Verfahren zur Gewinnung von Nucleinsäure aus Hefe oder hefeähnlichen Mikroorganismen Es ist bekannt, Nucleinsäure aus Hefe mit Hilfe von Natronlauge oder Kochsalzlösungen durch Spaltung der Protein-Nucleinsäure-Verbindungen zu gewinnen. Die Ausbeuten betrugen höchstens etwa 2,4 bis 4°%0 Nucleinsäure, bezogen auf Hefetrockensubstanz. Bei Verlängerung der Extraktionsdauer ließ sich die Ausbeute nicht steigern, da hierbei die Gefahr einer Spaltung der Nucleinsäure durch die lange Erhitzung gegeben war. Daneben wurden auch Verfahren bekannt, wonach man aus der Hefe zunächst die Nucleinsäure in der Form ihres Eisensalzes oder eines anderen Schwermetallsalzes abschied, dieses mit wäßriger Natriumacetatlösung auslaugte, vom abgeschiedenen Eisenhydroxyd absaugte und aus dem Filtrat die Nucleinsäure mit Salzsäure unter Zusatz von Alkohol fällte (Deutsche Patentschrift 448 947).According to these procedures, for which until today only living ones have been successful or fresh killed yeast was used, the nucleic acids go into solution and can be precipitated from this by acidification.
