Abstract
Substituierte Zimtsäureguanidide, Verfahren zu ihrer Herstellung, ihre Verwendung als Medikament oder Diagnostikum sowie sie enthaltendes Medikament. Verbindungen der Formel (I), in denen R(1), R(2), R(3), R(4), R(5), R(6), und R(7) die in den Ansprüchen angegebenen Bedeutungen haben, sind hervorragende Herz-Kreislauf-Therapeutika. Sie werden durch Umsetzung einer Verbindung (II) mit Guanidin erhalten.

Enthalten die Verbindungen der Formel I ein oder mehrere Asymmetriezentren, so können diese sowohl S als auch R konfiguriert sein. Die Verbindungen können als optische Isomere, als Diastereomere, als Racemate oder als Gemische derselben vorliegen.
Die Doppelbindungsgeometrie der Verbindungen der Formel I kann sowohl E als auch Z sein. Die Verbindungen können als Doppelbindungs-Isomere im Gemisch vorliegen.
Die bezeichneten Alkylreste, bzw. teilweise oder vollständig fluorierten Alkylreste können sowohl geradkettig als auch verzweigt vorliegen.
Als N-haltige Heterocyclen mit 1 , 2, 3, 4, 5, 6, 7, 8 oder 9 C-Atomen gelten insbesondere Pyrrolyl, Imidazolyl, Pyrazolyl, Triazolyl, Tetrazolyl, Oxazolyl, Isoxazolyl, Thiazolyl, Isothiazolyl, Pyridyl, Pyrazinyl, Pyrimidinyl, Pyridazinyl, Indolyl, Isoindolyl, Benzimidazolyl, Indazolyl, Chinolyl, Isochinolyl, Phthalazinyl, Chinoxalinyl, Chinazolinyl, Cinnolinyl.
Besonders bevorzugt sind die N-haltigen Heterocyclen Pyrrolyl, Imidazolyl, Pyrazolyl, Pyridyl, Pyrazinyl, Pyrimidinyl und Pyridazinyl.
Ganz besonders bevorzugt ist Pyridyl.
Die Erfindung betrifft weiterhin ein Verfahren zur Herstellung der Verbindungen I, dadurch gekennzeichnet, daß man eine Verbindung der Formel II
mit Guanidin umsetzt, worin R(1) bis R(7) die angegebene Bedeutung besitzen und L für eine leicht nucleophil substituierbare leaving group steht.
Leaving groups sind zum Beispiel: -OMe, -OEt, -OPh, -SPh, -SMe, 1 -Imidazolyl.
Die aktivierten Säurederivate der Formel II, worin L eine Alkoxy-, vorzugsweise eine Methoxygruppe, eine Phenoxygruppe, Phenylthio- Methylthio-, 2-Pyridylthiogruppe, einen Stickstoffheterocyclus, vorzugsweise 1 -Imidazolyl, bedeutet, erhält man vorteilhaft in an sich bekannter Weise aus den zugrundeliegenden Carbonsäurechloriden (Formel II, L = Cl), die man ihrerseits wiederum in an sich bekannter Weise aus den zugrundeliegenden Carbonsäuren (Formel II, L = OH) beispielsweise mit Thionylchlorid herstellen kann.
Neben den Carbonsäurechloriden der Formel II (L = Cl) lassen sich auch weitere aktivierte Säurederivate der Formel II in an sich bekannter Weise direkt aus den zugrundeliegenden Benzoesäurederivaten (Formel II, L = OH) herstellen, wie beispielsweise die Methylester der Formel II mit L = OCH3 durch Behandeln mit gasförmigem HCI in Methanol, die Imidazolide der Formel II durch Behandeln mit
Carbonyldiimidazol [L = 1-lmidazolyl, Staab, Angew. Chem. Int. Ed. Engl. 1 , 351 - 367 (1962)], die gemischten Anhydride II mit CI-COOC2H5 oder Tosylchlorid in
Gegenwart von Triethylamin in einem inerten Lösungsmittel, wie auch die Aktivierungen von Benzoesäuren mit Dicyclohexylcarbodiimid (DCC) oder mit O- [(Cyano(ethoxycarbonyl)methylen)amino]-1 ,1 ,3,3-tetramethyluronium-tetrafluoborat ("TOTU") [Proceedings of the 21. European Peptide Symposium, Peptides 1990, Editors E. Giralt and D. Andreu, Escom, Leiden, 1991]. Eine Reihe geeigneter Methoden zur Herstellung von aktivierten Carbonsäurederivaten der Formel II sind unter Angabe von Quellenliteratur in J. March, Advanced Organic Chemistry, Third Edition (John Wiley & Sons, 1985), S. 350 angegeben.
Die Umsetzung eines aktivierten Carbonsäurederivates der Formel II mit Guanidin erfolgt in an sich bekannter Weise in einem protischen oder aprotischen polaren aber inerten organischen Lösungsmittel. Dabei haben sich bei der Umsetzung der Benzoesäuremethyl- oder Benzoesäureethylester (II, L = OMe, OEt) mit Guanidin Methanol, Isopropanol, DMF oder THF von 20°C bis zur Siedetemperatur dieser Lösungsmittel bewährt. Bei den meisten Umsetzungen von Verbindungen II mit salzfreiem Guanidin wurde vorteilhaft in aprotischen inerten Lösungsmitteln wie THF, DMF, Dimethoxyethan, Dioxan gearbeitet. Aber auch Wasser kann unter Gebrauch einer Base wie beispielsweise NaOH als Lösungsmittel bei der Umsetzung von II mit Guanidin verwendet werden.
Wenn L = Cl bedeutet, arbeitet man vorteilhaft unter Zusatz eines Säurefängers, z. B. in Form von überschüssigem Guanidin zur Abbindung der Halogenwasserstoffsäure.
Ein Teil der zu Grunde liegenden Benzoesäurederivate der Formel II ist bekannt und in der Literatur beschrieben. Die unbekannten Verbindungen der Formel II können nach literaturbekannten Methoden hergestellt werden. Die erhaltenen Alkenylcarbonsäuren werden nach einer der oben beschriebenen
Verfahrensvarianten zu erfindungsgemäßen Verbindungen I umgesetzt.
Die Einführung einiger Substituenten gelingt durch literaturbekannte Methoden des Palladium-vermittelten cross-couplings von Arylhalogeniden bzw. Aryltriflaten mit beispielsweise Organostannanen, Organoboronsäuren oder Organoboranen oder Organokupfer- bzw. zink-Verbindungen.
Carbonsäureguanidine I sind im allgemeinen schwache Basen und können Säure unter Bildung von Salzen binden. Als Säureadditionssalze kommen Salze aller pharmakologisch verträglichen Säuren in Frage, beispielsweise Halogenide, Insbesondere Hydrochloride, Lactate, Sulfate, Citrate, Tartrate, Acetate, Phosphate, Methylsulfonate, p-Toluolsulfonate.
Die Verbindungen I sind substituierte Acylguanidine.
Aus der Europäischen Offenlegungsschrift 755 919 (HOE 94/F 168) sind ähnliche Zimtsäureguanidide bekannt, jedoch entsprechen diese bekannten Verbindungen nicht allen gewünschten Anforderungen.
Mit den Verbindungen der Formel I konnte eine verbesserte Wasserlöslichkeit erreicht werden, was sich vor allem in einer erhöhten Ausscheidung über die Niere auswirkt. Außerdem zeigen die Verbindungen der Formel I eine sehr gute inhibierende
Wirkung am NHE3 (Na+ / H+-Austauscher-Subtyp 3).
Dies ist durch die erfindungsgemäßen Verbindungen gelungen, welche keine unerwünschten und nachteiligen salidiuretischen, jedoch sehr gute antiarrhythmische Eigenschaften aufweisen, wie sie beispielsweise für die
Behandlung von Krankheiten wichtig sind, die durch Sauerstoffmangel bewirkt werden. Die Verbindungen sind infolge ihrer pharmakologischen Eigenschaften als antiarrhythmische Arzneimittel mit cardioprotektiver Komponente zur Infarktprophylaxe und der Infarktbehandlung sowie zur Behandlung der angina pectoris hervorragend geeignet, wobei sie auch präventiv die pathophysiologischen Vorgänge beim Entstehen ischämisch induzierter Schäden, insbesondere bei der
Auslösung ischämisch induzierter Herzarrhythmien, inhibieren oder stark vermindern. Wegen ihrer schützenden Wirkungen gegen pathologische hypoxische und ischämische Situationen können die erfindungsgemäßen Verbindungen der
Formel I infolge Inhibition des zellulären Na+/H+-Austauschmechanismus als Arzneimittel zur Behandlung aller akuten oder chronischen durch Ischämie ausgelösten Schäden oder dadurch primär oder sekundär induzierten Krankheiten verwendet werden. Dies betrifft ihre Verwendung als Arzneimittel für operative Eingriffe, z.B. bei Organ-Transplantationen, wobei die Verbindungen sowohl für den Schutz der Organe im Spender vor und während der Entnahme, zum Schutz entnommener Organe beispielsweise bei Behandlung mit oder deren Lagerung in physiologischen Badflüssigkeiten, wie auch bei der Überführung in den Empfängerorganismus verwendet werden können. Die Verbindungen sind ebenfalls wertvolle, protektiv wirkende Arzneimittel bei der Durchführung angioplastischer operativer Eingriffe beispielsweise am Herzen wie auch an peripheren Gefäßen. Entsprechend ihrer protektiven Wirkung gegen ischämisch induzierte Schäden sind die Verbindungen auch als Arzneimittel zur Behandlung von Ischämien des Nervensystems, insbesondere des ZNS, geeignet, wobei sie z.B. zur Behandlung des Schlaganfalls oder des Hirnödems geeignet sind. Darüber hinaus eignen sich die erfindungsgemäßen Verbindungen der Formel I ebenfalls zur Behandlung von Formen des Schocks, wie beispielweise des allergischen, cardiogenen, hypovolämischen und des bakteriellen Schocks.
Weiterhin induzieren die Verbindungen eine Verbesserung des Atemantriebes und werden deshalb zur Behandlung von Atmungszuständen bei folgenden klinischen Zuständen und Krankheiten herangezogen: Gestörter zentraler Atemantrieb (z. B. zentrale Schlafapnoen, plötzlicher Kindstod, postoperative Hypoxie), muskulärbedingte Atemstörungen, Atemstörungen nach Langzeitbeatmung, Atemstörungen bei Adaptation im Hochgebirge, obstruktive und gemischte Form der Schlafapnoen, akute und chronische Lungenkrankheiten mit Hypoxie und Hyperkapnie.
Zusätzlich erhöhen die Verbindungen den Muskeltonus der oberen Atemwege, so i daß das Schnarchen unterdrückt wird.
Eine Kombination eines NHE-Inhibitors mit einem Carboanhydrase-Hemmer (z. B. Acetazolamid), wobei letzterer eine metabolische Azidose herbeiführt und dadurch bereits die Atmungstätigkeit steigert, erweist sich als vorteilhaft durch verstärkte Wirkung und verminderten Wirkstoffeinsatz.
Es hat sich gezeigt, daß die erfindungsgemäßen Verbindungen eine milde abführende Wirkung besitzen und demzufolge vorteilhaft als Abführmittel oder bei drohender Darmverstopfung verwendet werden können, wobei die Verhinderung der mit Verstopfungen im Darmbereich einhergehenden ischämischen Schäden besonders vorteilhaft ist.
Weiterhin besteht die Möglichkeit, der Gallenstein-Bildung vorzubeugen.
Darüber hinaus zeichnen sich die erfindungsgemäßen Verbindungen der Formel I durch starke inhibierende Wirkung auf die Proliferationen von Zellen, beispielsweise der Fibroblasten- Zellproliferation und der Proliferation der glatten Gefäßmuskelzellen, aus. Deshalb kommen die Verbindungen der Formel I als wertvolle Therapeutika für Krankheiten in Frage, bei denen die Zellproliferation eine primäre oder sekundäre Ursache darstellt, und können deshalb als Antiatherosklerotika, Mittel gegen diabetische Spätkomplikationen, Krebserkrankungen, fibrotische Erkrankungen wie Lungenfibrose, Leberfibrose oder Nierenfibrose, Organhypertrophien und -hyperplasien, insbesondere bei Prostatahyperplasie bzw. Prostatahypertrophie verwendet werden.
Die erfindungsgemäßen Verbindungen sind wirkungsvolle Inhibitoren des zellulären Natrium-Protonen-Antiporters (Na/H- Exchanger), der bei zahlreichen Erkrankungen (Essentielle Hypertonie, Atherosklerose, Diabetes usw.) auch in solchen Zellen erhöht ist, die Messungen leicht zugänglich sind, wie beispielsweise in Erythrocyten, Thrombocyten oder Leukozyten. Die erfindungsgemäßen Verbindungen eignen sich deshalb als hervorragende und einfache wissenschaftliche Werkzeuge, beispielsweise in ihrer Verwendung als Diagnostika zur Bestimmung und
Unterscheidung bestimmter Formen der Hypertonie, aber auch der Atherosklerose, des Diabetes, proliferativer Erkrankungen usw.. Darüber hinaus sind die Verbindungen der Formel I für die präventive Therapie zur Verhinderung der Genese des Bluthochdrucks, beispielweise der essentiellen Hypertonie, geeignet.
Es wurde außerdem gefunden, daß NHE- Inhibitoren eine günstige Beeinflussung der Serumlipoproteine zeigen. Es ist allgemein anerkannt, daß für die Entstehung arteriosklerotischer Gefäßveränderungen, insbesondere der koronaren Herzkrankheit, zu hohe Blutfettwerte, sogenannte Hyperlipoproteinämien, einen wesentlichen Risikofaktor darstellen. Für die Prophylaxe und die Regression von atherosklerotischen Veränderungen kommt daher der Senkung erhöhter Serum- Lipoproteine eine außerordentliche Bedeutung zu. Die erfindungsgemäßen Verbindungen können daher zur Prophylaxe und zur Regression von atherosklerotischen Veränderungen herangezogen werden, indem sie einen kausalen Risikofaktor ausschalten. Mit diesem Schutz der Gefäße gegen das
Syndrom der endothelialen Dysfunktion sind Verbindungen der Formel I wertvolle Arzneimittel zur Prävention und zur Behandlung koronarer Gefäßspasmen, der Atherogenese und der Atherosklerose, der linksventrikulären Hypertrophie und der dilatierten Kardiomyopathie, und thrombotischer Erkrankungen.
Die genannten Verbindungen finden deshalb vorteilhaft Verwendung zur Herstellung eines Medikaments zur Prävention und Behandlung von Schlafapnoen und muskulär bedingter Atemstörungen; zur Herstellung eines Medikaments zur Prävention und Behandlung des Schnarchens; zur Herstellung eines Medikaments zur Blutdrucksenkung; zur Herstellung eines Medikaments mit abführender Wirkung zur Prävention und Behandlung intestinaler Verstopfungen; zur Herstellung eines Medikaments zur Prävention und Behandlung von Erkrankungen, die durch Ischämie und Reperfusion von zentralen und peripheren Organen ausgelöst werden wie das akute Nierenversagen, der Schlaganfall, endogene Schockzustände, Darmerkrankungen etc.; zur Herstellung eines Medikaments zur Behandlung von Hypercholesterinämie; zur Herstellung eines Medikaments zur Prävention der Atherogenese und der Atherosklerose; zur Herstellung eines Medikaments zur
Prävention und Behandlung von Krankheiten, die durch erhöhte Cholesterinspiegel ausgelöst werden; zur Herstellung eines Medikaments zur Prävention und Behandlung von Krankheiten, die durch endotheliale Dysfunktion ausgelöst werden; zur Herstellung eines Medikaments zur Behandlung des Befalls durch Ektoparasiten; zur Herstellung eines Medikaments zur Behandlung der genannten Leiden in Kombinationen mit blutdrucksenkenden Stoffen, bevorzugt mit Angiotensin Converting Enzyme (ACE)-Hemmem und Angiotensin-Rezeptorantagonisten. Eine Kombination eines NHE-Inhibitors der Formel I mit einem blutfettspiegelsenkenden Wirkstoff, bevorzugt mit einem HMG-CoA-Reduktaseinhibitor (z. B. Lovastatin oder Pravastatin), wobei letzterer eine hypolipidämische Wirkung herbeiführt und dadurch die hypolipidämischen Eigenschaften des NHE-Inhibitors der Formel I steigert, erweist sich als günstige Kombination mit verstärkter Wirkung und vermindertem Wirkstoffeinsatz.
Beansprucht wird die Gabe von Natrium-Protonen-Austausch-Hemmern der Formel I als neuartige Arzneimittel zur Senkung erhöhter Blutfettspiegel, sowie die Kombination von Natrium-Protonen-Austausch-Hemmern mit blutdrucksenkenden und/oder hypolipidämisch wirkenden Arzneimitteln.
Arzneimittel, die eine Verbindung I enthalten, können dabei oral, parenteral, intravenös, rektal oder durch Inhalation appliziert werden, wobei die bevorzugte Applikation von dem jeweiligen Erscheinungsbild der Erkrankung abhängig ist. Die Verbindungen I können dabei allein oder zusammen mit galenischen Hilfsstoffen zur Anwendung kommen, und zwar sowohl in der Veterinär- als auch in der Humanmedizin.
Welche Hilfsstoffe für die gewünschte Arzneimittelformulierung geeignet sind, ist dem Fachmann auf Grund seines Fachwissens geläufig. Neben Lösemitteln, Gelbildnern, Suppositorien-Grundlagen, Tablettenhilfsstoffen, und anderen Wirkstoffträgern können beispielsweise Antioxidantien, Dispergiermittel,
Emulgatoren, Entschäumer, Geschmackskorrigentien, Konservierungsmittel, Lösungsvermittler oder Farbstoffe verwendet werden.
Für eine orale Anwendungsform werden die aktiven Verbindungen mit den dafür geeigneten Zusatzstoffen, wie Trägerstoffen, Stabilisatoren oder inerten Verdünnungsmittel vermischt und durch die üblichen Methoden in die geeigneten Darreichungsformen gebracht, wie Tabletten, Dragees, Steckkapseln, wäßrige, alkoholische oder ölige Lösungen. Als inerte Träger können z. B. Gummi arabicum, Magnesia, Magnesiumcarbonat, Kaliumphosphat, Milchzucker, Glucose oder Stärke, insbesondere Maisstärke, verwendet werden. Dabei kann die Zubereitung sowohl als Trocken- als auch als Feuchtgranulat erfolgen. Als ölige Trägerstoffe oder als Lösemittel kommen beispielsweise pflanzliche oder tierische Öle in Betracht, wie Sonnenblumenöl oder Lebertran.
Zur subkutanen oder intravenösen Applikation werden die aktiven Verbindungen, gewünschtenfalls mit den dafür üblichen Substanzen wie Lösungsvermittler, Emulgatoren oder weiteren Hilfsstoffen in Lösung, Suspension oder Emulsion gebracht. Als Lösungsmittel kommen z. B. in Frage: Wasser, physiologische Kochsalzlösung oder Alkohole, z. B. Ethanol, Propanol, Glycerin, daneben auch Zuckerlösungen wie Glucose- oder Mannitlösungen, oder auch eine Mischung aus den verschiedenen genannten Lösungsmitteln.
Als pharmazeutische Formulierung für die Verabreichung in Form von Aerosolen oder Sprays sind geeignet z. B. Lösungen, Suspensionen oder Emulsionen des Wirkstoffes der Formel I in einem pharmazeutisch unbedenklichen Lösungsmittels, wie insbesondere Ethanol oder Wasser, oder einem Gemisch solcher Lösungsmittel.
Die Formulierung kann nach Bedarf auch noch andere pharmazeutische Hilfsstoffe wie Tenside, Emulgatoren und Stabilisatoren sowie ein Treibgas enthalten. Eine solche Zubereitung enthält den Wirkstoff üblicherweise in einer Konzentration von etwa 0,1 bis 10, insbesondere von etwa 0,3 bis 3 Gew.-%.
Die Dosierung des zu verabreichenden Wirkstoffs der Formel I und die Häufigkeit der Verabreichung hängen von der Wirkstärke und Wirkdauer der verwendeten

Verbindungen ab; außerdem auch von Art und Stärke der zu behandelnden
Krankheit sowie von Geschlecht, Alter, Gewicht und individueller Ansprechbarkeit des zu behandelnden Säugers.
Im Durchschnitt beträgt die tägliche Dosis einer Verbindung der Formel I bei einem etwa 75 kg schweren Patienten mindestens 0,001 mg/kg, vorzugsweise 0,01 mg/kg, bis höchstens 10 mg/kg, vorzugsweise 1 mg/kg Körpergewicht. Bei akuten Ausbrüchen der Krankheit, etwa unmittelbar nach Erleiden eines Herzinfarkts, können auch noch höhere und vor allem häufigere Dosierungen notwendig sein, z. B. bis zu 4 Einzeldosen pro Tag. Insbesondere bei i.v. Anwendung, etwa bei einem Infarktpatienten auf der Intensivstation können bis zu 200 mg pro Tag notwendig werden.

Experimenteller Teil
Allgemeine Vorschriften zur Herstellung von Alkenylcarbonsäureguanididen (I)
Variante 1 A: aus Alkenylcarbonsäure-alkylestern (II, L = O-Alkyl)
5,0 eq. Kalium-tertiär-butylat werden in DMF vorgelegt, mit 6,0 eq. Guanidin- hydrochlorid versetzt und 30 min bei Raumtemperatur gerührt. Zu dieser Lösung werden 1 ,0 eq. des Zimtsäure-alkylesters gegeben und bei Raumtemperatur gerührt, bis mittels Dünnschichtchromatogramm oder LCMS keine Umsatzsteigerung mehr festgestellt werden kann. Zur Aufarbeitung wird das
Lösungsmittel unter vermindertem Druck abdestilliert. Der Rückstand wird in H2O aufgenommen und mit 1 N HCI tropfenweise versetzt, bis ein Niederschlag ausfällt. Dieser wird abgesaugt und mit EE gewaschen. Das so erhaltene HCI-Salz des Alkenylcarbonsäureguanidids kann durch Behandlung mit Basen in das freie Zimtsäureguanidin übergeführt werden. Hieraus lassen sich wiederum, durch
Behandeln mit wässrigen, methanolischen oder etherischen Lösungen organischer oder anorganischer Säuren andere pharmakologisch verträgliche Salze herstellen.
Variante 1 B: aus Alkenylcarbonsäuren (II, L = OH) 1 ,0 eq. des Zimtsäurederivats wird in DMF vorgelegt, 1 ,1 eq. Carbonyldiimidazol zugegeben und 3 h bei Raumtemperatur gerührt. Zu dieser Lösung gibt man eine, wie unter 1 A beschriebene, frisch hergestellte Lösung der Guanidinbase und rührt weiter bei Raumtemperatur, bis vollständiger Umsatz erreicht ist, bzw. keine Umsatzsteigerung mehr festgestellt werden kann. Die Aufarbeitung erfolgt analog der in 1 A beschriebenen Vorgehensweise.
Die im Folgenden angegebenen Retentionszeiten (Rt) beziehen sich auf LCMS-
Messungen mit den folgenden Parametern: stationäre Phase: Merck Purospher 3μ, 2 x 55 mm mobile Phase: 95% H2θ(0.05% TFA)→ 95% Acetonitril; 4 min; 95%
Acetonitril; 1.5 min - 5% Acetonitril; 1 min; 0.5 ml/min.
Beispiel 1 :
4-[2,6-Difluoro-4-(3-guanidino-2-methyl-3-oxo-propenyl)-phenoxy]-benzolsulfonamid;
HCI-Salz.
Zwischenprodukt 1 : 2-MethyI-3-(3,4,5-trifluoro-phenyl)-acrylsäure-ethylester.
1 ,0 eq. 3,4,5-Trifluorbenzaldehyd wird in DMF gelöst und mit 1 ,1 eq. 2-(Triphenyl - phosphanylidene)-propionsäure-ethylester bei Raumtemperatur gerührt, bis mittel Dünnschichtchromatographie vollständiger Umsatz festgestellt werden kann. Es wird vom Lösungsmittel befreit und der Rückstand in Diisopropylether verrührt. Der unlösliche Rückstand wird abfiltriert, das Filtrat i. Vak. eingeengt und an Kieselgel Chromatographien. (EE / Heptan 1 :1 ), wobei die Titelverbindung in Form eines farblosen Feststoffs erhalten wird. (Schmp.: 66 - 68 °C).
Zwischenprodukt 2: 3-[3,5-Difluoro-4-(4-sulfamoyl-phenoxy)-phenyl]-2-methyl-acrylsäure-ethylester.
1 ,5 eq. 4-Hydroxy-benzolsulfonamid werden in Dimethylacetamid mit der äquimolaren Menge NaH versetzt und 1 h bei Raumtemperatur deprotoniert. Eine Lösung von 1 ,0 eq. des Zwischenprodukts 1 in Dimethylacetamid wird zugetropft und das Reaktionsgemisch bei 150 °C gerührt, bis mittels DC oder LCMS kein Edukt mehr nachgewiesen werden kann. Zur Aufarbeitung wird das Lösungsmittel i. Vak. entfernt, der Rückstand in H2O aufgenommen und 2 x mit EE extrahiert. Die organischen Phasen werden mit MgSθ4 getrocknet und eingeengt.

Zwischenprodukt 2: 4-Amino-N-dimethylaminomethyIene-benzolsuIfonamid
wird in dem Fachmann bekannter Weise, ausgehend von 4-Amino-benzolsulfonamid und DMF-dimethylacetal, synthetisiert (s. z. Bsp.: J. Med. Chem., 1983, 26, 1174 - 1187). (MS-ES+: 228,0).
Zwischenprodukt 3:
3-{4-[4-(Dimethylaminomethylene-sulfamoyl)-phenylamino]-phenyl}-2-methyl- acrylsäure-ethylester.
Unter Argon legt man 1 ,4 eq. CS2CO3, 0,03 eq. Pd(OAc)2, sowie 0,045 eq. 2,2-Bis- (diphenylphosphino)-1 ,1'-binaphthalin in wasserfreiem Toluol vor. Zu dieser Lösung werden 1 ,0 eq. des Zwischenprodukts 1 , sowie 1 ,2 eq. des Zwischenprodukts 2 gegeben und bei 100 °C unter Schutzgasatmosphäre gerührt, bis vollständiger Umsatz festgestellt werden kann (DC oder LCMS). Zur Aufarbeitung wird mit Ether versetzt und der entstandene Niederschlag abfiltriert. Das Filtrat wird eingeengt und an Kieselgel gereinigt, wobei das gewünschte Produkt als farbloses Öl isoliert werden kann. (MS-ES+: 416,3).
Zwischenprodukt 4: 2-Methyl-3-[4-(4-sulfamoyl-phenylamino)-phenyl]-acrylsäure.
Das Zwischenprodukt 3 wird in 2 N NaOH 4 bis 5 h bei 40 bis 60 °C erhitzt. Anschließend wird i. Vak. eingeengt, der Rückstand in H2O aufgenommen und mit
2N HCl ein pH-Wert von 6 bis 7 eingestellt, wobei das Produkt in Form eines leicht gelben Feststoffs ausfällt. Absaugen und Trocknen über P2O5 liefert das gewünschte Zimtsäurederivat in einer Ausbeute von 90 %. (MS-ES+: 333,2).

Ausgehend von Zwischenprodukt 4 wird die Titelverbindung durch die in 1 B beschriebene Vorgehensweise hergestellt. Das Produkt wird nach HPLC als gelblicher kristalliner Feststoff in einer Ausbeute von 40 bis 50 % isoliert. (MS-ES+: 374,2; LCMS-Rt = 3,761 min).

Die Synthese der Titelverbindung wird nach analoger Vorschrift zu Zwischenprodukt 3, Beispiel 4 hergestellt, wobei 77 % des gewünschten Produktes isoliert werden können. (MS-ES+: 416,3).
Zwischenprodukt 4: 2-Methyl-3-[4-(3-sulfamoyl-phenylamino)-phenyl]-acrylsäure.
Die Schutzgruppenabspaltung und Esterspaltung des Zwischenproduktes 3 erfolgt nach der in Beispiel 4, Zwischenprodukt 4 angegebenen Vorschrift, wobei Zwischenprodukt 4 in 87 %-iger Ausbeute in Form eines farblosen Feststoffs isoliert werden kann. (MS-ES+: 333,1 ).

Zwischenprodukt 1 :
4-Hydroxy-benzolsulfonylchloride
wird nach literaturbekanntem Verfahren synthetisiert (R. W. Campbell, H. W. Hill, jr.; J. Org. Chem., 38, 1973, 1047.)
Zwischenprodukt 2: N-(2-Dimethylamino-ethyl)-4-hydroxy-benzolsulfonamid
1 ,0 eq. des Zwischenproduktes 1 werden in Dichlormethan vorgelegt und bei Raumtemperatur 2,2 eq. N,N-Dimethylamino-ethan-diamin, gelöst in Dichlormethan, zugetropft und bei Raumtemperatur gerührt. Nachdem vollständiger Umsatz festgestellt wurde, wird vom Lösungsmittel befreit und der Rückstand in H2O aufgenommen. Es wird 2 x mit EE extrahiert und die wässrige Phase i. Vak. eingeengt. Der Rückstand wird einmal mit Aceton gewaschen und aus heißem Isopropanol umkristallisiert, wobei das Sulfonamid als farbloser Feststoff isoliert wird. (Schmp.: 165 - 168 °C).
Zwischenprodukt 3:
2-Methyl-3-(3,4,5-trifluoro-phenyl)-acrylsäure-ethylester.

Zwischenprodukt 4:
3-{4-[4-(2-Dimethylamino-ethylsulfamoyl)-phenoxy]-3,5-difluoro-phenyl}-2-methyl- acrylsäure-ethylester.
1 ,5 eq. Zwischenprodukt 2 werden in Dimethylacetamid mit 1 ,75 eq. NaH versetzt und unter Feuchtigkeitausschluß 30 min bei 75 °C gerührt. Dazu wird eine Lösung von 1 ,0 eq. des Zwischenprodukts 3 in Dimethylacetamid gegeben und bei 150 °C gerührt, bis keine Umsatzsteigerung mehr festgestellt werden kann (DC, LCMS). Zur
Aufarbeitung wird vom Lösungsmittel befreit und der Rückstand in H2O aufgenommen. Die wässrige Phase wird 2 x mit EE extrahiert, mit MgS04 getrocknet und eingeengt. Das so erhaltene Rohprodukt wird durch Chromatographie an Kieselgel gereinigt.
4-[2,6-Difluoro-4-(3-guanidino-2-methyl-3-oxo-propenyl)-phenoxy]-N-(2- dimethylamino- ethyl)-benzolsulfonamid, HCI-Salz. Zwischenprodukt 4 wird nach der unter 1 A beschriebenen Methode in das entsprechende Zimtsäureguanidid übergeführt, das in Form eines gelblichen Feststoffs isoliert werden kann. (MS-ES+: 482,2; LCMS-Rt = 3,750 min).
Beispiel 7
N-(3-{3,5-Difluoro-4-[4-(4-methyl-piperazin-1-sulfonyl)-phenoxy]-phenyl}-2-methyl- acryloyl)-guanidin; Trifluoracetat
Zwischenprodukt 1 : 4-Hydroxy-benzolsulfonylchlorid
wird nach literaturbekanntem Verfahren synthetisiert (R. W. Campbell, H. W. Hill, jr.; J. Org. Chem., 38, 1973, 1047.)
Zwischenprodukt 2: 4-(4-Methyl-piperazin-1-suIfonyl)-phenol
1 ,0 eq. des Zwischenproduktes 1 werden in Dichlormethan vorgelegt und bei Raumtemperatur 2,0 eq. N-Methyl-piperazin, gelöst in Dichlormethan, zugetropft und bei Raumtemperatur gerührt. Nachdem vollständiger Umsatz festgestellt wurde, wird vom entstandenen Niederschlag abfiltriert. Das Filtrat wird mit MgS04 getrocknet und vom Lösungsmittel befreit. Das so erhaltene Rohprodukt kann ohne weitere Reinigung in die nächste Stufe eingesetzt werden. (Schmp.: 225 - 227 °C).
Zwischenprodukt 3: 2-Methyl-3-(3,4,5-trifluoro-phenyl)-acrylsäure-ethylester.

Zwischenprodukt 4:
3-{3,5-Difluoro-4-[4-(4-methyl-piperazin-1-sulfonyl)-phenoxy]-phenyl}-2-methyl- acrylsäure-ethylester.
1 ,0 eq. des Zwischenproduktes 2 werden in Dimethyacetamid gelöst und bei Raumtemperatur mit 1 ,2 eq. NaH versetzt. Nach 30 min bei 75 °C wird eine Lösung von 0,7 eq. des Zwischenproduktes 3 in Dimethylacetamid zugetropft und bei 150 °C gerührt. Nach vollständigem Umsatz wird vom Lösungsmittel befreit. Der Rückstand wird in H2O aufgenommen, auf einen pH von 7 bis 8 gestellt und 2 x mit EE extrahiert. Die organischen Phasen werden mit MgSθ4 getrocknet und eingeengt.
Der Rückstand wird an Kiesel gereinigt, wobei die Titelverbindung in einer Ausbeute von 50 bis 60 % isoliert wird.
N-(3-{3,5-Difluoro-4-[4-(4-methyl-piperazin-1 -sulfonyl)-phenoxy]-phenyl}-2-methyl- acryloyl)-guanidin; Trifluoracetat
Zwischenprodukt 4 wird nach der unter 1 A beschriebenen Methode in das entsprechende Zimtsäureguanidid übergeführt, das in Form eines gelblichen Feststoffs isoliert werden kann. Abschließende Reinigung über HPLC liefert das entsprechende TFA-Salz. (MS-ES+: 494,3; LCMS-Rt = 3,809 min).

Methylisocyanat in Aceton zugetropft und weiter zum Rückfluss erhitzt. Nachdem mittels LCMS keine Umsatzsteigerung mehr feststellbar ist, wird von unlöslichen
Bestandteilen abfiltriert und i. Vak. eingeengt. Der Rückstand wird in H2O aufgenommen und mit 2 N HCI auf pH 1 gestellt, wobei die Titelverbindung ausfällt. Absaugen und Trocknen i. Vak. liefert den gewünschten Sulfonylharnstoff in guter Ausbeute. (MS-ES+: 455,1).
4-[2,6-Difluoro-4-(3-guanidino-2-methyl-3-oxo-propenyl)-phenoxy]-benzolsulfon-N- (carbox-N'-methyl-arnid)-amid; HCI-Salz.

4-Acetylamino-benzolsulfonylchlorid wird in Dichlormethan vorgelegt und bei Raumtemperatur mit zwei Äquivalenten N,N-Dimethylethylendiamin versetzt und solange bei Raumtemperatur gerührt bis vollständiger Umsatz festgestellt werden
kann (LCMS). Zur Aufarbeitung wird das Lösungsmittel abdestilliert, der ölige Rückstand in Dichlormethan oder Essigester aufgenommen und mit NaHCθ3-
Lösung. gewaschen. Die Phasen werden getrennt und die wässrige noch einmal mit Dichlormethan oder Essigester extrahiert. Die organischen Phasen werden mit Na2S04 getrocknet und eingeengt. Das so erhaltene Rohprodukt wird ohne weitere
Reinigung weiter umgesetzt. (MS-CI+: 286,0; LCMS-Rt = 2,542 min)
Zwischenprodukt 3: 4-Amino-N-(2-dimethylamino-ethyl)-benzolsulfonamid
2,3 g des Zwischenproduktes 2 werden in 20 ml Methanol gelöst. Nach Zugabe von 5,0 eq. Natriummethanolat wird solange unter Rückfluss erhitzt bis die Reaktionskontrolle vollständigen Umsatz anzeigt. Um die Reaktion zu beschleunigen wurden weitere 10 bis 15 eq. Natrium-methanolat zugegeben. Nach beendeter Reaktion wird vom Lösungsmittel befreit und der Rückstand in H2O aufgenommen und mit konz. HCI auf pH 8 eingestellt. Die wässrige Phase wird dreimal mit Essigester extrahiert. Die organischen Phasen werden mit Na2Sθ4 getrocknet und eingeengt, wobei die Titelverbindung in Form eines gelblichen Feststoffs erhalten wird. (MS-ES+: 244,1 ; LCMS-Rt = 0,468 min)
Zwischenprodukt 4:
3-{4-[4-(2-Dimethylamino-ethylsuIfamoyl)-phenylamino]-phenyl}-2-methyl-acrylsäure- ethylester
Ausgehend von Zwischenprodukt 1 und Zwischenprodukt 3 verläuft die Synthese der Titelverbindung analog der Vorschrift zu Zwischenprodukt 3, Beispiel 4, wobei als Lösungsmittel ein Gemisch aus Toluol / DMF (1 :1) verwendet wird. Nach beendeter Reaktion wird vom Lösungsmittel befreit und der Rückstand in Dichlormethan aufgenommen. Es wird von den unlöslichen Bestandteilen abfiltriert, eingeengt und der Rückstand an Kieselgel gereinigt. (MS-ES+: 432,3; LCMS-Rt = 4,407 min)
N-(2-Dimethylamino-ethyl)-4-[4-(3-guanidino-2-methyl-3-oxo-propenyl)- phenylaminoj-benzolsulfonamid; HCI-Salz

Die Titelverbindung wird durch Palladium-katalysierte Umsetzung von Zwischenprodukt 3 mit Zwischenprodukt 1 nach der in Beispiel 10, Zwischenprodukt 4 beschriebenen Methode synthetisiert. Nach Reinigung an Kieselgel kann das gewünschte Produkt in 71 % Ausbeute isoliert werden. (MS-ES+: 458,4; LCMS-Rt = 4,547 min)

Zwischenprodukt 2: N-(4-Methylsulfamoyl-phenyl)-acetamid
10,5 g (45 mmol) 4-Acetylamino-benzolsulfonylchlorid werden in 100 ml 40 %iger Methylamin-Lösung 3 Stunden bei 50 °C gerührt. Danach wird die Lösung dreimal mit Essigester extrahiert. Die organischen Phasen werden mit Na2S04 getrocknet
und eingeengt. Der Rückstand wird mit Dichlormethan verrührt und die unlöslichen Bestandteile abfiltriert. Einengen des Filtrats liefert das gewünschte Sulfonamid in ausreichender Reinheit und nahezu quantitativer Ausbeute. (MS-CI+: 229,1 ; LCMS- Rt = 3,249 min)
Zwischenprodukt 3: 4-Amino-N-methyl-benzolsulfonamid
Ausgehend von Zwischenprodukt 2 erfolgt die Abspaltung der Acetylgruppe nach der in Beispiel 10, Zwischenprodukt s beschriebenen Methode. (MS-CI+: 187,1 ; LCMS-Rt = 1 ,358 min)
Zwischenprodukt 4: 2-Methyl-3-[4-(4-methylsulfamoyl-phenylamino)-phenyl]-acrylsäure-ethylester
Die Umsetzung von Zwischenprodukt 1 mit Zwischenprodukt 3 erfolgt nach der in Beispiel 10, Zwischenprodukt 4 beschriebenen Weise, wobei der gewünschte Ethylester in Form eines gelblichen Öls erhalten wird. (MS-CI+: 375,1 ; LCMS-Rt = 5,064 min)
4-[4-(3-Guanidino-2-methyl-3-oxo-propenyl)-phenylamino]-N-methyl-benzol- sulfonamid; HCI-Salz
Zwischenprodukt 4 in nach der unter Variante 1 A beschriebenen allgemeinen Methode mit Guanidin umgesetzt, wobei die Titelverbindung in Form eines gelben Feststoffs erhalten wird. Die Ausbeute beträgt 88 %. (MS-ES+: 388,1 ; 329,1 ; LCMS- Rt = 4,126 min).

Testbeschreibung
In diesem Test wurde die Erholung des intrazellulären pHs (pHj) nach einer
Ansäuerung ermittelt, die bei funktionsfähigem NHE auch unter bicarbonatfreien Bedingungen einsetzt. Dazu wurde der pHj mit dem pH-sensitiven
Fluoreszenzfarbstoff BCECF (Calbiochem, eingesetzt wird die Vorstufe BCECF-AM) bestimmt. Die Zellen wurden zunächst mit BCECF beladen. Die BCECF- Fluoreszenz wurde in einem "Ratio Fluorescence Spectrometer" (Photon Technology International, South Brunswick, N.J., USA) bei Anregungswellenlängen von 505 und 440 nm und einer Emissionswellenlänge von 535 nm bestimmt und mittels Kalibrierungskurven in den pHj umgerechnet. Die Zellen wurden bereits bei der BCECF-Beladung in NH4CI-Puffer (pH 7,4) inkubiert (NH4CI-Puffer: 115 mM
NaCI, 20 mM NH4CI, 5 mM KCI, 1 mM CaC^, 1 mM MgSθ4, 20 mM Hepes, 5 mM
Glucose, 1 mg/ml BSA; mit 1 M NaOH wird ein pH von 7,4 eingestellt). Die intrazelluläre Ansäuerung wurde durch Zugabe von 975 μl eines NH4CI-freien
Puffers (s. u.) zu 25 μl Aliquots der in NH4CI-Puffer inkubierten Zellen induziert. Die nachfolgende Geschwindigkeit der pH-Erholung wurde bei NHE1 2 Minuten, bei NHE2 5 Minuten und bei NHE3 3 Minuten registriert. Für die Berechnung der inhibitorischen Potenz der getesteten Substanzen wurden die Zellen zunächst in Puffern untersucht, bei denen eine vollständige bzw. überhaupt keine pH-Erholung
stattfand. Zur vollständigen pH-Erholung (100%) wurden die Zellen in Na+-haltigem Puffer inkubiert (133,8 mM NaCI, 4,7 mM KCI, 1 ,25 mM CaCl2, 1 ,25 mM MgCl2, 0,97 mM Na2HPθ4, 0,23 mM NaH2P04, 5 mM Hepes, 5 mM Glucose, mit 1 M NaOH wird ein pH von 7,0 eingestellt). Für die Bestimmung des 0%-Wertes wurden die Zellen in einem Na+-freien Puffer inkubiert (133,8 mM Cholinchlorid, 4,7 mM KCI, 1 ,25 mM CaCl2, 1 ,25 mM MgCI2, 0,97 mM K2HPO4, 0,23 mM KH2PO4, 5 mM
Hepes, 5 mM Glucose, mit 1 M NaOH wird ein pH von 7,0 eingestellt). Die zu testenden Substanzen wurden in dem Na+-haltigem Puffer angesetzt. Die Erholung des intrazellulären pHs bei jeder getesteten Konzentration einer Substanz wurde in Prozent der maximalen Erholung ausgedrückt. Aus den Prozentwerten der pH- Erholung wurde mittels des Programms Sigma-PIot der ICso-Wert der jeweiligen
Substanz für die einzelnen NHE-Subtypen berechnet.

R(35)
H oder Alkyl mit 1 , 2, 3, 4 oder 5 C-Atomen; oder
R(30) und R(31) gemeinsam 4 oder 5 Methylenguppen, wobei unabhängig voneinander eine oder mehrere CH2- Gruppen durch O, NR(35), C=O, S oder C=S ersetzt sein können; oder R(31 ) und R(35) gemeinsam 4 oder 5 Methylengruppen; R(32), R(33) und R(34) unabhängig voneinander H, F, Cl, Methyl, CF3; wobei die N-haltigen Heterocyclen N- oder C-verbrückt sind und nicht substituiert oder substituiert sind mit 1 - 3 Substituenten ausgewählt aus der Gruppe bestehend aus F, Cl, CF3, Methyl, Methoxy und
NR(36)R(37); R(36) und R(37) unabhängig voneinander H, CH3, C2H5 oder CF3; und die jeweils anderen Substituenten R(1), R(2), R(3), R(4) und R(5) unabhängig voneinander H, F, Cl, SO2NH2, SO2CH3, NR(24)R(25), CN, (C-|-
C-4)-Alkyl, CF3, C2F5, O-(Cι-C4)-Alkyl, OCF3, OC2F5, (C3-C6)- Cycloalkyl oder (C3-C6)- Cycloalkyl-(C-|-C4)- alkyl;
R(24) und R(25) unabhängig voneinander H, Alkyl mit 1 , 2, 3 oder 4 C-Atomen; R(6) und R(7) unabhängig voneinander Wasserstoff, F, Cl, Br, I, CN, CH3 , C2H5, CF3 oder
Cycloalkyl mit 3, 4, 5 oder 6 C-Atomen; sowie deren pharmazeutisch verträgliche Salze.
5. Verfahren zur Herstellung einer Verbindung I nach Anspruch 1 , dadurch gekennzeichnet daß man eine Verbindung der Formel II
worin R(1) bis R(7) wie in Anspruch 1 definiert sind und L für eine leicht nucleophil substituierbare leaving group steht, mit Guanidin umsetzt.
6. Verwendung einer Verbindung I nach Anspruch 1 zur Herstellung eines Medikaments zu Behandlung von Arrhythmien.
7. Methode zum Behandeln von Arrhythmien, dadurch gekennzeichnet, daß man eine wirksame Menge einer Verbindung I nach Anspruch 1 mit den üblichen Zusatzstoffen versetzt und in einer geeigneten Darreichungsform verabreicht.
8. Verwendung einer Verbindung I nach Anspruch 1 zur Herstellung eines Medikaments zur Behandlung oder Prophylaxe des Herzinfarkts.
9. Verwendung einer Verbindung I nach Anspruch 1 zur Herstellung eines Medikaments zur Behandlung oder Prophylaxe der Angina Pectoris.
10. Verwendung einer Verbindung I nach Anspruch 1 zur Herstellung eines Medikaments zur Behandlung oder Prophylaxe von ischämischen Zuständen des Herzens.
11. Verwendung einer Verbindung I nach Anspruch 1 zur Herstellung eines
Medikaments zur Behandlung oder Prophylaxe von ischämischen Zuständen des peripheren und zentralen Nervensystems und des Schlaganfalls.
12. Verwendung einer Verbindung I nach Anspruch 1 zur Herstellung eines Medikaments zur Behandlung oder Prophylaxe von ischämischen Zuständen peripherer Organe und Gliedmaßen.
13. Verwendung einer Verbindung I nach Anspruch 1 zur Herstellung eines Medikaments zur Behandlung von Schockzuständen.
14. Verwendung einer Verbindung I nach Anspruch 1 zur Herstellung eines Medikaments zum Einsatz bei chirurgischen Operationen und Organtransplantationen.
15. Verwendung einer Verbindung I nach Anspruch 1 zur Herstellung eines
Medikaments zur Konservierung und Lagerung von Transplantaten für chirurgische Maßnahmen.
16. Verwendung einer Verbindung I nach Anspruch I zur Herstellung eines Medikaments zur Behandlung von Krankheiten, bei denen die Zellproliferation eine primäre oder sekundäre Ursache darstellt.
17. Arzneimittel, gekennzeichnet durch einen wirksamen Gehalt einer Verbindung der Formel I nach einem oder mehreren der Ansprüche 1 bis 3.
