Abstract

Изобретение предлагает способ анализа, который позволяет идентифицировать элемент ДНК с повышенной чувствительностью и определить уровень транскрипции элемента ДНК. Настоящее изобретение относится к способу декодирования последовательности оснований нуклеиновой кислоты, соответствующей концевому участку РНК, включающему стадию получения-декодирования способа получения с использованием матричной РНК по меньшей мере одной нуклеиновой кислоты, выбранной из группы, состоящей из нуклеиновой кислоты РНК 5'-концевого участка матричной РНК или комплементарной нуклеиновой кислоты ДНК, соответствующей 5'-концевому участку матричной РНК; нуклеиновой кислоты РНК 3'-концевого участка матричной РНК или комплементарной нуклеиновой кислоты ДНК, соответствующей 3'-концевому участку матричной РНК; нуклеиновой кислоты РНК частичного участка антисмысловой цепи или смысловой цепи полноразмерной матричной РНК; и их сочетаний, или нуклеиновой кислоты, последовательность которой содержит две или более связанных последовательностей, где матричная РНК, используемая на стадии получения-декодирования, представляет собой растущую цепь РНК. Элемент ДНК можно идентифицировать и проанализировать с повышенной точностью и повышенной чувствительностью путем определения информации о декодированной последовательности нуклеиновой кислоты и картирования элемента ДНК в геноме.
