Abstract
Der Erfindung betrifft Beta-Aminoketone der allgemeinen Formel (I), worin R1-R4 die in der Beschreibung angegebenen Bedeutungen haben zu deren - insbesondere stereoselektiver - Herstellung, Arzneimittel enthaltend erfindungsgemässer Beta-Aminoketone zur Herstellung von Arnzneimitteln zur Behandlung von Schmerz.
Description
BETA-AMINOKETONEN ZUR BEHANDLUNG VON SCHMERZ
Der Erfindung betrifft Beta-Aminoketone, ein Verfahren zu deren - insbesondere stereoselektiver - Herstellung, Arzneimittel enthaltend erfindungsgemäße Beta-Aminoketone sowie die Verwendung erfindungsgemäßer Beta-Aminoketone zur Herstellung von Arzneimitteln zur Behandlung von Schmerz.
Die Behandlung chronischer und nicht chronischer Schmerzzustände hat in der Medizin eine große Bedeutung. Es besteht ein weltweiter Bedarf an gut wirksamen Schmerztherapien für eine patientengerechte und zielorientierte Behandlung chronischer und nicht chronischer Schmerzzustände, wobei hierunter die erfolgreiche und zufriedenstellende Schmerzbehandlung für den Patienten zu verstehen ist. Dies zeigt sich in der großen Anzahl von wissenschaftlichen Arbeiten, die auf dem Gebiet der angewandten Analgetik bzw. der Grundlagenforschung zur Nociception in letzter Zeit erschienen sind.
Ein bekanntes Therapeutikum zur Behandlung starker Schmerzen ist Tramadolhydrochlorid - (1 RS,2RS)-2-[(Dimethylamino)methyl]-1 -(3-meth- oxyphenyl)cyclohexanol, Hydrochlorid. Aminomethyl-Aryl-Cyclohexanol- Derivate wie das Tramadol ((1RS, 2RS)-2-Dimethylaminomethyl-1-(3- methoxy-phenyl)-cyclo-hexanol, Hydrochlorid) können entsprechend eine analgetische Wirkung besitzen, aber auch hydroxylierte Tramadol-Derivate, wie sie z.B. in EP 753506 A1 beschrieben sind, oder sie können als Intermediate zur Herstellung von analgetisch wirksamen Substanzen
verwendet werden (wie z.B. 4- oder 5-substituierte Tramadol-Analoga, die in der EP 753 506 A1 oder EP 780 369 A1 beschrieben sind). Gerade Tramadol nimmt unter den zentralwirksamen Analgetika insofern eine Sonderstellung ein, daß dieser Wirkstoff eine starke Schmerzhemmung ohne die für Opioide bekannten Nebenwirkungen hervorruft (J. Pharmacol. Exptl. Ther. 267, 331 (1993)), wobei sowohl die Enantiomeren von Tramadol als auch die Enantiomeren der Tramadolmetaboiite an der analgetischen Wirkung beteiligt sind (J. Pharmacol. Exp. Ther. 260, 275 (1992)).
Es besteht aber noch Bedarf an weiteren Schmerzmitteln.
Gegenstand der Erfindung sind daher Beta-Aminoketone gemäß der allgemeinen Formel I

Besonders bevorzugt ist es, wenn diese erfindungsgemäßen Beta- Aminoketone in einer stereoselektiv reinen Form, insbesondere in einer anti-Konformation gemäß Formel la oder einer syn-Konformation gemäß Formel Ib

Die gezeigten Verbindungen sind wirksame Analgetika.
Im Sinne dieser Erfindung versteht man unter Alkyl- bzw. Cykloalkyl-Resten gesättigte und ungesättigte (aber nicht aromatische), verzweigte,
unverzweigte und cyclische Kohlenwasserstoffe, die unsubstituiert oder ein- oder mehrfach substituiert sein können. Dabei steht Cι-2-Alkyl für C1- oder C2-Alkyl, Cι-3-Alkyl für C1-, C2- oder C3-Alkyl, C1-4-Alkyl für C1-, C2-, C3- oder C4-Alkyl, C1-5-Alkyl für C1-, C2-, C3-, C4- oder C5-Alkyl, C1-6-Alkyl für C1-, C2-, C3-, C4-, C5- oder C6-Alkyl, C1-7-Alkyl für C1-, C2-, C3-, C4-, C5-, C6- oder C7-Alkyl, Cι-8-Alkyl für C1-, C2-, C3-, C4-, C5-, C6-, C7- oder C8-Alkyl, Cι-10-Alkyl für C1-, C2-, C3-, C4-, C5-, C6-, C7-, C8,- C9- oder C10-Alkyl und C- δ-Alkyl für C1-, C2-, C3-, C4-, C5-, C6-, C7-, C8,- C9-, C10-, C11-, C12-, C13-, C14-, C15-, C16-, C17- oder C18-Alkyl. In Bezug auf Cycloalkyl umfaßt der Begriff auch gesättigte Cycloalkyle, in denen ein oder 2 Kohlenstoffatome durch ein Heteroatom, S, N oder O ersetzt sind. Unter den Begriff Cycloalkyl fallen aber insbesondere auch ein- oder mehrfach, vorzugsweise einfach, ungesättigte Cycloalkyle ohne Heteroatom im Ring, solange das Cycloalkyl kein aromatisches System darstellt. Vorzugsweise sind die Alkyl- bzw. (gegebenenfalls auch mehrerer) Wasserstoffreste(s) durch F, Cl, Br, I, NH2, SH oder OH, wobei unter „mehrfach substituiert" bzw. „substituiert" bei mehrfacher Substitution zu verstehen ist, daß die Substitution sowohl an verschiedenen als auch an gleichen Atomen mehrfach mit den gleichen oder verschiedenen Substituenten erfolgt, beispielsweise dreifach am gleichen C-Atom wie im Falle von CF3 oder an verschiedenen Stellen wie im Falle von -CH(OH)-CH=CH-CHCI2. Besonders bevorzugte Substituenten sind hier F, Cl und OH. In Bezug auf Cycloalkyl kann der Wasserstoffrest auch durch OC-]-3-Alkyl oder Cι_3-Alkyl (jeweils ein- oder mehrfach substituiert oder unsubstituiert), insbesondere Methyl, Ethyl, n-Propyl, i- Propyl, CF3, Methoxy oder Ethoxy, ersetzt sein.
Unter dem Begriff (CH2)3-6 ist -CH2-CH2-CH2-, -CH2-CH2-CH2-CH2-, -CH2- CH2-CH2-CH2-CH2- und CH2-CH2-CH2-CH2-CH2-CH2- zu verstehen, unter (CH2)1-4 ist -CH2-, -CH2-CH2-, -CH2-CH2-CH2- und -CH2-CH2-CH2-CH2- zu verstehen, etc.
Unter einem Aryl-Rest werden Ringsysteme mit mindestens einem armomatischen Ring aber ohne Heteroatome in auch nur einem der Ringe verstanden. Beispiele sind Phenyl-, Naphthyl-, Fluoranthenyl-, Fluorenyl-, Tetralinyl- oder Indanyl, insbesondere 9H-Fluorenyl- oder Anthracenyl- Reste, die unsubstituiert oder einfach oder mehrfach substituiert sein können.
Unter einem Heteroaryl-Rest werden heterocyclische Ringsysteme mit mindestens einem ungesättigten Ring verstanden, die ein oder mehrere Heteroatome aus der Gruppe Stickstoff, Sauerstoff und/oder Schwefel enthalten und auch einfach oder mehrfach substituiert sein können. Beispielhaft seien aus der Gruppe der Heteroaryle Furan, Benzofuran, Thiophen, Benzothiophen, Pyrrol, Pyridin, Pyrimidin, Pyrazin, Chinolin, Isochinolin, Phthalazin, Benzo[1 ,2,5 ]thiadiazol, Benzothiazol, Indol, Benzotriazol, Benzodioxolan, Benzodioxan, Carbazol, Indol und Chinazolin aufgeführt.
Dabei versteht man im Zusammenhang mit Aryl und Heteroaryl unter substituiert - wenn nicht ausdrücklich anders definiert - die Substitution des Aryls oder Heteroaryls mit R23, OR23 einem Halogen, vorzugsweise F und/oder Cl, einem CF3, einem CN, einem NO2, einem NR 4R25, ejnem
C^.g-Alkyl (gesättigt), einem C-j.ß-Alkoxy, einem C .ß-Cycloalkoxy, einem
C .ß-Cycloalkyl oder einem C2_ρ-Alkylen.
Dabei steht der Rest R" für H, einen C^-jQ-Alkyl-, vorzugsweise einen
C-]_g-Alkyl-, einen Aryl- oder Heteroaryl- oder für einen über C<|_3-Alkyl, gesättigt oder ungesättigt, oder eine Ci. -Alkylen-Gruppe gebundenen
Aryl- oder Heteroaryl-Rest, wobei diese Aryl und Heteroarylreste nicht selbst mit Aryl- oder Heteroaryl-Resten substituiert sein dürfen,

Unter dem Begriff Salz ist jegliche Form des erfindungsgemäßen Wirkstoffes zu verstehen, in dem dieser eine ionische Form annimmt bzw. geladen ist und mit einem Gegenion (einem Kation oder Anion) gekoppelt ist bzw. sich in Lösung befindet. Darunter sind auch Komplexe des Wirkstoffes mit anderen Molekülen und Ionen zu verstehen, insbesondere Komplexe, die über ionische Wechselwirkungen komplexiert sind. Insbesondere versteht man darunter physiologisch verträgliche Salze mit Kationen oder Basen und physiologisch verträgliche Salze mit Anionen oder Säuren.
Unter dem Begriff des physiologisch verträglichen Salzes mit Kationen oder Basen versteht man im Sinne dieser Erfindung Salze mindestens einer der erfindungsgemäßen Verbindungen - meist einer (deprotonierten) Säure - als Anion mit mindestens einem, vorzugsweise anorganischen, Kation, die physiologisch - insbesondere bei Anwendung im Menschen und/oder Säugetier - verträglich sind. Besonders bevorzugt sind die Salze der Alkali- und Erdalkalimetalle aber auch mit NH4 +, insbesondere aber (Mono-) oder (Di-) Natrium-, (Mono-) oder (Di-) Kalium-, Magnesium- oder Calzium- Salze.
Unter dem Begriff des physiologisch verträglichen Salzes mit Anionen oder Säuren versteht man im Sinne dieser Erfindung Salze mindestens einer der erfindungsgemäßen Verbindungen - meist, beispielsweise am Stickstoff, protoniert - als Kation mit mindestens einem Anion, die physiologisch - insbesondere bei Anwendung im Menschen und/oder Säugetier - veträglich sind. Insbesondere versteht man darunter im Sinne dieser Erfindung das mit einer physiologisch verträglichen Säure gebildete Salz, nämlich Salze des jeweiligen Wirkstoffes mit anorganischen bzw. organischen Säuren, die physiologisch - insbesondere bei Anwendung im Menschen und/oder Säugetier - verträglich sind. Beispiele für physiologisch verträgliche Salze bestimmter Säuren sind Salze der: Salzsäure,
Bromwasserstoffsäure, Schwefelsäure, Methansulfonsäure, Ameisensäure, Essigsäure, Oxalsäure, Bernsteinsäure, Weinsäure, Mandelsäure, Fumarsäure, Milchsäure, Zitronensäure, Glutaminsäure, 1,1-Dioxo-1,2- dihydrol λ6-benzo[d]isothiazol-3-on (Saccharinsäure),
Monomethylsebacinsäure, 5-Oxo-prolin, Hexan-1-sulfonsäure,
Nicotinsäure, 2-, 3- oder 4-Aminobenzoesäure, 2,4,6-Trimethyl- benzoesäure, α-Liponsäure, Acetylglycin, Acetylsalicylsäure, Hippursäure und/oder Asparaginsäure. Besonders bevorzugt ist das Hydrochlorid-Salz.
Es ist für die vorgenannten erfindungsgemäßen Beta-Aminoketone besonders bevorzugt, wenn

In der pharmazeutischen Industrie sind zunehmend stereoisomerenreine Verbindungen von besonderer Bedeutung. Diese können völlig unterschiedliche Wirkungen entfalten. Daher ist ein besonderer Kern dieser Erfindung ein Verfahren, mit dem genau dies erreicht werdenm kann, nämlich eine hoch stereoselektive Synthese. Die Verfahren können aber natürlich auch unselektiv zur Herstellung der erfindungsgemäßen Verbindungen verwandt werden.
Ein weiterer Gegenstand der Erfindung ist ein Verfahren zur Herstellung von erfindungsgemäßen Beta-Aminoketonen der allgemeinen Formel I nach Reaktionschema III:

in Schritt c: die Temperatur bei < -78°C gehalten wird, und/oder als unpolares organisches Lösungsmittel THF verwendet wird, und/oder zur Einstellung eines ph < 7 NH F verwendet wird und TBAF oder HF eingesetzt werden.
Ein weiterer Gegenstand ist ein Verfahren zur Herstellung der erfindungsgemäßen Verbindungen mit freier NH2-Gruppe, bei dem nach Abschluß eines der Verfahren III, lila, lllb, IV, IVa, IVb oder IVc, bei dem R3 tert. Butyl entspricht, die hergestellten Verbindungen mit Trifluoressigsäure umgesetzt werden und das Lösungsmittel entfernt wird, so daß eine erfindungsgemäße Verbindung mit freier NH2-Gruppe in Form des Triflats entsteht.
Ein weiterer Gegenstand ist ein anderes Verfahren zur Herstellung der erfindungsgemäßen Verbindungen mit freier NH2-Gruppe, bei dem nach Abschluß eines der Verfahren III, lila, lllb, IV, IVa, IVb oder IVc, bei dem R3 nicht tert. Butyl entspricht, die hergestellten Verbindungen mit HCI umgesetzt werden und das Lösungsmittel entfernt wird.
Die erfindungsgemäßen Substanzen sind toxikologisch unbedenklich, so daß sie sich als pharmazeutischer Wirkstoff in Arzneimitteln eignen. Ein weiterer Gegenstand der Erfindung sind daher Arzneimittel enthaltend wenigstens ein erfindungsgemäßes Beta-Aminoketon sowie gegebenenfalls weitere Wirkstoffe, Hilfstoffe und/oder Zusatzstoffe.
Die erfindungsgemäßen Arzneimittel enthalten neben mindestens einer erfindungsgemäßen substituierten Verbindung gegebenenfalls geeignete Zusatz- und/oder Hilfsstoffe, so auch auch Trägermaterialien, Füllstoffe, Lösungsmittel, Verdünnungsmittel, Farbstoffe und/oder Bindemittel und können als flüssige Arzneiformen in Form von Injektionslösungen, Tropfen oder Säfte, als halbfeste Arzneiformen in Form von Granulaten, Tabletten, Pellets, Patches, Kapseln, Pflaster oder Aerosolen verabreicht werden. Die Auswahl der Hilfsstoffe etc. sowie die einzusetzenden Mengen derselben hängen davon ab, ob das Arzneimittel oral, peroral, parenteral, intravenös, intraperitoneal, intradermal, intramuskulär, intranasal, buccal, rektal oder örtlich, zum Beispiel auf die Haut, die Schleimhäute oder in die Augen, appliziert werden soll. Für die orale Applikation eignen sich Zubereitungen in Form von Tabletten, Dragees, Kapseln, Granulaten, Tropfen, Säften und Sirupen, für die parenterale, topische und inhalative Applikation Lösungen, Suspensionen, leicht rekonstituierbare Trockenzubereitungen sowie Sprays. Erfindungsgemäße Verbindungen in einem Depot, in gelöster Form oder in einem Pflaster, gegebenenfalls unter Zusatz von die Hautpenetration fördernden Mitteln, sind geeignete perkutane Applikationszubereitungen. Oral oder perkutan anwendbare Zubereitungsformen können die erfindungsgemäßen Verbindungen verzögert freisetzen. Prinzipiell können den erfindungsgemäßen Arzneimitteln andere dem Fachmann bekannte weitere Wirkstoffe zugesetzt werden.
Die an den Patienten zu verabreichende Wirkstoffmenge variiert in Abhängigkeit vom Gewicht des Patienten, von der Applikationsart, der Indikation und dem Schweregrad der Erkrankung. Üblicherweise werden
0,005 bis 1000 mg/kg, bevorzugt 0,05 bis 5 mg/kg wenigstens einer erfindungsgemäßen Verbindung appliziert.
Ein weiterer Gegenstand der Erfindung die Verwendung eines erfindungsgemäßen Beta-Aminoketons zur Herstellung eines Arzneimittel zur Behandlung von Schmerz, Anxiety, Depression oder Epilepsie, insbesondere Schmerz.
Im folgenden wird die Erfindung weiter durch Beispiele erläutert, ohne sie darauf zu beschränken.

Die eingesetzten Chemikalien und Lösungsmittel wurden kommerziell bei den herkömmlichen Anbietern erworben (Acros, Avocado, Aldrich, Fluka, Lancaster, Maybridge, Merck, Sigma, TCI etc. oder synthetisiert).

Allgemeine Arbeitsvorschrift für Schritt a (lllb) (AAV1)
Zu 1.2 Äquivalenten LDA (in 5 ml abs. THF und 1.15 ml abs. HMPA pro mmol Diisopropylamin) wurde unter Schutzgasatmosphäre bei -78 °C ein Äquivalent Keton X (verdünnt mit 1 ml abs. THF pro mmol) zugetropft. Anschliessend (8 - 12 Minuten) wurden 1.4 Äquivalente Tnmethylsilylchlorid unverdünnt zugegeben. Nach Erwärmen auf Raumtemperatur (ca. 1 Stunde) wurde überschüssige Base durch Zugabe von gesättigter, wässriger Natriumhydrogencarbonat-Lösung zerstört. Der Silylenolether wurde mit n-Pentan extrahiert und die organische Phase mit gesättigter, wässriger Natriumchlorid-Lösung gewaschen. Trocknen über Magnesiumsulfat, Filtration und Entfernen des Lösungsmittels am Rotationsverdampfer lieferte den rohen Silylenolether Xla.
Allgemeine Arbeitsvorschrift für Schritt b (Schema lllb, Methode A) (AAV2)
Zu einer Lösung von 1.1 Äquivalenten α-(Λ/-Alkoxycarbonyl)amin Sulfon Xlla in abs. Dichlormethan (5 ml abs. CH2CI2 pro mmol) gab man 1.2 Äquivalente KH und rührte für eine Stunde bei Raumtemperatur. Nach Abkühlen der Lösung auf -78 °C gab man tropfenweise 1.5 Äquivalente einer 1.0 N Titantetrachlorid-Dichlormethan-Lösung hinzu. Nach 20 Minuten kühlte man auf -90 °C ab und gab tropfenweise über 15 Minuten ein Äquivalent des (S,Z)-Silylenolethers Xla verdünnt mit abs. Dichlormethan (1 ml abs. CH2CI2 pro mmol) hinzu. Nach Rühren für 75 Minuten, wobei die Temperatur wieder auf -78 °C ansteigt, wurde die Reaktion durch tropfenweise Zugabe einer wässrigen, gesättigten Natriumhydrogencarbonat-Lösung beendet. Die auf Raumtemperatur erwärmte Lösung wurde mit Dichlormethan extrahiert. Die vereinigten organischen Phasen wurden mit gesättigter, wässriger Natriumchlorid-Lösung gewaschen. Nach Trocknen über Magnesiumsulfat, Filtration und Entfernen des Lösungsmittels am Rotationsverdampfer wurde das Produkt Xllla mittels Säulenchromatographie gereinigt.
Allgemeine Arbeitsvorschrift für Schritt b (Schema lllb, Methode B) (AAV3)
Zu einer Lösung von 1.1 Äquivalenten α-(Λ/-Alkoxycarbonyl)amin Sulfon Xlla in abs. Dichlormethan (5 ml abs. Nach Abkühlen auf -90 °C gab man tropfenweise über 15 Minuten ein Äquivalent des (S,Z)-Silylenolethers Xla verdünnt mit abs. Dichlormethan (1 ml abs. CH2CI2 pro mmol) hinzu. Nach Rühren für 75 Minuten, wobei die Temperatur wieder auf -78 °C anstieg, wurde die Reaktion durch tropfenweise Zugabe wässriger,
gesättigter Natriumhydrogencarbonat-Lösung beendet. Die auf Raumtemperatur erwärmte Lösung wurde mit Dichlormethan extrahiert. Die vereinigten organischen Phasen wurden mit gesättigter, wässriger Natriumchlorid-Lösung gewaschen. Allgemeine Arbeitsvorschrift für Schritt d (Schema IVa, IVc) (AAV4)
Zu 2.5 Äquivalenten LDA in abs. Tetrahydrofuran (in 5 ml abs. THF und 1.15 ml abs. HMPA pro mmol Diisopropylamin) wurde unter Schutzgasatmosphäre bei -78 °C ein Äquivalent Keton V oder X (1 ml abs. THF pro mmol) zugetropft und für fünf Minuten metalliert. 1.5 Äquivalente α-(/V- Alkoxycarbonyl)amin Sulfon VII oder Xlla wurden in wenig abs. Tetrahydrofuran (1 ml abs. THF pro mmol) gelöst und tropfenweise zur Reaktionslösung gegeben. Nach Rühren für 25 Minuten bei -78 °C versetzte man tropfenweise mit wässriger, gesättigter Natriumhydrogencarbonat-Lösung. Die wässrige Phase wurde zweimal mit π-Pentan extrahiert, und die vereinigten organischen Phasen wurden mit gesättigter, wässriger Natriumchlorid-Lösung gewaschen. Nach dem Trocknen über Magnesiumsulfat und Entfernen des Lösungsmittels am Rotationsverdampfer wurde das Produkt Vlllb oder Xlllb mittels Säulenchromatographie gereinigt. Weitere Aufreinigung und Isolierung des Überschussdiastereomeren erfolgte durch präparative HPLC.
Allgemeine Arbeitsvorschrift für Schritt c (Schema lllb, IVa, IVc) (AAV5)
Ein Äquivalent ß-Aminoketon Xllla, Vlllb oder Xlllb wurde in abs. Tetrahydrofuren (10 ml abs. THF pro mmol) gelöst und bei -78 °C mit fünfzig Äquivalenten Ammoniumfluorid und 1.5 - 1.7 Äquivalenten einer 1.0 N Tetrabutylammoniumfluorid-Lösung versetzt. Nach Rühren bei -78 °C für
1.0 - 1.5 Stunden wurde auf Raumtemperatur erwärmt. Die Aufarbeitung erfolgte durch Zugabe von pH-6 Puffer, Extraktion in n-Pentan oder Diethylether, Trocknen über Magnesiumsulfat und Entfernen des Lösungsmittels am Rotationsverdampfer. Die Reinigung erfolgte durch Kristallisation aus n-Pentan, Diethylether oder Dichlormethan bei -20 °C. Die ß-Aminoketone lla, Ib und llb wurden als farblose Feststoffe erhalten.

TMSCI eingesetzt wird;

in Schritt b: die Temperatur bei < -78°C gehalten wird, und/oder als polares organisches Lösungsmittel CH2CI verwendet wird, und/oder
als Lewis-Säure TiCI4 oder SnCI4, vorzugsweise TiCI verwendet wird; und/oder besonders bevorzugt zusätzlich KH eingesetzt wird.

in Schritt a: die Temperatur bei < -78°C gehalten wird, und/oder als unpolares organisches Lösungsmittel THF verwendet wird, und/oder als Base LDA verwendet wird; und
TMSCI und HMPA eingesetzt werden;
und/oder
in Schritt b: die Temperatur bei < -78°C gehalten wird, und/oder als polares organisches Lösungsmittel CH2CI2 verwendet wird, und/oder als Lewis-Säure TiCI4 oder SnCI , vorzugsweise TiCI4 verwendet wird; und/oder
besonders bevorzugt zusätzlich KH eingesetzt wird.
und/oder
in Schritt c: die Temperatur bei < -78°C gehalten wird, und/oder als unpolares organisches Lösungsmittel THF verwendet wird, und/oder zur Einstellung eines ph < 7 NH4F verwendet wird und TBAF oder HF eingesetzt werden.
21. Verfahren zur Herstellung von Beta-Aminoketonen der allgemeinen
Formel I gemäß Anspruch 1 , nach Reaktionschema IV:

, worin GSi = tert.-Hexyl-(CH2)2Si ist, dadurch gekennzeichnet, daß in Schritt d: die Temperatur bei < 0°C gehalten wird, ein unpolares organisches Lösungsmittel verwendet wird,

in Schritt c: die Temperatur bei < 0°C gehalten wird, ein unpolares organisches Lösungsmittel verwendet wird, TBAF oder HF eingesetzt werden.
22. Verfahren zur stereoselektiven Herstellung von syn-Beta-Aminoketonen der allgemeinen Formel Ib gemäß Anspruch 2, nach Reaktionschema

, worin GSi = tert.-Hexyl-(CH2)2Si ist, dadurch gekennzeichnet, daß in Schritt d: die Temperatur bei < -70°C gehalten wird, ein unpolares organisches Lösungsmittel verwendet wird,
HMPA eingesetzt wird, und
die Reaktion in Gegenwart einer Base stattfindet;
in Schritt c: die Temperatur bei < -70°C gehalten wird, ein unpolares organisches Lösungsmittel verwendet wird, TBAF oder HF eingesetzt werden und die Reaktion bei einem pH < 7 stattfindet.
23. Verfahren zur Herstellung von Beta-Aminoketonen der allgemeinen Formel II gemäß Anspruch 7 nach Reaktionschema IVb:

, worin GSi = tert.-Hexyl-(CH2)2Si ist, dadurch gekennzeichnet, daß in Schritt d: die Temperatur bei < 0°C gehalten wird,
ein unpolares organisches Lösungsmittel verwendet wird, und die Reaktion in Gegenwart einer Base stattfindet;
in Schritt c: die Temperatur bei < 0°C gehalten wird, ein unpolares organisches Lösungsmittel verwendet wird, TBAF oder HF eingesetzt werden..
24. Verfahren zur stereoselektiven Herstellung von syn-Beta-Aminoketonen der allgemeinen Formel llb gemäß Anspruch 8 nach Reaktionschema IVc:

, worin GSi = tert.-Hexyl-(CH2)2Si ist, dadurch gekennzeichnet, daß in Schritt d: die Temperatur bei < -70°C gehalten wird, ein unpolares organisches Lösungsmittel verwendet wird, HMPA eingesetzt wird,
und die Reaktion in Gegenwart einer Base stattfindet;
in Schritt c: die Temperatur bei < -70°C gehalten wird, ein unpolares organisches Lösungsmittel verwendet wird, TBAF oder HF eingesetzt werden und die Reaktion bei einem pH < 7 stattfindet.
25. Verfahren gemäß einem der Ansprüche 21 bis 24, dadurch gekennzeichnet, daß
in Schritt d: die Temperatur bei < -78°C gehalten wird, und/oder als unpolares organisches Lösungsmittel THF verwendet wird, HMPA eingesetzt wird und/oder als Base eine Lithiumbase, bevorzugt LDA eingesetzt wird.
und/oder
in Schritt c: die Temperatur bei < -78°C gehalten wird, und/oder als unpolares organisches Lösungsmittel THF verwendet wird, und/oder zur Einstellung eines ph < 7 NH4F verwendet wird und TBAF oder HF eingesetzt werden.
26. Verfahren zur Herstellung einer Verbindung gemäß einem der Ansprüche 3, 4, 9 oder 10, dadurch gekennzeichnet, daß nach Abschluß eines Verfahrens nach einem der Ansprüche 16 bis 25, bei dem R3 tert. Butyl entspricht, die hergestellten Verbindungen mit Trifluoressigsäure umgesetzt werden und das Lösungsmittel entfernt wird, so daß die Verbindung gemäß einem der Ansprüche 3, 4, 9 oder 10 in Form der Triflate entsteht.
27. Verfahren zur Herstellung einer Verbindung gemäß einem der Ansprüche 3, 4, 9 oder 10, dadurch gekennzeichnet, daß nach Abschluß eines Verfahrens nach einem der Ansprüche 16 bis 25, bei dem R3 nicht tert. Butyl entspricht, die hergestellten Verbindungen mit HCI umgesetzt werden und das Lösungsmittel entfernt wird.
28. Arzneimittel enthaltend mindestens ein Beta-Aminoketon gemäß einem der Ansprüche 1 bis 15 sowie gegebenenfalls weitere Wirkstoffe, Hilfstoffe und/oder Zusatzstoffe.
29. Verwendung eines Beta-Aminoketons gemäß einem der Ansprüche 1 bis 15 zur Herstellung eines Arzneimittel zur Behandlung von Schmerz, Anxiety, Depression oder Epilepsie, insbesondere Schmerz.
