Claims (8)
1. Fremgangsmåde til fremstilling af en evolveret stamme af mikroorganismer til fremstilling af 1,2-propandiol fra en kulstofkilde, hvilken fremgangsmåde omfatter: - dyrkning af en initial stamme under selektionstryk i et passende vækstmedium, hvor den initiale bakteriestamme omfatter en attenuation af ekspressionen af fp/A-genet og en attenuation af ekspressionen af mindst ét i omdannelsen af methylglyoxal til lactat involveret gen for at fremme evolutionen i den initiale stamme, - derefter udvælgelse og isolering af den evolverede stamme med forhøjet 1,2-propandiol-produktionshastighed, - derefter rekonstruering af et funktionelt ip/A-gen i den evolverede stamme.
2. Fremgangsmåde ifølge krav 1, hvorved det i omdannelsen af methylglyoxal til lactat involverede gen er udvalgt fra gruppen bestående af: gloA, aldA og aldB og kombinationer deraf.
3. Fremgangsmåde ifølge kravene 1 til 2, hvorved den initiale stamme endvidere omfatter attenuation af ekspressionen af mindst ét af de fra gruppen bestående af IdhA, pfIA, pilB, adhE, eddog eda udvalgte gener.
4. Fremgangsmåde ifølge kravene 1 til 3, hvorved den initiale stamme endvidere omfatter attenuation af mindst ét gen udvalgt fra gruppen bestående af arcA og ndh.
5. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 4, hvorved den evolverede stamme er udvalgt og isoleret på basis af dens 1,2-propandiol-produktionshastighed, forhøjet med mindst 20 % i sammenligning med produktionshastigheden for den initiale stamme.
6. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 5, hvorved den initiale stamme er udvalgt fra gruppen bestående af bakterier, gær og svampe.
7. Fremgangsmåde ifølge krav 6, hvorved den initiale stamme er udvalgt fra gruppen bestående af Enterobacteriaceae, Bacillaceae, Clostridiaceae, Streptomycetaceae og Corynebacteriaceae.
8. Fremgangsmåde ifølge krav 7, hvorved den initiale stamme er enten Escherichia coli eller Clostridium acetobutylicum.
