Den foreliggende opfindelse angår en fremgangsmåde til fremstilling af hidtil ukendte indolderivater.
Uanset de fremskridt, der er gjort i løbet af de sidste 4 årtier i udviklingen af midler til behandling af 5 inflammatoriske tilstande, er der stadig et behov for effektive midler uden de bivirkninger, der er forbundet med de terapeutiske midler, der nu anvendes til dette formål.
Indolderivater fremstillet ifølge opfindelsen har vist sig at have interessante og nyttige farmakodynamiske 10 egenskaber uden uønskede bivirkninger, nemlig antiinflam-matorisk hhv. analgetisk aktivitet.

Disse salte har de samme egenskaber som moderforbindelserne, når de indgives til dyr, og kan anvendes på samme måde. Hensigtsmæssige 20 baser, der danner disse salte, omfatter f.eks. natriums, kaliums, calciums og magnesiums hydroxider, lavere alkoxider, carbonater og bicarbonater såvel som baserne ammoniak, tri-ethylamin og benzylamin. De således opnåede additionssalte er funktionelle ækvivalenter til den sure moderforbindelse 25 i henseende til terapeutisk anvendelse, dvs. de baser, der anvendes ved fremstilling af saltene, er farmaceutisk acceptable.
Der fås også isomere af forbindelserne med formel I stammende fra de asymmetriske centre indeholdt deri.
30 De nyttige antiinflammatoriske og analgetiske virk ninger hos indolderivaterne fremstillet ifølge opfindelsen kan påvises ved standardfarmakologiske prøver, f.eks. ved de prøver, der beskrives af R.A. Turner i "Screening Methods in Pharmacology", Academic Press, New York og London, 1965, 35 side 152-163 og side 100-117.

Antiinflammatorisk og analcretisk aktivitet.
Den antiinflammatoriske virkning hos forbindelserne fremstillet ifølge den foreliggende opfindelse kan påvises 5 ved en modifikation af Freund's adjuvantprøve, nemlig prøven for adjuvantinduceret akut ødem som beskrevet af J.R. Boissier et al., Therapie, 25, 43 (1970). Denne prøve er kendt for at passe udmærket sammen med data opnået fra kliniske resultater med mennesker. Boissier et al har påvist denne 10 sammenhæng med sådanne klinisk aktive forbindelser som f.eks. phenylbutazon, mefenaminsyre, indomethacin, aspirin, hydro-cortison og prednisolon.
Mere specielt eksemplificeret kan påvises en væsentlig antiinflammatorisk virkning for forbindelserne angivet neden-15 for ved orale doser på 100 mg/kg eller mindre ved denne akutte ødemprøve. Ved denne prøve behandles hanrotter med forsøgsforbindelsen 1 time før indgivelse af Freund's adjuvant i poten (dag 0). Rotterne behandles derpå med samme dosis af forsøgsforbindelsen i de næste tre dage. Den an-20 tiinflammatoriske virkning for forsøgsforbindelsen måles ved reduktion af inflammation i foden, jf. Turner nævnt ovenfor, og udtrykkes som en procentvis inhibering af adjuvant indgivet til kontrolrotter på 3. dagen.

Manglen på bivirkninger for forbindelserne fremstillet ifølge den foreliggende opfindelse kan påvises ved standardforsøg for akut toksicitet (jfr. Turner ovenfor) og ved forlænget indgivelse af forbindelsen til varmblodede dyr.
5 Når indolderivaterne fremstillet ifølge den forelig gende opfindelse anvendes som antiinflammatoriske og anal-getiske midler i varmblodede dyr, f.eks. rotter, kan de indgives oralt, enten alene eller i dosisform, f.eks. kapsler eller tabletter, kombineret med farmakolotisk acceptable 10 strækkemidler, såsom stivelse, mælkesukker osv. De kan også indgives oralt i form af opløsninger i hensigtsmæssige bæremedier f.eks. vegetabilske olier.
Doseringen af indolderivaterne fremstillet ifølge den foreliggende opfindelse varierer alt efter den forbin-15 delse, der vælges, og indgivelsesmåden. Ydermere varierer den alt efter det forsøgsdyr, der behandles. I almindelighed indgives forbindelserne fremstillet ifølge den foreliggende opfindelse ved et koncentrationsniveau, der giver beskyttende virkninger uden nogen skadelige bivirkninger. Disse antiin-20 f lamator isk/analgetisk effektive koncentrationsniveauer opnås sædvanligvis inden for et terapeutisk område fra 1,0 mg til 500 mg/kg pr. dag, med et foretrukket område fra 10 til 100 mg/kg pr. dag.

Almindeligvis fås sammenlignelige produktudbytter ved denne fremgangsmåde, når enten ketosyren eller den tilsvarende ketoester anvendes. I tilfælde, hvor det er foretrukket at fremstille syreforbindelser med formlen VII, hvori 15 R8 0 1 11 16 ...
Y er -C—C-, og Rxo er hydrogen, er det imidlertid af hensyn R9 20 til udbyttet at foretrække først at kondensere den rette β--ketoester med formlen VI fremfor den tilsvarende /3-ketosyre og derpå at hydrolysere det opnåede esterprodukt til opnåelse af den ønskede syreforbindelse.
25 I øvrigt er det almindeligvis ved den praktiske ud øvelse af fremgangsmåden ifølge den foreliggende opfindelse ofte mere hensigtsmæssigt at fremstille syreforbindelserne med formlen VII under anvendelse af ketoesteren i stedet for ketosyren ved denne fremgangsmåde og derpå hydrolysere 30 det opnåede esterprodukt til den ønskede syre, simpelt hen fordi ketoestrene i almindelighed er lettere tilgængelige enten kommercielt eller syntetisk.
Hydrolyse af forbindelserne med formel VII, hvori R16 er lavere alkyl, til deres tilsvarende syrer med formlen 35 VII udføres hensigtsmæssigt ved behandling med et passende alkali, f.eks. kaliumhydroxid eller natriumcarbonat, i vandig methanol eller vandig ethanol.

Ved udøvelse af kondensationen (II + VI -> VII) har det vist sig hensigtsmæssigt at anvende et opløsningsmiddel som reaktionsmedium. Ethvert opløsningsmiddel, der er indifferent under reaktionsbetingelserne, kan anvendes.
5 Hensigtsmæssige opløsningsmidler omfatter benzen, toluen, diethylether, dioxan, tetrahydrofuran, methylenchlorid og carbontetrachlorid. Benzen og tetrahydrofuran er særlig hensigtsmæssige og praktiske til denne anvendelse. Forskellige hensigtsmæssige syrekatalysatorer kan anvendes til 10 denne kondensation, f.eks. den katalysatortype, der anvendes ved Fischers indolsyntese, dvs. p-toluensulfonsyre, phosphor-pentoxid, bortrifluorid, zinkchlorid, saltsyre eller svovlsyre. p-Toluensulfonsyre, bortrifluorid og phosphorpentoxid hører til de foretrukne syrekatalysatorer. Mængden af anvendt 15 syrekatalysator er ikke specielt kritisk og kan variere fra 0,01 mol-ækvivalenter til 100 mol-ækvivalenter; et område fra 0,1 til 10 mol-ækvivalenter foretrækkes imidlertid. Reaktionstiden kan variere fra 10 minutter til 60 timer, idet det foretrukne område er fra 30 minutter til 24 timer.
20 Reaktionstemperaturen kan variere fra -20°C til reaktionsblandingens kogepunkt. Den foretrukne temperatur ligger fra 20°C til 120°C.
α-, β-, ύ— og 6-ketosyrerne og -ketoesterne med formlen VI er enten kendte, f.eks. ethylpyruvat, levulinsyre, 25 ethyl-a,a-dimethylacetoacetat, β,Ø-dimethyllevulinsyre og benzoyleddikesyre, eller de kan fremstilles ved kendte metoder beskrevet i de almindeligt tilgængelige organisk-ke-miske lærebøger. En detaljeret redegørelse af egenskaberne og fremstillingen af sådanne a-, β-, ύ- og 5-ketosyrer og-30 ketoestere findes f.eks. i ovennævnte "Rodd's Chemistry of the Carbon Compounds'1, bind Id, side 226-274.
De følgende eksempler tjener til yderligere illustration af fremgangsmåden ifølge opfindelsen.

Eksempel 1 1-Methvl-l,3.4.9-tetrahvdropyranoΓ3.4-blindol-l-eddikesvre (I; R1 = CH3, R2, R3, R4, R5, R6a-R6b og R7 = Η, X = 0, Y = CH2C0 og Z = OH) 5 23,4 g ethylacetoacetat (0,18 mol) sættes til en opløsning af udgangsmaterialet med formlen II, 10,0 g tryp-tophol (0,06 mol) i 200 ml benzen. Efter henstand i 10 minutter tilsættes 1,3 g p-toluensulfonsyre og ca. 5 g hydra-tiseret alkali-aluminiumsilicat (molekylsigte nr. 4). Blan-10 dingen opvarmes under tilbagesvaling i 30 minutter, yderligere 600 mg p-toluensulfonsyre tilsættes, og tilbagesvalingen fortsættes i 2\ time. Molekylsigterne samles, og benzenopløsningen vaskes successivt med 5%'s natriumbicar-bonat og vand, tørres med natriumsulfat og inddampes under 15 formindsket tryk til tørhed, hvilket giver en olie. Olien underkastes chromatografi på silicagel. Eluering med 5% ether i benzen giver som rodukt esteren l-methyl-1,3,4,9-tetrahydropyrano[3,4-b]indol-l-eddikesyre-ethylester som en olie u£axl3 1715 cm-1.
20 Hydrolyse af denne ester til den i overskriften nævnte forbindelse udføres på følgende måde: Esteren opløses i 230 ml methanol. Hertil sættes 10 g Κ0Η i 30 ml H20, og opløsningen henstår natten over ved stuetemperatur. Methanolen inddampes, der tilsættes vand, og opløsningen vaskes med 25 benzen. Den vandige fase gøres sur med 6 N HC1 og ekstraheres med benzen. Denne organiske fase vaskes med vand, tørres med natriumsulfat og inddampes til tørhed, hvilket som produkt giver en olie, som krystalliseres fra benzen indeholdende en smule petroleumsether, hvilket giver den i over- CHClo 30 skriften nævnte forbindelse, smeltepunkt 150-152eC, umay3 3325 og 1705 cm"1.
En ækvivalent mængde methylacetoacetat kan erstatte ethylacetoacetat ved fremgangsmåden ifølge dette eksempel.
I dette tilfælde fås esteren 1-methyl-1,3,4,9-tetrahydropy-35 rano[3,4-b]indol-eddikesyre-methylester, smeltepunkt 87--90°C, efter omkrystallisation fra benzen-hexan.

Eksempel 2 1-Methyl-l,3,4,9-tetrahvdropyranor3,4-blindol-l-propionsyre (I? R1 = CH3, R2, R3, R4, R5, R6a-R6d og R7 = Η, X = O, Y = CH2CH2CO og Z = OH) 5 En blanding af udgangsmaterialet med formlen II, 500 mg tryptophol, 580 mg levulinsyre, 75 ml benzen, 1,7 g phos-phorpentoxid og ca. 0,5 g diatoméjord ("Celite") omrøres med magnetomrører ved stuetemperatur i 15 minutter og derpå ved 70°C i Ih time. Reaktionsblandingen filtreres. Filtratet 10 vaskes tre gange med 5 N NaOH; den kombinerede vandige fase vaskes to gange med ether og gøres derpå sur med kold 50%'s HC1. Den vandige fase ekstraheres med chloroform. Chloroform-ekstrakten tørres (Na2S04) og inddampes til tørhed. Remanensen omkrystalliseres fra ethylacetat-petroleumsether, hvilket 15 som produkt giver den i overskriften nævnte forbindelse, smeltepunkt 104-110°C, NMR (CDC13) S 1,47 (3H), 2,18 (4H), 2,74 (2H), 3,96 (2H), 7,18 (4H), 7,85 (IH), 9,60 (IH).
Den i overskriften nævnte forbindelse kan også fås ved fremgangsmåden i eksempel 1, idet ethylacetoacetat er-20 stattes med en ækvivalent mængde ethyllevulinat. I dette tilfælde fås l-methyl-l,3,4,9-tetrahydropyrano[3,4-b]indol- ΛΤΤΛ1 -1-propionsyreethylester, smeltepunkt 116-118°C, oj5ax 3 1716 cm-1, efter omkrystallisation fra benzen-petroleums-ether, som esteren inden hydrolyse.

Eksempel 3 1-Methvl-l,3,4,9-tetrahvdrothiopvrano Γ3.4-blindol-l-eddike-svre (I, R1 = CH3, R2, R3, R4, R5, R6a-R6d og R7 = Η, X = S, Y = 30 CH2CO og Z = OH)
Indol-3-ethanthiol (1,5 g) og methylacetoacetat blandes med 50 ml benzen, og opløsningen opvarmes i 30 minutter (badtemperatur 70-80“C). 0,15 g p-toluensulfonsyre tilsættes, og reaktionsblandingen opvarmes under tilbagesvaling og 35 omrøring i 12 timer. Vand dannet i reaktionsblandingen i løbet af dette tidsrum opsamles ved hjælp af en vandsepara-

Inddampning af benzenopløsningen giver 1-methyl-l ,3,4,9-tetrahydrothiopyrano [ 3,4-b] in-5 dol-l-eddikesyremethylester som et halv-faststof, wmax 3 1715 cm-1 som det ønskede esterprodukt.
Denne ester behandles derpå med vandig alkoholisk KOH på samme måde som beskrevet for esterne i eksempel 1 og 2, hvilket som produkt giver den i overskriften nævnte for-iO bindel se, smeltepunkt 147-149eC NMR (CDC13) δ 1,86 (S, 3H) 3,06 og 8,12 (6H), 7,35 (multiplet, 4H), 8,71 (IH), 10,31 (IH) efter omkrystallisation fra benzen-hexan.
Fremgangsmåderne ifølge eksempel 1 eller 3 kan følges til fremstilling af andre forbindelser med formlen I, hvori 15 R1, R2, R3, R4, R5, R6a-R6d, X og Y har de ovenfor angivne betydninger, R7 er hydrogen, og Z er hydroxy. 1. Fremgangsmåde til fremstilling af 1-substituerede 1,3,4,9-tetrahydropyrano-(eller thiopyrano)-[3,4-b]indol-1--alkansyrer, deres estere eller salte med formlen
5 E6a R5 R4 R R hvori R1 er valgt blandt alkyl med 1-6 carbonatomer, alkenyl med 2-6 carbonatomer, alkynyl med 2-6 carbonatomer, cyclo-alkyl med 3-6 carbonatomer, phenyl, benzyl og 2-thienyl,
15 R2, R3, R4 og R^ har samme eller forskellig betydning og er hver især valgt blandt hydrogen og alkyl med 1-6 carbonatomer, R6a, R6b, R6c, og R6d har samme eller forskellig betydning og er valgt blandt hydrogen, alkyl med 1-6 carbonatomer, hydroxy, alkoxy med 1-4 carbonatomer, benzyloxy, 20 alkanoyloxy med 2-6 carbonatomer, nitro, halogen, mercapto, alkylthio med 1-6 carbonatomer, trifluormethyl, amino og sulfamoyl, R7 er valgt blandt hydrogen, alkyl med 1-6 carbonatomer og alkenyl med 2-6 carbonatomer, X er oxygen eller svovl, Y er en alkanoylbro med formlen 25 0 II -c-, 30 R8 O R8 R10 O R8 R10 R12 O f II I I II I I I II —C—C—, —C—C-C— eller —C-C-C-C—
