(Ges. v, 15. 7.1951)(Ges. V, 15.7.1951)
It is known that dem /? - Phenylisopropylmethylamine in its various modifications has a central arousal effect.
It has been found that by saturating the phenyl radical in this compound with hydrogen new substances are created that are free of side effects, in particular with lower toxicity Brew production, are and any group of people, especially those with cardiovascular stress, can be administered safely.
B. das bekannte Methyl-y-cyclohexylpropylamin, C6H11 ·It is true that hydroaromatic amines whose secondary nitrogen atom is in an aliphatic side chain have already become known. However, these substances do not have a noteworthy pharmacological effect. So is z. B. the well-known methyl-y-cyclohexylpropylamine, C 6 H 11 ·
CH2-CH2 CH2-NH-CH3, in seiner Weckwirkung bei gleicher Toxizität den erfindungsgemäß hergestellten Produkten unterlegen, wie aus folgenden Versuchswerten hervorgeht:CH 2 -CH 2 CH 2 -NH-CH 3 , in its waking effect with the same toxicity, inferior to the products manufactured according to the invention, as can be seen from the following test values:
Die Abkürzung der Schlaf dauer beträgt bei Methyly-cyclohexylpropylamin nach 100 mg/kg 11 %, beiThe sleep produced in the mouse by 100 mg / kg isoamylethylbarbituric acid ("Amytal") interrupted by the substances to be compared. The new process products are mild stimulants that z. B, in the therapy of rhinitis for decongesting the swelling of the mucous membrane, as over-the-counter drugs, provide valuable services. Their advantages over similarly acting known agents from the series of sympathomimetics, in particular over the structurally related / J-phenylisopropylmethylamm, are primarily the lack of side effects. Among these side effects, particular emphasis should be placed on the risk of addiction, which is so inherent in the last-named product that this drug was not only made subject to mandatory prescription but also to the Narcotics Act. The ^ -Phenylisopropylmethylamin is an agent that provides valuable services in the hands of the doctor in many cases of illness, but can not be given to the layman without hesitation, because its euphoric effect is the great danger of addiction. In contrast, the new process products can be safely used by any layperson, especially in the case of the common colds that recur every season. The inhaled by a simple Tascheninhaler amounts practicing ^ -Cyclohexylisopropylmethylamin containing 1.8 amounted mg per liter of air of 30 0 C, even no influence on the circulatory system and the central nervous system, unlike the Phenylanalogen and amphetamine, which in this dosage already cause a certain central arousal and increase in blood pressure. The difference in the onset of action of the new compounds compared to that of / 3-phenylisopropylmethylamine can be demonstrated in a series of further tests, it being shown again and again that the cyclohexyl derivative exerts stimulating effects on individual central nervous centers even in doses at which an increase in the general activity of the entire central nervous system is not yet detectable. This applies in particular to i. for the stimulating effect on breathing, which is even more evident than on normal animals after the use of substances with specifically effective paralysis on the respiratory center, such as morphine. The effective threshold doses, which cause an increase in respiratory volume and respiratory rate, are 0.5 mg / kg for the phenyl analog when administered intravenously to the untreated animal and approximately 1 mg / kg after morphine.

This is a dosage range in which this substance is already a general one when given intravenously Causes stimulation of the central nervous system. The corresponding values for the cyclohexyl derivative are again 0.5 mg / kg on the untreated animal and 2.0 mg / kg after administration of morphine when administered intravenously. The products of the process thus influence normal breathing Animal in the same dosage range as the phenyl analog. There is therefore reason for that Assumption that the process products certain centers of the central nervous system in specific Way, and do so in doses that do not affect other parts of the central nervous system have a demonstrable effect. In contrast, with phenyl analogs, the respiratory center is excited not independent of the general excitation of the central nervous system.
2. Further differences between the phenyl and the cyclohexyl compounds are the influence metabolism; both substances increase it, but the phenyl compound only dissolves within the motility-increasing doses an increase in oxygen consumption. In the case of the cyclohexyl derivative A quarter of the activity-increasing dose is enough to induce a strong increase in metabolism. Even through that Bromisovalerylurea pretreatment lowered metabolism is the cyclohexyl derivative in the same small doses effective, while the phenyl compound only turns with the safe central excitatory Giving affects in the same way.
This also shows I-cyclohexylisopropylniethylamine even with attempts to influence the diuresis, that it is already effective at one dose, which is still below the generally central excitation threshold threshold dose. That’s enough 5 mg / kg per os in order to achieve an increased excretion of water in the diuresis experiment with certainty. In contrast, you need gifts from the ^ -phenyl compound, which in turn are already in the area of general activity-enhancing doses are falling; only 4 mgAcg per os are sufficient to prevent urine secretion to get started more often. The point of attack for this influencing is probably the Diuresis also in a centrally located stimulation center, so that these findings also provide a further indication of the presence specific points of attack in the ^ -Cyclohexylverbindungen are in the organism, since these special
Mechanisms of action are caused by gifts that do not have general excitation of the central nervous system cause.
Diese Versuchsergebnisse beweisen, daß die Verfahrensprodukte eine erregende Wirkung auf das gesamte Zentralnervensystem erst in großen Dosen ausüben, die sich am normalen und am narkotisierten Tier bei subkutaner Gabe und Zufuhr per os in einer Zunahme der Motilität eindeutig zu erkennenThese test results prove that the process products have an exciting effect on the The entire central nervous system should only be exercised in large doses, which differ on the normal and on the anesthetized Animal clearly recognizable in an increase in motility when administered subcutaneously and per os
ίο gibt. Compared to /? - phenylisopropylmethyl amine, however, the effective threshold doses are considerably higher, so that in the case of sole evaluation According to these findings, the cyclohexyl derivative can be described as a less effective stimulant.
Meanwhile, the process products solve in different Testing already stimulating effects in individual centers of the central nervous system a dosage range that is safely below that which excites the entire central nervous system Gifts lies. This applies to the respiratory center, to influencing the metabolism and diuresis. In contrast, with the phenyl compound, all of these areas of activity always coincide with the motility-enhancing doses together; the cyclohexyl derivative thus differs significantly from the phenyl analogue in certain qualities of action, their analysis and characterization provide important evidence that the cyclohexyl compound In contrast to the phenyl compound, certain centers of the central nervous system are preferred before it has a general central stimulus effect. On the basis of these findings, the Process product ascribed a different activity character compared to the phenyl compound which proves to be beneficial from a therapeutic point of view.
Since the products of the process are also not very toxic, especially when given peroirals, their application is compared to that of the phenyl derivative, since it is on the one hand, despite a lower central excitation Effect have a more favorable therapeutic index, on the other hand already in Doses achieve a safe therapeutic effect that does not have general excitation of the central nervous system trigger.
Therefore another important application for yff-Cyclohexylisopropylmethylamin as an appetite suppressant Means for weight loss cures. Here, too, jö-cyclohexylisopropylmethylamine is among the competing products, especially the /? - phenylisopropylmethylamine and its norbase especially due to the lack of unwanted central arousal and circulatory effects, as well as superior due to the lack of danger of addiction.
The effect of the new / J-cyclohexylisopropylmethylamine is thus different from that of its phenyl analogue in that it is a sure influence certain centers of the central nervous system without producing a general central excitation Effect permitted. With the new product and its modifications are therefore ζ. Β at Cold conditions, weight loss cures or influencing the diuresis at least to achieve equivalent therapeutic effects, but different from those of its phenyl analogs to the extent that they are advantageous in that they do not have any undesirable side effects, such as Sudht risk and increase in blood pressure, are accompanied.
