Abstract
Es wird eine neue pflanzliche Proteinfraktion mit Phospholipase D-Aktivitat beansprucht, die aus Vertretern der Familie Papaveraceae and insbesondere Papaver somniferum stammt und die sich aus zwei Proteinunterfraktionen A und B zusammensetzt, deren Molekularmasse insbesondere bei 116,4 kDa bzw. 114,1 kDa liegt, die einen isoelektrischen Punkt pl von 8,7 bzw. 6,7 besitzen and die ein hydrolytisches Aktivitatsoptimum bei PH 8,0 bzw. 5,5 aufweisen. Die beiden Unterfraktionen werden vor allem durch Zn2+-lonen aktiviert and stellen zwei Isoenzyme tar. Eingesetzt wird these Proteinfraktion vor allem zur Hydrolyse und/oder Transphosphatidylierung von Phospholipiden und deren Lyso-Formen.
Description
Pflanzliche Proteinfraktion mit Phospholipase D-Aktivität
Beschreibung
Gegenstand der vorliegenden Erfindung ist eine pflanzliche Proteinfraktion mit Phospholipase D-Aktivität.
Phospholipase D (PLD), bei der es sich um eine Phosphatidylcholin- Cholinhydrolase (EC 3.1.4.4) handelt, ist ein wichtiges Enzym des Phospholipidmetabolismus und ist in der Natur weit verbreitet.
Phospholipase D wird einer Enzymklasse zugerechnet, deren Vertreter in heterogenen Systemen wasserunlösliche Substrate umsetzen können, da sie an der Grenzfläche zwischen einem Lipid und Wasser Reaktionen katalysieren können. Dieses amphiphile Verhalten der Phospholipase D hat sie für die Wissenschaft besonders interessant erscheinen lassen, so dass in den vergangenen Jahrzehnten zahlreiche unterschiedliche Phospholipasen D (-Spezies) aus den unterschiedlichsten Quellen isoliert werden konnten. Insbesondere wird PLD eine Beteiligung an zellregulatorischen Aktivitäten im Zusammenhang mit dem interzellulären Signalaustausch zugeschrieben. Über ihre Hydrolyse-Aktivität hinaus können PLD-Enzyme auch Phosphatidyl-Reste auf Alkohole übertragen. Allgemein werden PLD- Varianten deshalb für den biokatalytischen Austausch von Kopfgruppen der Phospholipide eingesetzt.
So sind z.B. Enzymfraktionen aus Piastiden von Zuckerrübe, Spinat oder Kohlblättern bekannt, sowie aus Chromoplasten von Karotten. Entsprechende Fraktionen mit Phospholipase D-Aktivität wurden ebenfalls aus Mitochondrien und Mikrosomen unreifer Erdnusssamen gewonnen, aber auch aus Ricinus-Samen, Arabidopsis-Spezies und Tomate.
Auch aus entfettetem Baumwollsamenmehl ist es gelungen, ein entsprechendes Enzym zu extrahieren.
Neben pflanzlichen Quellen dienen auch Mikroorganismen als Quelle, wobei insbesondere Corynebakterien (Corynebacterium ovis), Escherichia coli, Bäckerhefezellen und Streptomyceten (Streptomyces hachijoensis) zu nennen sind.
Phospholipase konnte aber auch aus Säugetierzellen isoliert werden, wie z. B. aus menschlichen Eosinophilen und Rattenhirnmikrosomen.
Hinsichtlich des Molekulargewichts bietet sich bezüglich der bekannten Phospholipasen D ein heterogenes Bild:
So weist das aus Baumwollsamen isolierte lösliche Enzym ein Molekulargewicht von 71 000 3 000 Da auf; Phospholipase D aus Erdnusssamen besitzt ein Molekulargewicht von 200 000 10 000 Da und PLD aus menschlichen Eosinophilen ein Molekulargewicht von ca. 60 000 Da.
Entsprechende bakterielle Enzyme, wie sie bspw. aus Corynebacterium ovis isoliert werden können, besitzen ein Molekulargewicht von annähernd 90 000 Da.
Hinsichtlich des isoelektrischen Punktes sind für Phospholipase D aus Erdnusssamen pl-Werte von 4,65 bekannt, wo hingegen der pl eines Rohextraktes aus menschlichen Eosinophilen zwischen 4,8 und 5,0 liegt und durch zusätzliche Aufreinigung einen Wert zwischen 5,8 und 6,2 annehmen kann.
Die Aufreinigung von PLD aus Weißkohl in zwei Schritten beschreibt R. Lambrecht et al („A facile purification procedure of phospholipase D from
cabbage and its characterization"; (1992) Biol. Chem. Hoppe Seyler Vol 373 (2) 81-88). Diese Methode umfasst eine Ammoniumsulfat-Präzipitation und eine darauffolgende Ca+-vermittelte Affinitätschromatographie.
Der Veröffentlichung von I. Schaffner et al („Genomic structure, cloning and expression of two phopholipase D isoenzymes from white cabbage."; Eur. J. Lipid Sei. Technol. 104, 79-87 (2002); entsprechend Dissertation (2001)) kann entnommen werden, wie rekombinante Phospholipase D-aktive Isoenzyme aus Weißkohl über Klonierungsverfahren erhalten werden können und wie man diese Isoenzyme hinsichtlich ihrer spezifischen Hydrolyseaktivität in Abhängigkeit vom pH-Wert und von der Ca2+- Konzentration sowie hinsichtlich ihrer Transposphatidylierungseigenschaften charakterisieren kann.
Einen allgemeinen Überblick zum Kenntnisstand bezüglich Phospholipase D gibt der Übersichts-Beitrag von Michael Heller in Advanced Lipid Research, 1978, Band 16, Seiten 267 bis 326.
A. Lerchner et al beschreiben in „Identification of two isoenzymes of phospholipase D from opium poppy" (Direct Submission (2001) NCBI GenBank, accessions nos. AAL48261 - AAK48264 und Multipler Sequenzvergleich) zwei trunkierte Phospholipase D1 Polypeptide sowie zwei weitere trunkierte Phospholipase D2 Polypeptide aus Papaver somniferum. Wie aus dem multiplen Sequenzvergleich entnommen werden kann, haben die Teilaminosäure-Sequenzen der Proteine D1 und D2 eine
Sequenzidentität von 98% zueinander. Zudem besitzen die beschriebenen Teilsequenzen eine hohe Homologie (70-84%) zu den gut charakterisierten Phospholipase D-Varietäten des -Typs. Da die Sequenz-Ermittlung am 5'-Ende nicht vollständig ist, ist es allerdings nicht möglich, die Phospholipase D1 - und D2-Polypeptide einem definierten Enzym zuzuordnen.
Der Datenbankeintrag von A. Lerchner et al „Identification of two isoenzymes of phospholipase D from opium poppy" (Direct Submission (2001 ) NCBI GenBank, accession nos. AF451979 - AF451982) beschreibt Nukleinsäuresequenzen zu den eben bezeichneten beiden Polypeptiden PLD1 und PLD2.
Von Mohn-Samen ist bekannt, dass sie in der Lage sind, in außergewöhnlich hohem Ausmaß sekundäre Metabolite zu bilden. So können bspw. schon nach wenigen Tagen des Anquellens Alkaloide wie Thebain im Mohn-Samen nachgewiesen werden, was sie vor allem auch im Hinblick auf die Opium- Gewinnung interessant macht.
Da Phospholipase D (PLD) eine immer wichtigere Rolle bei der industriell genutzten katalytischen Hydrolyse von Glycerophospholipiden wie z.B. Phosphatidylcholin (PC) zu Phosphatidsäure (PA) spielt, aber auch bei Transphosphatidylierungs-Vorgängen zum Kopfgruppenaustausch der Phospholipide, hat sich für die vorliegende Erfindung die Aufgabe gestellt, neue pflanzliche und aus Vertretern der Familie Papaveraceae stammende • Proteinfraktionen mit Phospholipase D-Aktivität zu isolieren.
Gelöst wurde diese Aufgabe durch eine entsprechende Proteinfraktion, die dadurch gekennzeichnet ist, dass a) sie aus zwei Protein-Unterfraktionen A und B besteht und b) sie durch Zn2+-lonen aktivierbar ist sowie c) die Unterfraktionen A und/oder B Kohlenhydratanteile aufweisen, und wobei die Protein-Unterfraktion A ausschließlich eine Hydrolyse-Aktivität besitzt.
Unter den nachfolgend verwendeten Begriff "Proteinfraktion" fallen definitionsgemäß alle tatsächlichen Proteinfraktionen und Proteine sowie deren mögliche Varianten und auch sämtliche Enzyme und Enzym- Varianten, alle mit entsprechender PLD-Aktivität.
Überraschend konnte bei dieser pflanzlichen Proteinfraktion festgestellt werden, dass sie zwei Isoenzymeinheiten enthält, die beide ein relativ enges Molekularmassenspektrum aufweisen und deren Aktivitätsoptima zum einen im stark Sauren, zum anderen aber im leicht Basischen liegen. Außerdem war auf Grund der bislang bekannten PLDs nicht zu erwarten, dass Isoenzyme mit PLD-Aktivität aus Mohn Zink-aktivierbar sind, was insbesondere im Hinblick auf deren Verwendung bei der Herstellung von Phospholipiden von Vorteil ist, die bekanntlich mit Ca-Ionen zumeist unlösliche Komplexe bilden. Auf Grund der bislang vorliegenden Erkenntnisse mit Phospholipasen D aus pflanzlichen Quellen konnte außerdem nicht davon ausgegangen werden, dass in Vertretern der Familie der Papaveraceaen Proteinfraktionen mit entsprechenden Aktivitäten aufgefunden werden.
Die vorliegende Erfindung beansprucht insbesondere eine Proteinfraktion, die aus Papaver somniferum (Schlafmohn) stammt und ganz besonders bevorzugt aus sich entwickelnden Keimlingen und/oder aus Endospermen. Vorstellbar ist natürlich auch eine Variante, bei der die Proteinfraktionen indirekt aus Papaver stammen, indem diese nämlich mit Hilfe rekombinanter Methoden erhalten werden und vor allem mit Hilfe rekombinanter Mikroorganismen, die die Gene für die entsprechende Proteinfraktion enthalten.
Wie bereits ausgeführt, richtet sich ein erfindungswesentlicher Aspekt auf die Tatsache, dass die beanspruchte pflanzliche Proteinfraktion zwei Isoenzyme enthält. Diesbezüglich bevorzugt die vorliegende Erfindung eine Proteinfraktion, deren Unterfraktion A eine Molekularmasse zwischen 116 und 118 kDa, einen isoelektrischen Punkt pl zwischen 8,5 und 8,9 und ein hydrolytisches Aktivitätsoptimum bei pH-Werten zwischen 7,8 und 8,2 besitzt und die Unterfraktion B eine Molekularmasse zwischen 112 und 115 kDa, einen isoelektrischen Punkt pl zwischen 6,5 und 6,9 und ein hydrolytisches
Aktivitätsoptimum bei pH-Werten zwischen 5,0 und 6,0.
Hinsichtlich des isoelektrischen Punktes kann durch weitere Aufreinigung isolierter Fraktionen ein definierter Wert erreicht werden.
Die Proteinfraktion der vorliegenden Erfindung ist aus diesem Grund auch insbesondere dadurch gekennzeichnet, dass die Unterfraktion A einen isoelektrischen Punkt pl von 8,7 und eine Molekularmasse von 116,4 kDa sowie ein hydrolytisches Aktivitätsoptimum bei pH 8,0 aufweist. Die entsprechenden bevorzugten Werte der Unterfraktion B betragen hinsichtlich der Molekularmasse 114,1 kDa, hinsichtlich des isoelektrischen Punktes pl 6,7 sowie ein hydrolytisches Aktivitätsoptimum, das bei pH 5,5 liegt. Auch diese Merkmale werden von der vorliegenden Erfindung umfasst.
Wie bereits angegeben, sind Proteinfraktionen mit Phospholipase D-Aktivität üblicherweise Calciumionen-abhängig. Diese ausgeprägte Abhängigkeit hat sich allerdings für die beanspruchte pflanzliche Proteinfraktion aus Papaveraceen, die zwingend Zn +-lonen aktivierbar ist, nicht bestätigt. Allerdings kann das Aktivitätsoptimum dieser Proteinfraktion auch in Gegenwart von Calcium-Ionenkonzentrationen erreicht werden, die dann üblicherweise zwischen 40 M und 100 mM liegen, wobei entsprechende Enzymaktivitäten bei Konzentrationen zwischen 2 und 20 mM und zwischen 5 und 15 mM auftreten.
Bezüglich der Unterfraktion B beansprucht die vorliegende Erfindung eine Protein-Variante, deren Aktivierbarkeitsoptimum in Gegenwart von Zn +- lonenkonzentrationen auftritt, die zwischen 1 ,0 und 10 mM und besonders bevorzugt bei 5 mM liegen.
Als u.a. erfindungswesentlich sieht die vorliegende Erfindung vor, dass die Unterfraktionen A und/oder B Kohlenhydratanteile aufweisen, so dass sie also in glykosylierter Form als N-verknüpfte Glykoproteine vorliegen, und
dass es sich bei den Unterfraktionen A und B um Isoenzyme handelt.
Entsprechend der überraschend unterschiedlichen Aktivitätseigenschaften der Unterfraktionen A (ausschließlich Hydrolyse) und B (ausgeprägte * Transphosphatidylierung) umfasst die vorliegende Erfindung auch eine Variante der Proteinfraktion, bei der die Transphosphatidylierungs-Aktivität insgesamt stärker ausgeprägt ist als ihre Hydrolyse-Aktivität, was vor allem durch die Einzelaktivitäten der Unterfraktionen erklärbar ist und auch im Hinblick auf die bislang bekannten PLD-Varianten von Bedeutung ist, denen gegenüber die neuen Proteinfraktionen bis 100 mal ausgeprägtere Transphosphatidylierungs-Aktivitäten bezogen auf die entsprechenden Hydrolyse-Aktivitäten aufweisen.
Neben der Proteinfraktion selbst beansprucht die vorliegende Erfindung auch die Verwendung dieser Proteinfraktion zur Hydrolyse und/oder
Transphosphatidylierung von Phospholipiden und/oder deren Lyso-Formen, wobei insbesondere die Synthese von Phosphatidylcholin, Phosphatidylethanolamin, Phosphatidylglycerin, Phosphatidylinosit, Phosphatidsäure, Phosphatidylserin, deren Lyso-Formen und beliebige Mischungen beansprucht werden. Erwähnenswert ist in diesem Zusammenhang auch die Tatsache, dass die erfindungsgemäße Proteinfraktion in der Lage ist, Phosphatidylinosit zu hydrolysieren bzw. am Pl einen Kopfgruppenaustausch durchzuführen, was von den bislang bekannten PLDs so ebenfalls nicht bekannt ist. Dabei ist die Reaktionsführung insgesamt als nicht kritisch anzusehen, doch haben sich als Reaktionsmedien organische und/oder wässrige Phasen und als Phospholipid-Quelle Phosphatidylcholin und Phosphatidylethanolamin als sehr geeignet erwiesen.
Zusammenfassend lässt sich feststellen, dass mit dieser neuen pflanzlichen Proteinfraktion mit Phospholipase D-Aktivität aus Vertretern der Familie der Papaveraceae die Isolierung einer PLD gelungen ist, die insgesamt eine sehr
ausgeprägte Transphosphatidylierungs-Aktivität besitzt und deren beide isoenzymatische Unterfraktionen hydrolytische Aktivitäten insbesondere gegenüber Phosphatidylcholin aufweisen; beide Unterfraktionen sind zwar wie von PLDs bekannt durch Calciumionen aktivierbar, im Gegensatz zu bekannten Phospholipase D-Varianten aus anderen Pflanzen kann die neue Proteinfraktion aber auch und vor allem durch Zn2+-lonen aktiviert werden.
Die neue Proteinfraktion unterscheidet sich somit grundlegend von den bislang bekannten pflanzlichen PLDs und ihre Eigenschaften weichen deutlich von den PLDs ab, deren Gensequenzen bereits ermittelt worden sind.
Die nachfolgenden Beispiele verdeutlichen die charakteristischen Merkmale der beanspruchten pflanzlichen Proteinfraktion mit Phospholipase D-Aktivität.
Beispiele
Aufarbeitung pflanzlichen Materials (Enzvmαewinnunα) Mohn-Samen (Papaver somniferum) wurden auf einer 10 mm dicken Polyurethan-Schaumschicht in Petrischalen, die mit einem Nylongewebe abgedeckt waren, in destilliertem Wasser zum Keimen gebracht. Der Keimungsprozess wurde in Dunkelheit bei 25 °C und 70 bis 80 % relativer Luftfeuchtigkeit durchgeführt. Am zweiten Tag nach dem Anquellen wurde von den Keimlingen das Endosperm entfernt.
Diese aus insgesamt 10 g frischem Mohn-Samen gewonnenen Endospermen wurden mit einer geringen Menge kalten Acetons in einem Mörser verrieben und anschließend mit 300 ml kalten Acetons, das 300 g festes C02 enthielt, homogenisiert. Anschließend wurde das so erhaltene Präzipitat mit kaltem Aceton so lange gewaschen, bis das Filtrat farblos und transparent war. Der vakuumgetrocknete Rückstand war in Pulverform für mehrere Monate bei 4 °C stabil.
2 g dieses Acetonpulvers wurden in 50 ml einer Mischung aus 0,1 molarem Natriumacetat-Puffer/10 mM CaCIsB mM Cysteinhydrochlorid (pH 5,5) homogenisiert und bei 12 000 g bei 4 °C für 10 Minuten zentrifugiert. Die daraus resultierenden Extrakte wurden mit (NH4)2S04 (60 % Sättigung) behandelt und bei 26 000 g und 4 °C für 45 Minuten zentrifugiert. Der Niederschlag wurde anschließend in einer möglichst geringen Menge einer Mischung aus 0,1 M Natriumacetat-Puffer/10 mM CaCI_/6 mM
Cysteinhydrochlorid (pH 5,5) aufgenommen. Nach einer Dialyse gegen eine Mischung aus 0,01 molarem Natriumacetat-Puffer/50 mM CaCI2 (pH 5,5) wurde die Enzymiösung auf eine Octyl-Sepharose CL-4B-Säule aufgetragen. Mit folgenden Lösungen wurden die Proteine mit einer Flussrate von 9 ml/h dreistufig eluiert: 0,01 M Natriumacetat-Puffer/50 mM CaCI2; 0,005 M Natriumacetat-Puffer/30 mM CaCI2 (pH 5,5); 0,005 M Natriumacetat- Puffer/0,1 mM Ethylendiamin-Tetraessigsäure (EDTA), pH 5,5. Die
Enzymaktivität wurde mit Phosphatidyl-p-Nitrophenol (PpNP) bei pH-Werten von 5,5 und 8,0 bestimmt und die vereinten aktiven Fraktionen wurden mit Hilfe einer 100 kDa-Membran aufkonzentriert.
Proteinbestimmunq
Der Proteingehalt wurde entsprechend der Standardmethode von M.M. Bradford (Anal. Biochem. 72, 1976, 248 - 254) mit Rinderserum- Albumin als Standard bestimmt.
Hydrolytische Aktivität in wässriαen Systemen
Die hydrolytische Aktivität der so erhaltenen pflanzlichen Proteinfraktion mit Phospholipase D-Aktivität wurde in einem wässrigen System ermittelt, indem das aus PpNP freigesetzte p-Nitrophenol bestimmt wurde (Methode nach P. D'Arrigo, V. Piergianni, D. Scarcelli, S. Servi, "A Spectrophotometric Essay for Phospholipase D", Anal. Chim. Acta, 304, 1995, 249 - 254).
Zur Charakterisierung der jeweiligen Aktivitäten der PLD-Unterfraktionen A und B wurden die Reaktionen bei unterschiedlichen pH-Werten in Gegenwart von 10 mM CaC , in Gegenwart unterschiedlicher Konzentrationen von CaCI2 bei pH-Werten von 5,5 bzw. 8,0 sowie in Gegenwart unterschiedlicher Konzentrationen von ZnCI2 bei einem pH-Wert von 5,5 durchgeführt.
Transphosphatidylierung und hydrolytische Aktivität in einem Zwei-Phasensvstem
Entsprechend der Methode gemäß N. Dittrich und R. Ulbrich-Hofmann ("Transphosphatidylation by immobilised Phospholipase D in aqueous media", Biotechnol. Appl. Biochem. 34, 2001 , 189 - 194) wurden die Transphosphatidylierungs- und Hydrolyseaktivitäten im Zwei-Phasensystem bestimmt. Die entsprechenden Reaktionsmedien setzten sich aus einem Phosphatidylcholin enthaltenden Diethylether, Glycerin (zur Bestimmung der Transphosphatidylierungs-Aktivität) oder Wasser (zur Bestimmung der
Hydrolyse-Aktivität) sowie einem Gemisch aus Tris-HCI/CaCI2 oder einem Gemisch aus Natriumacetat-Puffer/CaCl2 und einem gereinigten Enzym (PLD-A und PLD-B) zusammen. Die jeweiligen Reaktionen wurden bei 30 °C in Reaktionsgefäßen, die mit Teflon-Silikon-Septen verschlossen waren, bei einer Schüttelfrequenz von 250/Min. durchgeführt. Während der Reaktion wurden Aliquote der organischen Phase(n) mittels HPTLC analysiert. Die Phospholipidgehalte wurden densitometrisch bei 550 nm gegenüber Standardmischungen aus Phosphatidylcholin, Phosphatidsäure und Phosphatidylglycenn bestimmt, wobei aus der Zunahme an Phosphatidylglycenn oder Phosphatidsäure die Enzymaktivität berechnet wurde.
Bestimmung von Enzvmmerkmalen
Die Bestimmung der Molekularmasse der gereinigten Proteine erfolgte mit Hilfe einer Elektrophorese in Gegenwart von SDS mit einer Bio-Rad Mini Protein II Gel-Elektrophoresezelle unter Verwendung von Polyacrylamidgelen.
Die isoelektrischen Punkte der PLD-Unterfraktionen A und B wurden mit Hilfe eines PhastGel IEF 3 - 9 on FastSystem Separation and Controll Unit
(Pharmacia LKB Biotechnology) bestimmt, wobei pl lEF-Marker (liquid mix 3 - 10) für die Kalibrierung eingesetzt wurden. Die Proteine wurden mit Coomassie Brilliant Blue G-250 gefärbt.
Zur Glycoprotein-Bestimmung wurde ein SDS-PAGE-Gel, das PLD-A, PLD-B und Peroxidase (als Standard für ein glycosiliertes Protein) sowie Aldolase (als Standard eines nicht-glycosilierten Proteins) enthielt, mit einer Nitrocellulose-Membran bei 300 V und 5 mA/cm2 über eine Dauer von 180 Minuten in Kontakt gebracht. Nach dem Proteintransfer wurde der Gesamtkohlenhydrat-Gehalt mit einem ECL-Glycoprotein-Detektionsmodul bestimmt.
Mit einem 492 cLC-Protein-Sequencer (PE Applied Biosystems) wurden Sequenzierungen N-terminierter Aminosäuren durchgeführt.
Ergebnisse: Die mit Hilfe einer hydrophoben Interaktionschromatographie zur
Aufreinigung von PLD entsprechend R. Lambrecht, R. Ulbrich-Hofmann ("A facile purification procedure of phospholipase D from cabbage and its characterization", Biol. Chem. Hoppe-Seyler 373, 1992, 81 - 88) erhaltenen zwei Enzymformen waren bei pH 8,0 (PLD-A) bzw. pH 5,5 (PLD-B) aktiv. Durch den Austausch von CaCI2 durch ZnCI2 in Pufferlösungen, konnten die gleichen Aufreinigungsergebnisse erzielt werden. Beide Enzymunterfraktionen waren im SDS-PAGE-Gel homogen. Tabelle 1 veranschaulicht die Aufreinigungsdaten, wobei die Aufreinigungsfaktoren der beiden Isoenzyme 84,7 (PLD-A) bzw. 94,1 (PLD-B) betrugen.
Proteinbestimmung der beiden Unterfraktionen
Mit Hilfe der SDS-PAGE-Methode wurden die Molekularmassen der PLD- Unterfraktion-A und der PLD-Unterfraktion-B mit 116,4 bzw. 114,1 kDa bestimmt. Deren isoelektrischen Punkte lagen bei 8,7 (PLD-A) und 6,7 (PLD- B). Es konnte nachgewiesen werden, dass sowohl PLD-A als PLD-B in glycosilierter Form vorlagen, da eine positive Deglycosilierungsreaktion mit Hilfe von N-Glycosidase F das Vorhandensein eines N-gebundenen Kohlenhydrats für beide Unterfraktionen ergab. Da die N-terminale Sequenzierungsmethode an beiden Unterfraktionen versagte, ist davon auszugehen, dass in beiden Fällen eine N-terminale Modifizierung vorliegt.
pH-Aktivitätsprofile
Gegenüber PpNP ergaben sich hinsichtlich der hydrolytischen Aktivitäten der PLD-Unterfraktionen A und B und als Funktion des pH-Wertes signifikante Unterschiede. Die Unterfraktion A besitzt ein scharfes pH-Optimum bei pH 8,0, wo hingegen bei diesem pH-Wert die Aktivität der Unterfraktion B
unausgeprägt ist. Dem gegenüber liegt das pH-Optimum der Unterfraktion B bei pH 5,5, bei dem die Unterfraktion A kaum Aktivität zeigt. Unter jeweils optimalen Bedingungen zeigt die Unterfraktion B eine gegenüber der Unterfraktion A um 38 % ausgeprägtere Aktivität.
Einfluss von Metallionen
Hinsichtlich der von PLD-Varianten bekannten Aktivierung durch Ca2+-lonen wurde auch bezüglich der neuen Unterfraktionen A und B aus Schlafmohn eine Calciumionen-Abhängigkeit festgestellt, wobei ein Aktivitätsmaximum mit einer 10 mM CaCI2-Konzentration erzielt werden konnte. Mit Mg2+-lonen konnte eine nur sehr geringfügige Aktivierung der beiden Untereinheiten erzielt werden, wohingegen Zn2+-lonen die PLD-Unterfraktionen A und vor allem B stärker aktivierten als Calciumionen. Mit einer optimalen Zn2+- lonenkonzentration (5 mM) konnte die Unterfraktion B um mehr als das vierfache stärker aktiviert werden, als mit den optimalen Ca2+- lonenkonzentrationen.
Transphosphatidylierunos- und hydrolytische Aktivitäten in einem 2-Phasen- Svstem In einem biphasischen System bestehend aus einem Natriumacetat- (pH 5,5) oder Tris-HCI-Puffer (pH 8,0) und 40 mM CaCI2, Diethylether mit Phosphatidylcholin als Substrat und Glycerin als Akzeptor-Alkohol, wurden die Transphosphatidylierungs-Potentiale der Unterfraktionen A und B mittels HPTLC und densitometrischer Quantifizierung der Reaktionsprodukte bestimmt. Die PLD-Unterfraktion B besaß bei pH 5,5 ein großes
Transphosphatidylierungs-Potential, da nach 240 Minuten mehr als 80 % des Phosphatidylcholins in das Transphosphatidylierungs-Produkt Phosphatidylglycenn umgewandelt worden waren, wohingegen unter diesen Reaktionsbedingungen keinerlei Phosphatidsäure gefunden werden konnte. Bei einem pH-Wert von 8,0 konnten für PLD-B weder eine
Transphosphatidylierung noch eine Hydrolyse festgestellt werden. Die PLD- Unterfraktion A zeigte weder bei pH 5,5 noch pH 8,0 eine
Transphosphatidylierungs-Aktivität, wie erwartet zeigte sie aber eine ausgeprägte Hydrolyse-Aktivität bei pH 8,0.

Die hydrolytische PLD-Aktivität der Proteinfraktion wurde gegenüber PpNP bei pH 8,8 und 5,0 bestimmt.

Claims
1. Pf lanzliche, aus Vertretern der Familie Papaveraceae stammende Proteinfraktion mit Phospholipase D-Aktivität, dadurch gekennzeichnet, dass a) sie aus zwei Protein-Unterfraktionen A und B besteht und b) sie durch Zn2+-lonen aktivierbar ist sowie c) die Unterfraktionen A und/oder B Kohlenhydratanteile aufweisen, und wobei die Protein-Unterfraktion A ausschließlich eine Hydrolyse- Aktivität besitzt.
2.Proteinfraktion nach Anspruch 1 , dadurch gekennzeichnet, dass sie aus Papaver somniferum stammt und ganz besonders bevorzugt aus sich entwickelnden Keimlingen und/oder aus Endospermen.
3. Proteinfraktion nach einem der Ansprüche 1 oder 2, dadurch gekennzeichnet, dass die Unterfraktion A eine Molekularmasse zwischen 116 und 118 kDa, einen isoelektrischen Punkt pl zwischen 8,5 und 8,9 und ein hydrolytisches Aktivitätsoptimum bei pH-Werten zwischen 7,8 und 8,2 und die Unterfraktion B eine Molekularmasse zwischen 112 und 115 kDa, einen isoelektrischen Punkt pl zwischen 6,5 und 6,9 und ein hydrolytisches Aktivitätsoptimum bei pH-Werten zwischen 5,0 und 6,0 besitzen.
4. Proteinfraktion nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass die Unterfraktion A eine Molekularmasse von 116,4 kDa, einen isoelektrischen Punkt pl von 8,7 und ein hydrolytisches Aktivitätsoptimum bei pH 8,0 aufweist.
5. Proteinfraktion nach einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass die Unterfraktion B eine Molekularmasse von 114,1 kDa, einen isolektrischen Punkt pl von 6,7 und ein hydrolytisches Aktivitätsoptimum bei pH 5,5 aufweist.
6. Proteinfraktion nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass die Unterfraktion B ein Aktivierbarkeitsoptimum bei Zn2+-lonenkonzentrationen zwischen 1 ,0 und 10 mM und besonders bevorzugt bei 5 mM besitzt.
7. Proteinfraktion nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass es sich bei den Unterfraktionen A und B um Isoenzyme handelt.
8. Proteinfraktion nach einem der Ansprüche 1 bis 7, dadurch gekennzeichnet, dass ihre Transphosphatidylierungs-Aktivität stärker ausgeprägt ist als ihre Hydrolyse-Aktivität.
9Nerwendung der Proteinfraktion nach einem der Ansprüche 1 bis 8, zur Hydrolyse und/oder Transphosphatidylierung von Phospholipiden und/oder deren Lyso-Formen.
10Nerwendung nach Anspruch 9 zur Synthese von Phosphatidylcholin, Phosphatidylethanolamin, Phosphatidylglycenn, Phosphatidylinosit, Phosphatidsäure, Phosphatidylserin und deren Lyso-Formen.
11.Verwendung nach einem der Ansprüche 9 oder 10 in Form einer Hydrolyse von Phosphatidylinosit und/oder eines Kopfgruppenaustausches an Phosphatidylinosit.
