Abstract
Представлен способ in vivo истощения аденозин 5'-монофосфата (АМФ) в клетках млекопитающих, применяемый при лечении определенных форм рака. В соответствии со способом популяцию клеток получают от хозяина и анализируют на утрату активности метилтиоаденозинфосфорилазы (МТАзы). МТАза расщепляет метилтиоаденозин до аденина для эндогенного использования его включения во внутриклеточный пул АМФ. Предпочтительным способом анализа утраты активности МТАзы является способ гибридизации с целью детекции гомозиготной утраты кодирующего гена с помощью терапевтически эффективного количества агента, который ингибирует активность аденилсукцинатсинтетазы, конвертирующей инозин 5'-монофосфат в АМФ, истощая таким образом субстраты опухолевых клеток для образования АМФ de novo. L-аланозин является предпочтительным ASS-ингибирующим агентом для использования в способе, соответствующем изобретению. На фиг. 1 представлена нуклеотидная последовательность геномной МТАзы (SEQ ID No 1), в которой подчеркнуты экзоны.Международная заявка была опубликована вместе с отчетом о международном поиске.
