Abstract
Description
The invention relates to a slide according to the Preamble of claim 1.
Such a slide is known from US-PS-4 501 496. The floor areas consist of a row from parallel to the bottom of the base plate, quadra tables, each surrounded by a channel are surrounded by a recess stands as the task for a drop of liquid serves. There is a recess in each floor area notch formed open to the canal, which the rivers directs liquid against the floor area. Between groups of floor areas in the row are transverse ridges exercise areas against which a cover strip made of stiff, transparent glass or plastic mate rial, the sample chambers with the bottom areas certain height defined. To connect the reason plate with the cover strip serves a three edges the frame surrounding the base plate, in which the Cover strip is tightly fitted. With this known slide is disadvantageous that the cover strip and / or the base plate due to the on can bend the tensioning seat of the cover strip, so that when microscoping blurs occur on the Reliability of the investigation can affect nen. There is also the risk that the counter will be falsified results from bending of the cover strip result because the sample chambers differed absorb liquid volumes and the meter nisse are not representative. Furthermore, it can sem slides happen that because the cover strip only rests loosely on the base plate, the channels do not are tightly closed to each other so that the samples different chambers mix what also falsified the result of the investigation. Training of the catchment area as a notch also hides the Ge drive from inaccuracies because the notch does not the entire sample chamber between the bottom area and Ab deck area with an even liquid film can supply. The user is at the liquid distribution by pressing against the cover strip need to help, causing counting solid particles can migrate into the channel in the liquid. A Another disadvantage of this slide arises for the Laboratory workers due to leaks between the base plate and masking strips so that the samples con can damage.
A slide known from DE-OS-23 18 837 consists from a base plate to which the cover areas are connected the bottom areas of the sample chambers to the Base plate are molded. Such slides are expensive because they are used to achieve sample chambers volume must be worked extremely precisely, d. H. high precision tools are required.
Another slide is in the EP publication 0 210 071 A2. In this case, both the base plate and the cover strip as stiff Profile parts designed, the base plate of the Bottom area pushed out upwards par tien and the cover strip on this matching Has depressions. When the Pro Fil parts create floor and cover areas semicircular shaped sample chambers. Crossing lengthways and crossways running ribs on the underside of the cover rich cover strip are as energy directional effective when the two profile parts ver along the crossing ribs are welded. In the individual square fields between the ribs are the sample chambers, that are not blocked off from the fields. As a pull aid is used in each case an incline ramp, which is between the longitudinal edge of the base plate and straight outer End of the floor area extends. The curved edges the sample chamber areas in the base plate and the Ab Cover strips are not connected to each other and each of three ribs of the cover strip and the bow of the Specimen chamber bounds a cavity in field by notches in the curved edge of each Cover area of the masking strip of excess rivers liquid can enter the sample chamber. Everyone Cavity exercises on the sloping ramp in the pro liquid drawn into the chamber has a certain suction kung, which, if the course of the investigation is rapid, for the even distribution of the sample in the sample came mer is cheap, but in the event of interruptions in the sub depending on the length of the delay, that the liquid from the sample chamber into the hollow space disappears and the liquid to be counted volume in the sample chamber is no longer correct. Here this results in inaccuracies in the sample sub search. These are also favored by the fact that two injection molded plates profile parts are inhomogeneous and when welding connection shrink, reducing the precision of the ab stands between floor area and cover area is affected. The cross-shaped welding of the two rigid profile parts is also unfavorable because the fields different voltages from each other through which the overall structure warps can. The cover strip becomes wavy, causing the on volume of the individual sample chambers changes and observation disorders when microscoping on tre When not in use, the undulations of the Cover strip for stacked slides to Ver scratches of the masking tape that make the observation field when microscoping and distort the examination complicate or even falsify.
The object of the invention is an object to train carriers of the generic type so that he with inexpensive manufacturability exact, representative Test results guaranteed.
This object is achieved in that in one with respect to the flat top of the bottom plate lowered longitudinal end of central zone min at least a series of recesses is embedded, which each form the bottom area of a sample chamber and to each of which there is an outward slant ramp connects that in the central zone the Bodenbe rich bordering on supporting boundary bars forms that are below the top of the base plate te end and that the transparent cover strip straight-edged section of film is exclusive is connected to the boundary bars and with a straight longitudinal edge the inclined ramps of each row from Ver deeply protrudes a bit.
Such a slide can be inexpensive from a rigid base plate and an unprofiled, produce straight-edged film section, the Surface of the film section in relation to the upper side of the base plate is sunk, so that at Stape slides do not scratch the viewing window zen can. Clear, undistorted perspective made easier microscopy and improves counting accuracy. This advantage is further increased by the fact that Foil due to its connection only with the Limiting webs of the floor areas no tension exerts on the base plate so that it remains straight with the result that all sample chambers are exactly the same Record liquid volumes and the count result is representative. Each sample chamber is self contained closed and is only connected to the sloping ramp dung. Passages to regions of the base plate are missing outside the sample chambers, so that even with longer ones Interruption of the examination in all sample chambers the same wetting films are present, the exact ones Guarantee counting results. As for the masking tape only the cutting costs are incurred literally very high quality film sections used that are homogeneous in terms of thickness and density and distortions, e.g. B. by streaking shut down. Also the absence of tension of the entire An order is a consequence of the homogeneity of a film, the by far exceeding that of injection molded parts. Only the base plate is injection molded poses; it takes over all functions of the object carrier gers. Base plate and section of film are expedient sometimes made of sterilizable plastic, such as glass rem polycarbonate or others.
In an advantageous embodiment of the invention is pre see that the bottom areas are asymmetrical semicircle have that the boundary webs of the arch shape follow and that one behind the curved boundary towards the length of the central zone web, which is connected to the film section ends connected to crossbars, each in the because the last curved boundary web of a series of Skip deepening. With only one row of longitudinal recesses is located on the outer edge of the sloping ramp opposite edge the base plate. With two parallel rows of vertie the longitudinal web runs between the two facing boundary webs of the floor areas of the two rows. In some of the crossbars, preferably the diametrically opposite crossbars, of two parallel rows of wells is min least a ventilation opening is formed. The ventilation openings prevent air pockets from being trapped when welding the webs to the film section, see above that the surface of the slide has no bulges Has.
All bars of the base plate are on the upper edge a weak rib serving as a welding aid see. This disappears after welding and the The edge of the webs forms the support for the corresponding the foil areas.
Because the straight-edged film section with a straight The sloping ramps of each row of depressions run along the longitudinal edge The catchment areas of the Sample chambers free of tension and the liquid drop has no slope instead of into the sample chamber, to flow along the edge and this as a gas outlet to block required passage. Feeding the Liquid into the sample chamber is made according to the invention improved by the fact that on one side of the floor cheshed-in line trace is formed, which extends from the outer edge of the sloping ramp to the Limiter of the floor area extends and fine The most striations form in the interior of the sample chamber to lead. With asymmetrical semicircular shape of the Bodenbe the line trace on the asymmetrical side is rich intended. This is particularly the case when a etched counter field on the floor area cheap, because then the counting fields continue from the moving aid removed and into an even wetting zone between floor area and cover area. The Attaching the counting fields to the base plate is green stig, because when microscoping they are simultaneously with the Wetting film of the liquid is sharp. The size the counting field changes with the height of the samples chamber. The lower the sample chamber height, the larger the counting field. The sample chamber is preferably made in this way placed so that it always contains 1 µl volume in the counting field. The preferred chamber height is in a range between 0.130 to 0.140 mm. The film thickness can be before pulls 0.08 to 0.4 mm, preferably 0.25 mm.
As protection against dripping liquid from the Inclined ramps end the inclined ramps of each row Wells outside on a common, high standing Border part, the top edge of which is in the plane of the top the base plate.
The film section is preferably on the Pro benkkammer facing area with a dried Wet surfactant solution. This way, a quick le, even distribution of the rivers to be examined liquid sample reached in the sample chamber without their relevant properties change.

A base plate 10 of a slide is made of crystal-clear, sterilizable plastic, such as polycarbonate, and the blank is provided with a flat, unprofi-treated underside 11 and a central zone 12 lowered with respect to the flat top 13 . The central zone 12 extends from a transverse side of the elongated, rectangular base plate 10 to the other and at each of the two longitudinal edges there is a longitudinal, straight elevation which defines the upper side 13 . The height between the underside 11 and the surface 13 is preferably 2.2 mm. The height of the elevations with respect to the base of the central zone 12 is dimensioned at 0.5 mm. The height of the elevation of the central zone 12 over the bottom of the cavities 30 and 31 is 0.7 mm.
The profiled base plate 10 , which is shown in plan view in FIG. 5, is produced from this blank by injection molding.
In the top of the central zone 12 , two parallel rows of depressions 20 and 21 are embedded, which lie next to one another in the longitudinal direction of the base plate 10 . All recesses 20 and 21 are identical. They each consist of a floor area 22 parallel to the flat underside 11 of the base plate 10 , which is surrounded by an asymmetrically semicircularly curved limb 23 and at the straight, outwardly facing edge 24 of which an inclined ramp 25 closes at the level of Bottom area 22 drops outwards. The closed arches of the boundary webs 23 of the two rows of recesses 20 and 21 face each other. Between them extends on the longitudinal central axis of the base plate 10, a longitudinal web 28 which extends from one narrow end of the base plate 10 to the other and from which cross webs 26 , 27 extend only at the ends on both sides. The transverse webs 26 , 27 and the curved boundary webs 23 enclose a flat cavity 30 , 31 which has a straight and a toothed side. In the crosspiece 26 at one end and the crosspiece 27 at the other end of the Grundplat te 10 is a ventilation opening 32 , which connect the cavities 30 , 31 with the outside air.
The curved boundary webs 23 , the longitudinal web 28 and the transverse webs 26 , 27 are lower than the upper side 13 defined by the elevations. In the same plane as the top 13 end edge parts 34 , which delimit the outer edge of the inclined ramps 25 and start from the same height gusset parts 36 , which are irregularly trapezoidal in plan view and with their lower ends at the outer ends of the curved limiting webs 23 connect that are slightly lower.
All webs 23 , 26 , 27 and 28 are provided on the upper edge with a flat rib 33 , which serves as a welding aid and when welding a film section 40 disappears. The film section 40 is quite angular, has the same length as the base plate 10 and is as wide as the lowered central zone 12 between the two elevations ( FIGS. 1 and 2). In the area of the inclined ramps 25 , the straight longitudinal edge of the film section 40 protrudes somewhat towards the outside, so that the dividing line 24 is surmounted a short distance. The longitudinal edges of the film section 40 lie on the runners 23 a of the arcuate boundary webs 23 , the height of which corresponds to the height of all webs.
On the surface of the bottom region 22 of each recess 20 and 21 , a square, grid-shaped counting field 41 is etched. A greatly enlarged view of a counting field 41 is shown in FIG. 6. Double lines 46 are present in the outer frame. In addition, every fourth line of the counting field 41 rasterized by single lines 47 is a double line 48 . This makes counting easier when determining the sediment. Advantageously, the 9 × 9 grid checks 49 and lines 46, 47, 48 are dimensioned as indicated in the drawing. The result is 9 × 0.303 mm = 2.727 mm; 2 x 0.04 mm = 0.08 mm; 2 × 0.05 mm = 0.10 mm a counting field 41 of the size 2.907 mm.
The straight-edged film section 40 covers the Bo den areas with a uniform distance ( Fig. 4) and bil det with these sample chambers 45 , into which a drop placed on the inclined ramp 25 of the liquid to be examined is drawn in via the pulling aid. The film section 40 retains its wave-free, smooth shape as a result of the welding only with the webs 23 , 26 , 27 , 28 , so that an exact microscope without distortion is possible.
The preferably described embodiment of the film section, wetted with a dried surfactant solution solution on the surface facing the test chambers for example by immersing the film on one side in a surfactant solution or by spraying it and then drying with, for example, warm Air, radiation or another energy source receive.
Anionic, non-ionic and / or amphoteric surfactants or preferably the mixture used surfactants. Can be used as anionic surfactants Carboxylates such as sodium palmitate or potassium oleate, Sul fates such as sodium 1-decyl sulfate and sulfonates such as natri um-p-1-tetradecylbenzenesulfonate can be used. When Non-ionic surfactants can, for example, glycerol dioleate, sorbitan mononopalmitate etc. can be used. As amphoteric surfactants, for example betaines or Sulfobetaines such as N-carboxymethyl-N, N-dimethyltetradecy laminium salt can be used.
The surfactant mixture is preferably in one weight ratio "anionic surfactants: non-ionic surfactants: amphoteric surfactants "such as" 15-30: 1-7: 1-7 "in particular "20: 5: 5" used.
The surfactant solution is, for example, in a conc tration of 0.05-0.5%, preferably 0.1-0.2% set.
