Verfahren zur Herstellung von am Stickstoff nicht substituierten Amidinen und deren Salzen mit anorganischen oder organischen Säuren, wobei entsprechende Carbonsäureamidoxime mit Zn in Gegenwart von Carbonsäuren umgesetzt werden.

Verfahren zur Herstellung von Amidinen
Beschreibung
Die vorliegende Erfindung beschreibt ein neues Verfahren zur Herstellung von am Stickstoff nicht substituierten Amidinen und deren Salzen mit anorganischen oder organischen Säuren, dadurch gekennzeichnet, daß man entsprechende Carbonsäureamidoxime mit Zn in Gegenwart von Carbonsäuren wie Ameisensäure, Essigsäure, Propionsäure umsetzt.
Zur Herstellung von Amidinen, die als bedeutendes Strukturelement in einer Reihe biologisch aktiver Moleküle wie Fibrinogen-Antagonisten (L. Alig et al . , J. Med. Chem.1992, 32, 4407), Thrombin-Inhibitoren (H. Vieweg et al.', Pharmazie 1982, 37, 178) oder auch Faktor Xa-Inhibitoren (T. Nagahara et al., J. Med. Chem. 1993, 36, 1811) vorkommen, existieren bereits eine Vielzahl von Synthesemethoden.
Eine der bewährtesten Methoden ist die zweistufige, als Pinner- Reaktion (A. Pinner, Chem. Ber. 1885, 18, 2845) bekannte, Transformation von Nitrilen über Iminoester in Amidine. In der Regel benutzt man große Überschüsse an Chlorwasserstoff in Alkoholen zur Bildung der Iminoester. Die Reaktionszeiten sind oft sehr lang (J. B. Medwid, J. Med. Chem. 1990, 33, 1237). Durch begleitende Abspaltung säurelabiler Schutzgruppen oder Umesterungs- reaktionen eignet sich die Methode nicht zur Herstellung komplexerer Moleküle. Die alternative basenkatalysierte Herstellung von Iminoestern (F.C. Schaefer et al . , J. Org. Chem., 26, 412, 1961) bleibt beschränkt auf elektronenarme Nitrile.
Eine Variante der Pinner-Reaktion ist die Umsetzung von Nitrilen über Thioamide und S-Alkylthioiminoester zu Amidine (H. Bredereck, Chem. Ber. 1957, 90, 1837). Diese Methode findet breite Anwendung, wenn es darum geht, die Amidinfunktionalität in komplexen Verbindungen wie den synthetischen Thrombin-Inhibitoren aufzubauen (B. Voigt, Pharmazie, 1983, 38, 835). Allerdings kann die Methode nur für die Herstellung kleiner Labormengen Ver- wendung finden. Die eingesetzten Chemikalien, Schwefelwasserstoff (übelriechend und hochtoxisch), Methyljodid oder Dirnethylsulfat (karzinogene Alkylierungsmittel) sowie das als Begleitprodukt gebildete Methylmercaptan (übelriechend und hochtoxisch) erfordern umfangreiche Sicherheitsmaßnahmen und Entsorgungsmöglichkeiten, die nur in Spezialanlagen zu gewährleisten sind. Zudem müssen die Produkte in aller Regel chromatographisch gereinigt werden.
Eine weitere Möglichkeit zur Herstellung von Amidinen besteht in der hydrogenolytischen Spaltung der N-O-Bindung von Carbonsäure- amidoximen (H.Jendralla et al . , Tetrahedron 1995, 51, 12047; B.D. Judkins, Synth. Commun. 1996, 26, 4351) mit Wasserstoff und 5 Palladium/Kohle-Katalysatoren. Für die Synthese von Amidinen, die im Molekül hydrierfähige Funktionalitäten wie CC-Doppelbindungen, benzylsubstituierte 0- bzw. N-Atome etc. enthalten können, ist diese Methode nicht einsetzbar.
0 In letzter Zeit wurden eine Reihe von synthetischen Arzneimitteln, wie z.B. Integrin-Rezeptor-Antagonisten, dort insbesondere Fibrinogen-Rezeptor-Antagonisten, oder Serinprotease- inhibitoren, dort insbesondere Thrombininhibitoren, Faktor Xa- Inhibitoren, Faktor Villa-Inhibitoren, Faktor IXa-Inhibitoren, 5 Urokinaseinhibitoren, Tryplaseinhibitoren, Complementinhibitoren (z.B. Cis und Cιr) beschrieben, die als Strukturelement substituierte Thienylamidine beinhalten: Faktor Xa-Inhibitoren: RPR 120844 (Thromb. Haemostasis 81, No 1, 157-160, (1999)); Urokinaseinhibitoren: (Fujisawa, WO 9811089); Thrombininhibitoren: 0 Eli Lilly WO95/23609 (Bsp. 65), BASF, WO 98/06741 (Bsp. 1-9 etc) , Boehringer Ingelheim DE 19754490 (Bsp. 98-103 etc)). Die un- substituierten 2- bzw. 3-Thiophencarboxamidine konnten nach der oben beschriebenen Pinner-Reaktion in guten Ausbeuten hergestellt werden (S. Gronowitz, Acta Chem. Scand B 31, 1977, 771).

Die durch die o.g. Patentanmeldungen offenbarten komplex substituierten Thienylamidine wurden im allgemeinen über die oben beschriebene Thioamid-Route in kleinen Mengen zugänglich gemacht. Ein befriedigender Zugang zur Herstellung größerer Mengen
30 dieser Verbindungen ist angesichts der erwähnten Nachteile dieser Methode bislang nicht beschrieben. Auch die katalytische Hydrierung von entsprechenden Carbonsäureamidoximen kommt wegen der allgemein bekannten Eigenschaft von schwefelhaltigen Verbindungen, Hydrierkatalysatoren zu vergiften, nicht in Betracht

Die einfache Addition von Ammoniak unter Druck in Gegenwart von Ammoniumsalzen (F.C. Schaefer et al . , J. Org. Chem., 27, 1255, 40 1962) führt bei Thiophencarbonitrilen nur zu schlechten Umsätzen.
Der Erfindung lag unter anderem die Aufgabe zugrunde, eine Methode zu finden, die auch die Herstellung von substituierten Amidinen in einfacher Art und Weise, mit guten Ausbeuten und im 45 technischen Maßstab ermöglicht. Besonders interessieren Aryl- und He arylamidinsynthesen, insbesondere solche von Thienylamidinen.
Gegenstand der Erfindung ist ein neues allgemein anwendbares Verfahren zur Herstellung von am Stickstoff nicht substituierten Amidinen und deren Salzen mit anorganischen oder organischen Säuren, dadurch gekennzeichnet, daß man ein entsprechendes Carbonsäureamidoxim mit Zn in Gegenwart von Carbonsäuren wie Ameisensäure, Essigsäure, Propionsäure, bevorzugt Essigsäure, umsetzt.
Nach dem neuen Verfahren lassen sich auch substituierte Thienyl- amidine, bevorzugt solche, in denen die Amidinfunktion entweder in der 2-Position oder 3-Position und ein weiterer Substituent in einer der drei anderen Positionen zum Schwefel steht, herstellen, wobei der Substituent seinerseits über ein C-, N- oder O-Atom an den Thiophenrest gebunden ist.
Insbesondere lassen sich nach dem erfindungsgemäßen Verfahren gemäß Anspruch 1 Amidine der Formel I

weitere Reste für R sind in den publizierten Integrinrezeptor- Antagonisten und Serinproteaseinhibitoren Anmeldungen angegeben, wobei R jeweils dem Molekülfragment entspricht, das an dem heterocyclischen alkylaminosubstituierten Amidinrest hängt,
herstellen.
Dazu setzt man Carbonsäureamidoxime der Formel II
in denen n und R die gleiche Bedeutung haben wie in Formel I, mit Zn in Gegenwart von Carbonsäuren wie Ameisensäure, Essigsäure, Propionsäure, bevorzugt Essigsäure, zu den entsprechenden Amidinen der Formel I um.
Verbindungen der Formel I können ein oder mehrere Asymmetriezentren enthalten. Nach dem erfindungsgemäßen Verfahren können sowohl reine Enantiomere bzw. Diastereomere als auch Mischung der Enantiomere bzw. Diastereomere hergestellt werden. Die
Konfiguration der Asymetriezentren bleibt bei der Reaktion nach dem erfindungsgemäßen Verfahren erhalten.
Das Verfahren zur Herstellung von Verbindungen der Formel I eröffnet somit einen einfachen Zugang zu Thrombin-Inhibitoren der WO 98/06741, dort repräsentiert durch die allgemeine Formel III
mit D in der Bedeutung
Die Strukturelemente A, B, E in den Bedeutungen der WO 98/06741 entsprechen besonders bevorzugten Verbindungen der Formel I, in der die Einheit A-B-E durch den Rest R repräsentiert wird.
Die Herstellung der zur Durchführung des erfindungsgemäßen Verfahrens benötigten Carbonsäureamidoxime erfolgt nach bekannten Methoden (F. Eloy et al . , Chem. Rev. 1962, 62, 155) durch Umsetzung der entsprechenden Nitrile mit Hydroxylamin oder vorzugs- weise seiner Salze, besonders bevorzugt Hydroxylamin-Hydrochlor- id. Dabei wird das entsprechende Hydroxylaminderivat bevorzugt im 1- bis 3fachen Überschuß eingesetzt. Die Reaktion wird in einem inerten Lösemittel durchgeführt. Bevorzugte Lösemittel sind Alkohole wie Methanol, Ethanol, n-Propanol, iso-Propanol oder n-Butanol, ggf im Gemisch mit Methylenchlorid, Toluol oder auch Wasser. Werden Salze des Hydroxylamins eingesetzt, so muß zur Freisetzung des Hydroxylamins mindestens ein Äquivalent einer stärkeren Base als Hydroxylamin selbst zugegeben werden. Die Base kann auch im Überschuß bezogen auf das Hydroxylaminsalz eingesetzt werden. Als Basen kommen sowohl anorganische Basen wie Natriumcarbonat (F. Tiemann, Chem. Ber. 17, 1884, 126), Metallalkoholate wie Natriummethylat oder Kalium-tert .-butylat (B.D. Judkins, Synth. Commun. 1996, 26, 4351), besonders bevorzugt aber auch tert. Amine wie Triethylamin (H. Jendralla et al . , Tetrahedron 1995, 51, 12047) oder Diisopropylethylamin in
Betracht. Die Reaktion erfolgt bereits bei Raumtemperatur, zur Beschleunigung kann aber auch bis zum Siedepunkt der Reaktionslösung erhitzt werden, in der Regel jedoch nicht höher als 80°C.
Die Isolation der Carbonsäureamidoxime ist unterschiedlich. Im Fall der Krictallisation aus der Reaktionslösung werden die Produkte durch Filtration isoliert. In anderen Fällen wird auf- konzentriert, mit einem nicht mit Wasser mischbaren Lösemittel aufgenommen, in dem die Produkte gut löslich sind. Diese Lösung wird mit Wasser, das ggf. mit Mineralsäure auf pH 2 angesäuert wird, gewaschen und aufkonzentriert . Die Rückstände können in einem geeigneten Lösemittel in üblicher Weise zur Kristallisation gebracht werden. Es ist aber in der Regel auch möglich die Produkte ohne weitere Reinigung weiter umzusetzen.
Die auf diese Weise hergestellten Verbindungen der Formel II stellen zum Teil neue Verbindungen dar und sind somit ebenfalls Gegenstand der vorliegenden Erfindung. Besonders bevorzugt sind die folgenden Verbindungen:

Die Synthese der Amidine nach dem erfindungsgemäßen Verfahren er- folgt vorzugsweise durch Überführung der Carbonsäureamidoxime in eine Essigsäurelösung, wo man sie durch Zugabe von metallischem Zink, das gewöhnlich als feines Pulver eingesetzt wird, zur Reaktion bringt. Die Reaktion wird üblicherweise in einer reinen Essigsäurelösung durchgeführt. Es ist jedoch auch möglich die Reaktion in z.B. Propionsäurelösung oder in einem Lösemittelgemisch durchzuführen. In Frage kommen z.B. Alkohole wie Methanol, Ethanol, n-Propanol, iso-Propanol und n-Butanol. Das Zink
wird im Überschuß bis zu 10 Äquivalenten eingesetzt. Die Reaktion kann in einem Temperaturbereich von -10°C bis 70°C durchgeführt werden, bevorzugt ist ein Temperaturbereich zwischen 20 und 50°C. Der Umsatz kann mittels Dünnschichtchromatographie kontrolliert werden. Üblich ist eine Reaktionszeit von 2 bis 5 Stunden. Es ist aber auch möglich bei 20°C über längere Zeit nachzurühren.
Die Aufarbeitung ist einfach. Es wird von überschüssigem Zink und den entstandenen Zinksalzen abfiltriert und aufkonzentriert . Die Reaktionsprodukte können entweder auf dieser Stufe isoliert und charakterisiert werden oder unmittelbar weiter umgesetzt werden. Insbesondere die Abspaltung von etwaigen Schutzgruppen wie tert.- Butylester oder N-tert. -Butoxycarbonylgruppen läßt sich vorteilhaft nach Abtrennung der festen Zn-Rückstände ohne Zwischen- isolierung durchführen.
Reduktionen von Carbonsäureamidoximen mit Zn zu Amidinen sind bislang nicht beschrieben. Gegenüber der bekannten Reduktion von Carbaldoximen mit Zn zu Aminomethylenverbindungen (G.D. Pandey et al. Indian J. Chem. Sect. B 1980, 19, 160; E. Testa et al . Chimia 1957, 11, 310; S. Negi et al. Synthesis, 1996, 991) ist es überraschend, daß nach dem erfindungsgemäßen Verfahren die Spaltung einer N,0-Bindung ohne Reduktion einer im selben Strukturelement vorliegenden C,N-Doppelbindung erreicht werden kann.
Die folgenden Beispiele dienen der Illustration des erfindungsgemäßen Verf hrens .
Beispiel 1
Synthese von 2-N-Boc-aminomethyl- (4-amidino) -thiophen
a) 2-N-Boc-aminomethyl- ( 4-amidoximo ) -thiophen
70 g (294 mmol) 2-N-Boc-aminomethyl- (4-cyano) -thiophen wurden zusammen mit 51,1 g ((735 mmol) Hydroxylammoniumchlorid in 700 ml Ethanol vorgelegt. Anschließend wurden man zwischen 20°C und 30°C 113,8 g (882 mmol) Diisopropylethylamin zugegeben und über 12 h bei 20°C bis 30°C nachgerührt. Zur Aufarbeitung wurde das Ethanol im Vakuum abdestilliert. Der Rückstand wurde in 1000 ml Methylenchlorid (CH2C12) aufgenommen. Die organische Phase wurde einmal mit 500 ml und ein zweites Mal mit 250 ml Wasser gewaschen. Nach Auf- konzentrieren der organischen Phase wurde das Produkt durch Zugabe von Methyl-tert . -butylether (MTBE) ausgefällt. Auswaage: 78 g
"C-NMR (DMSO; ppm) : 148,3 (Amidoxim) CnHι7N303S (271,34); berechnet: (M+H)+ = 272; gefunden: (M+H)+ = 272
b) 2-N-Boc-aminomethyl- (4-amidino) -thiophen
5 g (18,4 mmol) des nach a) hergestellten Carbonsäureamido- xims wurden bei 20°C in einem Gemisch aus 25 ml Essigsäure und 25 ml Methanol gelöst. Anschließend wurde unter ansteigen der Innentemperatur auf 30-35°C portionsweise 2,4 g (36,9 mmol) Zinkpulver zugegeben. Nach 12 h Nachrührzeit bei 25 bis 40°C war nach DC (CH2C12 /Methanol 9:1) kein Ausgangs- produkt mehr zu erkennen. Nach Abfiltrieren der Feststoffe wurde der Filterrückstand mit Methanol nachgewaschen Die gesammelten Filtrate wurden bei ca. 50°C im Vakuum auf- konzentriert . Der Rückstand wurde in CH2C1 aufgelöst. Die der CHC12-Phase wurde mit Wasser, das mit Natronlauge auf pH 12 eingestellt wurde, gewaschen. Die Wasserphase wurde nach der Phasentrennung mit CH2C12 nachextrahiert. Die vereinigten CH2C12 -Phasen wurden neutral gewaschen auf- konzentriert . Auswaage: 2,4 g erstarrter Schaum I3c-NMR (DMSO; ppm): 158,8 (verbreitert, Amidin)

Beispiel 2
Synthese von N-Boc-3 , 4-Dehydroprolyl-2- ( 4-amidino) -thienylmethylamid
a) N-Boc-3, 4-Dehydroprolyl-2- (4-amidoximo) -thienylmethylamid
50 g (177 mmol) N-Boc-3 , 4-Dehydroprolyl-2- (4-cyano) -thienylmethylamid wurden zusammen mit 30,8 g ((442,5 mmol) Hydroxyl- ammoniumchlorid in 590 ml Ethanol vorgelegt. Anschließend wurden zwischen 20°C und 30°C 113,8 g (882 mmol) Diisopropyl- ethylamin zugegeben und über 12 h bei 20°C bis 30°C nachgerührt. Danach war der Umsatz lt. DC (CH2C1 /Methanol 9:1) vollständig. Zur Aufarbeitung wurde das Ethanol im Vakuum größtenteils abdestilliert. Der Rückstand wurde in 120 ml CH2C12 aufgenommen. Die organische Phase wurde mit 180 ml Wasser gewaschen. Die Wasserphase wurde einmal mit 60 ml CHC12 nachextrahiert. Die vereinigten CH2C1 -Phasen wurden nochmals mit 30 ml Wasser gewaschen, filtriert und eingeengt. Auswaage: 66,3 g erstarrter Schaum. 13C-NMR (DMSO; ppm): 147,9 (Amidoxim)

b) N-Boc-3, 4-Dehydroprolyl-2- (4-amidino) -thienylmethylamid
41,4 g (113 mmol) des nach a) hergestellten Carbonsäure- amidoxims wurden bei 40°C in 400 ml Essigsäure gelöst. Anschließend wurde bis zu einer Innentemperatur von max. 50°C portionsweise 41,1 g (630 mmol) Zinkpulver zugegeben. Nach 2,5 h Nachrührzeit bei 25 bis 40°C war nach DC
(CHC1 /Methanol 9:1) kein Ausgangsprodukt mehr zu erkennen. Nach Abfiltrieren der Feststoffe wurde der Filterrückstand mit Essigsäure nachgewaschen Die gesammelten Filtrate wurden bei ca. 50°C im Vakuum aufkonzentriert und zwischen 180 ml MTBE und 370 ml Wasser verteilt. Die Wasserphase wurde mit
Natronlauge auf pH 7 gestellt. Nach Abtrennung der MTBE-Phase wurden 180 ml CHC12 zugegeben und die Wasserphase mit Natronlauge auf pH 12 gestellt. Nach Abtrennung der CH2C12-Phase wurde noch zweimal mit 150 ml CHC1 nachextrahiert. Die ver- einigten CH2C1 -Phasen wurden einmal mit Wasser nachgewaschen und aufkonzentriert. Auswaage: 34,6 g erstarrter Schaum. 13C-NMR (DMSO; ppm): 159,9 (Amidin) Cι6H22N4θ3S (350,44); berechnet: (M+H)+ = 351; gefunden: (M+H)+ = 351
Beispiel 3
Synthese von N- (t-Butoxycarbonyl-methylen)-N-Boc-D-Chg-3, 4-
Dehydroprolyl-2- (4-amidino) -thienylmethylamid
a) N- (t-Butoxycarbonyl-methylen) -N-Boc-D-Chg-3 , 4-Dehydroprolyl- 2- (4-amidoximo) -thienylmethylamid
365,8 g (0,623 mol) N- ( t-Butoxycarbonyl-methylen) -N-Boc-D- Chg-3, 4-Dehydroprolyl-2- (4-cyano) -thienylmethylamid wurden zusammen mit 108,3 g ((1,559 mol) Hydroxylammoniumchlorid in 1,83 1 Ethanol vorgelegt. Anschließend wurden zwischen 20°C und 30°C 241,3 g (1,870 mol) Diisopropylethylamin zugegeben und über 12 h bei 20°C bis 30°C nachgerührt. Danach war der Umsatz lt. DC (CH2C12 /Methanol 9:1) vollständig. Zur Aufarbeitung wurde das Ethanol im Vakuum größtenteils abdestilliert. Der Rückstand wurde in 730 ml MTBE aufgenommen. Die MTBE-Phase wurde einmal mit 1,1 1 Wasser gewaschen, die Wasserphase einmal mit 370 ml MTBE reextrahiert. Die vereinigten MTBE-Phasen wurden nochmals mit 180 ml Wasser ge- waschen und danach bei 0,5 bar durch azeotrope Destillation getrocknet und auf 730 ml Restvolumen aufkonzentrier . Das Produkt wird durch Eintropfen der MTBE-Lösung in 2,2 1 Petrolether (60/40) gefällt und im Trockenschrank bei 50°C 12 h getrocknet. Auswaage: 371,5 g leicht gelbes bis farbloses Pulver 13C-NMR (DMSO; ppm): 147,9 (Amidoxim)
C3oH45N507S (619,79); berechnet: (M+H)+ = 620; gefunden: (M+H)+ = 620
b) N- (t-Butoxycarbonyl-methylen)-N-Boc-D-Chg-3 , 4-Dehydroprolyl- 2- (4-amidino) -thienylmethylamid, Essigsäuresalz
5 g (8,1 mmol) des nach a) hergestellten Carbonsäureamidoxims wurden bei 30°C in 50 ml Essigsäure gelöst. Anschließend wurde bei einer Innentemperatur von 50 bis 55°C portionsweise 2,9 g (44,4 mmol) Zinkpulver zugegeben. Nach einer Nachrührzeit von 2 h bei ca. 50°C war nach DC (CH2C12/Methanol 9:1) kein Ausgangsprodukt mehr zu erkennen. Nach Abfiltrieren der Feststoffe wurde der Filterrückstand mit Essigsäure nach- gewaschen. Die gesammelten Filtrate wurden bei ca. 50°C im Vakuum aufkonzentriert .
Auswaage: 4,3 g leicht gelbes bis farbloses Pulver 13C-NMR (DMSO; ppm): 159 (verbreitert, Amidin) , 1H-NMR (DMSO; ppm): 1,87 (CH3C02-) C30H45N5O6S (603,79); berechnet: (M+H)+ = 604; gefunden: (M+H) + = 604
N- ( -Butoxycarbony1-methylen) -N-Boc-D-Chg-3 , 4-Dehydroprolyl- 2- (4-amidino) -thienylmethylamid, Base
62 g (100 mmol) des nach a) hergestellten Carbonsäure- amidoxims wurden bei 35°C in 620 ml Essigsäure gelöst. Anschließend wurde bei einer Innentemperatur von 35 bis 40°C portionsweise 36 g (550 mmol) Zinkpulver zugegeben. Nach einer Nachrührzeit von 4 h bei ca. 40°C war nach DC (CHCl2/Methanol 9:1) kein Ausgangsprodukt mehr zu erkennen. Nach Abfiltrieren der Feststoffe wurde der Filterrückstand mit Essigsäure nachgewaschen. Die gesammelten Filtrate wurden bei ca. 50°C im Vakuum aufkonzentriert und zwischen 200 ml MTBE und 410 ml Wasser verteilt. Die Wasserphase wurde mit Natronlauge auf pH 7 gestellt. Nach Abtrennung der Wasser- phase wurde die MTBE-Phase zweimal mit 200 ml Wasser nachextrahiert und verworfen. Die vereinigten Wasserphasen wurden anschließend mit Natronlauge auf pH 12 gestellt und zweimal mit mit je 330 ml MTBE extrahiert. Die vereinigten MTBE- Phasen wurden bei ca. 0,7 bar durch azeotrope Destillation getrocknet und auf ca. 270 ml Restvolumen aufkonzentriert . Das Produkt wird durch Eintropfen der MTBE-Lösung in 740 ml n-Heptan gefällt und im Trockenschrank bei 50°C 12 h getrocknet . Auswaage: 57,2 g leicht gelbes bis farbloses Pulver C3oH45N506S (603,79); berechnet: (M+H)+ = 604; gefunden: (M+H)+ = 604
Claims
1. Verfahren zur Herstellung von am Stickstoff nicht substi- tuierten Amidinen und deren Salzen mit anorganischen oder organischen Säuren, wobei entsprechende Carbonsäureamidoxime mit Zn in Gegenwart von Carbonsäuren umgesetzt werden.
2. Verfahren gemäß Anspruch 1 zur Herstellung von substituierten Aryl- oder Heteroarylamidinen, insbesondere von Thienyl- amidinen.
3. Verfahren gemäß Anspruch 1 zur Herstellung von substituierten Thienylamidinen, wobei die Thienylamidingruppe ein Struktur- fragment eines Arzneimittels ist.
4. Verfahren gemäß Anspruch 1 zur Herstellung von substituierten Thienylamidinen der Formel I
worin n = 1-3 sein und
R die folgenden Bedeutungen haben kann
Cι-C6-Alkyl-CO,
Cι-C6-Alkoxy-CO,
Benzyloxy,
Aryl oder Heteroaryl, wobei diese Aromaten gegebenenfalls weiter ein oder mehrfach substituiert sein können,
ein oc-Aminosäurefragment in der R- oder S-Konfiguration, deren N-Atom in der üblichen Weise geschützt oder ungeschützt vorliegt, ein, gegebenenfalls am N-Terminus ein oder mehrfach substituiertes Dipeptidfragment, bestehend aus den natürlichen Aminosäuren, den entsprechenden D-Aminosäuren oder den natürlichen Aminosäuren sehr ähnlichen Aminosäuren in der L- oder D-Form,
herstellen,
dadurch gekennzeichnet, daß man Carbonsäureamidoxime der Formel II
in denen n und R die gleiche Bedeutung haben wie in Formel I, mit Zn in Gegenwart einer Carbonsäure umsetzt.
5. Verfahren nach einem der Ansprüche 1 bis 4, wobei die Carbonsäure Essigsäure ist.
6. Verbindungen der Formel II gemäß Anspruch 4.
