Abstract
В традиционных подходах к селекции растений химический мутагенез можно использовать для случайного введения нуклеотидных замен в геном растения, т.е. без возможности контролировать сайты нуклеотидных изменений. Когда дело касается поиска заданной нуклеотидной замены, статистическая вероятность является крайне низкой из-за сложности генома. В настоящем изобретении, тем не менее, продемонстрировано, как разработать новое альтернативное применение цифровой полимеразной цепной реакции (dPCR), предпочтительно капельной dPCR (ddPCR), для обнаружения конкретных нуклеотидных замен в мутантных генах. Вся платформа включает способ скрининга с библиотекой подвергнутых мутагенезу организмов, системы на основе цифровой ПЦР и установку для размножения и анализа выявленных мутантных организмов.
