Abstract
Es wird eine Aufnahmevorrichtung mit einem Anregungsmodul (2, 10; 9, 10), das die Probe (3) zur Abgabe von Druckwellen anregt, einem Akustikmodul (9, 10) zur Detektion der erzeugten Druckwellen, und einem Steuermodul (10) das basierend auf den Daten des Akustikmoduls (9, 10) ein akustisches Bild ermittelt, bereitgestellt, wobei die Aufnahmevorrichtung ferner ein Abbildungsmodul (5) zum optischen Abbilden der Probe (3) aufweist und das Steuermodul (10) basierend auf der optischen Abbildung der Probe (3) eine Probengrenze und/oder eine Segmentgrenze innerhalb der Probe (3) bestimmt und bei der Ermittlung des akustischen Bildes die bestimmte Proben- und/oder Segmentgrenze berücksichtigt.
Description
Aufnahmevorrichtunq und Aufnahmeverfahren
Die vorliegende Erfindung betrifft eine Aufnahmevorrichtung mit einem Anregungsmodul, das eine Probe zur Abbildung von Druckwellen anregt, einem Akustikmodul zur Detektion der erzeugten Druckwellen, und einem Steuermodul, das basierend auf den Daten des Akustikmoduls ein akustisches Bild ermittelt.
Das Anregungsmodul kann beispielsweise die Probe (bzw. einen Teil davon) mit einem Laserpuls (beispielsweise ns-Puls) beleuchten. Zumindest ein Teil der eingebrachten optischen Energie wird von Strukturen in der Probe absorbiert, was zu einer lokalen Aufheizung und einer nachfolgenden thermoelastischen Expansion und somit zu einer Schallwelle führt. Die Schallwelle wird mittels des Akustikmoduls detektiert und kann daher zur Erzeugung eines ortsaufgelösten Bildes dienen. Die so erzeugten akustischen Bilder können Artefakte aufweisen, die z. B. auf eine Brechung der zu detektierenden Schallwellen an einer Grenze zwischen der Probe und dem umgebenden Medium zurückzuführen sind. Ferner können Inhomogenitäten innerhalb der Probe, die beispielsweise aufgrund unterschiedlicher Gewebearten vorliegen, zu Artefakten im erzeugten akustischen Bild führen.
Ausgehend hiervon ist es Aufgabe der Erfindung, eine Aufnahmevorrichtung der eingangs genannten Art bereitzustellen, bei der das ermittelte akustische Bild möglichst wenig Artefakte aufweist. Ferner soll ein entsprechendes Aufnahmeverfahren bereitgestellt werden. Erfindungsgemäß wird die Aufgabe durch eine Aufnahmevorrichtung mit einem Anregungsmodul, das die Probe zur Abgabe von Druckwellen anregt, einem Akustikmodul zur Detektion der erzeugten Druckwellen und einem Steuermodul, das basierend auf den Daten des Akustikmoduls ein akustisches Bild ermittelt, gelöst, wobei die Aufnahmevorrichtung ferner ein Abbildungsmodul zum optischen Abbilden der Probe aufweist und das Steuermodul basierend auf der optischen Abbildung der Probe eine Probengrenze und/oder eine
Segmentgrenze (oder auch Segmentgrenzen) innerhalb der Probe bestimmt und bei der Ermittlung des akustischen Bildes die bestimmte Proben- und/oder Segmentgrenze(n) berücksichtigt. Unter einer Proben- und/oder Segmentgrenze bzw. Segmentgrenzen wird hier insbesondere die gesamte Proben- bzw. Segmentgrenze oder auch nur ein Teil der Proben- und/oder Segmentgrenze verstanden.
Durch diese optische Ermittlung der Proben- und/oder Segmentgrenze(n) kann diese Grenze bei der Ermittlung des akustischen Bildes berücksichtigt werden (z. B. in den entsprechenden Berechnungsalgorithmen), so daß die Artefakte im erzeugten ortsaufgelösten akustischen Bild reduziert sein können.
Insbesondere kann das Steuermodul die Proben- und/oder Segmentgrenze(n) basierend auf einer zweidimensionalen optischen Aufnahme oder einer dreidimensionalen optischen Aufnahme bestimmen.
Ferner kann das Steuermodul die bestimmte Proben- und/oder Segmentgrenze(n) bei einem zur Ermittlung des akustischen Bildes verwendeten akustischen Propagationsmodell der Probe berücksichtigen. Damit ist es möglich, die Artefakte im akustischen Bild zu reduzieren.
Das Abbildungsmodul zum optischen Abbilden kann wie bei einem herkömmlichen Mikroskop ausgebildet sein. Insbesondere kann die Aufnahmevorrichtung auch ein Beleuchtungsmodul zum Beleuchten der Probe aufweisen. Auch das Beleuchtungsmodul kann wie bei einem herkömmlichen optischen Mikroskop ausgebildet sein.
Insbesondere kann für die optische Aufnahme jede bekannte optische Bildgebungstechnik eingesetzt werden, wie z. B. die Durchlichtmikroskopie, die Auflichtmikroskopie, die optische Projektionstomographie und/oder die Mikroskopie mit Lichtblattbeleuchtung für eine optische Schnittaufnahme. Dabei kann z. B. auf Phasen-, Fluoreszenz- und/oder Absorptionskontrast zurückgegriffen werden.
Das Abbildungsmodul kann z. B. als Laser-Scanning-Mikroskop ausgebildet sein. Ferner kann das Abbildungsmodul ein Objektiv aufweisen. Das Objektiv kann insbesondere ein Immersionsobjektiv sein.
Das Anregungsmodul kann zur Beaufschlagung der Probe mit elektromagnetischer Strahlung ausgebildet sein, um die Druckwellen zu erzeugen. Insbesondere kann die Anregung über das Abbildungsmodul zum optischen Abbilden durchgeführt werden. Es ist auch möglich, daß ein Drucksensor des Akustikmoduls Teil des Anregungsmoduls ist und zur Erzeugung von auf die Probe gerichtete Schallwellen eingesetzt wird.
Bei der erfindungsgemäßen Aufnahmevorrichtung kann das Abbildungsmodul ein Objektiv ausweisen, wobei das Objektiv eine probenseitige Frontlinse umfaßt, die einen optisch genutzten Mittelbereich festlegt, und das Akustikmodul einen ringförmigen Drucksensor aufweist, der im Bereich des probenseitigen Endes des Objektives so angeordnet und dessen Innendurchmesser so gewählt ist, daß er, in Richtung der optischen Achse des Objektives gesehen, den optisch genutzten Mittelbereich nicht überdeckt. Mit einer solchen Aufnahmevorrichtung wird vorteilhaft erreicht, daß die optische Detektion nicht beeinflußt wird, da der optisch genutzte Mittelbereich durch den Drucksensor nicht überdeckt wird. Andererseits kann durch die Anordnung des Drucksensors im Bereich des probenseitigen Endes des Objektives sichergestellt werden, daß die notwendige akustische Kopplung zwischen Drucksensors und Probe bereitgestellt werden kann.
Der Drucksensor kann an einem Dämpfungskörper befestigt sein, der wiederum am Objektivtubus befestigt ist. Damit können unerwünschte Schallreflexionen am Objektivtubus vermindert werden. Ferner ist es möglich, den Drucksensor vom Objektiv beabstandet anzuordnen, wobei hierunter insbesondere eine Anordnung verstanden wird, bei der keine direkte Verbindung zwischen Drucksensor und Objektiv vorliegt. Beispielsweise kann der Drucksensor an einem Deckglas oder Probenträger angeordnet sein. Es ist auch eine Anordnung an einer Wandung in einer Probenkammer möglich.
Bei der erfindungsgemäßen Aufnahmevorrichtung kann das Objektiv einen Objektivtubus aufweisen und der Drucksensor kann am Objektivtubus befestigt sein. Dadurch wird ein sehr kompakter Aufbau realisiert. Der Drucksensor kann einen Piezokeramik-Transducer aufweisen. Mit einem solchen Transducer ist eine gute Schalldetektion möglich.
Ferner kann der Drucksensor eine optisch detektierbare Eigenschaft aufweisen.
Das Objektiv kann als Immersionsobjektiv ausgebildet sein. Dadurch ist es möglich, daß das Immersionsmedium auch mit dem Drucksensor in Kontakt steht, wodurch eine gute optische Kopplung möglich ist.
Das Anregungsmodul kann so ausgebildet sein, daß es die Probe mit elektromagnetischer Strahlung beaufschlagt, um Druckwellen zu erzeugen. Bei der elektromagnetischen Strahlung kann es sich insbesondere um Strahlung aus dem Bereich von 300 nm bis 3 μιτι, bevorzugt 300 nm bis 1300 nm, 300 nm bis 1000 nm, 300 nm bis 700 nm, 700 nm bis 3 μηη, 700 nm bis 1300 nm oder 700 nm bis 1000 nm handeln. Insbesondere handelt es sich um gepulste Laserstrahlen. Die Pulslänge kann im Bereich von ns liegen.
Der Drucksensor kann Teil des Anregungsmoduls sein und zur Erzeugung von auf die Probe gerichteten Schallwellen eingesetzt werden. In diesem Fall dient der Drucksensor zur Erzeugung von Druck- bzw. Schallwellen und zur Detektion der aus der Probe zurückkommenden Schallantwort.
Die erfindungsgemäße Aufnahmevorrichtung kann als Mikroskop ausgebildet sein und kann weitere dem Fachmann bekannte Einheiten und Module zum Betrieb des Mikroskops umfassen.
Die Aufgabe wird ferner gelöst durch ein Aufnahmeverfahren, bei dem eine Probe zur Abgabe von Druckwellen angeregt wird, die erzeugten Druckwellen detektiert werden und basierend auf den detektierten Druckwellen ein akustisches Bild ermittelt wird, wobei ferner eine optische Abbildung der Probe durchgeführt wird und basierend auf der optischen Abbildung der Probe eine Probengrenze und/oder eine Segmentgrenze (oder auch Segmentgrenzen) innerhalb der Probe bestimmt und bei der Ermittlung des akustischen Bildes die bestimmte Proben- und/oder Segmentgrenze(n) berücksichtigt wird. Das erfindungsgemäße Aufnahmeverfahren kann so weitergebildet werden, daß die in Verbindung mit der erfindungsgemäßen Aufnahmevorrichtung (einschließlich der beschriebenen Weiterbildung) beschriebenen Verfahrensschritte ausgeführt werden. Ferner kann die erfindungsgemäße Aufnahmevorrichtung so weitergebildet werden, daß die in Verbindung mit dem erfindungsgemäßen Aufnahmeverfahren (einschließlich der Weiterbildungen) beschriebenen Verfahrensschritte durchführbar sind.
Es versteht sich, daß die vorstehend genannten und die nachstehend noch zu erläuternden Merkmale nicht nur in den angegebenen Kombinationen, sondern auch in anderen
Kombinationen oder in Alleinstellung einsetzbar sind, ohne den Rahmen der vorliegenden Erfindung zu verlassen.
Nachfolgend wird die Erfindung beispielsweise anhand der beigefügten Zeichnungen, die auch erfindungswesentliche Merkmale offenbaren, noch näher erläutert. Es zeigen:

Bei der in Fig . 1 gezeigten Ausführungsform ist die erfindungsgemäße Aufnahmevorrichtung 1 als Mikroskop ausgebildet und umfaßt ein Beleuchtungsmodul 2 zum Beleuchten einer Probe 3 und ein ein Objektiv 4 aufweisendes Abbildungsmodul 5 zum Abbilden der Probe 3.
Das Objektiv 4 ist als Immersionsobjektiv ausgebildet. Daher ist in der schematischen Darstellung von Fig . 1 neben der Probe 3, die zwischen einem Deckglas 6 und einem Objektträger 7 liegt, ein Immersionsmedium 8 zwischen dem Deckglas 6 und dem dem Deckglas 6 zugewandten Ende des Objektives 4 eingezeichnet.
Ferner umfaßt das Mikroskop 1 einen ringförmigen Drucksensor 9, der am dem Deckglas 6 bzw. der Probe 3 zugewandten Ende des Objektives 4 angeordnet ist, ein Steuermodul 10 und eine Ausgabeeinheit 1 1 .
Mittels des Steuermoduls 10 kann das Beleuchtungsmodul 2 so angesteuert werden, daß es gepulste elektromagnetische Strahlung im Bereich von z. B. 300 nm - 3 μιτι (nachfolgend auch Anregungsstrahlung genannt) erzeugt, die über eine im Beleuchtungsmodul 2 enthaltene
Ablenkeinheit 12 und das Objektiv 4 in die Probe 3 (z. B. als Fokusspot) fokussiert und in dieser bewegt wird. Ein Teil der dabei eingebrachten Energie wird von Strukturen in der Probe 3 absorbiert, was zu einer lokalen Aufheizung und nachfolgenden thermoelastischen Expansion und somit zu einer Druck- bzw. Schallwelle führt.
Die Schallwelle wird, wenn die Probe 3 z. B. eine biologische Probe ist, bei der Ausbreitung durch die Probe sehr wenig gestreut und kann daher zur Erzeugung eines ortsaufgelösten Bildes dienen, wobei eine hohe Eindringtiefe bei der Bildgebung von beispielsweise größer als 1 mm möglich ist. Zur Detektion der Schallwellen dient der ringförmige Drucksensor 9.
Wie in der vergrößerten Schnittdarstellung des probenseitigen Endes des Objektives 4 in Fig. 2 ersichtlich ist, umfaßt das Objektiv 4 einen Objektivtubus 13, in dem mehrere Linsen 14 und eine probenseitige Frontlinse 15 angeordnet sind. Die Frontlinse 15 legt einen optisch genutzten Mittelbereich 16 fest, der zur Beaufschlagung der Probe mit der gepulsten Anregungsstrahlung und der für eine herkömmliche optische Abbildung der Probe 3 über das Objektiv verwendet wird. Am frontseitigen Ende des Objektives 4 ist der ringförmige Drucksensor 9 angeordnet, wobei der Innendurchmesser und die Position des Drucksensors 9 so gewählt sind, daß, in Richtung der optischen Achse 17 des Objektives 4 gesehen, der Drucksensor 9 nicht den optisch genutzten Mittelbereich 16 überdeckt. Der Drucksensor 9, der auch als Ultraschallsensor bezeichnet werden kann, kann aus einer Piezokeramik gebildet sein, so daß eine gute Detektion der Schallwellen möglich ist. Durch die Anordnung des Drucksensors 9 am frontseitigen Ende des Objektives 4 ist der Drucksensor 9 während des Betriebes des Mikroskops 1 in Kontakt mit dem Immersionsmedium 8, so daß eine gute akustische Ankopplung von der Probe 3 an den Drucksensor 9 vorliegt.
Der Drucksensor 9 ist, wie schematisch in Fig. 1 gezeigt, mit dem Steuermodul 10 verbunden. Das Steuermodul 10 kann basierend auf den Meßdaten des Drucksensors 9 Bilddaten erzeugen, so daß eine photoakustische Bildgebung realisiert ist. Die Bilddaten können beispielsweise über die Ausgabeeinheit 1 1 angezeigt werden.
Durch die erfindungsgemäße Anordnung des Drucksensors 9 liegt keine Einschränkung der Funktion des Objektives 4 vor, so daß eine herkömmliche Lichtmikroskopie mit dem Objektiv 4 weiterhin möglich ist. Dies kann zur Aufnahme eines Vorschaukontrastbildes, eines Fluoreszenzkontrastbildes, etc. dienen.
Ferner kann die hohe numerische Apertur des Immersionsobjektives 4 genutzt werden, um einen sehr kleinen Fokus der Anregungsstrahlung in der Probe 3 zur Anregung der Druckwellen zu erzeugen. Es ist somit eine lokalisierte Anregung der Probe 3 mit der gepulsten
Anregungsstrahlung (beispielsweise Laserstrahlung mit ns-Pulsen) möglich, womit eine hohe räumliche Auflösung im photoakustischen Bildgebungsmodus erreicht wird. Die die Druckwellen erzeugende Anregungsstrahlung (insbesondere Laserstrahlung) kann die Probe 3 in einer Ebene senkrecht zur optischen Achse 17 abrastern. Dies kann beispielsweise durch einen in der Pupille des Objektives 4 angeordneten Scanspiegel (nicht gezeigt) der Ablenkeinheit 12 verwirklicht werden, wie dies üblicherweise bei Laser-Scanning-Mikroskopen der Fall ist. Zusätzlich kann die gepulste Anregungsstrahlung die Probe 3 in Richtung der optischen Achse 17 abscannen, indem die Fokusebene der Anregungsstrahlung entsprechend eingestellt wird. Alternativ oder zusätzlich kann natürlich auch die Probe 3 entsprechend bewegt werden.
Durch die erfindungsgemäße Anordnung des ringförmigen Drucksensors 9 am frontseitigen Ende des Objektivtubus 13 liegt eine Kompatibilität mit bestehenden Mikroskopsystemen vor. Es muß lediglich das erfindungsgemäße Objektiv bei bestehenden Mikroskopsystemen eingesetzt werden.
Wie bereits ausgeführt wurde, kann das Steuermodul 10 basierend auf den Meßdaten des Drucksensors 9 akustische Bilddaten erzeugen.
Die erzeugten akustischen Bilddaten können Artefakte aufweisen, die z. B. auf eine Brechung der zu detektierenden Druck- bzw. Schallwellen an der Grenze zwischen der Probe 3 und dem umgebenden Medium 8 zurückzuführen sind. Auch Inhomogenitäten innerhalb der Probe 3, die beispielsweise aufgrund unterschiedlicher Gewebearten vorliegen, können zu Artefakten im akustischen Bild führen. So ist z. B. die Schallgeschwindigkeit in Knochen-, Lungen- und Hirngewebe deutlich unterschiedlich, was im allgemeinen zu Brechung und Reflexion der Schallwellen an den Gewebegrenzen führt.
Daher wird bei der erfindungsgemäßen Aufnahmevorrichtung 1 zusätzlich eine optische Abbildung der Probe durchgeführt. Basierend auf der optischen Abbildung der Probe ermittelt das Steuermodul 10 die Probengrenze und/oder eine Segmentgrenze (oder auch Segmentgrenzen) innerhalb der Probe. Unter einer Segmentgrenze wird hier insbesondere eine Grenze verstanden, an der sich eine im wesentlichen konstante akustische Eigenschaft innerhalb der Probe ändert. Diese optisch bestimmte Proben- und/oder Segmentgrenze berücksichtigt das Steuermodul bei der Ermittlung des akustischen Bildes basierend auf den Daten des Drucksensors 9. So kann z. B. das Steuermodul 10 die Informationen bezüglich der Proben- und/oder Segmentgrenze bei der akustischen Bildbestimmung bzw. Bildrekonstruktion als Parameter und/oder Randbedingung nutzen. Beispielsweise können diese Grenzen bei einem eingesetzten akustischen Propagationsmodell zur Rekonstruktion des akustischen Bildes berücksichtigt werden.
Man kann somit sagen, daß erfindungsgemäß aus dem optischen Bild ein Bereich oder Bereiche mit zumindest annähernd konstanter akustischer Impedanz abgeleitet werden. Sofern aus dem optischen Bild keine direkten Rückschlüsse auf den konkreten Wert der akustischen Impedanz in einem bestimmten Bereich abgeleitet werden können, können auch Erfahrungswerte oder sonstige sinnvolle Werte verwendet werden. Diese Werte können in einer Datenbank abgelegt sein, die in dem Steuermodul 10 enthalten ist oder die für das Steuermodul 10 zugänglich ist. Diese hinterlegten Werte können beispielsweise von dem Steuermodul 10 automatisch aufgrund der Form und/oder Ausdehnung des jeweiligen Probensegments bzw. Probenbereichs ausgewählt werden. Dies wird dann bei der akustischen Bildrekonstruktion berücksichtigt, wodurch die Artefakte im akustischen Bild reduziert sind.
Die optische Abbildung der Probe kann auf unterschiedlichste Art und Weise durchgeführt werden. Sie kann, wie bereits beschrieben, im Gerät für die akustische Detektion durchgeführt werden. Sie kann jedoch auch in einem separaten Gerät durchgeführt werden.
Als optische Bildgebungstechnik kann z. B. eine Durchlichtmikroskopie, eine Auflichtmikroskopie, eine optische Projektionstomographie und/oder eine Mikroskopie mit Lichtblattbeleuchtung für eine optische Schnittaufnahme eingesetzt werden. Ferner kann beispielsweise auf Phasen-, Fluoreszenz- und/oder Absorptionskontrast zurückgegriffen werden. Die optischen Aufnahmen der Probe können zweidimensionale Aufnahmen oder dreidimensionale Aufnahmen sein. Zur Erzeugung von dreidimensionalen Aufnahmen können mehrere optische Aufnahmen durchgeführt werden. Dazu kann beispielsweise die Probe und/oder die Aufnahmevorrichtung 1 zwischen den Einzelaufnahmen selbst gedreht werden.
Die optische Abbildung der Probe kann unter einer Beleuchtung mit nur einer Wellenlänge, mehreren Wellenlängen oder eines Wellenlängenbereiches durchgeführt werden.
Neben der hier beschriebenen Art und Weise der akustischen Detektion sind auch weitere dem Fachmann bekannte akustische Detektionen möglich.
In Fig. 3 ist eine Ansicht von unten auf das frontseitige Ende des Objektives 4 gezeigt. Dort ist nochmals klar ersichtlich, daß der Drucksensor 9 die Frontlinse 15 umgibt und nicht überdeckt und somit auch den optisch genutzten Mittelbereich 16 der Frontlinse 15 nicht überdeckt.
Der Drucksensor 9 kann mit einer Schutzummantelung für eine bessere Reinigung bzw. einen Schutz versehen sein. Dabei kann es sich um eine Kunststoffummantelung handeln.
Ferner kann der Drucksensor 9 nicht direkt am Objektivtubus 13 befestigt sein, sondern es kann zwischen dem Drucksensor 9 und dem Objektivtubus 13 ein Dämpfungskörper (nicht dargestellt) angeordnet sein. Damit kann eine rückseitige Entkopplung des Drucksensors 9 vom Objektivtubus 13 zur Vermeidung von Schallreflexionen realisiert werden.
In Fig. 1 ist schematisch eine separate Verbindung vom Steuermodul 10 zum Drucksensor 9 dargestellt. Natürlich kann beispielsweise bei einer elektrischen Kontaktierung das Objektiv 4 auch so ausgebildet sein, daß die elektrischen Kontakte im Objektivflansch angeordnet sind. Der in Fig. 1 gezeigte Aufbau des erfindungsgemäßen Mikroskops ist nur beispielhaft zu verstehen. Neben dem in Fig. 1 gezeigten Auflichtaufbau sind natürlich auch andere Varianten möglich. So kann das Mikroskop auch als invertiertes Mikroskop ausgebildet sein, bei dem das Objektiv 4 unterhalb der Probe 3 angeordnet ist. Ferner ist es möglich, daß das Objektiv 4 seitlich in eine wassergefüllte Probenkammer 20 schaut, wie schematisch in Fig. 4 dargestellt ist. Bei dieser Ausführungsform ist eine Abdichtung zwischen dem Objektivtubus 13 und der Probenkammer 20 z. B. durch die schematisch eingezeichnete Ringdichtung 21 realisiert. Ferner kann neben der bereits beschriebenen Beleuchtung über das Objektiv 4 eine Durchlichtbeleuchtung mit einem optionalen Beleuchtungsmodul 22 (gestrichelt eingezeichnet) und/oder eine seitliche Lichtblattbeleuchtung mit einem weiteren optionalen Beleuchtungsmodul 23 (gestrichelt eingezeichnet) durchgeführt werden. Diese möglichen Beleuchtungen werden durch die Pfeile P1 und P2 angedeutet.
In Fig. 5 ist eine Abwandlung der Ausführungsform von Fig. 4 gezeigt. Bei dieser Abwandlung ist der ringförmige Drucksensor 9 nicht mehr direkt am Objektivtubus 13 befestigt, sondern in der dem Objektiv 4 zugewandten Probenkammerwand 22. Dabei ist der Drucksensor 9 wiederum so angeordnet, daß er, in Richtung der optischen Achse 17 gesehen, nicht den optisch genutzten Mittelbereich 16 der Frontlinse 15 überdeckt. Ferner ist er so in der Probenkammerwand 22 angeordnet, daß er in Kontakt mit dem Wasser in der Probenkammer 20 oder dem sonstigen Medium in der Probenkammer 20 steht, um die gewünschte gute Schallankopplung zu realisieren.
In Fig. 6 ist eine weitere Ausführungsform der erfindungsgemäßen Aufnahmevorrichtung 1 gezeigt, wobei die erfindungsgemäße Aufnahmevorrichtung 1 ein Optikmodul 24 zum optischen Abbilden der Probe 3 aufweist, wie durch den Doppelpfeil P3 angedeutet ist. Das Optikmodul 24 kann z. B. ein Beleuchtungsmodul und ein Abbildungsmodul enthalten.
Ferner umfaßt die Aufnahmevorrichtung 1 eine Halteeinrichtung 25, die vom Steuermodul 10 angesteuert wird. Das Steuermodul 10 steht auch in Verbindung mit dem Optikmodul 24 und dem Drucksensor 9. Wie durch den Pfeil P4 angedeutet ist, werden die Druckwellen mittels dem Drucksensor 9 detektiert.
Mittels der Halteeinrichtung 25 kann die Probe 3 gedreht werden, um die gewünschten optischen und/oder akustischen Aufnahmen durchzuführen.
Ferner kann noch optional eine Eingabeeinheit 26 vorgesehen sein, wie durch die schematisch dargestellte Computermaus angedeutet ist. Über die Eingabeeinheit können Eingaben an das Steuermodul 10 durchgeführt werden.
Bei den bisher beschriebenen Ausführungsformen wurde die Anregung der Schallwellen stets optisch durchgeführt. Es ist jedoch auch möglich, den Drucksensor 9 zur Schallerzeugung einzusetzen. Bei dieser Art der Detektion steuert das Steuermodul 10 den Drucksensor 9 so an, daß er für eine vorbestimmte Zeit Schallwellen in die Probe 3 sendet und die aus der Probe 3 zurückkommende Schallantwort detektiert. Die Frequenzen der Ultraschallwellen liegen beispielsweise bei 20 MHz oder größer. Wie bereits ausgeführt wurde, kann der Drucksensor 9 aus einer Piezokeramik gebildet sein. Man kann somit den piezoelektrischen Effekt zur Wandlung von Schallenergie in elektrische Signale zur Druckdetektion nutzen. Auch kann der piezoelektrische Effekt genutzt werden, um elektrische Signale in Drucksignale umzuwandeln, für den Fall, daß der Drucksensor 9 als Schallquelle eingesetzt wird.
Neben dieser Art der Druckdetektion ist auch jede andere mögliche Art der Druckdetektion möglich. So kann z. B. ein Fiber-Bragg-Sensor oder eine Wellenleiterstruktur für eine optische Detektion der Ultraschallwellen eingesetzt werden. In diesem Fall liegt eine optisch detektierbare druckabhängige Eigenschaft des Sensors vor, die optisch detektiert wird. Der Drucksensor kann als resonanter und/oder breitbandiger Drucksensor ausgelegt sein.
Das erfindungsgemäße Mikroskop 1 kann so ausgebildet sein, daß die Anregung der zu detektierenden Druckwellen optisch und/oder durch Schallwellen möglich ist.
Claims
1. Aufnahmevorrichtung mit
einem Anregungsmodul (2, 10; 9, 10), das die Probe (3) zur Abgabe von Druckwellen anregt, einem Akustikmodul (9, 10) zur Detektion der erzeugten Druckwellen, und
einem Steuermodul (10) das basierend auf den Daten des Akustikmoduls (9, 10) ein akustisches Bild ermittelt,
wobei die Aufnahmevorrichtung ferner ein Abbildungsmodul (5) zum optischen Abbilden der Probe (3) aufweist und das Steuermodul (10) basierend auf der optischen Abbildung der Probe (3) eine Probengrenze und/oder eine Segmentgrenze innerhalb der Probe (3) bestimmt und bei der Ermittlung des akustischen Bildes die bestimmte Proben- und/oder Segmentgrenze berücksichtigt.
2. Aufnahmevorrichtung nach Anspruch 1 , bei der das Steuermodul (10) basierend auf einer zweidimensionalen optischen Aufnahme die Proben- und/oder Segmentgrenze bestimmt.
3. Aufnahmevorrichtung nach Anspruch 1 , bei der das Steuermodul (10) basierend auf einer dreidimensionalen optischen Aufnahme die Proben- und/oder Segmentgrenze bestimmt.
4. Aufnahmevorrichtung nach einem der obigen Ansprüche, bei der das Steuermodul (10) die bestimmte Proben- und/oder Segmentgrenze bei einem zur Ermittlung des akustischen Bildes verwendeten akustischen Propagationsmodell der Probe (3) berücksichtigt.
5. Aufnahmevorrichtung nach einem der obigen Ansprüche, mit einem Beleuchtungsmodul (2) zum Beleuchten einer Probe (3).
6. Aufnahmevorrichtung nach einem der obigen Ansprüche, bei der das Abbildungsmodul (5) ein Objektiv (4) aufweist.
7. Aufnahmevorrichtung nach Anspruch 6, bei dem das Objektiv (4) als Immersionsobjektiv ausgebildet ist.
8. Aufnahmevorrichtung nach einem der obigen Ansprüche, bei dem das Anregungsmodul (2, 10) zur Beaufschlagung der Probe (3) mit elektromagnetischer Strahlung ausgebildet ist, um
Druckwellen zu erzeugen.
9. Aufnahmevorrichtung nach einem der obigen Ansprüche, bei dem ein Drucksensor (9) des Akustikmoduls Teil des Anregungsmoduls ist und zur Erzeugung von auf die Probe (3) gerichteter Schallwellen eingesetzt wird.
10. Aufnahmeverfahren, bei dem
eine Probe zur Abgabe von Druckwellen angeregt wird,
die erzeugten Druckwellen detektiert werden und
basierend auf den detektierten Druckwellen ein akustisches Bild ermittelt wird,
wobei ferner eine optische Abbildung der Probe durchgeführt wird und basierend auf der optischen Abbildung der Probe eine Probengrenze und/oder eine Segmentgrenze innerhalb der Probe bestimmt und bei der Ermittlung des akustischen Bildes die bestimmte Proben- und/oder Segmentgrenze berücksichtigt wird.
