansteckung nachweisbar. Aufgrund der geringen Sensitivität ist das Verfahren für die Schnelldiagnostik nicht geeignet.
Andere Verfahren zum direkten Erregernachweis (Mikroskopie, Kultur) spielen nur eine untergeordnete Rolle und sind durch aufwändige Probennahme, Durchführung bzw. lange Kulturdauer (mehrere Tage) von eingeschränktem Nutzen.
Laut eines Überblicks zur Legionellose des Robert-Koch-Instituts von Ende März 2015 ist der Antigen-Test aus dem Urin der am häufigsten verwendete Test (73,1%), gefolgt vom PCR Verfahren (12,8%), dem serologischen Nachweis (9,3%) und dem direkten Erregernachweis in Kultur mit 4,4% (Zugrunde lagen 841 Nennungen bei 806 Fällen).
Insgesamt ist festzustellen, dass für den zuverlässigen und schnellen Nachweis einer Legionellen-Infektion kein geeigneter Test für eine klinische Anwendung oder Diagnose beim Hausarzt zur Verfügung steht.
Die Legionärskrankheit (Legionellose) ist eine schwere und unbehandelt oft tödliche, bakterielle Lungenentzündung, die allerdings nur einen geringen T eil der atypischen Pneumonien darstellt. Zum Nachweis der bakteriellen Erreger, der Legionellen, soll ein Diagnostikverfahren auf der Grundlage eines Lateral-Flow-Tests (LFT) entwickelt werden, der die bisherigen diagnostischen Verfahren hinsichtlich der Sensitivität und Spezifität, wie auch der Testschnelligkeit übertrifft. Die bislang bei Pneumonien veranlasste und aufgrund der Seltenheit von Legionellosen unnötige Gabe von Makrolid- und Fluorchinolon-Antibiotika soll somit vermieden werden. Als Probenmaterial soll Speichel wegen der einfachen, nicht invasiven Verfügbarkeit verwendet werden. Dadurch kann u.a. die Ausbildung von Resistenzen verringert werden, sowie die Diagnostik bereits beim Hausarzt unterstützt werden.
Die Legionellose ist eine durch das Bakterium Legionella pneumophila verursachte schwere Form der Lungenentzündung. Jährlich werden in Deutschland etwa 200.000 Patienten mit Lungenentzündung im Krankenhaus behandelt. Diese werden standardmäßig hinsichtlich einer Legionellen-Infektion untersucht. Bis zum Vorliegen eines Ergebnisses werden vorbeugend Breitbandantibiotika verabreicht. Ziel des Projektes ist die Entwicklung eines zuverlässigen Schnelltests für den Nachweis von Legionellen in Speichel. Die vorliegende Erfindung soll ein schnelles Testresultat bei einfacher Handhabung und nicht-invasiver Probenahme liefern. Einsatzgebiet sollen auch Länder mit schwacher medizinischer Infrastruktur sein.
Es ist die Aufgabe der vorliegenden Erfindung ein Testsystem für den Nachweis einer Infektion mit Legionellen bereitzustellen, insbesondere ein Testsystem, welches einen zuverlässigen und schnellen Nachweis erlaubt.
Die Aufgabe wird erfindungsgemäß gelöst durch ein Testsystem mit den Merkmalen des Anspruchs 1. Danach ist vorgesehen, dass ein Testsystem bereitgestellt wird, zur Erkennung der Legionärskrankheit.
Die Legionärskrankheit kann alternativ als Legionellose bezeichnet werden.
Die Erfindung beruht auf der grundlegenden Idee, dass eine prophylaktische Vergabe von Antibiotika gegen die Legionellose, oder die Gabe von Antibiotika wegen falsch negativer Testergebnisse von unzuverlässigen Tests, verringert und/oder vermieden werden soll und somit der Entstehung von Antibiotikaresistenzen zumindest teilweise entgegengewirkt werden kann.
Das Testsystem kann alternativ und/oder zusätzlich für den Nachweis einer Infektion eines Subjekts mit Legionellen eingerichtet sein. Ein Subjekt kann dabei ein Säugetier, insbesondere ein Mensch sein. Insbesondere kann das Subjekt ein Patient sein. Insbesondere ist es möglich, dass das Testsystem auch eine Infektion eines Subjekts vor dem Ausbrechen der Erkrankung (Legionellose) nachweist. Auch hier ist ein wesentlicher Vorteil, dass eine prophylaktische Vergabe von Antibiotika gegen die Legionellose verringert und/oder vermieden werden soll und somit der Entstehung von Antibiotikaresistenzen zumindest teilweise entgegengewirkt werden kann.
In einer Ausführungsform ist oder umfasst das Testsystem einen Lateral-Flow-Test (LFT). Insbesondere ist unter einem Lateral Flow Test eine immunologische Methode zum qualitativen Nachweis von Stoffen mit entsprechenden Bindungspartnern zu verstehen. Ein derartiger Lateral-Flow Test kann auch an jedem erdenklichen Ort durchgeführt werden, insbesondere wird keine Laborumgebung und kein Laborpersonal oder medizinisch geschultes Personal benötigt. Ein Lateral-Flow Test erlaubt damit eine schnelle, einfache und kostengünstige Durchführung eines Tests zum Nachweis einer Infektion mit Legionellen oder zum Erkennen einer Legionellose. Somit wird auch eine Unterstützung des Hausarztes bei der Diagnose
ermöglicht. Die Testdauert von der Probennahme kann unter einer Stunde, insbesondere unter 30 Minuten betragen.
Das Testsystem kann zur Erkennung einer Legionärskrankheit und/oder zum Nachweis einer Infektion mit Legionellen in Speichel und/oder Sputum eingerichtet sein. Somit können eine vorteilhafte, anwenderfreundliche Verwendung des Testsystems oder ein anwenderfreundliches Durchführen des Verfahrens ohne invasive Entnahme einer Probe (z,B. Atemwegsproben, die über Bronchiallavage erhalten werden, Blut oder Lymphe) von dem zu untersuchenden Subjekt erfolgen. Vorteilhaft ist das erfindungsgemäße Testsystem auch gegenüber Testsystemen, welche für den Nachweis von Erregern in Urin und/oder Stuhl eingerichtet sind, denn für die Verwendung des erfindungsgemäßen Testsystems werden keine Toiletten und damit verbundene intime Tätigkeiten benötigt. Denkbar ist generell, dass die Speichelprobe oder Sputumprobe eine frisch entnommene Probe oder eine gelagerte, gekühlte oder gefrorene Probe umfasst.
Ein Einsatz von Blut, Atemswegsproben (insbesondere der unteren Atemwege), Lymphe, Urin und/oder anderen Körperflüssigkeiten und/oder Stuhl ist jedoch alternativ und/oder zusätzlich denkbar und/oder möglich. Insbesondere kann die Probe als unbehandelte Probe oder vorbehandelte Probe eingesetzt werden.
Insbesondere ist es möglich, dass die Erkennung und/oder der Nachweis zumindest teilweise über die Bindung mindestens eines Proteins, insbesondere eines Proteins der Bakteriengattung Legionella, an einen spezifischen Bindungspartner erfolgt. Insbesondere kann das Protein auch als Markerprotein bezeichnet werden. Dadurch wird eine hohe Spezifität des Testsystems erreicht.
In einer Ausführungsform ist das Testsystem ein Testsystem zum Nachweis einer Legoinellose in einer Speichelprobe, wobei das Testsystem einen Lateral-Flow-Test umfasst und wobei der Nachweis des Markers FLA durch das Testsystem für das Vorliegen der Legionellose spricht.
In einer Ausführungsform ist das Testsystem ein Testsystem zum Nachweis einer Legionellose in einer Speichelprobe, wobei das Testsystem einen Lateral-Flow-Test umfasst und wobei der Nachweis des Markers MIP durch das Testsystem für das Vorliegen der Legionellose spricht.
In einer Ausführungsform ist das Testsystem ein Testsystem zum Nachweis einer Legionellose in einer Speichelprobe, wobei das Testsystem einen Lateral-Flow-Test umfasst und wobei der Nachweis des Markers PAL durch das Testsystem für das Vorliegen der Legionellose spricht.
Es können auch zwei oder mehr Marker in einem Testsystem vereint sein.
Ein Protein kann dabei ein Peptid, Lipoprotein, Glykoprotein, Peptidoglycan, Filament, Antigen und/oder Epitop umfassen.
Generell ist es möglich, dass das mindestens eine Protein ein Protein der Art Legionella pneumophila, Legionella micadei, Legionella bozmanii, Legionella dumoffii und/oder Legionella longbeachae ist. Insbesondere können auch verschiedene und/oder alle (der etwa 57) Serotypen von Legionella pneumophila mit dem Testsystem erkannt und/oder nachgewiesen werden, insbesondere die Serotypen 1, 4 und/oder 6. Das hat den Vorteil, dass nicht nur eine Erkrankung/Infektion, die auf Legionella pneumophila Serotyp 1 - den häufigsten Verursacher für die Legionärskrankheit - zurückzuführen ist, nachgewiesen und/oder erkannt werden kann. Somit werden falsch negative Ergebnisse, durch Tests, welche spezifisch für nur einen Serotyp sind, vermieden.
Denkbar ist, dass das mindestens eine Protein oder auch mehrere Proteine Makrophage inflammatory Protein (MIP und/oder ( Legionella pneumophila) peptidoglycan-assoziiertes Lipoprotein (PAL) und/oder Flagellin (FLA) umfasst.
Macrophage inflammatory protein (MIP) ist ein 24-kDa Protein (pl 9.8). MIP kann MIP-1 alpha und/oder MIP-1 beta umfassen.
(Legionella pneumophila) Peptidoglycan-assoziiertes Lipoprotein (PAL) ist eines der am höchsten konservierten Proteine von Legionellen, und erreicht in diagnostischen Anwendungen Sensitivitäts- und Spezifitätswerte von ca. 90%.
Flagellin (FLA) ist ein Bestandteil des Flagellums von Legionella pneumophila und auch an der Invasion von Wirtszellen beteiligt.
Denkbar sind generell auch andere Markerproteine hoher Konservierung, Prävalenz und Immunogenität für den Nachweise einer Infektion mit Legionellen und/oder die Erkennung der Legionärskrankheit verwendet werden. Insbesondere sind Oberflächenproteine denkbar.
In einer Ausführungsform umfasst das Testsystem ein Testelement mit mikrofluidischen Strukturen, vorzugsweise einen chromatographischen Teststreifen. Das Testelement umfasst gegebenenfalls einen ersten Bereich, der zur Absorption von Elutionsmittel eingerichtet ist, einen zweiten Bereich, der für die Aufbringung einer Probe eingerichtet ist, einen dritten Bereich, der für den Nachweis, vorzugsweise für den optischen Nachweis, des Proteins, insbesondere von MIP, PAL und/oder FLA, eingerichtet ist, optional einen vierten Bereich, der für die Absorption überschüssigen Elutionsmittels eingerichtet ist. Das Testsystem kann ferner wahlweise ein Gehäuse umfassen. Ein derartiges Testsystem hat den Vorteil, dass es sehr kompakt ist und die Anwendung des Tests somit erheblich erleichtert wird. Das Gehäuse schützt zusätzlich vor äußeren Störfaktoren wie Beschädigungen des Tests oder Kontaminationen. Zudem ist nur ein sehr geringes Probevolumen von 1-20 pl Speichel, insbesondere 2-16 pl Speichel, insbesondere 4 mI Speichel notwendig.
In einer weiteren Ausführungsform kann das Testelement einen weiteren (fünften) Bereich umfassen, wobei der weitere (fünfte) Bereich zumindest vorübergehend den spezifischen Bindungspartner für das Protein, insbesondere MIP, PAL und/oder FLA umfasst.
Das T estsystem und/oder T estelement kann ein poröses Material umfassen, insbesondere ein Fasergewebe aus Zellulose, Glasfaser, Polymerfaser, Nylon, Seide, Wolle, Filz und/oder Baumwoll-Linter und/der einen Schwamm.
In einer weiteren Ausführungsform umfasst das Testsystem ferner ein Kontrollsystem, insbesondere ein optisches Kontrollsystem. Insbesondere kann das Kontrollsystem auch als Kontrolleinheit oder Kontrollelement bezeichnet werden. Das Kontrollsystem kann vorteilhaft anzeigen, ob das Testsystem so verwendet wurde, dass eine Erkennung einer Legionellose und/oder der Nachweis von Legionellen technisch möglich ist. In anderen Worten, die Wahrscheinlichkeit von falschen Ergebnissen aufgrund von falsch durchgeführten Tests unter Verwendung des Testsystems wird verringert. Insbesondere kann das Kontrollsystem den mit dem Markierungselement verbundenen spezifischen Bindungspartner des mindestens einen Proteins umfassen und/oder mindestens einen spezifischen Bindungspartner des spezifischen
Bindungspartners des mindestens einen Proteins. Insbesondere kann das Kontrollsystem mindestens einen Antikörper umfassen.
Das Testsystem kann ferner ein Probennahmeelement umfassen, welches für die Aufnahme einer das Protein enthaltenden Probe eingerichtet ist. Ein Probennahmeelement kann beispielsweise ein Speichelkollektor und/oder Sputum kollektor sein. Somit kann die Erkennung und/oder der Nachweis einer Legionellose und/oder einer Infektion mit Legionellen von der Probennahe bis zur Erkennung/dem Nachweis mit nur einem Testsystem durchgeführt werden, was eine vorteilhafte, sichere, schnelle und einfache Anwendung des Testsystems bedeutet.
Die Erfindung betrifft Ferner die Verwendung eines Testsystems zum Nachweis einer Infektion mit Legionellen.
Ferner betrifft die Erfindung ein Verfahren zum Nachweis von Legionellen in einer Probe, umfassend die folgenden Schritte: a) Bereitstellung eines Testsystems,
(b) Aufbringen der Probe auf ein Testelement, und
(c) Nachweis des Vorhandenseins mindestens eines Proteins, insbesondere MIP, PAL und/oder FLA in der Probe, wobei das Testsystem mindestens einen spezifischen Bindungspartner umfasst, an welchen das mindestens eine Protein, insbesondere MIP, PAL und/oder FLA bindet, wobei der Nachweis zumindest teilweise über die Bindung des mindestens einen Proteins, insbesondere MIP, PAL und/oder FLA, an den spezifischen Bindungspartner erfolgt.
In einer Ausführungsform ist das erfindungsgemäße Verfahren ein Verfahren zum Nachweis einer Legionellose in einer Speichelprobe, wobei ein Lateral- Flow-Test durchgeführt wird, und wobei die Detektion und/oder der Nachweis des Markers FLA für das Vorliegen der Legionellose spricht.
In einer anderen Ausführungsform ist das erfindungsgemäße Verfahren ein Verfahren zum Nachweis einer Legionellose in einer Speichelprobe, wobei ein Lateral-Flow-Test durchgeführt wird, und wobei die Detektion und/oder der Nachweis des Markers MIP für das Vorliegen der Legionellose spricht.
In einer anderen Ausführungsform ist das erfindungsgemäße Verfahren ein Verfahren zum Nachweis einer Legionellose in einer Speichelprobe, wobei ein Lateral-Flow-Test durchgeführt wird, und wobei die Detektion und/oder der Nachweis des Markers PAL für das Vorliegen der Legionellose spricht.
Es ist generell möglich, dass das Verfahren ferner das Lysieren der Bakterien in der Probe umfasst. Dies hat den Vorteil, das Verfahren und/oder das Testsystem ggf. empfindlicher, und/oder sensitiver wird.
Manche bakteriellen Proteine, beispielsweise MIP und PAL sind in/auf der Zellmembran der Bakterien (Legionellen) lokalisiert und teilweise vom Lipopolysaccharid überdeckt. Daher ist der Kontakt mit spezifischen Bindungspartnern am intakten Bakterium sterisch zumindest teilweise erschwert. Das Aufbrechen der Zellmembran (Lysieren) kann die Targets, also die Proteine (Legionellenproteine), an welche die spezifischen Bindungspartner binden sollen besser zugänglich machen.
Generell ist es möglich, dass die Probe eine Speichelprobe oder Sputum umfasst. In anderen Worten, das Testsystem kann zum Erkennen und/oder Nachweis von Legionellen in Speichel und/oder Sputum eingerichtet sein. Die Vorteile eines derartigen Testsystems sind bereits genannt.
Es ist denkbar, beispielsweise in einem weiteren Verfahrensschritt, dass um Abbau der Markerproteine in Speichel zu unterbinden, den Speichelproben Enzym-Inhibitoren zugesetzt werden (analog Fung et al. “Quantitative detection of Pf HRP2 in saliva of malaria patients in the Philippines”, Malaria Journal, 2012, 11:175). Dabei ist beispielsweise der Einsatz eines Inhibitoren-Cocktails aus Aprotinin, PMSF und Natriumorthovanadat denkbar. Denkbar sind generell auch andere Inhibitoren, insbesondere Proteaseinhibitoren.
Der spezifische Bindungspartner kann einen monoklonalen Antikörper, polyklonalen Antikörper affinitätsaufgereinigten Antikörper, ein Antikörperfragment, Peptid, Protein, Nanobody, Nukleinsäurefragment, Aptamer, Anticalin, Lektin, Affibody und/oder chemischen Liganden umfassen, wobei der spezifische Bindungspartner mit einem Markierungselement verbunden ist. In anderen Worten, es liegt ein Konjugat aus spezifischem Bindungspartner und Markierungselement vor. Durch die Verbindung eines spezifischen Bindungspartners für
mindestens ein Protein (Markerprotein) mit einem Markierungselement wird der Nachweis und/oder die Erkennung, insbesondere der optische Nachweis und/oder die optische Erkennung ermöglicht.
Das Markierungselement kann einen Farbstoff, ein Fluorochrom, Fluorophor, Luminophor, Enzym, Peptid, Biotin, Oligonukleotid, Protein-Tag, radioaktives Isotop, nicht radioaktives Nuklid, einen Nanopartikel, insbesondere Gold-Nanopartikel, fluoreszierenden Nanopartikel, magnetischen Nanopartikel und/oder Quantumdot umfassen. Die Fähigkeit eines für ein Markerprotein spezifischen Bindungspartners an ein entsprechendes Markerprotein zu binden und die Fähigkeit eines Markierungselements ein detektierbares und/oder erkennbares Signal zu geben kann also kombiniert vorliegen. Insbesondere kann eine Verbindung eines spezifischen Bindungspartners eines Markerproteins mit einen Nanopartikel, z.B. einem Gold- Nanopartikel, den Vorteil haben, per bloßem Auge erkennbar zu sein.
Ein „Nanopartikel“ bzw. „Nanoteilchen“ kann einen Verbund von wenigen bis einigen tausend Atomen oder Molekülen bezeichnen, dessen Größe typischerweise bei 1 bis 200 Nanometern liegt, ggf. aber auch bis 1000 Nanometer erweitert werden kann. Der Nanopartikel kann jedoch auch eine Größe <1nm haben.
Der Nanopartikel kann ein Kunststoff, Naturstoff oder metallischer Nanopartikel sein, insbesondere ein Gold-Nanopartikel (auch als Nanogold bezeichnet). Generell ist es auch möglich, dass Gold-Nanopartikel kolloidale Gold-Nanopartikel sind.
In einer besonders vorteilhaften Ausführungsform ist der spezifische Bindungspartner ein Antikörper und das Markierungselement ein Nanopartikel, insbesondere Gold-Nanopartikel (auch als Nanogold bezeichnet).
Insbesondere ist es denkbar, dass der spezifische Bindungspartner in löslicher Form vorliegt.
Der spezifische Bindungspartner kann insbesondere vorübergehend auf dem Testelement immobilisiert sein, insbesondere in einem definierten Bereich des Testelements.
In einer weiteren, vorteilhaften Ausführungsform liegt der spezifische Bindungspartner in getrockneter Form vor.
Bislang wurden zwei unterschiedliche Legionella-Markerproteine rekombinant hergestellt. Gegen diese Markerproteine wurden verschiedene Antikörper generiert und auf deren Einsetzbarkeit in der Lateral-Flow Technologie überprüft. Erste Versuchsaufbauten zeigen, dass der einfache und sensitive Nachweis beider Marker im Lateral-Flow Format möglich ist. Weitere Schritte werden die Optimierung des Tests hinsichtlich Sensitivität und eine Machbarkeitsstudie mit Patientenproben sein.
Ferner wurde ein weiteres Legionella Markerprotein rekombinant hergestellt.
Vorteile:
PoC-Speicheltest (Point-of-Care Speicheltest) kann die prophylaktische Vergabe von Antibiotika gegen die Legionellose verringern und somit der Entstehung von Resistenzen entgegenwirken.
Unterstützung des Hausarztes bei der Diagnose.
Einfache, anwenderfreundliche Handhabung und nicht-invasive Probenahme. Dadurch ist das Testsystem geeignet auch in Regionen mit wenig entwickelter medizinischer Infrastruktur eingesetzt zu werden.
Schnelle Diagnose, da das Messergebnis bereits nach maximal 30 Minuten vorliegt.
Weitere Einzelheiten und Vorteile der Erfindung sollen nun anhand eines in den Zeichnungen näher dargestellten Ausführungsbeispiels erläutert werden.

Fig. 13b beispielhaft die Auswahl von Antikörper-Paarungen für PAL für die Verwendung in einem erfindungsgemäßen Testsystem, anhand einer Testmembran;
Fig. 14 beispielhaft die Auswahl von Antikörper-Paarungen für MIP für die Verwendung in einem erfindungsgemäßen Testsystem;
Fig. 15 beispielhafte Bedingungen für die Konjugation von polyklonalen Antikörpern und Nanogold, und
Fig. 16 schematisch ein erfindungsgemäßes Verfahren zum Nachweis von Legionellen in einer Probe.
Fig. 1 zeigt eine schematische Zeichnung mit dem Einsatz des erfindungsgemäßen Testsystems 10. Hier wird Speichel eines möglichen Patienten im Bereich der Mundhöhle oder in sonst geeigneter Weise entnommen.
Insbesondere ist die Probennahme durch ein Probennahmeelement gezeigt.
Generell kann die Entnahme des Speichels durch ein Probennahmeelement, beispielsweise einen Wischer, oder das Testsystem selbst erfolgen.
Bei dem Testsystem 10 handelt es sich hier um einen Lateral- Flow-Test (LFT).
Alternativ kann beispielsweise eine Sputumprobe verwendet werden.
Fig. 2 zeigt weitere schematische Zeichnung mit dem Einsatz des erfindungsgemäßen Testsystems 10. Der Speicheltropfen wird beispielsweise auf einen Speichelkollektor aufgebracht und mit diesem auf den Lateral-Flow-Test LFT aufgebracht.
In anderen Worten, das Testsystem 10 umfasst hier einen Lateral-Flow-Test LFT.
Der Speichelkollektor kann auch als Probennahmeelement bezeichnet werden.
In anderen Worten, das Testsystem 10 umfasst hier eine Probennahmeelement.
Gezeigt ist hier ein Testsystem 10 mit zwei Teststreifen 12.
Generell kann ein Testsystem 10 auch nur einen Teststreifen 12 oder mehr als zwei Teststreifen 12 umfassen.
Fig. 3 zeigt schematisch die grundsätzliche Arbeitsweise des Testsystems 10, das hier als Lateral-Flow-Test (LFT) ausgebildet ist.
Für den hochsensitiven und schnellen PoC-Nachweis wird die bewährte DrugWipe®- Technologie der Securetec Detektions-Systeme AG verwendet. Der Speichel-Schnelltest
DrugWipe® wird seit mehr als 20 Jahren weltweit von Polizei und Zoll sehr erfolgreich zum Nachweis von Drogen in Speichel eingesetzt. Diese Technologie wird an den Nachweis von Legionellen im Speichel angepasst.
In anderen Worten, das Testsystem 10 ist ein Testsystem 10 zur Erkennung der Legionärskrankheit.
Das Testsystem 10 ist zur Erkennung der Legionärskrankheit und/oder zum Nachweis einer Infektion mit Legionellen in Speichel und/oder Sputum eingerichtet.
Der Nachweis kann zumindest teilweise über die Bindung mindestens eines Proteins M, insbesondere eines Proteins der Bakteriengattung Legionella, an einen spezifischen Bindungspartner B erfolgen.
Das Protein M kann auch als Markerprotein bezeichnet werden, vgl. Fig. 4.
Fig. 4 zeigt den Einsatz der Markerproteine.
Drei Markerproteine (M1-M3) wurden anhand von Verbreitung, Immunogenität and Homologie zwischen Legionella pneumophila Stämmen ausgewählt. M1 und M2 wurden als rekombinante Proteine in E. coli erzeugt und im mg-Maßstab aufgereinigt. Sie dienen der Gewinnung von Antikörpern.
M1 kann MIP (macrophage inflammatory protein) sein, insbesondere MIP 1-alpha (macrophage inflammatory protein 1-alpha).
M2 kann PAL sein.
M3 kann FLA sein.
Polyklonale Antikörper a-MIP und a-PAL) haben eine hohe Sensitivität und Spezifität gezeigt.
Sie bilden stabile Konjugate (Immunkonjugate) mit Goldpartikeln N.
MIP und PAL sind zudem stabil in Speichel.
Selbiges gilt für FLA.
Zu Versuchszwecken im Rahmen der Erprobung der Erfindung wurden MIP (23mg) und PAL (61mg) produziert mit >95% Reinheit und mit den entsprechenden Antikörpern (d.h. MIP und PAL) nachgewiesen.
Bisherige Ergebnisse:
Markerproteine erfolgreich rekombinant hergestellt (MIP, PAL, FLA);
- Antikörper entwickelt;
Nachweis von 10 ng/ml Markerprotein auf Lateral-Flow Teststreifen (Prototypen).
Fig. 5 zeigt die Charakterisierung von poly- und monoklonalen Antikörpern.
Bislang wurden je 7 Antikörper gegen M1 (MIP) und M2 (PAL) generiert und hinsichtlich Spezifität, Sensitivität und Stabilität selektiert.
Durch Ausschluss von Kreuzreaktionen wird gewährleistet, dass der LegioWipe keine anderen Erreger der Lungenentzündung und keine Mikro-organismen der normalen Mundflora erkennt.
Die sieben Antikörper umfassen jeweils polyklonale und monoklonale Antikörper.
Ferner wurden Antikörper gegen FLA generiert und hinsichtlich Spezifität, Sensitivität und Stabilität selektiert.
Fig. 6 zeigt den Aufbau eines Lateral-Flow Teststreifens 12.
Es wurde M1 (MIP) in Speichelsurrogat nachgewiesen.
Durch Konjugation der Antikörper mit Gold-Nanopartikeln entstehen rot gefärbte „Nanosonden“, welche den immunologischen Nachweis der Legionellen erlauben.
Erste Labormuster erreichten eine Sensitivität von ca. 10 ng/ml (440 pM) Markerprotein.
Weitere Labormuster erreichten eine Sensitivität von ca. 2 ng/ml - 20 ng/ml Markerprotein.
Modellversuche zeigten zudem, dass die Markerproteine in humanem Speichel stabil sind, vgl. Fig. 9.
Fig. 7 zeigt schematisch die Arbeitsweise eine LFT Systems, das beim erfindungsgemäßen Testsystem 10 verwendet wird.
Es handelt sich hierbei um ein Lateral-Flow-Test LFT im Sandwich Format.
Eine zu untersuchende Probe (in diesem Beispiel Speichel) wird auf das Testelement 12 aufgetragen (hier angezeigt durch den Wischer, welcher auch als Probennahmeelement bezeichnet werden kann).
Die Probe beginnt nach Zugabe des Laufmittels (hier Puffer) mit der Ausbreitung über das Testelement, 12 aufgrund von Kapillarkräften (Dünnschichtchromatographie).
Die Probe wandert mit der Flüssigkeit zu einem Bereich, wo sich Immunkonjugate (Antikörper B mit Nanogold N) befinden.
In diesem Ausführungsbeispiel liegen die Immunkonjugate zunächst in getrockneter Form vor. Die Flüssigkeit löst die Immunkonjugate vom Testelement 12.
Es wird eine spezifische Bindung jeweils eines Immunkonjugats an jeweils ein nachzuweisendes Protein in der Probe, sofern vorhanden, ermöglicht.
Die Flüssigkeit wandert weiter in den Kapillarbereich, wo in einem kleinen Abschnitt ein Antikörper immobilisiert wurde (kann auch aus Testlinie Antikörper bezeichnet werden), der an einer anderen Stelle auf der Oberfläche des nachzuweisenden Markerproteins bindet als es das Immunkonjugat macht, wodurch das nachzuweisende Markerprotein in diesem Bereich bindet und anreichert wird, während die Flüssigkeit weiterwandert.
Durch die Anreicherung des nachzuweisenden Proteins M und des daran gebundenen Immunkonjugats entsteht anhand des gebundenen Immunkonjugats mit dem Nanogold N eine Färbung auf dem Testelement 12, hier in Form einer Linie.
Die Erkennung und/oder der Nachweis des Proteins M erfolgen demnach zumindest teilweise über die Bindung mindestens eines Proteins M an einen spezifischen Bindungspartner B.
Die Flüssigkeit wandert weiter in den Kapillarbereich, wo in einem weiteren kleinen Abschnitt ein weiterer Antikörper immobilisiert wurde (kann auch als Kontrolllinienantikörper bezeichnet werden), der das Immunkonjugat (direkt) bindet und dieses dadurch in diesem Bereich anreichert, während die Flüssigkeit weiterwandert.
Durch die Anreicherung von Immunkonjugaten entsteht über das Nanogold N eine Färbung auf dem Testelement 12, hier in Form einer Linie.
Dies kann auch als Kontrollsystem 16 bezeichnet werden.
Alternativ ist ein Lateral-Flow-Test LFT im kompetitiven Format denkbar.
Denkbar ist auch ein Nachweis mittels eines nicht-kompetitiv aufgebauten LFT mit negativer Anzeige.
Das Testsystem 10 umfasst demnach ein Testelement 12 mit mikrofluidischen Strukturen, insbesondere einen chromatographischen Teststreifen 12.
In diesem Ausführungsbeispiel umfasst der Teststreifen 12 einen ersten Bereich, der zur Absorption von Elutionsmittel (in diesem Beispiel ein Puffer) eingerichtet ist,
In einem anderen Ausführungsbeispiel kann dieser erste Bereich fehlen.
In diesem Ausführungsbeispiel umfasst der Testreifen 12 einen zweiten Bereich, der für die Aufbringung einer Probe eingerichtet ist.
In diesem Ausführungsbeispiel umfasst der Testreifen 12 einen dritten Bereich, der für den Nachweis, vorzugsweise für den optischen Nachweis, des Proteins M eingerichtet ist,
In diesem Ausführungsbeispiel umfasst der Testreifen 12 einen vierten Bereich, der für die Absorption überschüssigen Elutionsmittels eingerichtet ist (Vlies).
In einem anderen Ausführungsbeispiel kann dieser vierte Bereich fehlen.
Der Teststreifen kann einen weiteren (fünften) Bereich umfassen, wobei der weitere (fünfte) Bereich zumindest vorübergehend den spezifischen Bindungspartner für das Protein M, insbesondere MIP und/oder PAL und/oder FLA umfasst.
Das Testsystem 10 umfasst zudem ein Gehäuse 14.
Insbesondere ist das Testelement 12 in ein Gehäuse 14 eingebaut.
In einem anderen Ausführungsbeispiel kann das Gehäuse 14 fehlen.
Nicht gezeigt in Fig. 7 ist, dass ein spezifische Bindungspartner B generell ein monoklonaler Antikörper, polyklonaler Antikörper affinitätsaufgereinigter Antikörper, ein Antikörperfragment, Peptid, Protein, Nanobody, Nukleinsäurefragment, Aptamer, Anticalin, Lektin, Affibody und/oder chemischer Liganden umfassen kann.
Nicht gezeigt in Fig. 7 ist auch, dass das Markierungselement N generell mindestens einen Farbstoff, ein Fluorochrom, Fluorophor, Luminophor, Enzym, Peptid, Biotin, Oligonukleotid, Protein-Tag, radioaktives Isotop, nicht radioaktives Nuklid, einen Nanopartikel, insbesondere Gold-Nanopartikel, fluoreszierenden Nanopartikel, magnetischen Nanopartikel und/oder Quantumdot umfassen kann.
Nicht gezeigt in Fig. 7 ist auch, dass der spezifische Bindungspartner B (für das nachzuweisende Protein M) generell in löslicher Form und/oder getrocknet (in getrockneter Form) vorliegt.
Der gezeigte Lateral- Flow-Test LFT kann zum Nachweis einer Infektion eines Subjekts mit Legionellen verwendet werden.
Nicht gezeigt in Fig. 7 ist, dass das mindestens eine oder auch mehrere Proteine M MIP und/oder PAL und/ oder FLA umfassen kann.
Nicht gezeigt in Fig. 7 ist auch, dass das Testsystem 10 geeignet ist zumindest teilweise ein Verfahren entsprechend Fig. 17 durchzuführen.
Fig. 8 zeigt die Produktion von Konjugation von Gold N und Antikörpern B.
Insbesondere ist die Produktion von stabilen Immunkonjugaten aus Antikörpern B (Schaf anti- PAL bzw. Schaf anti-MIP) und Nanogold N dargestellt.
Die Produktion von stabilen Immunkonjugaten aus anti-PAL kann analog dazu durchgeführt werden.
Fig. 9 zeigt die Stabilität von Antigenen in Speichel.
Die Stabilität von bakteriellen Antigenen wurde mittels Western Blot untersucht.
Insbesondere wurde die Stabilität von MIP und PAL im Speichel über einen Zeitraum von 10 min bis zu 18h (bei Raumtemperatur RT oder 37°C) untersucht.
Der Western Blot stellt ein Verfahren zum Nachweis von Proteinen, z.B. in einem Proteingemisch, dar. Im ersten Schritt wird das Proteingemisch mittels einer Gel- Elektrophorese in einer Trägermatrix (SDS-PAGE, Native-PAGE, isoelektrische Fokussierung, 2D-Gelelektrophorese, usw.) entsprechend der Protein-Größe, -Ladung oder anderer Eigenschaften aufgetrennt in einzelne Proteinbanden. Die getrennten Proteinbanden werden danach für den Western Blot aus dem Gel auf eine feste Trägermembran (z.B. aus Nitrozellulose, Nylon oder PVDF) transferiert. Dieser Vorgang wird Blotting genannt. Aufgrund Ladungswechselwirkungen bleiben die Proteine an der Membranoberfläche im Muster der elektrophoretischen Auftrennung haften und sind für die Antikörper-Bindung zur Detektion zugänglich. Der Western Blot wurde unter Standardbedingungen durchgeführt.
Die Antigene M MIP und PAL (rekombinante Proteine) wurden in Speichel Inkubiert (150 ng/pl).
Die Stabilität von MIP und PAL wurde mit und ohne Zugabe des unspezifischen Proteaseinhibitors PMSF untersucht.
Nach der Inkubation wurden 600ng Protein pro Linie aufgetragen.
Als Kontrollen wurden Speichel (ohne Zugabe von rekombinantem Protein, Negativkontrolle) und rekombinantes Protein (ng, Positivkontrolle) aufgetragen.
Ein Marker (Molekulargewichtsmarker) (wurde verwendet um die Markerproteine MIP und PAL eindeutig identifizieren zu können.
Die Antigene M sind in Speichel sowohl mit als auch ohne Proteaseinhibitor stabil, insbesondere auch bei 37°C.
Nicht gezeigt in Fig. 9 ist, dass die Stabilität von FLA im Speichel analog gemessen werden kann.
Fig. 10 zeigt die Spezifität von polyklonalem a-MIP und a-PAL.
Die Spezifität von polyklonalem a-MIP (anti-MIP) und a-PAL (anti-PAL) wurde mittels Western Blot (vgl. Fig. 9) untersucht.
Untersucht wurde insbesondere die Fähigkeit von a-MIP und a-PAL unterschiedliche Vertreter der oralen Mikroflora ( Haemophilus parainfluenzae, Staphylococcus epidermidis, Rothia mucilaginosa, Neisseria perflava, Streptocccus mitis, Streptococcus salvarius, Legionalla pneumophila) zu detektieren.
Die Erreger wurden als Proben in die Western Blot Analyse eingesetzt.
Ein Marker (Molekulargewichtsmarker) wurde verwendet um die Markerproteine M MIP und PAL eindeutig anhand ihrer Größe identifizieren zu können.
Als Positivkontrollen wurden die rekombinanten Proteine (Antigene) MIP bzw. PAL eingesetzt.
Als weitere Kontrolle wurde für a -PAL rekombinantes MIP und für a-MIP rekombinantes PAL eingesetzt.
Beide Antikörper zeigten keine oder nur eine minimale, zu vernachlässigende Kreuzreaktivität.
Nicht gezeigt in Fig. 10 ist, dass die Analyse der Spezifität von polyklonalem a-FLA analog stattfinden kann.
Fig. 11 zeigt ein beispielhaftes Labormuster eines erfindungsgemäßen Lateral-Flow-Tests LFT.
Gezeigt ist die Ermittlung der unteren Nachweisgrenze einfacher Lateral Flow Teststreifen LFT für MIP (Vierfachbestimmung).
Das Prinzip des Tests beruht im Wesentlichen auf dem in Fig. 7 beschriebenen System (ohne Kontrollsystem 16 und Probennahmeelement).
Ein rötlicher Fleck auf der Membran zeigt den spezifischen Nachweis des Markerproteins M MIP an.
Als Probe wurde Speichelsurrogat eingesetzt.
Die Immunkonjugate werden in diesem Fall als Punkte, nicht als Streifen angezeigt. Die empfindlichsten Teststreifen 12 erreichten eine Sensitivität von ca. 2 ng/ml - 20 ng/ml (440 pM) pro Markerprotein M in Speichelsurrogat.
In einem vergleichbaren Test wurde die Nachweisgrenze von PAL ermittelt. Diese liegt bei weniger als 2 ng/ml in Speichelsurrogat.
Fig. 12 zeigt Testsysteme 10 für MIP umfassend jeweils ein Kontrollsystem 16.
Die Testsysteme 10 funktionieren im Wesentlichen gemäß der Beschreibung von Fig. 7.
Gezeigt sind vier MIP-Entwicklungsmuster, jeweils nach Testende (ca. 20 Minuten).
Die Testsysteme 10 umfassen jeweils ein internes Kontroll System 16. Dieses bildet eine zusätzliche Bande/ Linie aus, wenn der Test (unabhängig vom Testergebnis) korrekt durchgeführt und korrekt abgelaufen ist.
Von links nach rechts sind Testsysteme 10 für folgende Proben gezeigt: humaner Speichel als Negativ-Probe, 5x106 Legionella pneumophila in Speichel, 5x105 Legionella pneumophila in Speichel, 5x104 Legionella pneumophila in Speichel.
Die Übertragung der Probe vom Probennahmeelement auf das Testelement 12 sowie die Chromatographie funktionierten ordnungsgemäß.
Das Kontrollsystem 16 lieferte nach 10 Minuten ein Signal (obere der beiden Linien).
Die untere Nachweisgrenze von MIP liegt bei ca. 1 x 105 intakten Legionella pneumophila Bakterien in humanem Speichel pro Testelement 12.
Bei allen Versuchen ist eine (rote) Kontrolllinie, bei den Positiv- Proben (größer/gleich Nachweisgrenze zusätzlich eine (rote) Testlinie zu erkennen.
Das Muster zeigt kein falsch-positives Signal in der Negativprobe (auch nach verlängerter Auslesezeit von bis zu zwei Stunden, nicht gezeigt).
Nicht gezeigt in Fig. 12 ist, dass in analogen Tests vergleichbare Ergebnisse für das Markerprotein M PAL oder FLA erhalten wurden.
Die untere Nachweisgrenze für PAL liegt bei ca. 1 x 107 intakten Legionella pneumophila Bakterien in humanem Speichel pro Teststreifen,
Fig. 13a und Fig. 13b zeigen beispielhaft die Auswahl von Antikörper-Paarungen für PAL für die Verwendung in einem erfindungsgemäßen Testsystem 10.
Ziel ist es, aus den Antikörpern bzw. Immunkonjugaten die Paare auszuwählen, mit denen ein Sandwich Assay für den Lateral Flow Test aufgebaut werden kann (sog. „matching pairs“) (Sandwich Assay, vgl. Fig. 7).
Alle verfügbaren Kombinationen (für PAL) von einem Testlinie Antikörper AK und einem Antikörper Nanogold-Konjugat (Testgold AK) wurden auf der Membran () aufgebaut und die Bindung an die Antigene (PAL) untersucht.
Es wurden dispensierte Testgolde verwendet und die Testlinien-AKs wurden gespottet.
Es wurden 10 ng/ml PAL in gepoolten Speichel als Proben verwendet.
Für den Analyten PAL deutet sich an, dass es (mindestens) zwei passende Antikörper- Paarungen gibt:
• Testgold AK #4 mit Testlinie AK #1 und umgekehrt Testgold AK #1 mit Testlinie AK #4
• Testgold AK #6 mit Testlinie AK#1 und umgekehrt Testgold AK#1 mit Testlinie AK #6
+++ steht für starke Bindung, ++ für mäßige Bindung, + für schwache Bindung, NSB steht für nicht spezifische Bindung (=falsch positiv, bedeutet AK konnte nicht getestet werden (vgl. Fig. 13a).
Fig. 13b zeigt beispielhaft eine entsprechende Membran für Testlinie AK#1 mit Testgold AK #4 und #6; alle Versuche in doppelter Ausführung, ist die Negativkontrolle (10 pl Wasser) „+“ ist die Positivprobe mit PAL (10 pl mit c = 100 ng/ml)
Testgold AK #1 ist mit Nanogold aggregiert. Daher ist es notwendig, ein stabiles Testgold mit diesem Antikörper zu finden, um sich alle Kombinationen offen zu halten
Nicht gezeigt in Fig. 13a oder 13b ist, dass unter Verwendung von Testgold Antikörpern (Immunkonjugaten) mit Gold-Nanopartikeln von anderen Herstellern weitere geeignete Antikörperpaarungen gefunden werden konnten.
Fig. 14 zeigt beispielhaft die Auswahl von Antikörper-Paarungen für MIP für die Verwendung in einem erfindungsgemäßen Testsystem 10.
Das Auswahlverfahren für MIP ist in den Grundzügen mit dem Auswahlverfahren für PAL, vgl. Fig. 13a und 13b, identisch.
Demnach ist das Ziel, aus Antikörpern bzw. Konjugaten die Paare auszuwählen, mit denen ein Sandwich Assay aufgebaut werden kann (sog. „matching pairs“). Alle verfügbaren MIP- Kombinationen wurden in Form von Spot-Tests aufgebaut. Dies wurde auf verschiedenen Membranen in Verbindung mit verschiedenen RP-Systemen gemacht.
Die für den Sandwich-Assay am besten geeigneten MIP-Paarungen sind:
• Testgold (TG) AK #1 mit Testlinie AK #4 und umgekehrt Testgold AK #4 mit Testlinie AK #1
• Testgold AK#6 mit Testlinie AK #4 und umgekehrt Testgold AK +#4 mit Testlinie AK #6
• Ferner: Testgold AK#4 mit Testlinie AK #2 und umgekehrt Testgold AK #2* mit Testlinie AK #4
Die Paarung AK#5/AK#6 kann ebenso eingesetzt werden, falls mit den obigen Paarungen wider Erwarten kein empfindlicher und stabiler Test aufgebaut werden kann.
Fig. 15 zeigt_beispielhafte Bedingungen für die Konjugation von polyklonalen Antikörpern und Nanogold.
Aus polyklonalem Anti- MIP- bzw. Anti-PAL-Antikörper und Nanogold wurden mehrere Bedingungen gefunden, unter welchen mehrere stabile Immunkonjugate hergestellt werden können (Puffer, pH-Wert, Nanogold Hersteller, Beladung).
Nicht gezeigt in Fig. 15 ist, dass_ein kompetitiver wie auch ein Sandwich-Test grundsätzlich mit den polyklonalen AK bzw. entsprechenden Immunkonjugaten funktionieren.
Fig. 16 zeigt schematisch ein erfindungsgemäßes Verfahren zum Nachweis von Legionellen in einer Probe.
Das Verfahren kann zumindest teilweise mit einem erfindungsgemäßen Testsystem 10, beispielsweise wie in Fig. 7 dargestellt, durchgeführt werden.
Das Verfahren umfasst mindestens die Schritte S1-S3.
In einem ersten Schritt S1 wird ein Testsystem 10 bereitgestellt.
Das Testsystem 10 umfasst ein Testelement 12.
In einem zweiten Schritt S2 wird eine Probe auf das Testelement 12 aufgetragen.
In einem dritten Schritt S3 wird das Vorhandensein mindestens Proteins M, insbesondere MIP, PAL und/oder FLA in der Probe nachgewiesen.
Das Testsystem 10 umfasst mindestens einen spezifischen Bindungspartner B, an welchen das mindestens eine Protein M bindet.
Der Nachweis erfolgt zumindest teilweise über die Bindung des mindestens einen Proteins M, insbesondere MIP, PAL und/oder FLA, an den spezifischen Bindungspartner B.
Ferner kann das Verfahren den Schritt der Bestimmung der Menge des mindestens einen Proteins umfassen.
Es ist außerdem möglich, dass das Verfahren ferner den Schritt des Lysierens der Probe umfasst.
Das Lysieren kann beispielsweise mit Detergenzien (z.B. Triton X-100), SDS und/oder Tween) oder mittels Ultraschall stattfinden.
Es ist zudem möglich, dass das Verfahren ferner den Schritt des Verflüssigens der Probe umfasst.
Das Verflüssigen kann zumindest teilweise durch Detergenzien und/oder Liquilzer erfolgen.
Ziel der Verflüssigung kann es sein, dass die Probe besser über einen Lateral-Flow-Test chromatographiert.
Die Probe kann beispielsweise Speichel oder Sputum sein.
Die untere Nachweisgrenze von MIP liegt bei ca. 1 x 105 intakten Legionella pneumophila Bakterien in humanem Speichel pro Testelement 12.
Die untere Nachweisgrenze für PAL liegt bei ca. 1 x 107 intakten Legionella pneumophila Bakterien in humanem Speichel pro Testelement 12.

Claims
1. Testsystem (10) zur Erkennung der Legionärskrankheit.
2. Testsystem (10) zum Nachweis einer Infektion eines Subjekts mit Legionellen.
3. Testsystem (10) nach Anspruch 1 oder Anspruch 2, dadurch gekennzeichnet, dass das Testsystem (10) ein Lateral-Flow-Test (LFT) ist oder umfasst.
4. Testsystem (10) nach einem der Ansprüche 1-3, dadurch gekennzeichnet, dass das Testsystem (10) zur Erkennung der Legionärskrankheit und/oder zum Nachweis einer Infektion mit Legionellen in Speichel oder Sputum eingerichtet ist.
5. Testsystem (10) nach einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass die Erkennung und/oder der Nachweis zumindest teilweise über die Bindung mindestens eines Proteins (M), insbesondere eines Proteins der Bakteriengattung Legionella, an einen spezifischen Bindungspartner (B) erfolgt.
6. Testsystem (10) nach Anspruch 5, dadurch gekennzeichnet, dass das mindestens eine Protein (M) ein Protein der Art Legionella pneumophila, Legionella micadei, Legionella. bozmanii, Legionella dumoffii und/oder Legionella longbeachae ist.
7. Testsystem (10) nach Anspruch 6, dadurch gekennzeichnet, dass das mindestens eine oder auch mehrere Proteine (M) MIP und/oder PAL und/ oder FLA umfasst.
8. Testsystem (10) nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass das Testsystem (10) umfasst: a) ein Testelement (12) mit mikrofluidischen Strukturen, vorzugsweise einen chromatographischen Teststreifen, umfassend:
(i) gegebenenfalls einen ersten Bereich, der zur Absorption von Elutionsmittel eingerichtet ist,
(ii) einen zweiten Bereich, der für die Aufbringung einer Probe eingerichtet ist,
(iii) einen dritten Bereich, der für den Nachweis, vorzugsweise für den optischen Nachweis, des Proteins (M), insbesondere von MIP, PAL und/oder FLA, eingerichtet ist,
(iv) optional einen vierten Bereich, der für die Absorption überschüssigen Elutionsmittels eingerichtet ist, und b) wahlweise ein Gehäuse (14).
9. Testsystem (10) nach Anspruch 8, dadurch gekennzeichnet, dass das Testelement (10) einen weiteren Bereich umfasst, wobei der weitere Bereich zumindest vorübergehend den spezifischen Bindungspartner (B) für das Protein (M), insbesondere MIP, PAL und/oder FLA umfasst.
10. Testsystem (10) nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass das Testsystem (10) ferner ein Kontroll System (16), insbesondere ein optisches Kontrollsystem (16) umfasst.
11. Testsystem nach einem der Ansprüche 5 bis 10, dadurch gekennzeichnet, dass das Testsystem ferner ein Probennahmeelement umfasst, welches für die Aufnahme einer das mindestens eine Protein (M) enthaltenden Probe eingerichtet ist.
12. Verwendung eines Testsystems (10) nach einem der vorhergehenden Ansprüche zum Nachweis einer Infektion mit Legionellen.
13. Verfahren zum Nachweis von Legionellen in einer Probe, umfassend die folgenden Schritte: a) Bereitstellung eines Testsystems (10),
(b) Aufbringen der Probe auf ein Testelement (12), und
(c) Nachweis des Vorhandenseins mindestens eines Proteins (M), insbesondere MIP, PAL und/oder FLA in der Probe, wobei das Testsystem (10) mindestens einen spezifischen Bindungspartner (B) umfasst, an welchen das mindestens eine Protein (M), insbesondere MIP, PAL und/oder FLA bindet, wobei der Nachweis zumindest teilweise über die Bindung des mindestens einen Proteins (M), insbesondere MIP, PAL und/oder FLA, an den spezifischen Bindungspartner (B) erfolgt.
14. Testsystem (10) nach einem der Ansprüche 8-11 oder Verfahren nach Anspruch 13, dadurch gekennzeichnet, dass die Probe eine Speichelprobe oder Sputum umfasst.
15. Testsystem (10) nach einem der Ansprüche 5-11 oder 14 oder Verfahren nach einem der Ansprüche 13 bis 14, dadurch gekennzeichnet, dass der spezifische Bindungspartner (B) einen monoklonalen Antikörper, polyklonalen Antikörper affinitätsaufgereinigten Antikörper, ein Antikörperfragment, Peptid, Protein, Nanobody, Nukleinsäurefragment, Aptamer, Anticalin, Lektin, Affibody und/oder chemischen Liganden umfasst, wobei der spezifische Bindungspartner mit einem Markierungselement verbunden ist.

Testsystem (10) oder Verfahren nach Anspruch 15, dadurch gekennzeichnet, dass das Markierungselement (N) mindestens einen Farbstoff, ein Fluorochrom, Fluorophor, Luminophor, Enzym, Peptid, Biotin, Oligonukleotid, Protein-Tag, radioaktives Isotop, nicht radioaktives Nuklid, einen Nanopartikel, insbesondere Gold-Nanopartikel, fluoreszierenden Nanopartikel, magnetischen Nanopartikel und/oder Quantumdot umfasst.
17. Testsystem (10) nach einem der Ansprüche 5 bis 11 oder 14 bis 16 oder Verfahren nach einem der Ansprüche 13 bis 16, dadurch gekennzeichnet, dass der spezifische Bindungspartner (B) in löslicher Form und/oder getrocknet vorliegt.
