Abstract
Die vorliegende Erfindung betrifft pharmazeutisch wirksame Verbindungen aus einem Isoflavon oder Isoflavon-Glycosid und, kovalent mit diesem verbunden, einer Aminosäure, einem Peptid oder Peptid-Derivat mit 2 bis 5 Aminosäuren, sowie pharmazeutisch akzeptable Salze oder Solvate dieser Verbindungen, sowie Verwendungen solcher Stoffe für die Herstellung von pharmazeutischen Zusammensetzungen, insbesondere für die Behandlung von Thrombozytenaggregation und für die Tumortherapie. Anwendungsgebiete der Erfindung sind demzufolge die Medizin und die pharmazeutische Industrie. Die erfindungsgemäßen Verbindungen haben die allgemeine Formel (I) X-Pep, wobei X ein Isoflavon oder Isoflavon-Glycosid ist, und Pep eine Aminosäure oder ein Peptid oder Peptid-Derivat mit 2 bis 5 Aminosäuren ist, und zwischen X und Pep entweder durch eine kovalente Bindung oder ein Linkersystem eine Verknüpfung besteht.
Description
Pharmazeutisch wirksame Verbindungen
Die vorliegende Erfindung betrifft pharmazeutisch wirksame Verbindungen aus einem Isoflavon oder Isoflavon-Glycosid und, kovalent mit diesem verbunden, einer Aminosäure, einem Peptid oder Peptid-Derivat mit 2 bis 5 Aminosäuren, sowie pharmazeutisch akzeptable Salze oder Solvate dieser Verbindungen, sowie Verwendungen solcher Stoffe für die Herstellung von pharmazeutischen Zusammensetzungen, insbesondere für die Behandlung von Thrombozytenaggregation und für die Tumortherapie. Anwendungsgebiete der Erfindung sind demzufolge die Medizin und die pharmazeutische Industrie.
Isoflavone, gelegentlich auch Isoflavonoide genannt, bilden eine Gruppe von zumeist gelblich gefärbten Pflanzenfarbstoffen, die sich vom Isoflavon ableiten. Im Rahmen dieser Erfindung werden zu den Isoflavonen auch deren Glycoside gezählt. Zu den bekannteren Isoflavonen zählen insbesondere Isoflavon, Daidzein, Genistein, Prunetin, Biochanin A, Orobol, Santal, Glycitein, Pratensein, Formononetin, Genistin, 6"-O- Malonylgenistin, 6"-O-Acetylgenistin, Daidzin, 6"-O-Malonyldaidzin, 6"-O-Acetyldaidzin, Glycitin, Ononin und Sissotrin und ferner Genistin, Daidzin, 6"-O-Malonylglycitin und 6"- O-Acetylglycitin. Die Malonyl-Glucoside des Genisteins machen den Hauptanteil der Isoflavone in Sojabohnen aus. In fermentierten Sojaprodukten liegen die Isoflavone hauptsächlich in ihrer jeweiligen Aglycon-Form Genistein, Daidzein und Glycitein vor.
Von einigen Isoflavonen ist bekannt, dass sie eine Estrogen-Wirkung beispielsweise auf Weidetiere zeigen. Von anderen Isoflavonen wird vermutet, dass sie eine antioxidative Wirkung im Menschen oder anderen Säugetieren besitzen, ein Beweis einer solchen Wirkung ist bisher nicht gelungen. Ebenfalls umstritten ist, ob und ggf. welche Isoflavone eine anti-karzinogene, anti-atherogene, anti-osteoporotische und/oder hypolipidemische Wirkung besitzen. Vielfach war es nämlich bisher nicht möglich, die den Isoflavonen zugeschriebenen Wirkungen durch Verabreichen des reinen Isoflavons, ggf. mit üblichen pharmazeutischen Träger- und Hilfsstoffen, zu reproduzieren. So ist es insbesondere nicht gelungen, eine antioxidative Wirkung von Genistein trotz Gabe hoher Dosen und Nachweis einer hohen Konzentration von Genistein in den Zielzellen eines Menschen zu erreichen.
Beispielsweise hemmen bekanntermaßen Isoflavone, insbesondere Genistein, die Thrombozytenaggregation dosisabhängig (1 ). Dabei ist der Einfluss des Isoflavons auf die Funktion der Thrombozyten komplexer Natur: Einerseits blockiert insbesondere Genistein diverse Membranrezeptoren (Adenosinrezeptor, von Willebrand Faktor) (2, 3), und auf Grund seiner charakteristischen chemischen Struktur insbesondere auch den Thromboxan A2 Rezeptor (4). Andererseits hemmt Genistein die Thrombozytenaggregation durch Modulation intrazellulärer Stoffwechselvorgänge: Hemmung der Tyrosinkinase im Stoffwechsel der Cyclooxygenase (5), Modulation des cAMP durch Hemmung der Phosphodiesterase (6), Reduktion intrazellulärer Verfügbarkeit von Peroxid als wichtigem Agens von Phospholipase C und Arachidonsäurestoffwechsel (7, 8).
Genistein ist ein aus Soja gewonnenes Isoflavon. Verschiedene Untersuchungen zeigen einen hemmenden Effekt von Genistein auf das Wachstum von bösartigen Zellen, wie zum Beispiel denen von hämatopoetischen Tumoren wie Leukämien und Lymphomen, Melanomen und epithelialen Tumoren wie Mamma-, Lungen-, Prostata- und Kopf-Hals-Plattenepithelkarzinomen (7-5)
Die wachstumshemmende Aktivität von Genistein beruht auf seiner antiproliferativen und proapoptotischen Wirkung (6). Sie zeigte sich aber aufgrund der eingeschränkten zellulären Aufnahme von Genistein in Zellkultursystemen erst bei relativ hohen Konzentrationen, die in vivo nur schwer zu erreichen sind (7).
Es wurde bereits vorgeschlagen, synergistische pharmazeutische Zusammensetzungen aus Isoflavonen oder Isoflavon-Glycosiden mit kurzkettigen Peptiden herzustellen (PCT/DE2006/001795). In diesen Zusammensetzungen war die Verfügbarkeit der Isoflavone durch verbesserte zelluläre Aufnahme erheblich gesteigert. Jedoch sind hierfür vergleichsweise hohe Konzentrationen an Substrat - insbesondere Peptid - notwendig. Einerseits kommt es bei oraler Applikation zu einer teilweisen Aufspaltung der verwendeten Peptide bei der Magen-Darmpassage, andererseits liegen die Plasmahalbwertzeiten für Peptide im Bereich von Minuten, was die Verfügbarkeit vermindert sowie unerwünschte Nebeneffekte hervorruft. Beispielsweise sind solche Nebeneffekte von Glutaminsäure (als Abbauprodukt von GIu-GIu) im Sinne von zentralnervösen Störungen beschrieben worden. Das hat zur Folge, dass eine präzise Steuerbarkeit von Plasmaspiegeln nach Applikation, wie sie für die Therapie bestimmter Erkrankungen unabdingbar ist, nicht erreicht wird.
Aufgabe der vorliegenden Erfindung war es daher, den oben beschriebenen Nachteilen abzuhelfen oder sie zumindest zu mildern. Insbesondere sollten Verbindungen bereitgestellt werden, die neben unterschiedlichsten Anwendungsmöglichkeiten insbesondere als Pharmakon i.S. eines Genisteinderivates verwendbar sind, wobei durch verbesserte intrazelluläre Verfügbarkeit bspw. eine dem Genistein bzw. den
Zusammensetzungen Isoflavone/kurzkettige Peptide überlegene Hemmung der
Thrombozytenaggregation oder eine verbesserte Anti-Tumor-Aktivität erreicht werden sollte. Soweit möglich sollten auch weitere Anwendungsgebiete von Isoflavonen erschlossen werden.
Diese Aufgabe wird durch Verbindungen gemäß Anspruch 1 , pharmazeutische Zusammensetzungen gemäß Anspruch 10, und die Verwendung solcher Verbindungen gemäß einem der Ansprüche 11 oder 14-16 gelöst. Die weiteren Ansprüche sind bevorzugte Ausgestaltungen und Ausführungsformen der Erfindung.
Vollkommen überraschend hat sich herausgestellt, dass Verbindungen gemäß der Formel (I),
(I) X-Pep ,
wobei X ein Isoflavon oder Isoflavon-Glycosid ist und Pep eine Aminosäure oder ein Peptid oder Peptid-Derivat mit 2 bis 5 Aminosäuren ist, und wobei zwischen X und Pep eine kovalente Verbindung besteht oder die Verknüpfung durch ein geeignetes Linkersystem (wie Ethylenglykol, Ethanolamin, höhere Homologe davon oder die entsprechenden Polyethylenglykolderivate) hergestellt wird, sowie pharmazeutisch akzeptable Salze oder Solvate dieser Verbindungen, die Wirkungen des Isoflavonbestandteils der Verbindung auf Organismen, insbesondere Säugetiere, verstärken oder biologische Wirkungen überhaupt erst zur Entfaltung bringen.
In bevorzugten Ausführungsformen besteht der Bestandteil X der Verbindung I aus einem Isoflavon oder Isoflavon-Glycosid einer Substanz der allgemeinen Formel (II) oder aus einem pharmazeutisch akzeptablen Salz oder Solvat einer solchen Substanz gebildet:
wobei die Reste R1 , R2, R3, R4, R5 und R6 jeweils unabhängig voneinander bedeuten können: Wasserstoff, Hydroxy, Methoxy oder Glycosid (GIc), wobei einer der Reste 1 - 6 in der Verbindung I durch den Rest Pep (Bedeutung siehe oben) ersetzt ist. Bevorzugte Verknüpfungsstellen sind R2 und R5.
Das Glycosid in Formel Il hat die allgemeine Formel III
wobei R unabhängig von R1 , R2, R3, R4, R5 und R6 ausgewählt ist aus der Gruppe bestehend aus Wasserstoff, Acetyl und Malonyl.
Besonders bevorzugt sind solche erfindungsgemäßen Verbindungen, in denen der Bestandteil X aus Isoflavon, Daidzein, Genistein, Prunetin, Biochanin A, Orobol, Santal, Glycitein, Pratensein, Formononetin, Genistin, 6"-O-Malonylgenistin, 6"-O- Acetylgenistin, Daidzin, 6"-O-Malonyldaidzin, 6"-O-Acetyldaidzin, Glycitin, Ononin oder Sissotrin gebildet ist.
In weiteren bevorzugten Ausführungsformen enthält der Bestandteil X zwei bis drei OH- Gruppen, welche frei oder ganz oder teilweise mit Monosacchariden, inkl. teilweise mit Essigsäure, Malonsäure, Zimtsäure, Cumarsäure, Caffeesäure, Ferulasäure acylierten Monosacchariden oder Disacchariden, inkl. teilweise mit Essigsäure, Malonsäure, Zimtsäure, Cumarsäure, Caffeesäure, Ferulasäure acylierten Disacchariden, Methyl oder Sulfat substituiert sind.
In einer besonders bevorzugten Ausführungsform ist X aus Genistein, Genistin, Daidzein oder Daidzin, bevorzugt Genistein, gebildet.
Der Peptidrest der Verbindung X-Pep (I) ist bevorzugt aus einem Peptid oder Peptid- Derivat mit 2 bis 5 Aminosäuren gebildet, wobei mindestens 2 Aminosäuren Seitenketten umfassen, die bei pH=7 negativ geladen sind.
In bevorzugten Ausführungsformen ist der Bestandteil Pep aus einem Dipeptid gebildet, das aus der Gruppe der Peptide mit den Sequenzen DD, EE, DE, ED; ausgewählt ist, wobei D=Asparaginsäure und E=Glutaminsäure ist. Derart kurze Peptidbestandteile und deren pharmazeutisch akzeptable Salze oder Solvate haben sich überraschend als besonders stark synergistisch wirksam, insbesondere in Kombination mit Genistein,
Genistin, Daidzein und Daidzin, erwiesen. Von den genannten Dipeptiden ist Glutaminsäuredipeptid (Peptid der Sequenz EE) besonders bevorzugt. Mit
Glutaminsäuredipeptid in Verbindung mit Genistein konnte eine besonders markante
Steigerung der thrombozytenaggregationshemmenden Wirkung sowie bei der
Hemmung des Tumorwachstums, im Vergleich zu Genistein, in vitro und auch in vivo bei Menschen beobachtet werden.
In weiteren bevorzugten Ausführungsformen ist der Bestandteil Pep aus einem Peptid gebildet, das aus der Gruppe der Peptide mit den Sequenzen

ausgewählt ist, wobei D=Asparaginsäure und E=Glutaminsäure ist.
Prinzipiell sind alle möglichen Kombinationen von X und Pep, die kovalent verknüpft sind, als erfindungsgemäße Verbindung geeignet. Beispielsweise hat sich für verschiedene Einsatzbereiche eine Verbindung bewährt, bei der das Isoflavon Genistein mit dem Peptid oder Peptid-Dehvat der Sequenz EE kovalent verknüpft ist, wobei E=Glutaminsäure ist.
Im Prinzip kann die kovalente Verknüpfung zwischen X und Pep innerhalb der erfindungsgemäßen Verbindung auf beliebige Weise, vorzugsweise über eine Carbamat-, Ether- oder Esterbindung, ausgebildet sein. Besonders bewährt hat sich, wenn das Peptid über seine N-terminale Aminogruppe oder über seine C-terminale Carboxylgruppe oder die daraus durch Reduktion erhältliche primäre Hydroxylfunktion direkt mit einer Hydroxylgruppe von X, die an einer der Positionen R1-R6 befindlich ist, unter Ausbildung einer kovalenten Carbamat-, Ether-, oder Esterbindung verbunden ist, oder über ein geeignetes Linkersystem das beide Funktionalitäten verbindet. Geeignet als Linkersystem sind Ethylenglykol, Ethanolamin, höhere Homologe davon oder die entsprechenden Polyethylenglykolderivate.
In einer besonders vorteilhaften Ausführungsform ist dabei die kovalente Verknüpfung über ein Amid, einen Ether oder einen Carbonsäureester ausgebildet, vorzugsweise wenn die Verknüpfung über R2 oder R5 von X erfolgt ist.
Die vorliegende Erfindung umfasst ferner die Verwendung dieser Verbindungen oder pharmazeutisch akzeptabler Salze oder Solvate davon zur Herstellung pharmazeutischer Zusammensetzungen zur Behandlung und/oder zum Schutz vor medizinischen Indikationen des menschlichen Organismus oder des Organismus anderer Säugetiere.
Bevorzugte medizinische Indikationen umfassen dabei die Prophylaxe und Therapie der thrombembolischen Erkrankungen, Herz-Kreislauf-Erkrankungen, Gefässerkrankungen und Gefässanomalien, Erkrankungen im Zusammenhang mit einer erhöhten Thromozytenzahl und / oder Thrombozytenaggregation, Stoffwechselerkrankungen und Krebserkrankungen
Besonders bevorzugte medizinische Indikationen umfassen dabei die Prophylaxe und Therapie von: Hypertonie, Hypercholesterinämie, Herzinfarkt und Reinfarkt, Infarkte und Reinfarkte anderer Organe, Schlaganfall, Lungenembolie, Beinvenenthrombose, arteriosklerotische Gefäßerkrankungen, durch erhöhte Thrombozytenzahl und oder Aggregation verursachte Erkrankungen, Diabetes mellitus, Hyperhomocysteinämie, Malignome, und oder Osteoporose, sowie die prä und postoperative Thromboseprophylaxe,
Die vorliegende Erfindung betrifft ferner die Verwendung der genannten Verbindungen und Wirkstoffkombinationen zur Herstellung pharmazeutischer Zusammensetzungen zur Behandlung und/oder zum Schutz vor medizinischen Indikationen des menschlichen Organismus oder des Organismus anderer Säugetiere.
Weiterhin umfasst die vorliegende Erfindung die Verwendung wenigstens einer erfindungsgemäßen Verbindung oder pharmazeutisch akzeptabler Salze oder Solvate davon als Zusatz zu pharmazeutischen Zusammensetzungen, um die Verfügbarkeit von Wirkstoffen in diesen pharmazeutischen Zusammensetzungen in Säugetieren zu verbessern.
Die vorliegende Erfindung umfasst insbesondere die Verwendung der erfindungsgemäßen Verbindungen oder pharmazeutisch akzeptabler Salze oder Solvate davon als Nahrungsmittelzusatz, als Zusatzstoff zum Schutz von Zellen in Fermentern oder Bioreaktoren, als Tiernahrung und als Pflanzenschutzmittel.
Besonders bevorzugt ist insbesondere die Verwendung der erfindungsgemäßen Verbindungen oder pharmazeutisch akzeptablen Salze oder Solvate davon zur

Ausgangspunkt der Erfindung ist die überraschende Feststellung, dass die neuen Verbindungen der Formel (I) über ein zelluläres Peptidtransportsystem deutlich effektiver in die Zelle transportiert werden. Damit ist gewährleistet, dass auch bereits niedrigere Konzentrationen dieser Verbindungen physiologische Effekte auslösen
können, insbesondere bei der Vorbeugung, Behandlung und Linderung von humaner Thrombozytenaggregation und Tumoren und damit verbundenen Indikationen. Somit sind auch wesentlich niedrigere Mengen erforderlich als bei der Verwendung des entsprechenden reinen Isoflavons oder Isoflavon-Glycosids.
Insbesondere wirken die Verbindungsbestandteile X und Pep durch ihre kovalente Verknüpfung auf vorteilhafte Weise synergistisch zusammen. Dabei können zusätzlich Synergieeffekte mit weiteren Wirkstoffen eintreten.
Dabei wird unter einem Wirkstoff eine Substanz verstanden, die bei Verabreichung an ein Säugetier, insbesondere einem Menschen, eine pharmazeutisch gewünschte Veränderung eines physiologischen Zustandes des behandelten Säugetiers hervorrufen kann. Unter einem Wirkstoff wird insbesondere die pharmazeutisch wirksame Bestandteil eines Arzneimittels verstanden, insbesondere auch der Wirkstoffbestandteil X der erfindungsgemäßen Verbindung. Soweit ferner im Rahmen dieser Erfindung Substanzen in ihrer Singularform bezeichnet werden, so sind damit auch Mischungen mehrerer der entsprechenden Substanzen gemeint, soweit nichts anderes gesagt ist. Die Erfindung betrifft daher auch solche pharmazeutischen Zusammensetzungen, bei denen der Wirkstoff ein Gemisch zweier oder mehrerer Verbindungen der Formel (I) ist.. Ferner werden erfindungsgemäß unter einem Wirkstoff und einem Peptid bzw. daraus durch kovalente Verknüpfung hervorgegangene erfindungsgemäße Verbindungen, auch deren jeweils pharmazeutisch akzeptable Salze oder Solvate gezählt.
Im Rahmen der vorliegenden Erfindung umfasst der Begriff Peptide lineare oder verzweigte Peptide, die sowohl aus den 20 gencodierten Aminosäuren, als auch aus nichtnatürlich auftretenden alpha-, beta- und gamma-Aminosäuren bestehen können.
Unter einem Peptid-Derivat wird im Rahmen der vorliegenden Erfindung ein Peptid verstanden, das in der Haupt und/oder in der Seitenkette durch mindestens einen linearen, zyklischen oder verzweigten, Halogen- oder Hydroxy- oder Amin-substituierten oder nicht-substituierten, gesättigten oder aliphatischen Alkyl-, Alkylether-,
Alkylthioether-, Alkoxy-, Acyl- oder Aryl-Rest modifiziert ist, wobei der Rest zwischen 1 und 20 Kohlenstoffatome enthält.
Die pharmazeutische bzw. therapeutische Wirkung, die durch die Verbindung erreicht wird, ist insbesondere auch die eines Antioxidationsmittels, wenn der Wirkstoff bzw. Wirkstoffbestandteil X aus einem Isofiavon oder einem Isoflavon-Glycosid gebildet ist. Die erfindungsgemäße Verbindung ermöglicht es erstmals, eine bisher vermutete oder nur bei hohen Konzentrationen beschriebene antioxidative Wirkung eines Isoflavons bei pharmazeutisch akzeptablen Konzentrationen des Wirkstoffs in den Zielzellen eines behandelten Säugetiers, insbesondere eines Menschen, zu erreichen. Durch die erfindungsgemäße Verbindung wird damit erstmals ein Mittel bereitgestellt, mit dem die bisher nur vermuteten förderlichen Wirkungen von Isoflavonen und Isoflavon- Glycosiden reproduzierbar erzielt werden können. Isoflavone, Isoflavon-Glycoside und deren pharmazeutisch akzeptable Salze oder Solvate können somit erstmals in einer pharmazeutischen Zusammensetzung mit Peptiden mit genau bekannter, gleichmäßiger und reproduzierbarer Beschaffenheit eingesetzt werden.
Insbesondere ist es bevorzugt, wenn die Menge an Wirkstoff und/oder die Menge an Peptid jeweils allein nicht ausreichend ist zum Erzeugen der pharmazeutischen bzw. therapeutischen Wirkung.
Besonders bevorzugt ist eine erfindungsgemäße Verbindung einer der zuvor beschriebenen Arten zum Herabsetzen der Verfügbarkeit von Wasserstoffperoxid in einem Säugetier, insbesondere einem Menschen. Wasserstoffperoxid ist ein auslösender oder zumindest fördernder Faktor beim Entstehen zahlreicher
Erkrankungen von Säugetieren, insbesondere des Menschen. Mit den herkömmlichen
Isoflavon-Zusammensetzungen ist es insbesondere beim Menschen nicht gelungen, die Verfügbarkeit von Wasserstoffperoxid herabzusetzen und dessen krankheitsfördernde oder krankheitsauslösende Wirkung zu beseitigen. Insbesondere ist es bisher nicht reproduzierbar gelungen, durch Verabreichen der Isoflavone und Isoflavon-Glycoside
Genistein, Daidzein, Genistin und/oder Daidzin die Verfügbarkeit von
Wasserstoffperoxid in den Körperzellen eines Menschen oder anderen Säugetiers physiologisch wirksam herabzusetzen. Die erfindungsgemäße Verbindung schafft hier erstmals Abhilfe.
Ferner ist eine erfindungsgemäße Verbindung einer der zuvor beschriebenen Arten bevorzugt, die eingerichtet ist zum Hemmen der Thrombozytenaggregation, zur
Vorbeugung und/oder Behandlung von Hypertonie, Hypercholesterinämie, Hyperhomocysteinämie, Diabetes mellitus, Herzinfarkt, Schlaganfall, atherosklerotischer Gefäßerkrankung, Malignomen, und Osteoporose.
Vorzugsweise liegen in der erfindungsgemäßen pharmazeutischen Zusammensetzung erfindungsgemäße Verbindungen in einer solchen Menge vor, die ausreicht, um bei Verabreichung an ein Säugetier, insbesondere einem Menschen, eine Hemmung der Thrombozytenaggregation, herbeizuführen. Eine solche Wirkung ist insbesondere bei Verabreichen von Daidzein, Daidzin, Genistein und/oder Genistin, ggf. mit üblichen pharmazeutischen Hilfsstoffen und Trägern, nicht reproduzierbar unter Verwendung physiologisch akzeptabler Konzentrationen des genannten Wirkstoffs bzw. der genannten Wirkstoffe gelungen. Die erfindungsgemäße pharmazeutische Zusammensetzung ist daher vorteilhafterweise geeignet als Ersatz herkömmlicher Acetylsalicylsäure- und/oder Clopidogrel-enthaltender pharmazeutischer Zusammensetzungen. Die erfindungsgemäßen pharmazeutischen
Zusammensetzungen ermöglichen es, ohne die bekannten Nebenwirkungen herkömmlicher pharmazeutischer Zusammensetzungen mit Acetylsalicylsäure- und/oder Clopidogrel-basierten Wirkstoffen eine gewünschte therapeutische Wirkung zu erzielen, insbesondere die Thrombozytenaggregation im Blut eines behandelten Menschen oder anderen Säugetiers zu hemmen.
Mit den genannten Stoffen, insbesondere mit Genistein, lassen sich erfindungsgemäße Verbindungen herstellen, die besonders geeignet sind zum Hemmen der Thrombozytenaggregation in einem Säugetier wie beispielsweise dem Menschen. Mit derartigen erfindungsgemäßen Zusammensetzungen können insbesondere die im Zusammenhang mit der Verwendung herkömmlicher Acetylsalicylsäure- und/oder Clopidogrel-basierter Thrombozytenaggregationshemmer auftretenden, unerwünschten Nebenwirkungen sowie ein Therapieversagen vermieden oder zumindest verringert werden.
Die therapeutische Wirksamkeit kann insbesondere umfassen oder bestehen aus einer antioxidativen Wirkung, insbesondere dem Herabsetzen der Verfügbarkeit von Wasserstoffperoxid in einem Säugetier, und insbesondere vorzugsweise der Hemmung der Thrombozytenaggregation.
Die erfindungsgemäße pharmazeutische Zubereitung ist vorzugsweise eingerichtet zur oralen oder parenteralen Applikation. Dazu kann die erfindungsgemäße pharmazeutische Zusammensetzung insbesondere in Form einer Tablette, Dragee, Saft oder sonstiger Lösung vorliegen. Die erfindungsgemäße pharmazeutische
Zusammensetzung kann vorzugsweise als pharmazeutisch akzeptable Träger- und
Hilfsstoffe Wasser und Glucose enthalten. Weitere geeignete Hilfs- und Trägerstoffe entnimmt der Fachmann der Veröffentlichung von Fiedler, H. P., Lexikon der Hilfsstoffe für Pharmazie, Kosmetik und angrenzende Gebiete, 4. Auflage, Aulendorf: ECV-Editio- Kantor-Verlag, 1996.
Vorzugsweise wird die erfindungsgemäße Zusammensetzung formuliert als feste oder flüssige Arzneiform, insbesondere Pulver, Puder, Granulate, Tabletten, insbesondere Filmtabletten, Pastillen, Sachets, Cachets, Dragees, Kapseln, Salben, Cremes, Hydrogele, Pasten, Pflaster, Lösungen, Emulsionen, insbesondere vom Öl-in-Wasser- Typ, Suspensionen wie beispielsweise Lotionen, Injektions- und Infusionszubereitungen.
Je nach Zubereitungsart enthält die erfindungsgemäße pharmazeutische Zubereitung die Verbindung gegebenenfalls in besonders ausgewählten Mengen. Üblicherweise wird eine erfindungsgemäße Zubereitung auch Eisen in den bisher bekannten, ggf. zubereitungsabhängigen Mengen enthalten. Wenn die erfindungsgemäße pharmazeutische Zusammensetzung die erfindungsgemäße Verbindung als Wirkstoff enthält, so beträgt dessen Menge zumindest 1 mg pro Darreichungseinheit (beispielsweise pro Tablette) und vorzugsweise bis zu 500 mg pro Darreichungseinheit.
Besonders bevorzugte erfindungsgemäße Zusammensetzungen enthalten insgesamt
10 mg bis 500 mg der Verbindungen pro Darreichungseinheit, insbesondere pro
Tablette, wobei Mengen von 100 mg bis 500 mg für eine Darreichung in Tablettenform bevorzugt sind. Bei Darreichung in parenteral zu verwendender Lösung beträgt die Konzentration an der erfindungsgemäßen Verbindung zumindest 0,1 mg/ml und vorzugsweise bis zu 100 mg/ml, besonders bevorzugt 10 bis 50 mg/ml.
Das neue Derivat Glu-Glu-Genistein (I1 X = Genistein, Pep = GIu-GIu) kann bspw. mittels der beiden Glutaminsäurereste über ein zelluläres Peptidtransportsystem
deutlich effektiver in die Zelle transportiert werden. Damit ist gewährleistet, dass auch bereits niedrigere Konzentrationen von Glu-Glu-Genistein physiologische Effekte wie eine Induktion der Apoptose (physiologischer Zelltod) auslösen können.
Die Erfindung wird nachfolgend anhand der Ausführungsbeispiele und der Figuren näher erläutert, wobei der Gegenstand der Erfindung nicht auf die Ausführungsbeispiele und die Figuren beschränkt ist.
Beispiel 1) Anti-Tumor-Effizienz von Glu-Glu-Genistein Das neue Derivat Glu-Glu-Genistein kann mittels der beiden Glutaminsäurereste über ein zelluläres Peptidtransportsystem deutlich effektiver in die Zelle transportiert werden. Damit ist zu erwarten, dass auch bereits niedrigere Konzentrationen von Glu-Glu- Genistein physiologische Effekte wie eine Induktion der Apoptose (physiologischer Zelltod) auslösen können.
Ziele
• Die verbesserte Anti-Tumor-Aktivität des neuen Genisteinderivats Glu-Glu- Genistein werden im Vergleich zu Genistein in 8 ausgewählten Zelllinien humaner Tumoren demonstriert. • Die Effekte werden in den Vorversuchen zunächst auf den Ebenen der Apoptoseinduktion, der Zytotoxität sowie der Proliferationshemmung untersucht.
Vorgehen
• Zelllinien: Mammakarzinom: MCF-7, MDA 435
Kolonkarzinom: SW-620, SW-480
Malignes Melanom: A-375, SK-Mel-13
Kutanes Plattenepithelkarzinom: SCC-12, SCC-13
• Die Zellen werden 2-3 Wochen vorkultiviert, bis sich ein gleichmäßiges Wachstum (logarithmische Phase) eingestellt hat.
• Für die durchzuführenden Experimente werden die Zellen in 6-well-Schalen ausgesät (jeweils 200.000 Zellen pro well). Die Behandlung erfolgt nach 24 Stunden.
• In Vorexperimenten werden zunächst 5 verschiedene Konzentrationen von Glu-Glu- Genistein und von Genistein eingesetzt. Die Wirkung auf Proliferation und Zelltod
wird mikroskopisch beurteilt. Beobachtungszeiten: 4h, 8h, 24h, 48h, 72h, 6 Tage. Die Konfluenz sowie der Anteil an abgelösten Zellen werden in Wachstumsprotokollen festgehalten. Aufgrund dieser Protokolle werden zwei geeignete Konzentrationen ausgewählt. • Ggf. muss ein zweites Pilotexperiment mit veränderten Bedingungen durchgeführt werden.
• Apoptosebestimmung: 24h vor der Aussaat bekommen die Zellen frisches Wachstumsmedium. Die Zellen werden zu den ermittelten Bedingungen in 6-well- Schalen ausgesät (i.d.R.: 200.000 Zellen pro well). Nach 24 h wird die Behandlung durchgeführt. Eingesetzte Konzentrationen und Behandlungszeiten richten sich nach den Ergebnissen der Vorexperimente. .Das Wachstum sowie die Effekte werden mindestens 1x täglich mikroskopisch kontrolliert und protokolliert. Nach Ablauf der Inkubationszeit werden die 6-well-Schalen zentrifugiert und das Zellpellet lysiert. Unter Verwendung von anti-Histon sowie anti-DNA-Antikörpern wird ein Sandwich- ELISA durchgeführt, der eine Aussage über das Maß der DNA-Fragmentierung erlaubt (Cell Death Detection ELISA, Fa. Roche). Die Apoptose wird in relativen Werten im Verhältnis zu unbehandelten Kontrollen dargestellt.
• Zytotoxizitätsbestimmung: 24h vor der Aussaat bekommen die Zellen frisches Wachstumsmedium. Die Zellen werden zu den ermittelten Bedingungen in 6-well- Schalen ausgesät (i.d.R.': 200.000 Zellen pro well). Nach 24 h wird die Behandlung durchgeführt. Eingesetzte Konzentrationen und Behandlungszeiten richten sich nach den Ergebnissen der Vorexperimente. Das Wachstum sowie die Effekte werden mindestens 1x täglich mikroskopisch kontrolliert und protokolliert. Nach Ablauf der Inkubationszeit wird der Zellkulturüberstand geerntet. In diesem wird mit einer enzymatischen Nachweismethode die Aktivität der Laktat-Dehydrogenase bestimmt (LDH-release-assay, Fa. Roche). Die Zytotoxizität wird in relativen Werten im Verhältnis zu unbehandelten Kontrollen dargestellt.
• Proliferationsassay: 24h vor der Aussaat bekommen die Zellen frisches Wachstumsmedium. Die Zellen werden in geringer Dichte in 6-well-Schalen ausgesät (i.d.R.: 50.000 Zellen pro well). Nach 24 h wird die Behandlung durchgeführt. Die Bestimmungen zur Zeilproliferation werden zu den Zeitpunkten 0, 1 , 2, 3, 5, 7 Tagen nach Behandlung durchgeführt. Zur Bestimmung der Zellzahl werden die anhaftenden Zellen fixiert und mit Kristallviolett gefärbt. Die Intensität der Färbung
wird photometrisch bestimmt. Die Zeilproliferation wird in Wachstumskurven dargestellt.
• Für alle drei Experimentserien (Apoptose, Zytotoxizität und Zeilproliferation) werden folgende Ansätze durchgeführt: a) unbehandelte Kontrollen, b) Genistein, Konzentration 1 , c) Genistein, Konzentration 2, d) GIu-GIu, Konzentration 2, e) GIu- Glu-Genistein, Konzentration 1 , f) Glu-Glu-Genistein, Konzentration 2. Alle Experimente werden mit jeweils Dreifachwerten durchgeführt. Die gesamte Serie an Experimenten wird 1x wiederholt.
• Die Ergebnisse werden mit Hilfe der Mathematik unscharfer Systeme (Leibnizsystem) ausgewertet und ein Abschlussbericht erstellt.
Durch den Einsatz repräsentativer Karzinomzelllinien und die Prüfung der für eine Antitumortherapie wesentlichen zellulären Funktionskaskaden ergeben sich stringente Hinweise zur Funktion von Glu-Glu-Genistein im Bereich der Tumortherapie.
Beispiel 2)
Durch eine erfindungsgemäße Verbindung lässt sich auf Grund verbesserter intrazellulärer Verfügbarkeit eine dem Genistein überlegene Hemmung der Thrombozytenaggregation erreichen. Dies geschieht über eine Koppelung an das Dipeptid GIu-GIu, so dass Gen-Glu-Glu i.S. eines Peptidomimetikums über spezifische Oligopeptidtransporter der Thrombozytenmembran vermehrt nach intrazellulär transportiert wird.
Versuche
• Plättchenreiches Plasma (PRP) wird mit in DMSO gelöstem Genistein in unterschiedlichen Konzentrationen versetzt: 0, 25, 50, 100, 200 μM. Anschließend Messung der Collageninduzierten Aggregation mittels der
Aggregometrie unter Zusatz von Collagen 1 ug/ml. Auswertung und Identifizierung einer beginnend effektiven Genisteinkonzentration (ca 50 μM)
• Testung von Genistein vs Gen-Glu-Glu wie oben beschrieben im Bereich der zuvor bestimmten beginnend effektiven Konzentration, beispielsweise 0, 25, 50, 75 μM. Es zeigt sich eine Überlegenheit von Gen-Glu-Glu in Bezug auf die
Hemmung der collageninduzierten Aggregation.
• Wiederholung des zweiten Versuchs, aber nach vorheriger Zugabe des indifferenten Dipeptides AIa-AIa in 100facher Konzentration, also 0, 2.5, 5, 7.5 mM. Es zeigt sich nunmehr eine Antagonisierung des spezifischen Gen-Glu-Glu Effektes, da die Peptidtransporter durch den Überschuss von AIa-AIa blockiert werden.

Herstellung von 7, 4'-Di-(tert.-Butyloxycarbonyloxy)-hydroxy-3-phenyl-4H- chromen-4-one
Zu einer Lösung von 8.1 g (29.7 mmol) Genistein 1 in DCM werden 4.9 ml_ (2 eq.) Pyridin und 13.05 g (60.7 mmol, 2.1 eq.) Di-tert.-butyl-dicarbonat gegeben und die Mischung 30 min bei Raumtemperatur gerührt. Das Lösungsmittel wird abgezogen, der ölige Rückstand wird zweimal mit Toluol koevaporiert. Der anfallende Feststoff wird am Hochvakuum getrocknet.
Ausbeute: 13 g 2 in Form eines weißen amorphen Feststoffs, der üblicherweise zu etwa 30% aus dem dreifach umgesetztem Genistein besteht.

Herstellung von 5-(2-aminoethoxy)-7-hydroxy-3-(4-hydroxyphenyl)-4H-chromen-4- one
Eine Lösung von 2 (8 g, 17.01 mmol), N-Boc-Ethanolamin (4.11 g 25.5 mmol, 1.5 eq.) und Triphenylphosphin (6.69 g, 25.5 mmol, 1.5 eq.) in DCM wird auf -15°C gekühlt, dann wird, verteilt über 5 min, eine Lösung von Diisopropylazodicarboxylat (5.159 g, 25.5 mmol, 1.5 eq.) in DCM zugetropft. Die Mischung wird auf RT gebracht und über
Nacht gerührt. Die Reaktionsmischung wird mit DCM verdünnt, zweimal mit Wasser gewaschen und über Natriumsulfat getrocknet. Das Lösungsmittel wird abgezogen, der Rückstand wird dreimal mit Toluol koevaporiert. Der erhaltene Rückstand wird mit möglichst wenig DCM gelöst und durch Zugabe von Diethylether gefällt. Ausbeute: 10.9 g eines weißen Feststoffes bestehend aus 3 und Tris-BocGenistein. Das Zwischenprodukt 3 (5.4 g) wird in 10 mL Methanol gelöst und bei RT mit 30 ml_ gesättigter methanolischer HCl (1.25 mol/l) behandelt. Nach kurzer Zeit beginnt die Ausfällung eines gelben Feststoffs, der nach 12h abgesaugt wird. Trocknen in Vakuum liefert reines 4 als HCI-SaIz, das ohne weitere Reinigung umgesetzt wird. Ausbeute: 1.4 g 4 als leuchtend gelbes feines Pulver.
Beispiel 6
Herstellung von 5-{2-GluGlu-2-aminoethoxy)-7-hydroxy-3-(4-hydroxyphenyl)-4H- chromen-4-one
Zu einer Lösung von 4 (1.22 g, 3.49 mmol) in DMF werden Boc-Glu(OtBu)Glu(OtBu)OH
(1.1 g, 2.59 mmol, 0.75 eq.) und PPA (50% in DMF, 2.14 mL) gegeben und die
Mischung durch Zugabe von N-Ethyldiisopropylamin auf pH 8-9 eingestellt. Es wird 12h bei RT gerührt.
Die Reaktionsmischung wird mit Ethylacetat verdünnt und dreimal mit Wasser gewaschen, danach wird die organische Phase über Natriumsulfat getrocknet und im
Vakuum eingeengt. Der Rückstand wird chromatographisch gereinigt
(Hexan/Ethylacetat 7:3 → 9:1→ reines Ethylacetat).
Ausbeute: 1.04 g (34%) 5 als weißer Schaum.
Das Zwischenprodukt 5 (1.04 g) wird mit 5 mL Ethylacetat aufgeschlämmt und mit 20 mL HCl in Ether (2M) versetzt, wobei sich die Mischung gelb färbt und klar wird. Nach
6h wird das ausgefallene Produkt abgesaugt, mit Ethylacetat und Diethylether gewaschen und anschließend im Hochvakuum getrocknet. Ausbeute: 820 mg des an chromatographisch reinem 6 als gelbes Pulver. 415 mg des Rohprodukts werden in Wasser gelöst und gefriergetrocknet. Ausbeute: 291 mg 6 in Form eines gelben Schaums.
Beispiel 7 Biologische Testung
Versuchsbeschreibung
Substanzen
Untersucht wurden jeweils folgende fünf Genistein-Glu-Glu Konjugate:
• Genistein-Glu-Glu Carbamat, die Koppelung erfolgt C-terminal über die OH- Gruppe in C7 Position des Genisteins, im folgenden C1
• Genistein-Glu-Glu gelinkerter Ether, Koppelung N-terminal, Methylgruppe als Link, Bindung über die OH-Gruppe in C5 Position des Genisteins, im folgenden L2
• Genistein-Glu-Glu Ether, Koppelung C-terminal über die OH-Gruppe in C5 Position des Genisteϊns, im folgenden A2
• Genistein-Glu gelinkerter Ether, Koppelung N-terminal, Bindung über die OH- Gruppe in Position C5, im folgenden L1
• Genistein-Glu Ether, Koppelung C-terminal, Bindung über die OH-Gruppe in C5 Position des Genisteins, im folgenden A1 • Natives Genistein
• Gemisch natives Genistein/ GIu-GIu
Materialien
Die verwendeten Substanzen Genistein, GIu-GIu1 DMSO, AIa-AIa stammen von der Fma Sigma-Aldrich, München. Zur Messung wurde der Platelet Aggregation Profiler Model PAP-8E der Firma Bio/Data Corporation, ein 8-Kanal Aggregometer mit rechnergestützter digitaler Kurvenerstellung, verwendet.
Präanalytik
Nach Blutentnahme aus großkalibriger Vene in ungepuffertes Citrat 10% wird das so gewonnene Vollblut bei ca 1500U/min 10 Minuten zentrifugiert. Das so gewonnene Plättchenreiche Plasma (PRP) wird 45 Minuten bei Raumtemperatur Ruhen gelassen. Gleichzeitig wird aus der gleichen Probe durch gewohntes Zentrifugieren Plättchenarmes Plasma zur Erstellung der jeweiligen Leerwerte gewonnen. Zur Messung werden Küvetten mit 0.25ml Füllvolumen verwandt.
Von den zu prüfenden Substanzen, einschließlich Genistein sowie den zur Verwendung kommenden Peptiden AIa-AIa und GIu-GIu werden im Verhältnis 1 mg auf 1 ml Stammlösungen erstellt, Genistein sowie die untersuchten Koppelungsprodukte in DMSO, GIu-GIu und AIa-AIa in H2O.
Vorversuch In mehreren Vorversuchen wird zum einen ein normales Aggregationsverhalten der nativen Probe nach Zugabe von 1 μg/ml Collagen (87% Plättchenaggregation, s.u.) sowie ein indifferentes Verhalten von DMSO im Bereich der verwendeten Konzentrationen gezeigt. In der Citratblutprobe wurden 230.000 μl Thrombozyten gemessen.
Messungen
Zunächst wurde die aggregationshemmende Wirkung von nativem Genistein ermittelt: Bei der Referenz (kein Zusatz) ergab sich eine Aggregation von 90%, entsprechend auch dem Vorversuch. Nach Zugabe von Genistein 50μM ergab sich eine Aggregation von 82% der Plättchen, unter Zugabe von Genistein 100μM eine Aggregation von 50% und bei einer Konzentration von 200μM eine nahezu vollständige Hemmung der Thrombozytenaggregation.
Die Ergebnisse decken sich im wesentlichen sowohl mit eigenen älteren Untersuchungen als auch mit den in der Literatur beschriebenen Effekten. Zur Problematik der Vergleichbarkeit wurde dabei schon Stellung bezogen.
Entsprechend der eigenen Vorgabe betrachteten wir nun wie auch erwartet eine Konzentration von 50μM als eine beginnend effektive Wirkkonzentration. Entsprechend
untersuchten wir die fünf Genisteinderivate in Bezug auf die collagen-induzierte Aggregation (wiederum 1 μg/ml) bei einer Konzentration von 50μM. Es ergaben sich folgende Ergebnisse:
Auffallend ist - neben einer erheblich ausgeprägteren aggregationshemmenden Wirkung aller getesteter Substanzen gegenüber dem nativen Genistein (FA 82%) - eine deutliche Desaggreagation im zeitlichen Verlauf, zu erkennen an der Rückläufigkeit der Kurven bzw Differenz zwischen MA und FA. Darüber hinaus ist die Geschwindigkeit der Aggregation, der so genannte „Slope" insbesondere bei L2 aber auch bei C1 gegenüber dem nativen Genistein deutlich vermindert.
Zum Vergleich erfolgte eine Aggregationsmessung nach Zugabe von Genistein 50μM und GIu-GIu 150μM im Sinne eines Gemisches. Die Messung erfolgte direkt, also ohne relevante Vorinkubation, sofern dies nicht schon der Meßmethode immanent ist. Auch hier zeigte sich eine dem Genistein überlegene Wirkung von MA 56% bzw FA 44% (Genistein MA 85%, FA 82%).
Um die Hypothese zu untermauern, dass es sich bei den beobachteten Phänomenen um einen verbesserten Membrantransport über spezifische Peptridtransporter auf der Thrombozytenmembran handelt, erfolgte eine erneute Messung der Aggregation unter
dem Einfluß von L2 nach vorheriger Zugabe des indifferenten, aber als Substrat der Peptidtransporter fungierenden Dipeptids AIa-AIa im lOfachen molaren Überschuß, also 1_2 50μM/Ala-Ala 500μM: Die zuvor beobachtete fast vollständige Aggregationshemmung konnte so teilweise antagonisiert werden.
Weitere Versuche
Im weiteren haben wir die Substanz L2 auf ihre aggregationshemmenden Eigenschaften hin untersucht. Zunächst wird die Aggregation nach Zugabe von L2 in einer Konzentration von 50 μM nach Zugabe von Collagen in ansteigenden Konzentrationen untersucht (beobachteter Effekt reversibel?):

Claims
1. Verbindung gemäß Formel (I),
(I) X-Pep
wobei
- X ein Isoflavon oder Isoflavon-Glycosid ist, und
- Pep eine Aminosäure oder ein Peptid oder Peptid-Derivat mit 2 bis 5 Aminosäuren ist, und zwischen X und Pep eine direkte kovalente Verknüpfung besteht, oder die Verknüpfung durch ein geignetes Linkersystem (wie Ethylenglykol, Ethanolamin, höhere Homologe davon oder die entsprechenden Polyethylenglykolderivate) hergestellt wird,
sowie pharmazeutisch akzeptable Salze oder Solvate dieser Verbindungen.
2. Verbindung nach Anspruch 1 , wobei das Isoflavon oder Isoflavon-Glycosid 2 - 4 OH-Gruppen enthält, welche teilweise mit Monosacchariden, inkl. teilweise mit Essigsäure, Malonsäure, Zimtsäure, Cumarsäure, Caffeesäure, Ferulasäure acylierten Monosacchariden oder Disacchariden, inkl. teilweise mit Essigsäure, Malonsäure, Zimtsäure, Cumarsäure, Caffeesäure, Ferulasäure acylierten Disacchariden, Methyl oder Sulfat substituiert sind.
3. Verbindung nach Anspruch 1 und 2, wobei X aus einem Isoflavon oder Isoflavon- Glycosid einer Substanz der allgemeinen Formel (II) oder aus einem pharmazeutisch akzeptablen Salz oder Solvat einer solchen Substanz gebildet wird,
wobei die Reste R1 , R2, R3, R4, R5 und R6 jeweils unabhängig voneinander bedeuten können: Wasserstoff, Hydroxy, Methoxy oder Glycosid (GIc) und einer der Reste 1 - 6 in der Verbindung I durch den Rest Pep (Bedeutung siehe oben) bevorzugt an den Verknüpfungsstellen R2 und R5 ersetzt ist.
4. Verbindung nach Anspruch 3, wobei das Glycosid GIc die allgemeine Formel III
aufweist, wobei R unabhängig von R1 , R2, R3, R4, R5 und R6 aus der Gruppe bestehend aus Wasserstoff, Acetyl und Malonyl ausgewählt ist.
5. Verbindungen nach Anspruch 1 bis 4, wobei das Isoflavon oder Isoflavon-Glycosid ausgewählt ist aus der Gruppe bestehend aus Isoflavon, Daidzein, Genistein,
Prunetin, Biochanin A, Orobol, Santal, Glycitein, Pratensein, Formononetin,
Genistin, 6"-O-Malonylgenistin, 6"-O-Acetylgenistin, Daidzin, 6"-O-Malonyldaidzin,
6"-O-Acetyldaidzin, Glycitin, Ononin und Sissotrin.
6. Verbindung nach Anspruch 1 , wobei das Isoflavon Genistein ist.
7. Verbindung nach Anspruch 1 , wobei X eine der folgenden Formeln IV - VII bedeutet
8. Verbindungen nach Anspruch 1 bis 7, wobei das Peptid oder Peptid-Derivat 2 bis 5 Aminosäuren umfasst, und wobei mindestens 2 Aminosäuren Seitenketten umfassen, die bei pH=7 negativ geladen sind.
9. Verbindungen nach Anspruch 1 bis 7, wobei das Peptid oder Peptid-Derivat ein
Dipeptid ist, ausgewählt aus der Gruppe der Peptide mit den Sequenzen DD, EE, DE, ED; wobei D=Asparaginsäure und E=Glutaminsäure ist.
10. Verbindungen nach Anspruch 1 bis 7, wobei das Peptid oder Peptid-Derivat ausgewählt ist aus der Gruppe der Peptide mit den Sequenzen
DDDDD, DDDDE, DDDED, DDDEE, DDEDD, DDEDE, DDEED, DDEEE, DEDDD, DEDDE, DEDED, DEDEE, DEEDD, DEEDE, DEEED, DEEEE, EDDDD, EDDDE, EDDED, EDDEE, EDEDD, EDEDE, EDEED, EDEEE, EEDDD, EEDDE, EEDED, EEDEE, EEEDD, EEEDE, EEEED, EEEEE,

11.Verbindungen nach Anspruch 1 , wobei das Isoflavon Genistein ist und das Peptid oder Peptid-Derivat die Sequenz EE hat; wobei E=Glutaminsäure ist.
12. Verbindung nach Anspruch 1 , dadurch gekennzeichnet, dass die kovalente Verknüpfung als Carbamat-, Ether- oder Esterbindung ausgebildet ist.
13. Pharmazeutische Zusammensetzungen umfassend Verbindungen gemäß
Anspruch 1-12.
14. Verwendung einer Verbindung gemäß Anspruch 1-13. zur Herstellung einer pharmazeutischen Zusammensetzung zur Behandlung und/oder zum Schutz vor einer medizinischen Indikation ausgewählt aus der folgenden Gruppe:
Herz-Kreislauf-Erkrankungen, Erkrankungen im Zusammenhang mit einer erhöhten Thrombozytenaggregation, Stoffwechsel-Erkrankungen,
Knochenerkrankungen oder Krebserkrankungen.
15. Verwendung gemäß Anspruch 14, wobei die medizinische Indikation ausgewählt ist aus der folgenden Gruppe:
Leukämie, Lymphom, Melanom, Mammakarzinom, Lungenkarzinom, Prostatakarzinom, Kopf-Hals-Plattenepithelkarzinom, Kolonkarzinom.
16. Verwendung nach Anspruch 14, wobei die medizinische Indikation ausgewählt ist aus der folgenden Gruppe:
Hypertonie, Hypercholesterinämie, Herzinfarkt, arteriosklerotische Gefäßerkrankungen, Schlaganfall, durch erhöhte Thrombozytenaggregation verursachte Krankheiten, Diabetes mellitus, Hyperhomocysteinämie, Malignome,
Osteoporose.
17. Verwendung einer Verbindung nach Anspruch 1 bis 12 als Nahrungsmittelzusatz.
18. Verwendung einer Verbindung nach Anspruch 1 bis 12 als Zusatzstoff zum
Schutz von Zellen in Fermentern oder Bioreaktoren.
19. Verwendung einer Verbindung nach Anspruch 1 bis 12 als Tiernahrung.
20. Verwendung einer Verbindung nach Anspruch 1 bis 12 als Pflanzenschutzmittel.
21. Herstellung von Verbindungen der Formel I, umfassend die Schritte: a) Bereitstellen eines Wirkstoffs, ausgewählt aus der Gruppe bestehend aus Isoflavon, Daidzein, Genistein, Prunetin, Biochanin A, Orobol, Santal, Glycitein, Pratensein, Formononetin, Genistin, 6"-O-Malonylgenistin, 6"-O- Acetylgenistin, Daidzin, 6"-O-Malonyldaidzin, 6"-O-Acetyldaidzin, Glycitin, Ononin und Sissotrin, und/oder eines pharmazeutisch akzeptablen Solvats oder Salzes des Wirkstoffs, wobei der Wirkstoff ggf. eine geschützte Seitengruppe aufweist, b) Bereitstellen einer Aminosäure oder eines Peptids oder Peptid-Derivats mit 2 bis 5 Aminosäuren Länge oder Bereitstellen einer Aminosäure und/oder eines Solvats oder Salzes des Peptids oder der Aminosäure, wobei zumindest eine der Aminosäuren eine Seitengruppe besitzt, die geschützt ist, und der N-Terminus des Peptids oder der Aminosäure geschützt ist und/oder der C- Terminus des Peptids oder der Aminosäure an einer Festphase gebunden ist, c) Mischen des Stoffs aus a) mit dem Stoff aus b) und einem Aktivierungsreagens, vorzugsweise zur Aktivierung von Carboxyl- oder
Hydroxylgruppen.
