Claims (10)
- PATENTKRAY1. Fremgangsmåde til detektering af et enkeltnukleotid-mål, kendetegnet ved, at den omfatter trinnet at fastgøre målet til en probe ved anvendelse af en polymerase og en ligase med henblik på dannelse af en anvendt probe, som helt er dobbelt-strenget, hvorved proben omfatter et ind-fangningssted, som består af et enkeltnukleotid, der er komplementært i forhold til målet, hvis ender er fastgjort til to forskellige dobbelt-strengede oligo-nukleotid-områder, hvorved i det mindste et af oligonukleotid-områderne omfatter detekterbare elementer med karakteristisk detekterings-egenskab, og hvorved de detekterbare elementer er arrangeret således på oligonukleotid-området, at detekterings-egenskaben i mindre grad er detekterbar, end når samme antal detekterbare elementer er bundet til et tilsvarende antal enkeltnu-kleotider.
- 2. Fremgangsmåde ifølge krav 1, kendetegnet ved, at den omfatter det ekstra trin at behandle den anvendte probe med et restriktions-enzym og en exonuklease for at frigive det/de detekterbare element/elementer i en form, i hvilken de kan detekteres.
- 3. Fremgangsmåde ifølge krav 2, kendetegnet ved, at den desuden omfatter trinnet at observere den detekterbare egenskab, som udvises af det/de frigivne detekterbare element(er).
- 4. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 3, kendetegnet ved, at det/de detekterbare element/elementer er fluoro-phorer.
- 5. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, kendetegnet ved, at den anvendte probe omfatter et restriktionsen-zym-genkendelsessted, der er blevet dannet ved hæftning af målet til indfang-ningsstedet.
- 6. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, kendetegnet ved, at målet bringes i kontakt med en blanding af fire forskellige biologiske prober, der hver omfatter et enkelt-strenget nukleotid-område, som omfatter et forskelligt nukleotid, der er udvalgt blandt (1) guanin, cytosin, adenin og thymin eller (2) guanin, cytosin, adenin og uracil.
- 7. Fremgangsmåde ifølge krav 6, kendetegnet ved, at hver af de fire prober omfatter et forskelligt, de-tekterbart element.
- 8. Fremgangsmåde ifølge krav 7, kendetegnet ved, at de forskellige, detekterbare elementer er fluoro-phorer.
- 9. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, kendetegnet ved, at målet omfatter eet blandt en strøm af enkelt-nukleotider, svarende til sekvensen af nukleotider i et DNA- eller RNA-prøveemne.
- 10. Anvendelse af en fremgangsmåde ifølge et hvilket som helst af de foregående krav med henblik på sekvensering af DNA eller RNA.
