Verfahren zur Herstellung hochwirksamer Stoffe aus Hypophysenhinterlappen
Die Gewinnung wirksamer Stoffe aus dem Hinterlappten der Hypophyse ist bereits aus
dem Schrifttum bekannt. So benutzte Fühner (Zeitschrift für die gesamte experimentelle
Medizin, Bd. i, S. 399/400, 1913, Therapeutische Halbmonatshefte 34/ig2o,
S. 437) wie auch andere Autoren zur Fällung der wirksam-en Substanzen wäßriger Hinterlappenauszüge
Phosphorwolframsäure. Dadurch wurde zwar ein wirksamer Niederschlag erhalten, es
gelang jedoch-nicht, aus einem solchen Nied-erschläg die einzelnen Wirkstoffe in
guter Ausbeute abzutrennen. (Vgl. Abderhalden, Handbuch der biologischen Arheitsmethoden,
Abt.V, Tei13, S.I027 bis 1029/1936.) Es ist ferner bekannt (Patent 264 1 ig), daß
man aus vorgereinigten eiweißfreien Hypophysenextrakten durch Fällung mit Schwermetallsalzen,
insbesondere Quecksilbersalzen, den wirksamen Anteil ausfällen und ihn nach Entfernen
des Metalls in fester Form erhalten kann. Nach diesem Verfahren wird jedoch. nur

Form und fast iooo/oiger Ausbeute erhält, -,vemi man den durch Ausfällung der Wirkstoffe
mittels Säureanhydriden der 6. Gruppe des periodischen Systüms und der sich
davon ableitenden H#eteropolysäur,en, wie- Phosphorwolframsäure, Wolframsäute, Molybdänsäupe,
Phosphormolybdänsäure u. a., bei tiefer Temperatur erhaltenen wirksamen Niederschlag
vom Fällmittel in der Weise befreit, daß man den Niederschlag mit ein-er Lösung
oder Suspension von Erdalkalihydroxyden, z. B. mit methylalkoholischerBariumhydroxydlösung
in wasserhaltigen organischen Lösungsmitteln für die Wirkstoffe bei Temperaturen
unter o', zweckmäßig bei solchen unter -io', behandelt.
Das auf
diese Weise in bis zu iooo/oiger Ausbeute erhaltene und gegebenenfalls durch mehrmaliges
Umfällen gereinigte Endprodukt zeigt die Wirkungen der in dem Ausgangs, material

schwacher Essigsäure (o,25%ig)
gekocht und abfiltriert, und dann wird der Niederschlag mit Wasser gewaschen, bis
das Filtrat ,etwa 2,5 1 beträgt. Nach Abkühlen des Filtrats auf o' wird es
mit der berechneten Menge Phosphorwolframsäure (80 ccm i o %i"--er Säure in i o
% iger Schwefelsäure gelöst) gefällt, nach einstündigem Stehen wird im Kühlraum
zentrifugiert und der Niederschlag mit einer geringen Menge verdünnter (etwa
3 %iger) Phosphorwolframsäurelösung gewaschen. Der Niederschlag wird dann
in 400 ccm bis auf - io' abgekühltem Methylalkohol aufgeschwemmt und mit
ebenfalls entsprechend abgekühlter methylalkoholischer gesättigter Barytlauge bis
zur deutlich alkalischen Reaktion (Phenolphtal-ein) versetzt, wozu etwa
100 Ccm gebraucht wer-,den. Nach 30 Minuten wird im Kühlraum zentrifugi,ert,
die Lösung abgegossen und der Niederschlag mit wenig Methylalkohol gewaschen; die
vereinigten methylalkoholischen Lösungen werden zwecks völliger Ausfällung des Bariums
mit verdünnter Schwefelsäure genau neutralisiert, wobei man zweckmäßig die Beendigung
der Fällung durch Zusatz einer kleinen Menge festen Natriumsulfats vornimmt. Der
Bariumsulfatniederschlag wird abfiltriert und das Filtrat evtl. nach schwachem Ansäu#ern
mit Essigsäure im guten Vakuum bis zur Trockne eingedampft. Der

das Eilt gefällt. Die abfil-
trierte Fällung wird in. völlig trockenem Zustand,e in ioccm Eises,sig (98%ig) g-.löst,
die Lösung klar filtriert, das Filtrat mit 4o ccm. Äther versetzt und die entstandene
Fällung mit Äther gewaschen und getrocknet. Ausheute: 5oo bis 250 mg mit
insgesamt 6o ooo i. E.; i mg enthält demnach 24o bis 120 i. E. der drei genannten
