Description
The subject of the invention is a microbiological detection system for yeasts, molds, lactic and acetic acid bacteria, in particular in non-alcoholic beverages and cleaning samples in the beer sector (screening test), consisting of a culture medium, disposable tubes and a light box for the background lighting of the tubes.
Various state-of-the-art methods are already known for detecting microorganisms in drinks that generally have a pH value of 4.5 or lower. These methods can be used to detect, for example, mold, yeast, lactic acid and acetic acid bacteria.
There are also mechanical evaluation methods such as fluorescence and conductivity measurements.
The current state of the art in microbiological culture medium technology has the disadvantage that the culture media used, such as agar culture media, have only a low level of transparency and that evaluation is made more difficult using the classic pour plate method. For example, in microbiological tests using the conventional pour plate method, each dish must be evaluated individually. An additional disadvantage of agar culture media (pour plate method) is that the hot culture media (approx. 47°C) is mixed with the material being examined, which can cause damage to the germs.
The object of the invention was therefore to provide an improved microbiological detection system that avoids the disadvantages mentioned above, reduces the detection time and also considerably facilitates the evaluation.
This task is solved by a microbiological detection system for yeasts, molds, lactic and acetic acid bacteria, especially in non-alcoholic beverages and cleaning samples in the beer sector (screening test), consisting of a nutrient medium, disposable tubes and a light box for the background lighting of the tubes.
Der Nährboden kann folgende Zusammensetzung besitzen:The nutrient medium can have the following composition:
- 10 bis 20 Gew.-% Hefeextrakt- 10 to 20 wt.% yeast extract
5 bis 10 Gew.-% Pepton5 to 10 wt.% peptone
- 35 bis 80 Gew.-% Zucker- 35 to 80% sugar by weight
1 bis 3 Gew.-% Stabilisierungsmittel
1 bis 2 Gew.-% Mineralstoffe.1 to 3 wt.% stabilizing agent
1 to 2% by weight of minerals.
In a particular embodiment, 30 to 60 wt.% glucose, 2 to 10 wt.% maltose and 2 to 10 wt.% sucrose are included. The nutrient medium can also contain lactic acid, with which the necessary pH value, which is preferably between 4.3 and 4.4, can be set.
Another component of the microbiological detection system are disposable tubes made of transparent plastic or glass, which have a diameter of 1 to 3 cm and a height of 5 to 10 cm and can be closed.
The microbiological detection system also contains a light box with shelves and lights arranged behind it, whereby the disposable tubes are placed next to each other on the shelf and can be backlit by the lights arranged behind them. With this arrangement, the individual sample tubes can be evaluated at a glance and do not have to be picked up individually to carry out an evaluation, as is the case with conventional techniques.
The culture medium according to the invention is produced by mixing the solid, dry individual components. A further component is a lactic acid solution of preferably 40% by weight, with which the pH value of the culture medium solution can be adjusted. To prepare the culture medium, the solid mixture is dissolved in demineralized water and heated to 50 ° C with constant stirring. The pH value is then adjusted to a value of 4.3 to 4.4 with the lactic acid solution and, after the pH has been adjusted, the mixture is pasteurized by heating it to 85°C for 60 seconds. The liquid culture medium produced in this way is slightly opal and can be stored for up to 4 weeks at 5°C.
For sample preparation, the test material is placed in sterile disposable tubes and then filled with the appropriate amounts of culture medium. The samples are then placed in a light box and stored at a temperature of 28 0 C.
Whereas previously such tests required a waiting time of 3 to 4 days, the microbiological detection system according to the invention can provide a reliable diagnosis of infections with yeast, mold and other bacteria after just 1 to 2 days. The microbiological detection system according to the invention also has the advantage that, in contrast to machine evaluation methods such as fluorescence measurement and conductivity measurement, a classic microbiological technique is used. This also means that the cultures can be examined under a microscope and cultivated further if necessary. The method according to the invention also makes it possible to easily identify the fermentation behavior of yeasts, so that statements can be made about the increase in pressure caused by CO 2 in the sealed beverage containers and an assessment of the product hazard can be made.
SoFurthermore, the evaluation using the culture medium technique is also considerably cheaper than mechanical evaluation methods.
The detection system according to the invention can reduce the duration of the microbiological examination by 30 to 50%. In the case of yeast contamination, the infection is visible after just 24 hours; in the case of a mold infection, the detection time is 24 to 48 hours.
In comparative tests with classical methods, it was found that there is a correlation of 99 to 100% between the detection system according to the invention and the classical cast plate method.
The faster evaluation option of the detection system according to the invention is also due to the high transparency of the culture medium compared to previously known culture media. This results from the smaller test quantities that have to be introduced and the resulting very low turbidity. The faster evaluation is made considerably easier by the clearer arrangement on the shelf with background lighting.
This makes it possible to evaluate the results without having to handle each individual sample.
