Claims (10)
- POLYPEPTIDER MED XYLANASEAKTIVITET1. Polypeptid, der har xylanaseaktivitet og omfatter en aminosyresekvens, hvilken aminosyresekvens er mindst 75 % identisk med aminosyresekvensen ifølge SEQ ID NO: 1, og hvilket polypeptid har i) én eller to aminosyremodifikationer i en position, der er udvalgt fra: 12 og 13; og ii) én eller flere yderligere aminosyresubstitutioner i en position, der er udvalgt fra: 15, 34, 54, 77, 81, 82, 99, 104,110, 113, 114,118, 122, 141, 154, 159, 162,164, 166, 175 og 179, hvor positionerne er bestemt som positionen, der svarer til positionen af B. subtilis-xylanasesekvensen ifølge SEQ ID NO: 1 ved alignment; hvor aminosyremodifikationen i position 12 og 13 er en aminosyresubstitution med tyrosin.
- 2. Polypeptid ifølge krav 1, der omfatter én eller flere aminosyresubstitutioner, der er udvalgt fra gruppen bestående af: 13Y, 15Y, 34K, 77V, 77M, 77Y, 77L, 77S, 811, 821, 99Y, 104W, 110A, 113D, 113A, 114F, 114D, 114Y, 118V, 122F, 122D, 154R, 159D, 162E, 162D, 164F, 166F, 175L, 175K, 175E, 175Y og 179Y, hvilken eller hvilke positioner bestemmes som den tilsvarende position af B. subtilis -aminosyresekvensen ifølge SEQ ID NO: 1.
- 3. Polypeptid ifølge et hvilket som helst af kravene 1-2, hvor polypeptidet er mindst 80, 85, 90, 95 eller 98 % identisk med den sekvens, med hvilken den har den højeste procentvise identitet, der er udvalgt fra SEQ ID NO: 1.
- 4. Polypeptid ifølge et hvilket som helst af kravene 1-3, der har en β-jelly roll-fold.
- 5. Polypeptid ifølge et hvilket som helst af kravene 1-4, hvilket polypeptid har et samlet antal aminosyrer på mindre end 250, såsom mindre end 240, såsom mindre end 230, såsom mindre end 220, såsom mindre end 210, såsom mindre end 200 aminosyrer, såsom i intervallet på mellem 160 til 240, såsom i intervallet på mellem 160 til 220 aminosyrer.
- 6. Polypeptid ifølge et hvilket som helst af kravene 1-5, hvilket polypeptid omfatter én eller flere aminosyresubstitutioner, der er udvalgt fra gruppen bestående af: 13Y, 110A, 122D, 113A, 13Y, 113D, 175L, 122F, 34K, 99Y, 104W, 154R, 159D, 175K, 811, 166F, 162E, 162D, 164F, 114D, 114Y, 114F, 118V, 175K, 77L, 77M, 77S, 77V og 77Y, hvilken eller hvilke positioner bestemmes som den tilsvarende af B. -aminosyresekvensen ifølge SEQ ID NO: 1.
- 7. Fremgangsmåde til fremstilling af et polypeptid ifølge et hvilket som helst af kravene 1-6, hvilken fremgangsmåde omfatter ekspression af en nukleotidsekvens, der koder for polypeptidet, og eventuelt isolering af og/eller oprensning af polypeptidet efter ekspression.
- 8. Nukleotidsekvens, der koder for et polypeptid ifølge et hvilket som helst af kravene 1-6.
- 9. Sammensætning, der omfatter polypeptidet ifølge et hvilket som helst af kravene 1-6, eller polypeptid fremstillet ifølge krav 7 eller nukleotidsekvens ifølge krav 8 sammenblandet med en ikke-toksisk bestanddel.
