Abstract

Un aspecto del presente invento está relacionado con una proteína de ?-mananasa aislada la cual tiene una secuencia de aminoácidos que es 90% similar a la secuencia de aminoácidos de la SEQ ID NO:1. El presente invento también se relaciona con un polinucleótido aislado que codifica la proteína de ?-mananasa, y un sistema de expresión aislado y una célula hospedera que contiene el polinucleótido. Otro aspecto del presente invento está relacionado con un método de producir de manera recombinante una proteína de ?-mananasa. Este método comprende proporcionar una célula hospedera que contenga el polinucleótido del presente invento y cultivar la célula hospedera bajo condiciones que sean efectivas para que la célula hospedera exprese la proteína de ?-mananasa. La proteína de ?-mananasa es recuperada. Un aspecto adicional del presente invento está relacionado con un método de degradación de mananos y polisacáridos en material vegetal. Este método comprende proporcionar material vegetal y contactar el material vegetal con la proteína de ?-mananasa del presente invento bajo condiciones efectivas para degradar los mananos y polisacáridos en el material vegetal. El presente invento se relaciona con una secuencia de polinucleótidos aislados la cual codifica una enzima de mananasa involucrada en la hidrólisis de polisacáridos de manano, incluyendo moléculas de manano ramificadas y no ramificadas enlazadas entre sí vía un enlace beta 1,4 glicosilo. El polinucleótido es aislado a partir de un genoma de insecto (broca del café, Hypothenemus hampei).
