The formula in which R is an alkylene group is known to also have a uterospasmolytic effect exhibit. However, this effect is much less pronounced with these known compounds. aside from that it is always associated with an undesirable lowering of blood pressure, surprisingly now it has been found that the compound of the invention, although having a stronger uterospasmolytic effect nevertheless lowers blood pressure to a much lesser extent.
Dabei wurden die Wirkungen der erfindungsgemäßen Verbindung mit denjenigen einer Verbindung der Formel II aThe uterospasmolytic effect was determined, among other things, in tests in which the Influence on the in uteri of rats by administration of pituitrin and acetylcholine as spasmogens generated spasms was examined. b) bezüglich der Toxizität kein Unterschied besteht, b) there is no difference in toxicity,
c) die uterospasmolytische und die bronchospasinolytische Wirkung der erfindungsgemäßen Verbindung zweimal so groß ist wie die der Verbindung der Formel Ha, so daß also zum Erreichen der gleichen spasmolytischen Wirkung wie der dieser bekannten Verbindung von der neuen Verbindung nur die Hälfte der Dosis erforderlich ist,c) the uterospasmolytic and the bronchospasinolytic Effect of the compound according to the invention is twice as great as that of the compound of the formula Ha, so that thus to achieve the same spasmolytic effect as the this known compound of the new compound only required half the dose is,
d) die (i-lytische Wirkung der erfindungsgemäßen Verbindung beträchtlich geringfügiger als die der bekannten Verbindungen ist,d) the (i-lytic effect of the compound according to the invention is considerably less than that of known connections is
e) die erfindungsgeniäße Verbindung eine geringfügigere blutdrucksenkende Wirkung hai: Die bekannten Verbindungen weisen eine zweimal so große blutdrucksenkende Wirkung wie die Verbindung nach der Erfindung auf.e) the connection according to the invention has a minor one antihypertensive effects hai: The known compounds exhibit a twice like that great antihypertensive effect like the compound of the invention.
From these results it can be concluded that the compound according to the invention is a more suitable one / i-Mimeticum as the compound of the formula Ha.
It is known that the levorotatory antipode of the compound of the formula Ha with a melting point of 123 to 124.5 ° C, whose amidosulfonate has a rotation value [α] £ 3 of about -3 ° C, has a uterospasmolytic effect which is twice greater than that of the racemate, while the lowering of blood pressure by the racemate is to be ascribed to the right-turning antipode (published documents of Belgian patent 612 995). Comparative experiments showed that the uterospasmolytic effect of the compound according to the invention is about OJ times the effect of this levorotatory antipode. Since this difference is so small that it is still within

Demgegenüber ist es ein bedeutender Vorteil, daß die erfindungsgemäße Verbindung in Razematform Verwendung findet, da die Auftrennung von Razematen in optische Antipoden bekanntlich sehr langwierig und kostspielig ist.half of the error limits of such comparative tests is the uterospasmolytic effect the connection according to the invention is practically as large as that of the left-handed antipode of the connection the formula Ha. Furthermore, comparative tests showed that the toxicity and the broncholytic, the u-lytic and antihypertensive effects are almost the same. In contrast, it is a significant one Advantage that the compound according to the invention is used in racemate form, since the separation from racemates to optical antipodes is known to be very tedious and expensive.

A solution of 44 g of 2-bromo-4'-benzyloxypropiophenone and 44 g of 2- (4-methoxyphenyl) ethylamine in 270 mol of ethanol was refluxed for 3 hours. Then the ethanol was in vacuo distilled off and the concentrate mixed with ether. The crystals obtained were filtered off with suction, whereupon the filtrate was mixed with an excess of 2η hydrochloric acid. This slowly crystallized that Hydrochloride of 4'-benzyloxy-2- [2- (4-melhoxyphenyl) ethylamino] propiophenone. This stuff was Also sucked off, washed with water and alcohol and dried in a vacuum !. After this Recrystallization from dilute alcohol yielded 25.5 g of a product having a melting point from 217 to 218 C.
Darauf wurde der Katalysator durch Filtrieren entfernt, und von der filtrierten Lösung wurde das Lösungsmittel im Vakuum ganz verdampft.12 g of the product thus obtained were dissolved in a mixture of 300 ml of ethanol and 9 ml of water. After 42 ml of 1% palladium chloride solution and 3.9 g of norit carbon were added to this solution, it was hydrogenated at room temperature and about 1.1 atm pressure until about 760 ml of hydrogen was taken up had been. The catalyst was then removed by filtration, and the filtered off Solution, the solvent was completely evaporated in vacuo.
The obtained from the hydrochloride of 4 '- hydroxy - 2 - [2 - (4 - methoxyphenyl) - ethylamino] propiophenone Existing residue was mixed with 30 ml of a 48% hydrobromic acid solution and the mixture boiled until no more methyl bromide was evolved, which is the case after about 45 minutes was. The reaction mixture was then stored in the refrigerator, whereupon the hydrobromide of 4 '- Hydroxy - 2 - [2 - (4 - hydroxyphenyl) - ethylamino] propiophenone crystallized. It was sucked off and converted into the hydrochloride in that the substance obtained is redissolved in water, the The solution is decolorized with a little norite charcoal and an equal volume of concentrated hydrochloric acid is added to it became. This caused the hydrochloride to crystallize. The yield was 9.6 g of a product with a melting point of 136 to 138 ° C. After this product has been recrystallized again, it was reduced to amino alcohol.
For this purpose, a solution of 3.2 g of the hydrochloride in 160 ml of distilled water was used 0.5 g of Norit and 8 ml of 1% palladium chloride solution and the mixture at room temperature and hydrogenated about 1.1 atm pressure until no more hydrogen was absorbed. The catalyst was then removed by filtration and the filtrate concentrated in vacuo. The concentrated one Solution of the reduced product, an excess of dilute ammonia was then added, whereby the base of 1- (4-hydroxyphenyl) -2- [2- (4-hydroxyphenyl) ethylamino] propanol when a viscous mass was eliminated. After the mixture has been kept in the refrigerator for some time, the product was filtered off with suction, washed with water and dried in vacuo. The base was a resinous one Mass with a melting point of about 88 to 90 C. Yield: 2.3 g.
Die Ausbeute betrug 10,3 g Hydrochlorid von 4'-Benzyloxy-2-[2-(4-benzyloxyphenyl) - äthylamino] - propiophenon mit einem Schmelzpunkt von 202 bis 204° C.A solution of 12.76 g of 2-bromo-4'-benzyloxypropiophenone, 9.08 g of 2- (4-benzyloxyphenyl) ethylamine and 4.44 g of triethylamine in 40 ml of ethanol was Cooked for 3 hours. The solution was partially evaporated and something crystallized out. The mixture 100 ml of water and 200 ml of ether were added and the undissolved substance (hydrobromide des Reaction product) removed by filtration. The two layers of the filtrate were separated The ether layer was washed once more with water and then with an excess of 4N hydrochloric acid mixed. After all the hydrochloride of the reaction product had crystallized, it was filtered off with suction, washed with water and ether and dried in vacuo next to KOH. The yield was 10.3 g Hydrochloride of 4'-benzyloxy-2- [2- (4-benzyloxyphenyl) - ethylamino] - propiophenone with a Melting point from 202 to 204 ° C.
A solution of 2.00 g of this product in 60 ml of 80% ethanol was hydrogenated until the two benzyl groups were removed by hydrogenolysis, which resulted from a considerable decrease in the rate of hydrogenation emerged. The catalyst was then filtered off and the alcohol from the filtrate was increased Part removed by being distilled off in vacuo. The concentrate obtained was mixed with water diluted and re-hydrogenated until no more hydrogen was absorbed.
Das charakteristische Ultraviolettabsorptionsspektrum zeigte ein Maximum bei 267,5 ΐτίμ ((3310).The catalyst was then filtered off and the aqueous solution evaporated in vacuo until it became cloudy. After some time, the hydrochloride of 1- (4-hydroxyphenyl) -2- [2- (4-hydroxyphenyl) ethylamino] propanol crystallized from this. It was sucked off sharply and dried in vacuo. Yield 0.55 g of a product with a melting point from 183 to 186 ° C (decomposition). The ultraviolet absorption characteristic spectrum showed a Maximum at 267.5 ΐτίμ ((3310).
Das Gemisch wurde 3 stunden gekocht und darauf im Vakuum eingedampft, bis etwa 300 g Rückstand zurückblieben.3.45 g of sodium borohydride were added to a mixture of 75 ml of methanol and 1.5 ml of 2η sodium hydroxide solution solved. This solution became a solution of 15 g of the hydrochloride of 4'-benzyloxy-2- [2- (4-benzoyloxyphenyl) ethylamino] propiophenone in 450 ml of methanol, the 1.2 g of sodium hydroxide (in a few milliliters Wassf r) was added, added. The mixture was boiled for 3 hours and then evaporated in vacuo, until about 300 g of residue remained.
Das charakteristische Ultraviolettabsorptionsspektrum entsprach demjenigen des Endprodukts aus Beispiel 1.Then another 300 ml of water were added and the mixture was evaporated again to about 250 g. The separated Product was then extracted with a total of about 500 ml of benzene (four extractions). The benzene solution was then dried over sodium sulfate and then dried to a residue of about 15 g evaporated. The fact that this residue was diluted with 25 ml of diethyl ether crystallized 7.4 g of 1- (4-benzyloxyphenyl) -2- [2- (4-benzyloxyphenyl) ethylamino] propanol with a melting point from 97.5 to 99 ° C. The product was recrystallized once more from ethanol and then through Hydrogenation debenzylated. This was done by hydrogenation a suspension of 3.88 g in 80 ml of about 60% ethanol and 2.2 ml of 3.04 η-hydrochloric acid to the above described way. After removal of the catalyst, the ethanolic solution was after dilution evaporated with 50 ml of water in vacuo to 5.5 g. This caused the hydrochloride to crystallize of 1- (4-hydroxyphenyl) -2- [2- (4-hydroxyphenyl) ethylamino] propanol the end. By diluting a solution of the crystallized product in about 9 ml of ethanol with about 40 ml of absolute ether 2.38 g of the hydrochloride mentioned are obtained with a melting point of 193 to 195 ° C. (decomposition) The characteristic ultraviolet absorption spectrum corresponded to that of the end product from Example 1.
