Verfahren zur Herstellung von substituierten Äthylendiaminen Es wurde
gefunden, daß man substituierte Äthylendiamine herstellen kann, wenn man Verbindungen
der allgemeinen Formel
wobei R einen aliphatischen, araliphatischen oder lieterocyclischen Rest, R1 die
Carbonsäureestergruppe oder Wasserstoff bedeutet, und Ac für einen Acylrest, beispielsweise
die Acetyl- oder Phenacetylgruppe steht, der Hydrierung unterwirft. Die Acylgruppe
und gegebenenfalls die Estergruppe können gegebenenfalls nach der Hydrierung durch
Verseifung abgespalten werden. Will man a, ß-Diaminosäuren gewinnen, so werden die
Carbonsäureester, wie sie beispielsweise nach der in -Berichte der Deutschen Chem.
Ges. u 82, Heft 4j5, S. 387 bis 389 (i949) enthaltenen Mitteilung -Synthesen von

herstellen, so verseift man zweckmäßig
die Estergruppe und spaltet
z. B. durch Erhitzen nach dem Ansäuern Kohlensäure ab. Die dabei entstandenen Verbindungen
der allgemeinen Formel
werden dann der Hydrierung unterworfen, worauf schließlich der Acylrest durch Erhitzen
mit Mineralsäuren, wie z. B. Bromwasserstoffsäure, abgespalten wird. a, ß-Diaminocarbonsäuren
können als solche als Heilmittel Verwendung finden, während die substituierten Äthylendiamine
beispielsweise als Zwischenprodukte für die Darstellung von Heilmitteln dienen.
Beispiele i. 4,6 g Natrium werden in 8o ccm Methvlalkohol gelöst, 46,4 g Phenacetylaminocyanessigsäuremethylester
und darauf 25 g Benzylchlorid zugegeben. Man kocht etwa i Stunde unter Rückfluß,
worauf die alkalische Reaktion verschwunden ist, versetzt mit Wasser und saugt den
kristallin abgeschiedenen Phenacetylaminobenzylcyanessigsäuremethylester ab. Zunächst
werden 43,5 g mit 200 ccm 2 n-Natrorlauge so lange auf dem Dampfbad erhitzt, bis
klare Lösung eingetreten ist. Man setzt dann 220 ccm 2 n-Salzsäure zrr und erwärmt
4 bis 5 Stunden auf dem Dampfbad, bis die Kohlensäureentwicklung zu Ende gekommen
ist. Die Abscheidung des entstandenen Phenacetyl-% amirrobenzylessigsäurenitrils

28g dieser Verbindung werden in der zehnfachen Menge Isopropylalkohol
gelöst und bei 7o° mit Raney-Nickel hydriert. Nach Aufnahme der berechneten Wasserstoffmenge
wird vom Nickel abgesaugt und der Isopropylalkohol im Vakuum abdestilliert. Der
Rückstand wird mit 75 ccm 46°!oiger Bromwasserstoftsäure 3 Stunden unter Rückfiuß
gekocht, dann wird mit der doppelten Menge Wasser versetzt und die abgeschiedene
Phenylessigsäure mit Äther ausgeschüttelt. Der wäßrige Auszug wird fast zur Trbckne
eingedampft, mit konzentrierter Natronlauge versetzt und die entstandene Base mit
Äther ausgezogen. Nach dem Abdestillieren des Äthers wird das i, 2-Diamino-3-phenylpropan
im Vakuum destilliert, wobei es bei 2o mm Druck bei 14o bis 142- übergeht. Das Dichlorhvdrat

der zehnfachen Menge Iso propylalkohol gelöst und mit Raney-Nickel in der vorgeheizten
Bombe bei ioo hydriert. Dann wir(' vom Nickel befreit, der Isopropylalkohol abdestilliert
und der Rückstand in verdünnter Salzsäure gelöst. Von wenig Ungelöstem wird abfiltriert
und die saure Lösung mit Sodalösung alkalisch gestellt. Die abgeschiedene Base wird
abgetrennt und mit der zehnfachen Menge 46°/oiger Bromwasserstoffsäure so lange
gekocht, bis alles in Lösung gegangen ist (etwa 3 bis 4 Stunden), dann wird mit
Wasser versetzt, ausgeäthert und der wäßrige Auszug im Vakuum auf ein kleines Volumen
eingedampft, wobei sich das Dibromhydrat der a-Benzvl-a, ß-diaminopropionsäure mit
einem Zersetzungspunkt von 250 abscheidet.2. zugegeben. Nachdem die Reaktion abgeklungen ist, kocht man noch kurze Zeit unter
Rückfluß, bis neutrale Reaktion eingetreten ist. Man versetzt mit Wasser und saugt
den Phenacetylaminop-methoxybenzylcyarressigsäuremethylester ab. 74 9 davon werden
wie in Beispiel 2 der Hydrierung unterworfen. Die weitere Aufarbeitung nach Beispiel
ergibt a-(p _Methoxybertzyl)-a, ß-diaminopropionsättrebromhydrat vom Zersetzungspunkt
