Description
It has been found that the hitherto unknown 2-phenyl-3-aminobutane by methods customary for amine syntheses, for example by condensation of 2-phenylbutanone- (3) with hydroxylamine or hydrazine and reduction of the oxime or azine thus obtained, or by conversion of the Carboxyl group of a · yS-dimethyldihydrocinnamic acid in a conventional manner, for example by AU construction according to Hofmann or Curtius, into the amino group. A particularly elegant conversion of the carboxyl group into the amino group consists, according to KF Schmidt, in the reaction of the acids with nitric acid in the presence of a suitable catalyst, for example conc. Sulfuric acid.
Compared to i-phenyl-2-aminopropane, which is known as a stimulant or i-phenyl-2-methylaminopropane, the new compound is characterized by a shorter duration of action. It is therefore better in hand, signs of fatigue z. B. temporarily in the evening without, if not unnecessarily administered large doses later falling asleep suffers. Also owns the 2-Phenyl-3-aminobutane has a milder effect than the known substances, so that even with considerable increase in the administered doses does not exceed a certain degree of effectiveness will. In this way, the temperature increase achieved at about 0.04 g per kg do not increase the dose significantly, while with the known compounds Administration of increasing doses an increase in temperature reaching the limit of viability can occur.

25 S a' /3-Dimethylhydrozimtsäure werden in . ioo cm3 Chloroform gelöst. The solution is concentrated with 50 cm 3. Sulfuric acid underlayer; 16 g of sodium azide are then added in portions to the reaction mixture, so slowly that the temperature of the reaction mixture remains at 40 to 45 °. This temperature is held for a further 4 x / ½ hours until the evolution of gas ceases and the reaction mixture is then poured onto ice. From the aqueous phase can be prepared by addition of alkali and extracting with ether 14.9 g (7 1 Vo of theory) of 2-phenyl-3-aminobutane isolate (Kp, 8 _ 14 = 87 to 90 0; Kp 8 = 72 0).
Man erhält in nahezu quantitativer Ausbeute 320 bis 330 g Säurechlorid (Kp4 = 96 bis 97°). 300 g of dimethylhydrocinnamic acid are heated to the boil for 4 to 5 hours in 800 cm 3 of benzene with 240 g of thionyl chloride. After the evolution of hydrogen chloride has ended, the mixture is distilled in vacuo. 320 to 330 g of acid chloride (boiling point 4 = 96 to 97 °) are obtained in an almost quantitative yield.
The acid amide is filtered off with suction and schmälzt recrystallized from formic acid 0 5o ° / 192 0th Yield: 93% of theory.
The ofjS-Dimethylhydrozimtsäureamid will now by means of the Hofmann degradation in the 2-phenyl-3-aminobutane converted (yield 70 to 80 ° / 0 calculated on the acid amide); Kp 8 = 72-73 0th
Bei 12 mm und 94 bis 960 gehen ungefähr 9 g (61 % der Theorie) 2-Phenyl-3~aminobutan über.14.8 g of 2-phenylbutanone- (3). are refluxed for 1 hour in 100 cm 3 80% alcohol with 7.5 g hydroxylamine chlorohydrate and 25 cm 3 20% sodium hydroxide solution. After cooling, it is acidified with dilute sulfuric acid, the oxime is suctioned off and washed with a little alcohol. • The oxime is dissolved in 100 cm 3 of alcohol by heating and reduced to the amine by adding 6 g of sodium in portions. The alcoholic solution is diluted with water, the base is extracted with ether and the ether residue is distilled in vacuo. At 12 mm and 94 to 96 0 , about 9 g (61% of theory) of 2-phenyl-3-aminobutane pass over.
