Opfindelsen angår en særlig fremgangsmåde til fremstilling af carboxylsyreamider og peptider, som er ejendommelig ved, at man omsætter forbindelser, der indeholderen fri aminogruppe, med forbindelser indeholdende en 5 fri carboxylgruppe, i nærværelse af anhydrider af .alkan-phosphonsyrerne.
Fremgangsmåden ifølge opfindelsen egner sig både til fremstilling af carboxylsyreamider og især også til fremstilling af peptider.
10 Som carboxylsyreamider kommer først og fremmest amider af aromatiske, aliphatiske og heterocycliske carboxylsyrer i betragtning, f.eks. eventuelt med alkyl-, alkoxy-, nitro-, dialkylamino-, acylamino- eller nitril-grupper substituerede benzoesyrer, naphthoesyrer eller 15 anthracensyrer, endvidere phenylalkylcarboxylsyrer med ligekædede eller forgrenede alkylgrupper med indtil 10 C-atomer, som eventuelt kan indeholde de ovennævnte sub-stituenter ved benzenkernen, samt N- eller S-holdige, heterocycliske forbindelser med 5- og 6-leddet ring, 20 som ligeledes kan bære de samme substituenter.
Som aminokomponenter til fremstilling af carboxyl-syreamiderne kan foruden ammoniak også aliphatiske, aromatiske eller heterocyclisk substituerede primære og sekundære aminer med de ovenfor nævnte substituenter 25 komme på tale.
Til fremstilling af aminocarboxylsyreamider eller peptider arbejder man med sådanne aminocarboxylsyre-derivater eller peptider, hvis carboxylgruppe er beskyttet, og omsætter disse med aminocarboxylsyrer eller pep-30 tider, som indeholder en fri carboxylgruppe, men hvis aminogruppe dog er beskyttet. Andre funktionelle grupper i aminosyrerne eller peptiderne beskyttes hensigtsmæssigt med de gængse peptid-beskyttelsesgrupper, og disse beskyttende grupper kan derpå ved fremgangsmåden 35 ifølge opfindelsen fraspaltes på sædvanlig måde.

Ved fremstillingen af carboxylsyreamider arbejdes der ved stuetemperatur eller også ved moderat forhøjet temperatur svarende til ca. 60°, og ved fremstillingen af aminocarboxylsyreamider eller peptider ved 5 temperaturer på -10°C til +40°C, fortrinsvis mellem 0° og stuetemperatur.
Til vandfraspaltningen efter fremgangsmåden ifølge opfindelsen er anhydriderne af de ligekædede eller forgrenede, eventuelt cycliske alkanphosphonsyrer med 10 kædelængder på 1 til 8 carbonatomer, fortrinsvis indtil 4 carbonatomer, egnede.
Som anhydrider af alkanphosphonsyrerne kan eksempelvis nævnes: methan-phosphonsyreanhydrid, ethan-phos-phonsyreanhydrid, n-propanphosphonsyreanhydrid og n-15 -b utan-phosphonsyre anhydrid.
Fremstillingen af alkanphosphonsyreanhydriderne kan gennemføres på i og for sig kendt måde, f-eks, som beskrevet i Houben-Weyl, Meth. d. Org. Chem. (1963), XII/1, s. 612.
20 Ved fremstillingen af .peptider arbejder man hensigtsmæssigt med særligt rene alkanphosphonsyre-an-hydrider. Sådanne anhydrider kan fremstilles ved omsætning af rene alkanphosphonsyredichlorider med 1 mol vand og efterfølgende fjernelse af det endnu i reaktionsblan-25 dingen resterende hydrogenchlorid ved anvendelse af formindsket tryk. Disse anhydrider kan derudover fortrinsvis fremstilles ved den fremgangsmåde, der er beskrevet i den tyske patentansøgning nr. P 2.811.628.1. Ved denne fremgangsmåde omdannes rene alkanphosphonsyrer ved ter-30 misk vandfraspaltning til anhydriderne. En efterfølgende rensning ved hjælp af en vakuumdestillation kan være hens ig-fesmæss ig.
Omsætningen ifølge opfindelsen gennemføres, når der skal syntetiseres peptider, på hensigtsmæssig måde 35 i neutralt eller højst svagt alkalisk medium.

Det enkleste er at ændre mediet til en pufferopløsning ved tilsætning af aliphatiske og cycloalipha-tiske, tertiære baser, såsom N-methylmorpholin, N-ethyl-morpholin eller trialkylaminer med op til 6 C-atomer pr.
5 alkylgruppe. De ifølge opfindelsen anvendte alkanphos-phonsyreanhydrider er for det meste godt opløselige i opløsningsmidler som f.eks. dimethylsulfoxid, DMF, DMA, diethylphosphit, 1-methyl-pyrrolidon, chloroform og methylenchlorid.
10 Til fremstilling af oligopeptider anvender man som udgangsmaterialer en aminosyre eller et peptid med en blokeret carboxylgruppe og en aminosyre eller et peptid med en blokeret aminogruppe. Til beskyttelse af carboxylgruppen og aminogruppen kan der anvendes alle 15 gængse beskyttelsesgrupper inden for peptidsyntesen. Til beskyttelse af carboxylgruppen anvendes f.eks. estere af ligekædede og forgrenede, aliphatiske alkoholer, såsom methanol, ethanol eller tert.butanol, jfr. Houben--Weyl, Methoden d. Org. Chemie 15/1 (1974) Synthese von 20 Peptiden, s. 315-350. Også estere af araliphatiske alkoholer, såsom benzylalkohol og diphenylmethylcarbinol, kan finde anvendelse.
Til beskyttelse af aminogrupperne kan man eksempelvis anvende carbobenzoxygruppen og carbo-tert.butyloxy-25 gruppen, jfr. Houben-Weyl, Methoden d. Org. Chemie 15/1 (1974), side 47 og 117 ff. Som opløsningsmiddel kan alle inden for peptidsyntesen gængse, vandfrie, indifferente opløsningsmidler finde anvendelse, såsom methylenchlorid, chlorofom, dimethylformamid, dimethylacetamid, dioxan, 30 diethylphosphit, 1-methyl-pyrrolidon og tetrahydrofuran.
Alkanphosphonsyreanhydriderne ifølge opfindelsen anvendes fortrinsvis i overskud (2-2,5 mol alkanphosphon-syreanhydrid pr. mol peptidbinding, der dannes).

Indtil nu er der ikke iagttaget nogen allergivirkning ved de syrer, der danner basis for syntesereagenset. Toksiciteten er ringe. Reagenset selv, som er enkelt at fremstille, giver især ved anvendelse af de-5 stillerede alkanphosphonsyreanhydrider ingen tungtopløse-lige biprodukter efter syntesen, som der f.eks. forekommer ved den hyppigt anvendte peptidbinding under anvendelse af dicyclohexylcarbodiimid.
I forhold til de hidtil beskrevne fremgangsmåder 10 for peptidsyntesen under anvendelse af aktiveringsmidler på basis af 3- eller 5-valent phosphor, som f.eks. peptidsynteserne ifølge phosphorazometoden (Liebigs Ann.
Chem., 580, s. 68, (1953)), syntesemetoderne under anvendelse af diethylchlorphosphit og tetraethylpyrrophos-15 phit (J. Am. Chem. Soc., 74, 5304, 5307 og 5309, (1952)) og syntesemetoden under anvendelse af polyphosphorsyre-estere (Ber., 91, (1958), s. 1073-1082 eller J. Org. Chem.
26, 2534, (1961)), har fremgangsmåden ifølge opfindelsen den fordel, at racemiseringen er ringere ved den sædvan-20 lige anvendelse af aminosyre- eller peptidester-hydro-chlorider.
Således viser f.eks. Z-Phe-Gly-OEt, fremstillet ved fremgangsmåden ifølge opfindelsen, en [a]D~værdi på -17° (c = 2, ethanol). Metoden under anvendelse af di-25 ethylchlorphosph.it (J. Am. Chem. Soc., 74, 5307 (1952)) giver et peptid med en [a]D~værdi på -16° (c =, ethanol).
Den i eksempel 4 beskrevne forbindelse Z-Asp (OBu^)-Phe--NH2 udviser en [a]D-værdi på -33,8° (c = 1, ethanol) i sammenligning med litteraturværdien på -30,7° (phosphor-30 azometode, Hoppe-Seyler§ Z-Physiol. Chem., 353, (1972, s . 1250)". Den som racemiseringstest kendte syntese af Z-Gly-Phe-Gly-OEt (J. Am. Chem. Soc., 80, (1958), s.
2902) gav ved fremgangsmåden ifølge opfindelsen L-for-bindelsen i et udbytte på 75% af det teoretiske, jfr.
35 eksempel 10. Andele (DL-peptider), tungtopløselige i ethanol, dannedes ikke. Derimod forekommer der 32% af

5 den i ethanol tungtopløselige DL-forbindelse og 32% af L-forbindelsen ved syntesen af tripeptidet med polyphos-phorsyreethylesterne ifølge J.Org. Chem., 26_, 2534, (1961).
En yderligere fordel ved fremgangsmåden ifølge 5 opfindelsen er syntesereagensets gode termiske stabilitet.
I forhold til polyphosphorsyreesterne må desuden først og fremmest destillerbarheden af alkanphosphonsyreanhydri-derne noteres som en fordel, hvilken destillerbarhed åbner vejen til definerede oligomere.
10 Den stærke opløsningsformidlende virkning af al- kanphosphonsyreanhydrideme og deres følgeprodukter under peptidsyntesen er desuden meget ønskelig, især ved syntesen af længere peptidkæder og tungtopløselige peptidsekvenser.
15 De ved fremgangsmåden fremstillede produkter an vendes som mellemprodukter eller til fremstilling af farmaceutisk virksomme peptider.
De følgende eksempler tjener til nærmere belysning af fremgangsmåden ifølge opfindelsen.

Eksempel 1
Carbobenzoxy-glycylglycinethylester
Til en opløsning af 10,5 g (0,05 mol) carbobenz-oxy-glycin sætter man under omrøring og god afkøling 25 efter hinanden ved 0°C 7,0 g (0,05 mol) H-Gly-CX^H,- ,HC1, 15 ml (0,118 mol) N-ethylmorpholin og 36 ml af en 55%1 s (W/V) opløsning af n-propanphosphonsyreanhydrid (0,1 mol) i methylenchlorid. Der opvarmes under omrøring til stuetemperatur. Efter 16 timers henstand ved stuetemperatur 3Q afdestillerer man opløsningsmidlet under formindsket tryk og optager remanensen i en blandning af 200 ml eddikesyreester og 100 ml 5%'s kaliumbisulfatopløsning. Eddikesyrees teropløsnin gen vaskes endnu to gange med 100 ml mættet natriumbicarbonatopløsning pr. gang, tørres over natrium-35 sulfat og inddampes under formindsket tryk.

Udbytte: 11,8 g Z-dipeptidester med et smp. på 81°C (80% af det teoretiske). Efter omkrystallisation fra eddikesyreester/petroleumsether: 82°C.
5 Eksempel 2 Z-Val-Tyr(But)His-OCH,,
Til en suspension af 11,5 g H-His-OCH^,HC1 i 100 ml dimethylformamid sætter man ved 0°C og under omrøring efter hinanden 30 ml N-ethylmorpholin, 21,5 g Z-Val-Tyr-OH og 10 40 ml af en 38%'s opløsning af n-propanphosphonsyrean- hydrid i methylenchlorid ved temperaturer op til 10°C.
Den praktisk talt klare reaktionsopløsning, som fremkommer efter, at den exoterme reaktion er løbet til ende, henstår natten over ved stuetemperatur, hvorefter man afdam-15 per opløsningsmidlerne under formindsket tryk ved stue temperatur og sætter en blanding af 100 ml mættet NaHCOg--opløsning og 200 ml eddikesyreethylester til remanensen.
Råproduktet overføres i eddikesyreesterfasen under omrystning, denne vaskes med lidt vand, tørres over na-20 triumsulfat og bringes til tørhed under formindsket tryk.
Den i remanensen tilbageblevne Z-tripeptidester stivner ved digereringen med diethylether.
Udbytte: 20 g med et smp. på 188-190°C; [aJD = -8,2° (c = 1, DMF). Af moderluden kan der ved afdampning af 25 opløsningsmidlet og omfældning fra eddikesyreethylester/- diethylether udvindes yderligere 3,5 g af peptidet. Samlet udbytte: 76% af det teoretiske.
Eksempel 3 30 Z-Pro-Ala-Lys-(Boc)-Phe-NH^ 19,1 g Η-Lys (Boc) -Phe-NE^HCl (0,044 mol) opløses i 100’ ml dimethylformamid, og der tilsættes ved 0°C under omrøring 26 ml N-ethylmorpholin, 16,0 g (0,005 mol) Z-Pro-Ala-OH og 22 g n-propanphosphonsyreanhydrid som 35 38%'s (W/V) opløsning i methylenchlorid. Man lader reak tionsblandingen henstå ved stuetemperatur i 48 timer,

7 hvorefter man bringer den til tørhed under formindsket tryk ved stuetemperatur, digererer remanensen med 100 ml 2 N natriumcarbonat-opløsning, 100 ml 10%'s vandig citronsyreopløsning og 100 ml destilleret vand og tørrer 5 under formindsket tryk over phosphorpentoxid.
Udbytte: 30,1 g = 87% af det teoretiske. [a]D = -27,0° (c = 1, DMF). Smp. 163°C.
Eksempel 4 10 Z-Asp(QBu^)-Phe-NH2 1,6 g (0,01 mol) H-Phe-NI^ opløses sammen med 28 ml (0,22 mol) N-ethylmorpholin ved 0°C i 20 ml di-methylformamid. Til denne opløsning sætter man under omrøring og afkøling 3,23 g (0,01 mol) Z-Asp(OBu1*) -OH og 15 11,4 ml 38%'s n-propanphosphonsyreanhydridopløsning i methylenchlorid. Den således fremstillede reaktionsopløsning lader man henstå natten over ved stuetemperatur.
Efter afdestillation af opløsningsmidlet og tilsætning af vand ekstraherer man med eddikesyreester, vasker med 20 vand, vandig natriumbicarbonatopløsning, 5% vandig KHSO^- -opløsning, koncentrerer den over natriumsulfat tørrede eddikesyreesteropløsning, og fælder slutproduktet med diethylether.
Udbytte: 4,0 g (85% af det teoretiske). Smp. 162°C [a]D = 25 -33,1°, (c = 1, CH3OH).
Eksempel 5 Boc-Met-Gly-OEt
Til 115 ml dimethylformamid sættes under omrøring 30 og udelukkelse af fugtighed ved 0°C og efter hinanden 24,8 g Boc-Met-OH, 14,0 g H-Gly-OEt,HCl, 60 ml N-ethylmorpholin og 67 ml af en 67%'s methylenchloridopløsning af n-propanphosphonsyreanhydrid. Man efterrører i yderligere 20 timer ved stuetemperatur, afdestillerer opløs-35 ningsmidlet under formindsket tryk ved stuetemperatur og sætter vand til remanensen. Den udfældede peptidester op-

8 tages i eddikesyreester, og eddikesyreesteropløsningen vaskes i nævnte rækkefølge med mættet, vandig NaHCO^--opløsning, 5%'s KHSO^-opløsning og vand. Acylpeptid-esteren udkrystalliserer af den over natriumsulfat tør-5 rede eddikesyreesteropløsning efter vidtgående afdampning af opløsningsmidlet under formindsket tryk ved stuetemperatur. Der vaskes med lidt absolut diethylether og filtreres ved frasugning.
Udbytte efter tørring under formindsket tryk over ?2°5: 10 29,5 g = 93% af det teoretiske. Smp. 53°C [a]p = -15,4°, (c = 1, DMF).
Eksempel 6
Boc-Tyr(Etoc)-Met-Gly-OCpH^ 15 23 g H-Met-Gly-CX^H^,HC1 opløses i 200 ml di me thylformamid, og der tilsættes under stærk afkøling ved 0°C 58 ml N-ethylmorpholin, 31,5 g Boc-Tyr(Etoc)-OH og 44 g n-propanphosphonsyreanhydrid, opløst i 60 ml methylenchlorid. Man lader opløsningen henstå i 20 timer 20 ved stuetemperatur, hvorefter man afdestillerer opløsningsmidlet under formindsket tryk ved stuetemperatur og oparbejder reaktionsblandingen som i eksempel 5.
Udbytte: 40,7 g (84% af det teoretiske). Smp. 137,5°C.
[a]D = -11,7°, (c = 1, DMF).

Eksempel 7 Z-Gly-Thr(Bufc)-Phe-OCH^ I 120 ml dimethylsulfoxid (p. A. Merck) opløses i nævnte rækkefølge 18,6 g (0,05 mol) H-Thr(Bu1")-Phe-30 -OCH^,HC1, 30 ml (0,238 mol) N-ethylmorpholin og 10,4 g (0,05) Z-Gly-OH, og man tilsætter derpå under isafkøling og udelukkelse af fugtighed 16,3 g methanphosphonsyre-anhydrid portionsvis under omrøring. Methanphosphonsyre-anhydridet går langsomt i opløsning. Man efterrører i 35 yderligere 24 timer ved stuetemperatur og hælder derefter reaktionsopløsningen i 500 ml mættet natriumbicarbonatop-

9 løsning. Reaktionsproduktet udfælder herved. Man dekanterer den ovenstående opløsning fra og optager fældningsproduktet i eddikesyreethylester. Eddikesyreesteropløs-ningen vaskes med vand, tørres over magnesiumsulfat, kon-5 centreres vidtgående under formindsket tryk, og slutpro duktet fældes med petroleumsether. Det udkrystalliserer natten over ved +4°C.
Eksempel 8 10 Z-Phe-cyclohexylamid 30 g (0,01 mol) Z-Phe-OH, 1,0 g (0,01 mol, 1,2 ml) cyclohexylamin og 5 ml N-ethylmorpholin opløses i 30 ml DMF, og der tilsættes derpå under isafkøling 8,8 g af en 50 vægtprocents n-propanphosphonsyreanhydridopløs-15 ning i methylenchlorid (0,0435 mol). Efter at den ekso-terme reaktion er løbet til ende (temperaturstigning til +10°C), lader man opløsningen opvarme til stuetemperatur under omrøring og oparbejder derefter reaktionsblandingen som beskrevet under eksempel 1.
20 Udbytte: 3,1 g (81% af det teoretiske). [a]D = -2,8°, (c = 1, DMF). Smp. 167°C.
Eksempel 9 Z-Ala-Tyr-Gly-Leu-Arg-Pro-Gly-NH^ 25 En blanding af 4,99 g (0,01 mol) Z-Ala-Tyr-Gly- -OH, 5,13 g (0,01 mol) H-Leu-Arg-Pro-Gly-NH2,2HCl, 20 ml dimethylformamid og 10 ml (0,078 mol) N-ethylmorpholin afkøles til -10°C under omrøring og udelukkelse af fugtighed. Derefter tildrypper man under fortsat omrøring 30 6,6 g (0,022 mol) n-hexanphosphonsyreanhydrid som 50%'s opløsning i methylenchlorid (p.A., destilleret over P2C>5) . Temperaturen må under tildrypningen ikke overstige +10°C. Den fremkomne opløsning efterrøres i yderligere 3 timer og henstår natten over. Derefter koncen-35 trerer man under højvakuum ved stuetemperatur til ca.

Eddikesyreesteropløsningen ekstraherer man endnu to gange med 50 ml mættet natriumbicarbonat-opløsning og to gange med 50 ml vand, tørrer over na-5 triumsulfat og isolerer produktet ved afdampning af eddikesyreesteropløsningen under formindsket tryk, hvorefter man udriver remanensen med absolut diethylether.
Udbytte: 7,46 g (81% af det teoretiske). Smp. 130°C (sønderdeling) Γa]D = -29,2°, (c = 1, DMF) .

Fremstilling af n-hexanphosphonsyreanhydridet: 200 g n-hexanphosphonsyre opvarmes i en glaskolbe med en påmonteret, ikke-spejlende 12 cm Vigreux--kolonne og en tilsluttet luftkøler med forlag til max.
15 340°C ved et tryk på ca. 0,3 Torr. I løbet af 10 timer afdestillerer hexanphosphonsyreanhydrid ved en overgangstemperatur på 225°C til 250°C. I en efter apparatet tilkoblet afkølingsfælde kondenserer ca. 22 g vand. Man får 170 g af anhydridet. Produktet fraktioneres endnu en 20 gang inden omsætningen.
Det er klart opløseligt i methylenchlorid, chloroform, dimethylformamid, dimethylsulfoxid, petroleums-ether med kogepunkt 40-80°C, ether, toluen, dioxan, cyclo-hexan og tetrahydrofuran.

Eksempel 10 Z-Gly-Phe-Gly-OEt 1,4 g (0,01 mol) H-Gly-Oet,HCl og 3,56 g (0,01 mol) Z-Gly-Phe-OH suspenderes under omrøring i 30 ml dimethyl-30 formamid, og efter afkøling til -5°C tilsættes der under omrøring og udelukkelse af fugtighed 5,6 ml (0,044 mol) N-ethylmorpholin. Derefter tildrypper man under omrøring og afkøling 8,8 g af en 50%'s methylenchloridopløsning af n-propanphosphonsyreanhydrid (0,021 mol). Reaktionsblan-35 dingens temperatur må derved ikke overskride +10°C. Derefter efterrører man endnu i 1 time og lader henstå ved stuetemperatur i 48 timer.

Man afdestillerer opløsningsmidlet under højvakuum og tilsætter 50 ml isvand. Det udfældede slutprodukt skilles fra ved sugning efter udkrystallisation, vaskes med natriumbicarbonatopløsning, citronsyreopløsning (5%1 s) 5 og vand, og tørres over ?2°5 u^er formindsket tryk.
Udbytte: 4,30 g (98% af det teoretiske). [a]D = -11,9°, (c = 2, ethanol). Smp. 119°C.
Eksempel 11 10 l-Phenyl-2,3-dimethyl-4-(4-oxyphenylpropionylaminopyra-zolon(5)
Til en under udelukkelse af fugtighed omrørt opløsning af 16,6 g (0,1 mol) p-oxyphenylpropionsyre, 21 g (0,1 mol) l-phenyl-2,3-dimethyl-4-aminopyrazolon(5) og 15 68 ml N-ethylmorpholin sætter man dråbevis 88 g af en 50%'s opløsning af propanphosphonsyreanhydrid i methylen-chlorid. Temperaturen holdes herved, eventuelt ved at drosle tilstrømningen af propanphosphonsyreanhydrid, under +40°C. Man efterrører i yderligere 6 timer og lader 20 derefter henstå ved stuetemperatur i 24 timer. Derefter fortynder man reaktionsopløsningen med 800 ml methylen-chlorid, vasker opløsningen tre gange med 50 ml 5%'s kaliumbisulfatopløsning, tre gange med 50 ml mættet natriumbicarbonatopløsning og tre gange med 50 ml vand.
25 Man tører over magnesiumsulfat. Produktet udkry stalliserer ved koncentreringen af den tørrede methylen-chloridopløsning under formindsket tryk ved stuetemperatur. En mindre restfraktion kan endda fås af det med petroleumsether fældede filtrat.
30 Samlet udbytte: 25,0 g = 70% af det teoretiske med et smp. på 218°C.

12 tighed i 3 portioner 17,6 g af en SOS's opløsning af n--propanphosphonsyreanhydrid i methylenchlorid. Temperaturen stiger derved til ca. +40°C. Man lader afkøle til stuetemperatur under omrøring og oparbejder efter 24 5 timers henstand, idet man tilsætter 350 ml methylenchlorid, og ekstraherer opløsningen 3 gange med 50 ml vand, 50 ml mættet natriumcarbonatopløsning og 50 ml kalium-bisulfatopløsning. Råproduktet udkrystalliserer af den over magnesiumsulfat tørrede methylenchloridopløsning.
10 Råproduktet skilles fra ved sugning sammen med lidt pe-troleumsether (kogepunkt: 40-60°) .
Udbytte: 3,8 g = 84% af det teoretiske. Smp. 111-112°C.
Efter omkrystallisation fra ethanol: 116°C (spåner) .

Thiophen-2-carboxylsyre^-pheny le thy lamid
Analogt med eksempel 12 får man af 2,6 g thiophen--2-carboxylsyre, 2,4 g β-phenylethylamin, 14 ml N-ethyl-20 morpholin opløst i 15 ml methylenchlorid og 17,6 g af en 50%'s opløsning af propanphosphonsyreanhydrid i methylenchlorid thiophen-2-carboxylsyre-2-phenylethylamid.
Udbytte: 3,5 g = 76% af det teoretiske. Smp. 108-109°C.
Efter omkrystallisation fra ethanol: 111°C.

Eksempel 14
Benzoesyre-4-nitroanilid
Til en blanding af 2,8 g 4-nitroanilin, 15,5 ml triethylamin, 2,2 g benzoesyre og 15 ml methylenchlorid 30 sætter man under omrøring og afkøling i 3 portioner i alt 17,6 g af en 50%'s opløsning af n-propanphosphonsyre-anhydrid. Man efterrører endnu i én time ved stuetemperatur, og herunder klarer blandingen. Efter 32 timers henstand ved stuetemperatur oparbejder man som beskrevet 35 under eksempel 1.
Udbytte efter omkrystallisation fra ethanol: 3,38 g = 70% af det teoretiske. Smp. 198°C.
