Description

en ligand, såsom en kompetitiv inhibitor for alkalisk phosphatase, f.eks. para-aminobenzyl-phosphonsyre (p-ABPA). Dextran-kuglerne ville først blive behandlet med (p-ABPA), og alkalisk phosphatase ville derefter blive bragt i kontakt med de således behandlede dextrankugler. Den kompetitive inhibitor p-ABPA virker som en ligand for fæstnelse af alkalisk phosphatase til dextrankuglerne, medens den på samme tid optager og beskytter det aktive sted af alkalisk phosphatase. Derefter derivatiseres eller modificeres den således fæstnede alkaliske phosphatase, f.eks. ved biotinylering. Efter biotinylering af den alkaliske phosphatase elueres den resulterende biotinylerede alkaliske phosphatase eller fjernes fra fæstnelse til det p-ABPA-behandlede faste substrat eller dextrankugler, hvortil det var bundet, til dannelse af en biotinyleret alkalisk phosphatase med det aktive eller funktionelle sted af denne intakt. Komplekset kunne, medens det stadig var bundet til dextrankuglerne, behandles yderligere med en anden forbindelse eller komponent, som også om ønsket kunne tilvejebringe et aktive eller funktionelt sted, f.eks. ved kontakt med avidin, der ville binde til den modificerende eller derivatiserende del, dvs. biotin, af den biotinylerede alkaliske phosphatase, og det således dannede avidin-biotiny-lerede, alkaliske phosphatasekompleks kunne derefter elueres. Dette kompleks, såsom et kompleks indbefattende avidin-bioti-nyleret alkalisk phosphatase kunne anvendes som en detektor i et detektionssystem til identifikationen og/eller lokaliseringen af en anden forbindelse, såsom en biotinyleret forbindelse, f.eks. et biotinyleret nucleotid eller biotinyleret DNA.
