Verfahren zur Darstellung des wirksamen Bestandteils des Calebassen-Curare
in kristallisierter, reiner Form Curarewirkung, d. h. die Wirkung von südamerikanischen
Pfeilgiften, ist pharmakologisch schon eingehend beschrieben worden; es sind auch
schon verschiedene Versuche gemacht worden, diese Wirkung therapeutisch zu verwenden.
Der wirksame Stoff der verschiedenen Curarearten oder vielleicht richtiger die wirksamen
Stoffe dieser Pfeilgift,. sind bisher noch nicht in einheitlicher, reiner Form gewonnen
worden, obwohl Versuche dazu schon wiederholt gemacht worden sind (vgl, R. B o e
h m , Heffters Handbuch der experimentellen Pharmakologie, Band z, i. Hälfte d9271,

Die Bearbeitung des Calebassen-Curare, einer der verschiedenen schon
lange bekannten Curaresorten, hat nun zu einem kristallisierter., einheitlichen
Salz des Stoffes, der die hohe Giftigkeit des Calebassen-Curare ausmacht, geführt.
Um das zu erreichen, braucht man eine Reinigung in mehreren Stufen. Die wesentlichen
Schritte sind Reinigung der Reineckate des wirksamen Anteils der Droge durch chromatographische
Adsorption, fraktionierte Fällung der schon vorgereinigte n salzsauren oder anderen
Salze und dann Bildung eines i3-anthrachinonsulfonsauren Salzes. Aus diesem läßt
sich eine Base abtrennen, die ein kristallisiertes jodmethylat liefert. Man erhält
aus der Base selbst ein kristallisiertes Pikrat. Sowohl das (3-anthrachinonsulfonsaure
wie die anderen Salze sind durch außerordentlich hohe Wirksamkeit im Tierversuch
ausgezeichnet. Es handelt sich um eine der wirksamsten Pflanzenbasen. Die Wirksamkeit
dieser Salze ist mehr als i omal größer als die der bisher beschriebenen besten
Präparate. Wesentlich für die Kenntnis des Wirkstoffes ist die Feststellung, daß
es sich nicht, wie man bisher glaubte, um eine quaternäre Base handelt. Die wässerige
Lösung der Base reagiert gegen Lackmus nur . mittelstark alkalisch,
gegen
Phenolphtalein undeutlich. Sie schmeckt ungemein bitter. Für die Abtrennung des
wirksamen Stoffes ist weiter wichtig wissen, daß die Base bei längerer Berilli"
rung mit überschüssiger Mineralsäure :;ich'" dunkel färbt; daher müssen saure Lösungi?@f'i
mit Vorsicht behandelt werden. Zur Keniazeichnung und zur Verfolgung der Reinigung
sind auch die schönen Farbreaktionen (Griinfärbung mit Salpetersäure, Blaufärbung
mit Bichromatschwefelsäure) geeignet. Beispiel Die wässerige Lösung des Rückstandes
eines Methanolauszugeswird, wie bekannt, mit einem mäßigen überschuß von Quecksilberchlorid
gefällt. Der Niederschlag wird in Alkohol bei 5o bis 6o° mit Schwefelwasserstoff
zerlegt und der Alkaloidanteil in wässeriger Lösung in das Reineckesalz übergeführt.
Das Reineckesalz wird in sehr verdünnter Acetonlösung durch eine Säule von Aluminiumoxyd
geschickt und die Acetonlösung durch reines Aceton verdrängt. In dem Aluminiumoxyd
bleiben beträchtliche Mengen störender, unwirksamer Verunreinigungen zurück. Die
noch immer rotbraun gefärbte Acetonlösung wird im Vakuum eingeengt, mit verdünnter
wässeriger Salzsäure versetzt und die R@eineckesäure durch Äther entfernt. Dann
wird die wässerige Lösung der rohen Hydrochloride im Vakuum zur Trockne gebracht.
Dieses Salzgemisch wird in 1 o Teilen Alkohol und 3 Teilen Aceton gelöst und langsam
mit der q.ofachen Menge Äther gefällt. Es scheidet sich der größte Teil. der zur
Fraktionierung eingesetzten Substanz als dunkelbraune, ,schmierig werdende Masse
ab. Durch weiteren Zusatz von 13 Teilen Äther wird noch k@irF hellgelbe Zwischenfraktion
ausgefällt. e'Lösung ist fetzt hellzelb zeworden. Sie
vorsichtig verdampft, der Rückstand in
Wasser gelöst und sehr langsam bis
@( ken Trübung mit einer kalt gesättigten,
wässerig-alkoholischen Lösung von (3-anthrachinonsulfonsaurem Natrium versetzt.
Dann erwähnt man gerade bis zur Lösung und läßt langsam abkühlen. Nach 1 Stunde
haben sich hellgelbe Nadeln in Büscheln abgeschieden, die völlig einheitlich aussehen
und sich aus heißem Alkohol, zu dem man die gleiche Menge Wasser tropfen läßt, leicht
umkristallisieren lassen. F. gegen 278" (unter Zersetzung). Aus den Vorstufen
der Hydrochloride lassen sich durch wiederholte Fraktionierung noch erhebliche Mengen
