Abstract

Die vorliegende Erfindung betrifft anorganisch-organische Hybridverbindungen zur Verwendung in der Medizin bzw. zur Verwendung als Arzneimittel, bestehend aus einem anorganischen Metallkation und einem organischen Platin-haltigen zytostatischen Anion, insbesondere auch einem cisPlatin-Derivat.

Description

Anorganisch-organische Hybridverbindungen mit organischen Platin-haltigen Anionen zur Verwendung in der Medizin
Die vorliegende Erfindung betrifft anorganisch-organische Hybridverbindungen zur Verwendung in der Medizin bzw. zur Verwendung als Arzneimittel, bestehend aus einem anorganischen Metallkation und einem organischen Platin-haltigen zytostatischen Anion, insbesondere auch einem cisPlatin-Derivat.
Die gezielte Freisetzung von Pharmazeutika (Drug Delivery) mit Nanopartikeln stellt eine große Herausforderung für interdisziplinäre Wissenschaften dieser Zeit dar. Aktuell wird hier eine große Zahl an unterschiedlichen Material- und Therapiekonzepten für unterschiedlichste Krankheitsbilder verfolgt. Weit verbreitet ist die Einkapselung von Wirkstoffen in Vesikeln, meso- oder nanoskaligen Hohlkugeln oder Polymerkapseln. Darüber hinaus werden Wirkstoffe an Nanopartikel wie S1O2, Au, Quantendots oder Polymernanopartikel gebunden. Alternativ können Wirkstoffe auch in Nanopartikel eingebettet werden, wobei S1O2 und Polymere die gebräuchlichsten Matrixmaterialien sind. Nachteile der derzeit diskutierten Materialien und Lösungsansätze sind die zum Teil sehr geringe Menge an Wirkstoff in Bezug auf die Nanopartikel insgesamt (z.B. 1% Wirkstoff eingekapselt in 99% S1O2 als Nanopartikelmatrix), die nicht vollständige Abbaubarkeit bzw. potentielle Toxizität der Nanopartikel nach Freisetzung des Wirkstoffs (beispielsweise bleiben Si02-Nanopartikel zurück). Darüber hinaus sind die Materialkonzepte und die Synthese der Materialien komplex und sehr aufwendig. Dies ist insbesondere für multifunktionelle Nanopartikel der Fall, die neben der Wirkstofffreisetzung auch eine analytische Erkennung ermöglichen.
Zusammenfassend können als Nachteile von Drug-Delivery-Systemen und Materialien gemäß dem Stand der Technik folgende Punkte angeführt werden:
aufwendige chemische Synthese, Mehrstufenprozesse;
komplexe Materialsysteme wie Kern-Schale-Strukturen und aufwendige Modifizierung der Partikeloberfläche;
geringe Menge an Wirkstoff bezogen auf die Gesamtmasse der Nanopartikel; toxische bzw. unter physiologischen Bedingungen nicht vollständig oder nur sehr langsam abbaubare/ausscheidbare Bestandteile;
Wirksamkeit eines komplexen Materialsystems nur gegen ein Krankheitsbild; Wirkstoff nur oberflächlich an ein Nanopartikel gebunden und daher in Suspension durch Partikelstöße leicht abreibbar;
Auftreten von erheblichen Nebenwirkungen; (zu schnelle oder zu langsame Freisetzung der jeweiligen Pharmazeutika.
Die gezielte Freisetzung von Platin-Derivaten, insbesondere cisPlatin-Derivaten, ist dabei eine besonders große Herausforderung. cisPlatin-Derivate gehören generell zu den wirksamsten und am meisten verwendeten Zytostatika, die jedoch folgende erhebliche Nachteile aufweisen;
Unspezifische und unselektive Aufnahme, so dass insbesondere alle schnell wachsenden Zellen betroffen sind,
Erhebliche Nebenwirkungen, die durch die unspezifische/unselektive Aufnahme begünstig werden, cisPlatin-Derivate werden nahezu ausschließlich in gelöster Form verwendet,
Nanopartikel enthalten bislang nur geringe Mengen an cisPlatin, bezogen auf die Gesamtmasse der Nanopartikel (d.h. <5 Gew-%), keine Platin-haltigen Anionen mit Phosphat-, Phosphonat-, Sulfonat- oder Carboxylat-Gruppen vorliegend, die für die Schwerlöslichkeit in Wasser und die Bildung von Nanopartikeln in Wasser essentiell sind.
Vor diesem Hintergrund ist es die Aufgabe der vorliegenden Erfindung, ein einfaches Materialkonzept bei gleichzeitig sehr breitem Anwendungsspektrum auf der Basis von entsprechenden Platin-Derivaten, insbesondere cisPlatin-Derivaten, zur Verwendung in der Medizin bereitzustellen. Diese Aufgabe wird durch die in den Ansprüchen gekennzeichneten Ausführungsformen gelöst.
Insbesondere werden anorganisch-organische Hybridverbindungen zur Verwen dung in der Medizin, aufgebaut als lonenverbindung aus einem anorganischen Metallkation, ausgewählt aus Ba2+, [ZrO]2+, [HfO]2+, Sc3+, Y3+, Gd3+, [GdO]+, [Gd(OH)]2+, [LaO]+, [La(OH)]2+, Fe3+, Bi3+, Ag+ oder einem Lanthanoid, und einem organischen Platin-haltigen Anion, welches mindestens eine Phosphat-, Pyro- phosphat-, Phosphonat-, Sulfat-, Sulfonat-, Carbonat- oder Carboxylatgruppe als funktionelle Gruppe enthält, wobei die Verbindung eine molare Löslichkeit von < 102 mol/l in Wasser aufweist und einen Partikeldurchmesser im Bereich von 1 bis 100 nm aufweist, bereitgestellt. Die vorliegende Erfindung betrifft insofern pharmazeutische Zusammensetzungen, umfassend mindestens eine solche anorganisch-organische Hybridverbindung.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung ist das Metallkation aus Ba2+, [ZrO]2+, [HfO]2+, [GdOf, [Gd(OH)]2+, [LaO]+, [La(OH)]2+ oder Ag+ ausgewählt.
Das organische Platin-haltige Anion enthält vorzugsweise mindestens eine Phosphat-, Pyrophosphat- oder Phosphonatgruppe als funktionelle Gruppe. Besonders bevorzugt ist das organische Platin-haltige Anion aus einem der folgenden ausgewählt:
[Pt2(POP)4] (POP: Pyrophosphat),
[cisPt(AEP)2]4 (AEP: Aminoethylphosphat), oder
[cisPt(PAA)2]4 (PAA: Phosphonoacetat).
Um Platin-Derivate, insbesondere auch cisPlatin-Derivate, für das Konzept der erfindungsgemäßen anorganisch-organischen Hybridnanopartikel überhaupt verfügbar zu machen, ist eine entsprechende Modifizierung von Platin-Derivaten, insbesondere cisPlatin-Derivaten, mit Phosphat-, Pyrophosphat-, Phosphonat-, Sulfat-, Sulfonat-, Carbonat- oder Carboxylatgruppen notwendig. Diese Modifizie- rung ist äußerst aufwendig. Die vorstehend aufgeführten Platin-haltigen Anionen sind neu bzw. kommerziell nicht erhältlich.
Die erfindungsgemäßen anorganisch-organischen Hybridnanopartikel erweisen sich als sehr vielversprechende Alternativen zu klassischen Platin-Derivaten, insbesondere cisPlatin-Derivaten. Vorteile dieser anorganisch-organischen Hybridverbindungen in der Form von Nanopartikeln sind die hohe Zellaufnahme, die hohe Wirksamkeit, die geringen Nebenwirkungen sowie die Möglichkeit der Kombination mehrerer Zytostatika in einem Nanopartikel sowie die multimodale Detektion über optische und magnetische Bildgebungsverfahren.
Insbesondere sind die erfindungsgemäßen anorganisch-organischen Hybridnanopartikel frei von entsprechenden, sie umgebenden Matrizes bzw. Einkapselungen, insbesondere beispielsweise einer Si02-Matrix, sowie frei von jeglichen Phosphorlipid-Matrizes bzw. -Einkapselungen.
In einer Ausführungsform der vorliegenden Erfindung können die anorganisch-organischen Hybridverbindungen weiterhin ein Fluoreszenzfarbstoffanion und/oder ein Wirkstoffanion umfassen, die jeweils eine Phosphat-, Phosphonat-, Sulfat-, Sulfonat-, Carbonat- oder Carboxylatgruppe als funktionelle Gruppe tragen, so dass die anorganisch-organische Hybridverbindung einen Wirkstoff freisetzen und/oder aufgrund der Fluoreszenz des Fluoreszenzfarbstoffanions in Zellen, Gewebe, Organen durch Lichtemission lokalisiert werden kann. Das in den erfindungsgemäßen anorganisch-organischen Hybridverbindungen zwingend vorgesehene Platin-haltige Anion kann aber selbst die Funktion als Wirkstoff, beispielsweise mit zytostatischen Eigenschaften, oder aufgrund dessen Fluoreszenz als Fluoreszenzfarbstoff einnehmen.
Neben dem organischen Platin-haltigen zytostatischen Anion, dessen Freisetzung und dem resultierenden zytostatischen Effekt können die erfindungsgemäßen anorganisch-organischen Hybridnanopartikel insofern sowohl mehrere zytostatische Anionen oder andere Wirkstoffanionen als auch Fluoreszenzfarbstoffanionen enthalten. Durch Fluoreszenzfarbstoffanionen ist sowohl eine optische und/oder pho- toakustische Bildgebung möglich. Wie vorstehend bereits ausgeführt, kann in einzelnen Fällen auch das Platin-haltige Anion selbst Fluoreszenz aufweisen.
Unter„Wirkstoffanion“ wird ein Stoff verstanden, der als Mittel zur Heilung oder zur Verhütung menschlicher oder tierischer Krankheiten bezeichnet wird, sowie ein Stoff, der für die Verwendung im oder am menschlichen oder tierischen Körper zur Erstellung einer ärztlichen Diagnose oder zur Wiederherstellung, Besserung oder Beeinflussung der menschlichen oder tierischen Körperfunktionen bestimmt ist.
Das in die erfindungsgemäße anorganisch-organische Hybridverbindung gegebenenfalls zusätzlich eingebaute, organische Wirkstoffanion unterliegt keiner wesentlichen Beschränkung, sofern es mindestens eine Phosphat-, Phosphonat-, Sulfat-, Sulfonat-, Carbonat- oder Carboxylatgruppe, vorzugsweise Phosphat-, Phospho- nat-, Sulfat- oder Sulfonatgruppe, als funktionelle Gruppe, die in Verbindung mit dem anorganischen Kation die Bildung einer in Wasser schwerlöslichen Verbindung und damit die Bildung von Nanopartikeln ermöglichen, aufweist. Solche Wirkstoffe sind dem Fachmann bekannt. Entsprechende Verfahren der Funktionalisie- rung solcher organischer Wirkstoffe sind dem Fachmann bekannt.
Besonders bevorzugt ist das zusätzlich eingebaute, organische Wirkstoffanion ausgewählt aus: Acetaminophenphosphat, Betamethasonphosphat, Dexame- thasonphosphat, 5‘-Fluor-2‘-deoxyuridin-5‘-monophosphat (FdUMP), Methylprednisolonphosphat, T riamcinolonphosphat, Estronphosphat, Estramustinphosphat, Codeinphosphat, Clindamycinphosphat, Fospropofol, Alendronat, Canrenoat, Doxycyclin Hydrat, Doxorubicin, Aztreonam, Tigemonam, Cefsulodin, Fosamprenavir, Tenofovir, Adefovir, Folsäure, Fosfomycin, a-T ocopherolphosphat oder Glucose-6-phosphat. Die erfindungsgemäße anorganisch-organische Hybridverbindung kann auch so verstanden werden, dass sie eine anorganische Matrix und eine organische Platin-haltige Verbindung umfasst, wobei die anorganische Matrix aus einer anorga- nischen Verbindung, ausgewählt aus der Gruppe, bestehend aus Metallphosphaten, einschließlich den Pyrophosphaten, Hydrogenphosphaten und Dihydrogen- phosphaten, Metalloxidphosphaten, Metallphosphonaten, Metallsulfaten, Metall- sulfonaten, Metallcarbonaten oder Metallcarboxylaten, aufgebaut ist, wobei die anorganische Verbindung ein Kation, ausgewählt aus Ba2+, [ZrO]2+, [HfO]2+, Sc3+, Y3+, Gd3+, [GdO]+, [Gd(OH)]2+, [LaO]+, [La(OH)]2+, Fe3+, Bi3+, Ag+ oder einem Lan- thanoid, enthält, wobei das organische Platin-haltige Anion (organische Pla tin-haltige Verbindung) ein oder mehrere funktionelle Gruppen, ausgewählt aus Phosphat-, Pyrophosphat-, Phosphonat-, Sulfat-, Sulfonat, Carbonat- oder Car- boxylatgruppen, aufweist, über welche die Platin-Einheit in die anorganische Matrix eingebaut ist. Insofern die Platinverbindung (sowie die optionale Wirkstoffverbindung und/oder der optionale Fluoreszenzfarbstoff) über deren funktionale Gruppe in die anorganische Matrix mittels ionischer Bindung eingebaut ist, ist demgemäß die funktionelle Gruppe, beispielhaft Phosphat, dann der anorganischen Matrix zuzurechnen, d.h. es liegt keine eigenständige Phosphatgruppe an der Platinverbindung vor.
Zusätzlich kann neben dem organischen Platin-haltigen Anion auch beispielsweise noch Hydrogenphosphat als Anion vorliegen (z.B.
[ZrO]2+2[(cisPt(AEP)2)o.5(HP04)i.o]4-).
In einer bevorzugten Ausführungsform der vorliegenden Erfindung enthält die an organisch-organische Hybridverbindung, wie vorstehend bereits ausgeführt, optional einen organischen Fluoreszenzfarbstoff, d.h. ein entsprechendes Anion davon, welcher eine oder mehrere funktionelle Gruppen, ausgewählt aus Phosphat-, Phosphonat-, Sulfat-, Sulfonat, Carbonat- oder Carboxylatgruppen, aufweist, über welche der Fluoreszenzfarbstoff(anion) in die lonenverbindung bzw. anorganische Matrix eingebaut ist. Dabei sind die organischen Fluoreszenzfarbstoffe, die als solche keine Phosphat-, Phosphonat-, Sulfat-, Sulfonat, Carbonat- oder Car- boxyiatgruppe aufweisen, mit mindestens einer von diesen funktionellen Gruppen modifiziert (z.B: Phenylumbelliferonphosphat (PUP), Methylfluoresceinphosphat (MFP), Resorufinphosphat (RRP), Dyomics-647-uridinphosphat (DUT)). Entsprechende Verfahren zum Funktionalisieren von solchen organischen Fluoreszenzfarbstoffen sind dem Fachmann bekannt.
Im Rahmen der vorliegenden Erfindung ist es möglich, die Detektion der erfindungsgemäßen anorganisch-organischen Hybridverbindungen aber nicht nur optisch über die Fluoreszenz eines gegebenenfalls eingebauten Fluoreszenzfarb- stoffanions, sondern in Gegenwart schwerer oder magnetischer anorganischer Kationen (z.B. Ba2+, [ZrO]2+, [HfO]2+, Gd3+, La3+, Fe3+, Bi3+) auch durch Röntgen absorption oder magnetische Messungen durchzuführen. So ist beispielsweise durch Verwendung paramagnetischer Kationen wie Gd3+ auch eine magnetische Bildgebung möglich.
In einer weiteren Ausführungsform der vorliegenden Erfindung ist die anorganisch-organische Hybridverbindung weiter mit einem oder mehreren tumorspezifischen Liganden (z. B. kleine organische Moleküle, Peptide, Antikörper und Nu- cleinsäuren) funktionalisiert, um beispielsweise Krebszellen spezifisch durch selektive Bindung an tumorspezifische oberflächenexprimierte Rezeptoren zu erreichen bzw. um die anorganisch-organischen Hybridnanopartikel und den enthaltenen Wirkstoff in vivo an einen spezifischen Wirkort zu schleusen und hier anzureichern. Aufgrund der wasserbasierten Synthese der anorganisch-organischen Hybridverbindungen ist diese Kopplung mit Antikörpern oder ähnlichen Molekülen besonders einfach und schonend.
Die erfindungsgemäßen anorganisch-organischen Hybridverbindungen sind schwerlöslich. Im Rahmen der vorliegenden Erfindung werden unter schwerlöslichen Verbindungen solche Verbindungen verstanden, die eine molare Löslichkeit von < 102 mol/l aufweisen. Vorzugsweise weisen die schwerlöslichen Verbindungen eine molare Löslichkeit von < 104 mol/l auf. Dies ist hinsichtlich der Synthese der erfindungsgemäßen anorganisch-organischen Hybridverbindungen vorteilhaft, da sich so die anorganisch-organische Hybridverbindung, welche das Platin-haltige Anion enthält, aus löslichen Vorläuferverbindungen ausfällen lässt.
Üblicherweise weist die anorganisch-organische Hybridverbindung eine röntgenamorphe Struktur auf. Dies ist hinsichtlich einer vereinfachten Synthese vorteilhaft, da amorphe Nanopartikel ohne größeren synthetischen Aufwand erhalten werden können.
Gegebenenfalls kann die anorganisch-organische Hybridverbindung weiter mit einem oder mehreren Kationen und/oder Anionen dotiert werden. Durch eine der artige Dotierung ist es möglich, die Lumineszenzeigenschaften der erfindungsge mäßen Hybridverbindung zu modifizieren, da nach Anregung eines Fluoreszenzfarbstoffes, wenn vorhanden, ein vollständiger oder partieller Energieübertrag auf die Dotierung erfolgt, so dass nachfolgend eine von der Dotierung stammende Emission zu beobachten ist. Es ist weiterhin möglich, dass die Dotierung eine veränderte Anregung der erfindungsgemäßen Hybridverbindung bewirkt. Die Dotierung kann in jedem geeigneten Konzentrationsbereich erfolgen. Vorzugsweise liegt die Dotierung in einem Konzentrationsbereich von 5 ppm bis 50 mol-% vor, besonders bevorzugt in einem Konzentrationsbereich von 0,1 bis 5,0 mol-%. Die anorganisch-organische Hybridverbindung ist vorzugsweise, wenn das anorganischen Metallkation kein Lanthanoid ist, mit einem Lanthanoid, ausgewählt aus La, Ce, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, Yb oder Lu, einem Übergangsmetall, ausgewählt aus Cr, Mn, Cu, Zn, Y, Ag oder Cd, einem Hauptgruppenelement, ausgewählt aus Sn, Sb, Pb oder Bi, oder einem komplexen Anion, ausgewählt aus [V04]3-, [M0O4]3- oder [WO4]3-, dotiert.
Die erfindungsgemäße Hybridverbindung kann jede geeignete Partikelgröße aufweisen. Üblicherweise ist die erfindungsgemäße Hybridverbindung nanoskalig und weist einen Partikeldurchmesser im Bereich von 1 bis 100 nm auf. Besonders bevorzugt ist ein Partikeldurchmesser im Bereich von 30 bis 70 nm. Darüber hinaus weist die erfindungsgemäße Hybridverbindung vorzugsweise eine nahezu monodisperse Größenverteilung im Bereich von ±30%, besonders bevorzugt im Bereich von ±5%, auf. Weiterhin weist die erfindungsgemäße Hybridverbindung vorzugsweise einen geringen Agglomerationsgrad auf, besonders bevorzugt mit einer Größenverteilung im Bereich von ±30%, noch mehr bevorzugt im Bereich von ±5%, auf. Im Stand der Technik sind geeignete Verfahren zur Bestimmung des Parti keldurchmessers und der monodispersen Größenverteilung bekannt.
Gemäß der vorliegenden Erfindung können jeweils unterschiedliche Anregungsund Emissionseigenschaften bei gleicher Hybridverbindung durch die Wahl und den Einbau eines entsprechenden organischen Fluoreszenzfarbstoffes eingestellt werden, wenn erforderlich. Vorzugsweise liegt die Anregung der erfindungsgemäßen Hybridverbindung im Bereich von 100 bis 800 nm und die Emission im Bereich von 200 bis 2000 nm. In der Regel erfolgt eine Anregung durch eine Leuchtdiode oder einen Laser, welche sichtbares bis nah-infrarotes Licht emittieren (d.h. 450 bis 800 nm) und eine Emission des organischen Fluoreszenzfarbstoffes bzw. der erfindungsgemäßen Hybridverbindung im sichtbaren Spektralbereich zwischen blau und infrarot (d.h. 450 bis 1400 nm). In einer anderen Ausführungsform erfolgt die Anregung in Form von UV-Licht (d.h. 100 bis 450 nm). Vorzugsweise nimmt bei der erfindungsgemäßen Hybrid Verbindung die Lumineszenzintensität unter Anregungsbedingungen über die Dauer der Anregung im Vergleich zu dem ungebundenen organischen Fluoreszenzfarbstoff weniger stark ab, besonders bevorzugt nimmt die Lumineszenzintensität über die Dauer der Anregung nicht ab, insbesondere bei Anregung mit einer Leuchtdiode. So nimmt bei Einwir- kung mit UV-Licht die Lumineszenzintensität vorzugsweise um nicht mehr als 10% ab, bei Einwirkung mit Tageslicht um nicht mehr als 1%.
Die vorliegende Erfindung betrifft ferner ein Verfahren zur Herstellung der erfindungsgemäßen Hybridverbindungen, umfassend die Schritte:
(a) das Bereitstellen einer Lösung einer Platin-haltigen Anion-Verbindung, die eine oder mehrere funktionelle Gruppen, ausgewählt aus Phosphat-, Pyrophos- phat-, Phosphonat-, Sulfat-, Sulfonat-, Carbonat- oder Carboxylatgruppen, aufweist,
(b) das Bereitstelien einer Lösung eines löslichen Metallsalzes, enthaltend Metallkationen, ausgewählt aus Ba2+, [ZrO]2+, [HfO]2+, Sc3+, Y3+, Gd3+, [GdO]+, [Gd(OH)]2+, [LaO]+, [La(OH)]2+, Fe3+, Bi3+, Ag+ oder einem Lanthanoid,
(c) das Vereinigen der beiden Lösungen unter Rühren, um die Hybridverbindung auszufällen, und
(d) das Isolieren und/oder Aufreinigen der ausgefällten Hybridverbindung.
Der Schritt (a) des erfindungsgemäßen Verfahrens umfasst das Bereitsteilen einer Lösung des organischen Platin-haltigen Anions. Ferner kann diese Lösung gegebenenfalls weiter mindestens ein Anion, ausgewählt aus Phosphat, Phosphonat, Sulfat, Sulfonat, Carbonat oder Carboxylat, enthalten. Wenn eingesetzt, so kann dieses Anion zusammen mit einem Kation in der Form eines gelösten Salzes vor liegen, beispielsweise als gelöstes Alkalimetallsulfat, Alkalimetallphosphat, Alkali- metallcarboxylat, Alkalimetallcarbonat. Vorzugsweise handelt es sich bei dem Alkalimetall um Natrium oder Kalium. Das Anion kann auch in Form der korrespon dierenden Säure in der Lösung vorliegen.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung wird zu der in Schritt (a) bereitgestellten Lösung weiter ein organischer Wirkstoff und/oder ein organischer Fluoreszenzfarbstoff, welche jeweils eine oder mehrere funktionelle Gruppen, ausgewählt Phosphat-, Phosphonat-, Sulfat-, Sulfonat-, Carbonat- oder Carboxylatgruppe, aufweist, zugegeben. Als Lösungsmittel kann jedes geeignete Lösungsmittel verwendet werden. Vorzugsweise wird als Lösungsmittel Wasser, isotones Wasser, ein physiologischer Puffer, ein Alkohol oder ein Gemisch aus mehreren dieser Lösungsmittel verwendet. Bevorzugte Alkohole zur Verwendung als Lösungsmittel sind Methanol, Ethanol, Propanol und Isopropanol.
Der Schritt (b) des erfindungsgemäßen Verfahrens umfasst das Bereitstellen einer Lösung eines löslichen Metallsalzes, welches Metallkationen enthält, die gleich oder verschieden sein können und aus Ba2+, [ZrO]2+, [HfO]2+, Sc3+, Y3+, Gd3+, [GdO]+, [Gd(OH)]2+, [LaO]+, [La(OH)]2+, Fe3+, Bi3+, Ag+ oder einem Lanthanoid ausgewählt sind. Als Lösungsmittel kann jedes geeignete Lösungsmittel verwendet werden. Vorzugsweise werden ebenfalls die vorstehend genannten Lösungsmittel, nämlich Wasser, isotones Wasser, ein physiologischer Puffer, Alkohole sowie Gemische aus mehreren dieser Lösungsmittel verwendet. In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung wird isotones Wasser oder ein physiologischer Puffer als Lösungsmittel verwendet. Als Metallsalz kann jedes Salz verwendet werden, das in dem verwendeten Lösungsmittel löslich ist. Geeignete Metallsalze sind dem Fachmann bekannt. Vorzugsweise können als Metallsalze die Halogenide, Nitrate und Sulfate der vorstehend genannten Metalle verwendet werden, sofern diese im jeweils verwendeten Lösungsmittel löslich sind.
Der Schritt (c) des erfindungsgemäßen Verfahrens umfasst das Vereinigen der beiden Lösungen unter Rühren. Auf diese Weise wird die erfindungsgemäße Hybridverbindung ausgefällt. Bei dem Schritt des Vereinigens können die beiden Lösungen jede geeignete Temperatur aufweisen. In einer bevorzugen Ausführungsform der vorliegenden Erfindung weist zumindest eine der beiden Lösungen oder weisen beide Lösungen eine Temperatur im Bereich von Raumtemperatur bis 85°C auf, besonders bevorzugt eine Temperatur im Bereich von 40° C bis 75°C. Das Vereinigen der beiden Lösungen erfolgt vorzugsweise rasch, d.h. innerhalb eines Zeitraums von nicht mehr als 10 Sekunden, vorzugsweise innerhalb eines Zeitraums von nicht mehr als 5 Sekunden. Der Schritt (d) des erfindungsgemäßen Verfahrens umfasst das Isolieren und/oder Aufreinigen der ausgefällten Hybridverbindung. Dieses Isolieren und/oder Aufreinigen kann durch alle geeigneten Verfahren erfolgen. Derartige Verfahren sind im Stand der Technik bekannt.
Vorzugsweise erfolgt das Isolieren und/oder Aufreinigen der Hybridverbindungs- partikel durch ein Verfahren, ausgewählt aus der Gruppe, bestehend aus Zentrifugationstechniken, Dialysetechniken, Phasentransfertechniken, Chromatogra- phietechniken, Ultrafiltrationstechniken, Waschtechniken und Kombinationen davon. Die vorstehend genannten Verfahren zur Isolierung und/oder Aufreinigung der Hybridverbindungspartikel können auch kombiniert und/oder mehrfach ausgeführt werden.

Beispiele:
Ausführungsbeispiel 1 : [ZrO]22+[Pt2(POP)4]4'
Das anorganisch-organische Hybrid [ZrO]22+[Pt2(POP)4] (POP: Pyrophosphat) wird durch Mischen zweier Lösungen dargestellt. Lösung 1 enthält Zr0Ckx8H20 (6,5 mg) in demineralisiertem Wasser (5,0 ml). Lösung 2 enthält Kaliumpyrophos- phatoplatinat(ll) (K4[Pt2(POP)4)], 20,3 mg) in demineralisiertem Wasser (50 ml). Lösung 2 wurde auf 50 °C erwärmt und stark gerührt (ca. 1000 U min 1). Anschließend wurde Lösung 1 schnell mit einer Spritze unter intensivem Rühren injiziert. Nach zweiminütigem Rühren wird der gelbe Feststoff durch Zentrifugieren (15 min bei 22.500 U min 1) abgetrennt. Die Nanopartike! werden zweimal in de- mineralisiertem Wasser (25 ml) resuspendiert und erneut zentrifugiert, um alle verbliebenen Salze zu entfernen. Schließlich werden stabile Suspensionen durch Resuspendieren der Nanopartikel in HEPES-Puffer (12 ml, 30 mmol/l, pH = 7,4) erhalten. Alternativ wird das Zentrifugat in demineralisiertem Wasser (3,1 ml) resuspendiert. Anschließend wird zu dieser Suspension unter Rühren eine Lösung aus Dextran 40 (3 ml, 1 ,6 mg/ml H2O) getropft. Alternativ können die Nanopartikel auch nur in demineralisiertem Wasser redispergiert werden. Das verwendete de- mineralisierte Wasser wird in allen Fällen zuvor mit Hilfe eines sterilen Spritzenfilters (PA, 0,20 pm) vor Verwendung staub- und keimfrei gemacht. Man erhält das anorganisch-organische Hybrid [ZrO]22+[Pt2(POP)4]4 (siehe Figur 1 ), welches [ZrO]2+ als anorganisches Kation sowie das zytostatische und grün emittierende Anion [Pt2(POP)4]4 enthält, als amorphe Nanopartikel mit einem Durchmesser von etwa 40 nm.
Kaliumpyrophosphatoplatinat (K4[Pt2(POP)4)] (siehe Figur 1) wird nach einer literaturbekannten Synthesevo rschrift hergestellt (siehe M. A. Filomena Dos Remedios Pinto, P. J. Sadler, S. Neidle, M. R. Sanderson, A. Subbiah, R. Kuroda, J. Chem. Soc. - Chem. Commun. 1980, 1 , 13)
Ausführungsbeispiel 2: [ZrO]2+2[cisPt(AEP)2]4'
Das anorganisch-organische Hybrid [ZrO]2+2[cisPt(AEP)2]4 (AEP: Aminoethylp- hosphat) wird durch Mischen zweier Lösungen dargestellt. Lösung 1 enthält Zr0Cl2><8H20 (52 mg) in demineralisiertem Wasser (5,0 ml). Lösung 2 enthält Ka- lium-cis-dichloridodiaminoethylphosphatoplatinat(ll) (K4[cisPt(AEP)2]4 , 50,0 mg) in demineralisiertem Wasser (50 ml). Lösung 2 wurde auf 50 °C erwärmt und stark gerührt (ca. 1000 U min-1). Anschließend wurde Lösung 1 schnell mit einer Spritze unter intensivem Rühren injiziert. Nach zweiminütigem Rühren wird der dunkelbraune Feststoff durch Zentrifugieren (15 min bei 22.500 U min 1) abgetrennt. Die Nanopartikel werden zweimal in demineralisiertem Wasser (25 ml) resuspendiert und erneut zentrifugiert, um alle verbliebenen Salze zu entfernen. Schließlich werden stabile Suspensionen durch Resuspendieren der Nanopartikel in HEPES-Puffer (12 ml, 30 mmol/l, pH = 7,4) erhalten. Alternativ wird das Zentrifugat in demineralisiertem Wasser (3,1 ml) resuspendiert. Anschließend wird zu dieser Suspension unter Rühren eine Lösung aus Dextran 40 (3 ml, 1 ,6 mg/ml H2O) getropft. Alternativ können die Nanopartike! auch nur in demineralisiertem Wasser redispergiert werden. Das verwendete demineralisierte Wasser wird in allen Fällen zuvor mit Hilfe eines sterilen Spritzenfilters (PA, 0,20 pm) vor Verwendung staub- und keimfrei gemacht. Man erhält das anorganisch-organische Hybrid [ZrO]2+2[cisPt(AEP)2]4 (siehe Figur 2), welches [ZrO]2+ als anorganisches Kation sowie das zytostatische Anion [cisPt(AEP)2]4 enthält, als amorphe Nanopartikel mit einem Durchmesser von etwa 30 nm.
Zur Darstellung von Kalium-cis-dichloridodiaminoethylphosphatoplatinat(ll) (K4cisPt(AEP)2p) werden 50 mg foPtCU und 34 mg Aminoethylphosphat (AEP) in 10 mL Wasser gelöst und der pH-Wert mittels NaOH auf 7-10 eingestellt. Die Lösung wird für 60 min auf 50°C erwärmt. Dabei findet ein Farbwechsel von hellorange zu dunkelbraun statt. Das braune Produkt K4[cisPt(AEP)2]4~ (siehe Figur 2) wird nachfolgend an der Luft getrocknet.
Ausführungsbeispiel 3: [Ag]+2[cisPt(SA)2]2'
Das anorganisch-organische Hybrid [Ag]+2[cisPt(SA)2]2 (SA: Succinat) wird durch Mischen zweier Lösungen dargestellt. Lösung 1 enthält AgNÜ3 (12 mg) in demineralisiertem Wasser (0,3 ml). Lösung 2 enthält c/s,c/s,frans-Diammindichloridodisuccinatoplatin(IV) (20,0 mg) in demineralisiertem Wasser (50 ml). Lösung 2 wurde auf 50 °C erwärmt und stark gerührt (c . 1000 U min-1). Anschließend wurde Lösung 1 schnell mit einer Spritze unter intensivem Rühren injiziert. Nach zweiminütigem Rühren wird der gelbe Feststoff durch Zentrifugieren (15 min bei 22.500 U min-1) abgetrennt. Die Nanopartikel werden zweimal in demineralisiertem Wasser (25 ml) resuspendiert und erneut zentrifugiert, um alle verbliebenen Salze zu entfernen. Schließlich werden stabile Suspensionen durch Resuspendieren der Nanopartikel in HEPES-Puffer (12 ml, 30 mmol/l, pH = 7,4) erhalten. Alternativ wird das Zentrifugat in demineralisiertem Wasser (3,1 ml) resuspendiert. Anschließend wird zu dieser Suspension unter Rühren eine Lösung aus Dextran 40 (3 ml, 1 ,6 mg/ml H2O) getropft. Alternativ können die Nanopartikel auch nur in demineralisiertem Wasser redispergiert werden. Das ver- wendete demineralisierte Wasser wird in allen Fällen zuvor mit Hilfe eines sterilen Spritzenfilters (PA, 0,20 pm) vor Verwendung staub- und keimfrei gemacht. Man erhält das anorganisch-organische Hybrid [Ag]+2[cisPt(SA)2]2 (siehe Figur 3), wel- ches [Ag]+ als anorganisches Kation sowie das zytostatische, orange emittierende Anion [cisPt(SA)2p enthält, als amorphe Nanopartikel mit einem Durchmesser von etwa 30 nm.
Synthese von cis-Diammindichloridoplatin(ll) (cisPlatin). Abgewandelt nach einer literaturbekannten Synthese werden 166 mg K2PtCl4, 60 mg KCl und 124 mg NH4OAC in 4 mL Wasser gelöst. Die rot-orange Lösung wird in der Mikrowelle innerhalb einer Minute auf 90°C erhitzt und weitere 14 min auf dieser Temperatur gehalten. Anschließend wird das Reaktionsgefäß (50 mL Kolben) im Eisbad ge- kühlt und es werden 16 mL Ethanol zugegeben, um das gelborange Produkt voll ständig auszufällen. Der Niederschlag wird durch Zentrifugation (10 min, 15000 U min 1) abgetrennt. Anschließend wird mit 4 mL DMF extrahiert, um ggf. entstandenes Platin abzutrennen (Zentrifugation, 10 min, 15000 U min 1). Das Produkt wird mit der vierfachen Menge Ethanol wieder aus der gelben Lösung ausgefällt und durch zweimaliges Waschen mit Ethanol aufgereinigt. Nach dem T rocknen wird in 0.1 M HCl bei 90 °C umkristallisiert. Das gelborange Produkt s-Diammindichloridoplatin(ll) (siehe Figur 3) kristallisiert beim Abkühlen aus, wird abzentrifugiert, zweimal mit Ethanol gewaschen und anschließend getrocknet.
Synthese von cis, cis, trans-Diammindichloridodihydroxidoplatin(l V) (cisPlatinOH). Abgewandelt nach einer literaturbekannten Synthese werden 100 mg cisPlatin in 5 mL Wasser suspendiert und mit einem Überschuss an 30%-igem H2O2 (0,6 mL) versetzt. Die Mischung wird unter Rühren für 6 h bei 70 °C und anschließend über Nacht bei Raumtemperatur gerührt. Es findet ein Farbwechsel von gelborange zu hellgelb statt. Das Wasser wird unter Vakuum entfernt, das hellgelbe Produkt c/s,c/s,frans-Diammindichlorido-dihydroxidoplatin(IV) (siehe Figur 3) durch zweimaliges Waschen mit Ethanol aufgereinigt und anschließend getrocknet.
Synthese von cis, cis, trans-Diammindichloridodisuccinatoplatin(IV) (cisPlatinSA). Abgewandelt nach einer literaturbekannten Synthese werden 100 mg (0.3 mmol) cisPlatinOH und 180 mg (1 ,8 mmol) Succinsäureanhydrid in 3 mL DMSO gelöst und unter Rühren für 24 h auf 70°C erhitzt. Anschließend wird das Lösungsmittel unter Vakuum und Erwärmen auf 50°C entfernt. Lösungsmittelreste werden durch mehrmalige Zugabe von einigen mL Wasser und anschließendes T rockenziehen im Vakuum entfernt. Das gelbe Produkt c/s,c/s,frar?s-Diammindichloridodisuccinatoplatin(IV) wird mit Aceton gewaschen und vor der Weiterverwendung getrocknet (siehe Figur 3).
Ausführungsbeispiel 4: [ZrO]2+2[cisPt(PAA)2]4'
Das anorganisch-organische Hybrid [ZrO]2+2[cisPt(PAA)2]4 (PAA: Phosphono- acetat) wird durch Mischen zweier Lösungen dargestellt. Lösung 1 enthält Zr0Cl2x8H20 (21 mg) in demineralisiertem Wasser (0,3 ml). Lösung 2 enthält cis,cis,trans-Diammindichloridodiphosphonoacetatoplatin(IV) (20,0 mg) in demineralisiertem Wasser (50 ml). Lösung 2 wurde auf 50 °C erwärmt und stark gerührt (ca. 1000 U min 1). Anschließend wurde Lösung 1 schnell mit einer Spritze unter intensivem Rühren injiziert. Nach zweiminütigem Rühren wird der gelbliche Fest stoff durch Zentrifugieren (15 min bei 22.500 U min 1) abgetrennt. Die Nanopartikel werden zweimal in demineralisiertem Wasser (25 ml) resuspendiert und erneut zentrifugiert, um alle verbliebenen Salze zu entfernen. Schließlich werden stabile Suspensionen durch Resuspendieren der Nanopartikel in HEPES-Puffer (12 ml, 30 mmol/l, pH = 7,4) erhalten. Alternativ wird das Zentrifugat in demineralisiertem Wasser (3,1 ml) resuspendiert. Anschließend wird zu dieser Suspension unter Rühren eine Lösung aus Dextran 40 (3 ml, 1 ,6 mg/ml H2O) getropft. Alternativ können die Nanopartikel auch nur in demineralisiertem Wasser redispergiert werden. Das verwendete demineralisierte Wasser wird in allen Fällen zuvor mit Hilfe eines sterilen Spritzenfilters (PA, 0,20 pm) vor Verwendung staub- und keimfrei gemacht. Man erhält das anorganisch-organische Hybrid [ZrO]2+2[cisPt(PAA)2]4 (siehe Figur 4), weiches [ZrO]2+ als anorganisches Kation sowie das zytostatische, orange emittierende Anion [cisPt(PAA)2]4 enthält, als amorphe Nanopartikel mit einem Durchmesser von etwa 30 nm.
Synthese von cis,cis,trans-Diammindichloridodiphosphonoacetatoplatin(IV) (cis- PlatinPAA). 100 mg cisPlatinOH und 168.0 mg Phosphonoessigsäure (PAA) wer- den in 3 mL DMSO gelöst und unter Rühren für 24 h auf 70°C erhitzt. Anschließend wird das Lösungsmittel unter Vakuum und Erwärmen auf 50°C entfernt. Lösungsmittelreste werden durch mehrmalige Zugabe von einigen mL Wasser und anschließendes Trockenziehen im Vakuum entfernt. Ausführungsbeispiel 5: [HfO]22+[Pt2{POP)4]4-
Das anorganisch-organische Hybrid [HfO]22+[Pt2( PO P)4]4 (POP: Pyrophosphat) wird durch Mischen zweier Lösungen dargestelit. Lösung 1 enthält Hf0Cl2><nH20 (5,3 mg) in demineralisiertem Wasser (5,0 ml). Lösung 2 enthält Kaliumpyrophos- phatoplatinat(fl) (K4[Pt2(POP)4)], 20,3 mg) in demineralisiertem Wasser (50 ml). Lösung 2 wurde auf 50 °C erwärmt und stark gerührt (ca. 1000 U min 1). Anschließend wurde Lösung 1 schnell mit einer Spritze unter intensivem Rühren inji ziert. Nach zweiminütigem Rühren wird der gelbe Feststoff durch Zentrifugieren (15 min bei 22.500 U min 1) abgetrennt. Die Nanopartikel werden zweimal in demineralisiertem Wasser (25 ml) resuspendiert und erneut zentrifugiert, um alle verbliebenen Salze zu entfernen. Schließlich werden stabile Suspensionen durch Resuspendieren der Nanopartikel in HEPES-Puffer (12 ml, 30 mmol/l, pH = 7,4) erhalten. Alternativ wird das Zentrifugat in demineralisiertem Wasser (3,1 ml) resuspendiert. Anschließend wird zu dieser Suspension unter Rühren eine Lösung aus Dextran 40 (3 ml, 1 ,6 mg/ml H20) getropft. Kaliumpyrophosphatoplatinat (K4[Pt2(POP)4)] wird nach einer literaturbekannten Synthesevorschrift hergestellt.
Ausführungsbeispiel 6: [Gd(OH)I2+[cisPt(SA)2J2- Das anorganisch-organische Hybrid [Gd(OH)]2+[cisPt(SA)2]2 (SA: Succinat) wird durch Mischen zweier Lösungen dargestellt. Lösung 1 enthält GdC x6H20 (13 mg) in demineralisiertem Wasser (0,3 ml). Lösung 2 enthält c/s,c/'s,frans-Diammindichloridodisuccinatoplatin(IV) (20,0 mg) in demineralisiertem Wasser (50 mi). Lösung 2 wurde auf 50 °C erwärmt und stark gerührt (ca. 1000 U min-1). Anschließend wurde Lösung 1 schnell mit einer Spritze unter intensivem Rühren injiziert. Nach zweiminütigem Rühren wird der farblose Feststoff durch Zentrifugieren (15 min bei 22.500 U min 1) abgetrennt. Synthese von cis-Diammindichloridoplatin(ll) (cisPlatin). Abgewandelt nach einer literaturbekannten Synthese werden 166 mg foPtCU, 60 mg KCl und 124 mg NH4OAc in 4 mL Wasser gelöst. Die rot-orange Lösung wird in der Mikrowelle innerhalb einer Minute auf 90°C erhitzt und weitere 14 min auf dieser Temperatur gehalten. Anschließend wird das Reaktionsgefäß (50 mL Kolben) im Eisbad gekühlt und es werden 16 mL Ethanol zugegeben, um das gelborange Produkt voll- ständig auszufällen. Der Niederschlag wird durch Zentrifugation (10 min, 15000 U min 1) abgetrennt. Anschließend wird mit 4 mL DMF extrahiert, um ggf. entstandenes Platin abzutrennen (Zentrifugation, 10 min, 15000 U min 1). Das Produkt wird mit der vierfachen Menge Ethanol wieder aus der gelben Lösung ausgefällt und durch zweimaliges Waschen mit Ethanol aufgereinigt. Nach dem T rocknen wird in 0.1 M HCl bei 90 °C umkristallisiert. Das gelborange Produkt c/s-Diammindichloridoplatin(ll) kristallisiert beim Abkühlen aus, wird abzentrifugiert, zweimal mit Ethanol gewaschen und anschließend getrocknet.
Synthese von cis, cis, trans-Diammindichloridodihydroxidoplatin(IV) (cisPlatinOH). Abgewandelt nach einer literaturbekannten Synthese werden 100 mg cisPlatin in 5 mL Wasser suspendiert und mit einem Überschuss an 30%-igem H2O2 (0,6 mL) versetzt. Die Mischung wird unter Rühren für 6 h bei 70 °C und anschließend über Nacht bei Raumtemperatur gerührt. Es findet ein Farbwechsel von gelborange zu hellgelb statt. Das Wasser wird unter Vakuum entfernt, das hellgelbe Produkt c/s,c/s,frans-Diammindichlorido-dihydroxidoplatin(IV) durch zweimaliges Waschen mit Ethanol aufgereinigt und anschließend getrocknet.
Ausführungsbeispiel 7: [Ba]2+2[cisPt(AEP)2]4'
Das anorganisch-organische Hybrid [Ba]2+2[cisPt(AEP)2]4 (AEP: Aminoethylphos- phat) wird durch Mischen zweier Lösungen dargestellt. Lösung 1 enthält BaCl2 x2H20 (34 mg) in demineralisiertem Wasser (5,0 ml). Lösung 2 enthält Ka- lium-cis-dichloridodiaminoethylphosphatoplatinat(ll) (K4[cisPt(AEP)2]4 , 50,0 mg) in demineralisiertem Wasser (50 ml). Lösung 2 wurde auf 50 °C erwärmt und stark gerührt (ca. 1000 U min 1). Anschließend wurde Lösung 1 schnell mit einer Spritze unter intensivem Rühren injiziert. Nach zweiminütigem Rühren wird der dunkelbraune Feststoff durch Zentrifugieren (15 min bei 22.500 U min-1) abgetrennt. Die Nanopartikel werden zweimal in demineralisiertem Wasser (25 ml) resuspendiert und erneut zentrifugiert, um alle verbliebenen Salze zu entfernen. Schließlich werden stabile Suspensionen durch Resuspendieren der Nanopartikel in HEPES-Puffer (12 ml, 30 mmol/l, pH = 7,4) erhalten. Alternativ wird das Zentrifugat in demineralisiertem Wasser (3,1 ml) resuspendiert. Anschließend wird zu dieser Suspension unter Rühren eine Lösung aus Dextran 40 (3 ml, 1 ,6 mg/ml H2O) getropft. Alternativ können die Nanopartikel auch nur in demineralisiertem Wasser redispergiert werden. Das verwendete demineralisierte Wasser wird in allen Fällen zuvor mit Hilfe eines sterilen Spritzenfilters (PA, 0,20 pm) vor Verwendung staub- und keimfrei gemacht. Zur Darstellung von Kalium-cis-dichloridodiaminoethylphosphatoplatinat(ll) (K4cisPt(AEP)2]4~) werden 50 mg K2PtCU und 34 mg Aminoethylphosphat (AEP) in 10 mL Wasser gelöst und der pH-Wert mittels NaOH auf 7-10 eingestellt. Die Lösung wird für 60 min auf 50°C erwärmt. Dabei findet ein Farbwechsel von hellorange zu dunkelbraun statt. Das braune Produkt K4[dsPt(AEP)2]4 wird nachfol- gend an der Luft getrocknet.
Ausführungsbeispiel 8: [ZrO]2+2[(c is Pt( AE P)2)o.99s( D UT)o.oos]4'
Das anorganisch-organische Hybrid [ZrO]2+2[(cisPt(AEP)2)o.995(DUT)o.oo5]4 (AEP: Aminoethylphosphat) wird durch Mischen zweier Lösungen dargestellt. Lösung 1 enthält Zr0Cl2><8H20 (52 mg) in demineralisiertem Wasser (5,0 ml). Lösung 2 enthält Kalium-cis-dichloridodiaminoethylphosphatoplatinat(ll) (K4[cisPt(AEP)2]4 , 50,0 mg) sowie das im nahen Infrarot emittierende farbstoffmodifizierte Nukleosid- triphosphat DY-647-dUTP (H4(DUT), 1 ,0 mg, Dyomics) in demineralisiertem Wasser (50 ml). Lösung 2 wurde auf 50 °C erwärmt und stark gerührt (ca. 1000 U min-1). Anschließend wurde Lösung 1 schnell mit einer Spritze unter intensivem Rühren injiziert. Nach zweiminütigem Rühren wird der braune Feststoff durch Zentrifugieren (15 min bei 22.500 U mim1) abgetrennt. Die Nanopartikel werden zweimal in demineralisiertem Wasser (25 ml) resuspendiert und erneut zentrifugiert, um alle verbliebenen Salze zu entfernen. Lösung 1 enthält Z1OCI2X8H2O (52 mg) in demineralisiertem Wasser (5,0 ml). Lösung 2 enthält Kalium-cis-dichloridodiaminoethylphosphatoplatinat(ll) (K4[cisPt(AEP)2]4 , 50,0 mg) und Natrium-5‘-Fluor-2‘-deoxyuridin-5‘-monophosphat (Na2(FdUMP), 82 mg) in demineralisiertem Wasser (50 ml). Lösung 2 wurde auf 50 °C erwärmt und stark gerührt (ca. 1000 U mim1). Anschließend wurde Lösung 1 schnell mit einer Spritze unter intensivem Rühren injiziert. Claims

Ansprüche
1. Anorganisch-organische Hybridverbindung zur Verwendung in der Medizin, aufgebaut als lonenverbindung aus einem anorganischen Metaflkation, ausgewählt aus Ba2+, [ZrO]2+, [HfO]2+, Sc3+, Y3+, Gd3+, [GdOf, [Gd(OH)]2+, [LaO]+, [La(OH)]2+, Fe3+, Bi3+, Ag+ oder einem Lanthanoid, und einem organischen Platin-haltigen zytostatischen Anion, welches mindestens eine Phosphat-, Pyrophosphat-, Phosphonat-, Sulfat-, Sulfonat-, Carbonat- oder Carboxylat- gruppe als funktionelle Gruppe enthält, wobei die Verbindung eine molare Löslichkeit von < 10-2 mol/l in Wasser und einen Partikeldurchmesser im Bereich von 1 bis 100 nm aufweist.
2. Anorganisch-organische Hybridverbindung zur Verwendung in der Medizin gemäß Anspruch 1 , wobei das Metallkation aus Ba2+, [ZrO]2+, [HfO]2+, [GdO]+, [Gd(OH)]2+, [LaO]+, [La(OH)]2+ oder Ag+ ausgewählt ist.
3. Anorganisch-organische Hybridverbindung zur Verwendung in der Medizin gemäß Anspruch 1 oder 2, wobei das organische Platin-haltige Anion mindestens eine Phosphat-, Pyrophosphat- oder Phosphonatgruppe als funktioneile Gruppe enthält.
4. Anorganisch-organische Hybridverbindung zur Verwendung in der Medizin nach einem der Ansprüche 1 bis 3, wobei das organische Platin-haltige Anion aus
[Pt2(POP)4] (POP: Pyrophosphat),
[cisPt(AEP)2]4 (AEP: Aminoethylphosphat), oder
[cisPt(PAA)2]4 (PAA: Phosphonoacetat)
ausgewählt ist.
5. Anorganisch-organische Hybridverbindung zur Verwendung in der Medizin nach einem der Ansprüche 1 bis 4, welche weiter ein Fluoreszenzfarbstoff- anion und/oder ein Wirkstoffanion, die jeweils eine Phosphat-, Phosphonat-, Sulfat-, Sulfonat-, Carbonat- oder Carboxylatgruppe als funktionelle Gruppe tragen, umfasst.

Anorganisch-organische Hybridverbindung zur Verwendung in der Medizin nach Anspruch 5, wobei das organische Fluoreszenzfarbstoffanion von Fluoreszenzfarbstoffen abgeleitet ist, ausgewählt aus der Gruppe, bestehend aus 1 ,1'-Diethyl-2,2'-cyaniniodid, 1 ,2-Diphenylacetylen, 1 ,4-Diphenylbutadien, 1 ,6-Diphenylhexatrien, 2,5-Diphenyloxazol, 2-Methylbenzoxazol, 4',6-Diamidino-2-pheny!indol (DAPI),

Anorganisch-organische Hybridverbindung zur Verwendung in der Medizin nach Anspruch 5, 6 oder 7, wobei das zusätzlich eingebaute, organische Wirkstoffanion aus Acetaminophenphosphat, Betamethasonphosphat, Dexamethasonphosphat, 5‘-Fluor-2‘-deoxyuridin-5‘-monophosphat (FdUMP), Methylprednisolonphosphat, T riamcinolonphosphat, Estronphosphat, Est- ramustinphosphat, Codeinphosphat, Clindamycinphosphat, Fospropofol, Alendronat, Canrenoat, Doxycyclin Hydrat, Doxorubicin, Aztreonam, Tige- monam, Cefsulodin, Fosamprenavir, Tenofovir, Adefovir, Folsäure, Fosfomy- cin, a-Tocopherolphosphat oder Glucose-6-phosphat ausgewählt ist.
9. Anorganisch-organische Hybridverbindung zur Verwendung in der Medizin nach einem der Ansprüche 1 bis 8, welche in röntgenamorpher Form vorliegt.
10. Anorganisch-organische Hybridverbindung zur Verwendung in der Medizin nach einem der Ansprüche 1 bis 9, dadurch gekennzeichnet, dass sie frei von entsprechenden, sie umgebenden Matrizes bzw. Einkapselungen, insbe sondere beispielsweise einer Si02-Matrix, sowie frei von jeglichen Phosphor lipid-Matrizes bzw. -Einkapselungen ist.
11. Anorganisch-organische Hybridverbindung zur Verwendung in der Medizin nach einem der Ansprüche 1 bis 10, welche weiter mit einem mit einem oder mehreren tumorspezifischen Liganden funktionalisiert ist.
12. Verfahren zur Herstellung einer anorganisch-organischen Hybridverbindung nach einem der Ansprüche 1 bis 11 , umfassend die Schritte
(a) das Bereitstellen einer Lösung einer Platin-haltigen Anion-Verbindung, die eine oder mehrere funktionelle Gruppen, ausgewählt aus Phosphat-, Py- rophosphat-, Phosphonat-, Sulfat-, Sulfonat, Carbonat- oder Carboxylat- gruppen, aufweist,
(b) das Bereitstellen einer Lösung eines löslichen Metallsalzes, enthaltend Metallkationen, ausgewählt aus Ba2+, [ZrO]2+, [HfO]2+, Sc3+, Y3+, Gd3+, [GdO]+, [Gd(OH)]2+, [LaOf, [La(OH)]2+, Fe3+, Bi3+, Ag+ oder einem Lanthano- id, (c) das Vereinigen der beiden Lösungen unter Rühren, um die Hybridverbindung auszufällen, und
(d) das Isolieren und/oder Aufreinigen der ausgefällten Hybridverbin- dung.
