Description

Process for the preparation of a culture medium for influenza bacilli and for the manufacture of vaccines from them. It is known that influenza bacilli can only be bred and developed with difficulty. The breeding happened usually in such a way that sputum from influenza patients or autopsy material can be obtained Inoculated blood plates.

It has now been found that it is better to breed influenza bacilli and can bring about more abundant growth if one proceeds in the following way: Streptococci and pneumococci are grown separately in a conventional manner. After about 24- b: s 48-hour growth, the cultures, just_a-1s, are separated for some time, about: 3 to 4 hours, in a shaking machine: ne for the purpose of pouring out: ns all - r soluble Substances shaken out at incubation temperature and then used for the purpose of After killing the bacilli, heated to 50 °. Made by both this way Cultures are roughly equal parts, e.g. B. each i to a ccm, m: t fresh venous blood, for example from a rabbit, mixed and processed into nutrient media with agar or the like. Influenza bacilli are grown in pure culture on the plates produced in this way. These then show a conspicuous, lush growth compared to the usual Methods obtained influenza bacilli. In this way it is much easier to get bigger cultures. -The influenza bacilli obtained in this way are for further processing particularly suitable for vaccines. As a result of the lush growth, they can be Rinse easily from the culture media by treatment with physiological saline solution and process them into vaccines.

It has also been found that the influenza bacilli thus obtained particularly useful vaccines of excellent healing properties result if one after they have been killed by depriving them of the nutrient medium, in a suspension of saline solution with weakly acidic substances, such as B. mixed acid or Tricresol, treated in such a way and for so long that only some of the bacilli are broken open while the other part remains. The setting and dilution of the vaccines treated in this way are carried out in such a way that the suspension is approximately .5oo Contains millions in 1 cc of influenza bacilli.

With the vaccines obtained in this way, unmistakably specific ones are obtained Medicinal effect for flu and flu pneumonia.

Im Zentralblatt für Bakteriologie, I. Abtlg. Bd. 28, S. 744, erwähnt C a n t a ni, daß Influenzabazillen aus Eiweißnährbödcn, denen Gcnokck_:en, D_ph:heriebazi=1_noder Staphylokokken lebend oder sterilisiert zugesetzt waren, erheblich besser wachsen sollen, während Streptokokken keine besondere Wirkung entfalten. In the Zentralblatt für Bakteriologie, I. Abtl. Vol. 28, p. 744, Cantani mentions that influenza bacilli from protein nutrients to which Gnokck_: en, D_ph: heriebazi = 1_n or Staphylococci = 1_n or Staphylococci were added alive or sterilized, should grow considerably better, while Streptococci do not have any particular effect.

The essential difference of the present method from the just mentioned is readily apparent. Because according to the present proceedings become streptococci, with which C a n t a n i has not achieved success, and pneumococci, which he has not mentioned and examined at all, bred separately for himself and shaken out to draw out the secretions, then heated to about 5o "to kill, mixed with blood and made nutrient media from the mass thus obtained, whereupon then the influenza bacilli are grown. This way of working is described by C a n t a n i does not, and he does not achieve the results achieved with the present method Success.
