Gegenstände der vorliegenden Erfindung sind ferner ein Verfahren zur Herstellung der Verbindungen der Formel (I)
sowie pharmazeutische Zusammensetzungen (Mittel bzw. Präparate) , die sie als aktive Komponenten bzw. Wirkstoffe
enthalten. Verbindungen der allgemeinen Formel

worin R, X und Y die oben angegebenen Bedeutungen haben,
sind bereits bekannt. Einige dieser Verbindungen werden in großem Umfange in der Therapie als antibakterielle
Mittel verwendet, insbesondere zur Behandlung von Harnwegsinfekten.
/ So weist beispielsweise die 1-Ethyl-6-fluoro-1,4-dihydro-4-oxo-(1-piperazinyl)-chinolin-3-carbonsäure
oder Norfloxacin (Formel II, worin R = Ethyl, Y = CH und X= CF ), wie sie in der BE-PS 870 917 beschrieben ist, ein breites Aktivitätsspektrum
und eine gute antibakterielle Aktivität auf, die höher ist als diejenige der Pipemidinsäure (Formel
II, worin R = Ethyl und X und Y = Stickstoff), die zusammen mit Nalidixinsäure als Vorläufer für die Klasse
von antibakteriellen Chinolin-Mitteln angesehen werden
können.

Nach Norfloxacin wurden auch andere Derivate dieser Reihe synthetisiert, die charakterisiert sind durch die
Anwesenheit eines Fluoratoms in der 6-Stellung, dazu gehören Cyprofloxacin (Formel II, worin R = Cyclopropyl, Y
= CH und X = CF), Pefloxacin (ein Norfloxacin-Analogon,
methyliert am Piperazinring), Ofloxacin und Enoxacin,
welche die meistversprechendsten Verbindungen zu sein scheinen.
3s wurde nun überraschend gefunden, daß Verbindungen der
obenangegebenen Formel (I) solche pharmakologischen Merkmale aufweisen, daß sie in der Therapie wertvoll sind in
Form geeigneter pharmazeutischer Präparate, wobei ihr antibakterieller Effekt und ihr breites Aktivitätsspektrum
15 besonders vorteilhaft sind.
Unter den Verbindungen der Formel (I) besonders bevorzugt
sind solche, in denen R eine Ethyl- oder Cyclopropylgruppe und X eine^C-F-Gruppe bedeuten, wobei Y entweder ein
Stickstoffatom oder eine^C-H- oder^C-F-Gruppe darstellt.
R1 und R" können beide Wasserstoff bedeuten oder einerder
beiden Reste bedeutet Wasserstoff und der andere ein Chlor-, Brom- oder Jodatom in der 2-, 3- oder 4-Stellung;
eine Methyl-, Trifluoromethyl- oder Methoxygruppe; einschließlich
der Doppelsubstitution an dem Phenylring,
wie z.B. in den 2,4-; 3,4-; 3,5-; 2,6-Stellungen mit Gruppen, die gleich oder verschieden sein können.
QQ Die Verbindungen der Formel (I) können mit geeigneten
organischen oder anorganischen Basen in ein Salz überführt werden; Beispiele für geeignete Salze sind die Natrium-,
Kalium-, Calcium-, Magnesium-, Ammonium-, Triethylamin-, Trometamol-, Urotropin-, Lysin-, Arginin-,
3g Procain-, Morpholinsalze und dgl.
Die Verbindungen der Formel (I) werden hergestellt durch Umsetzung von Verbindungen der obenangegebenen Formel
(II), worin R, X und Y die obenangegebenen Bedeutungen
haben, mit Phenylacetaldehyden der allgemeinen Formel R1

worin R1 und R" die oben in bezug auf die Formel (I) angegebenen
Bedeutungen haben.
Die Umsetzung wird durchgeführt unter Anwendung einer sauren Katalyse unter Verwendung katalytischer Mengen
von Schwefelsäure, Chlorwasserstoffsäure, p-Toluolsulfonsäure
und dgl. Das Reaktionslösungsmittel kann ausgewählt werden aus wasserfreien aromatischen Kohlenwasserstoffen,
wie Benzol, Toluol oder Xylol; wasserfreien halogenierten Kohlenwasserstoffen, wie Methylenchlorid
oder Chloroform; Äthern, wie Tetrahydrofuran, Ethylather;
aliphatischen Kohlenwasserstoffen, wie Hexan, Cyclohexan, Petroläther und dgl. Wasserfreie aromatische Kohlenwasserstoffe,
insbesondere Benzol und Toluol, sind am meisten bevorzugt.
Die Umsetzung wird in der Regel bei der Rückflußtemperatur
des Lösungsmittels durchgeführt für einen Zeitraum, der erforderlich ist, um eine geeignete Umwandlungsausbeute
zu erzielen.
Die Verbindungen der Formel (I) werden in Form von Säuren erhalten und unter Anwendung konventioneller Methoden,
beispielsweise durch Filtrieren, gewonnen.
Die entsprechenden Salze können erhalten werden durch Umsetzung mit Alkali- oder Erdalkalicarbonaten, -bicarbonaten
oder -hydroxiden oder mit den obengenannten Aminen.
Die Überführung in ein Salz kann in Wasser oder in anderen geeigneten Lösungsmitteln durchgeführt werden.
Die Erfindung wird durch die folgenden Beispiele näher } erläutert, ohne jedoch darauf beschränkt zu sein.

5,66 g (1,77 χ 1O~2 Mol) 1-Ethyl-i/4-dihydro-4-oxo-7-(1-piperazinyl)-chinolin-S-carbonsäure

(1,77 χ 10 Mol) 2-(2-Chlorophenyl)acetaldehyd und eine katalytische Menge p-Toluolsulfonsäure wurden in.150 ml wasserfreiem Toluol miteinander umgesetzt.

und mit Methanol heiß gewaschen. Auf diese Weise erhielt man 6,9 g (Ausbeute 85 %) 1-Ethyl-6-fluoro-1,4-dihydro-4-oxo-7-(4(2-(2-chlorophenyl)ethen-1-yl)-1-pipe

Die antibakterielle Aktivität der Verbindungen der Beispiele 1 bis 3 wurde bewertet im Vergleich zu derjenigen
von Norfloxacin durch Messung der minimalen Hemmkonzentration
gegenüber grampositiven und gramnegativen Bakterien. Stufenförmige Verdünnungen der Testverbindungen
in Dimethylsulfoxid (DMSO) wurden in Mueller-Hinton-Agar-Platten inokuliert auf Endkonzentrationen innerhalb
des Bereiches von 0,1 bis 100 jig/ml. Die Platten, die vorher
mit einer Brühenkultur der Bakterien inokuliert worden waren, wurden 48 Stunden lang bei 35 C inkubiert. Es
wurden Stämme verschiedener nationaler und internationaler Sammlungen zusammen mit Stämmen aus Krankenhausisolaten

Die vorliegende Erfindung bezieht sich auf alle industriellen Aspekte bezüglich der therapeutischen Verwendung
der Verbindungen der Formel (I) oder ihrer Salze als antibakterielle Arzneimittel.
Ein Hauptaspekt der vorliegenden Erfindung sind daher pharmazeutische Zusammensetzungen (Mittel bzw. Präparate)
für die orale, parenterale oder topische Verabreichung,
die als aktiven Bestandteil (Wirkstoff) therapeutisch wirksame Mengen der Verbindung der Formel (I) oder von
Salzen davon zusammen mit konventionellen Hilfsstoffen oder Verdünnungsmitteln enthalten.
Zu pharmazeutischen Präparaten für die orale Verabreichung gehören Kapseln, Tabletten, mit Zucker beschichtete Tabletten,
Granulate, Sirupe und Lösungen. Zu pharmazeutischen Präparaten für die parenterale Verabreichung gehören
sterile injizierbare Lösungen; zu Präparaten für die topische Verabreichung gehören Cremes ,kleine Eier (ovules),Waschmittel,
Lotionen und dgl.
Die pharmazeutischen Zusammensetzungen bzw. Präparate enthalten auch konventionelle Hilfsstoffe, wie z.B. Bindemittel,-
Konservierungsmittel, Stabilisatoren, Aromastoffe und dgl.
Insbesondere die Kapseln, Tabletten, mit Zucker beschichtete Tabletten und Granulate können Hilfsstoffe
enthalten, wie z.B. Laktose, Calciumphosphat, Mannit,
Stärke, Kaolin und dgl., Bindemittel, wie Traganthgummi,
Gelatine, Maisstärke und dgl.; Schmiermittel (Gleitmittel), wie z.B. Magnesium- oder Calciumstearat, Talk und dgl.;
Disaggregiermittel, wie z.B. Alginsäure, Maisstärke und dgl.; natürliche oder synthetische Lösungsmittel, Aroma-
35 stoffe und Färbemittel.

Die Sirupe und Lösungen für die orale Verabreichung können neben der in Wasser oder in einem wäßrigen Träger
gelösten aktiven Komponente (Wirkstoff) Süßungsmittel, wie z.B. Saccharose, Konservierungsmittel, wie Methyl-
und Propylparaben, Färbemittel und Aromastoffe enthalten.
Die topischen Präparate werden mit einer neutralen Fettbase oder mit wasserlöslichen oder selbstemulgierbaren
Hilfsstoffen formuliert.

Zusätzlich zu oder anstelle der oben aufgezählten Substanzen können
auch verschiedene andere Substanzen verwendet werden, wie für den Fachmann auf dem pharmazeutischen Gebiet an sich
bekannt. Es ist klar, auch wenn darauf hier nicht ausdrücklich hingewiesen wurde, daß alle diese pharmazeutischen
Formulierungen bzw. Präparate in den Rahmen der vorliegenden Erfindung gehören.
Die tägliche Dosis für die orale Verabreichung variiert in Abhängigkeit vom Alter, vom Körpergewicht und dem Zustand
des Patienten innerhalb des Bereiches von 100 bis 2 000 mg, ggf. aufgeteilt in 2 bis 4 Verabreichungsportionen.
Die bevorzugten Einheitsdosierungsformen enthalten

R eine C.-C.-Alkylgruppe oder eine C^-Cg-Cycloalkylgruppe;R' und R", die gleich oder verschieden sein können, Wasserstoff atome , Halogenatome, Methyl-, TrifIuoromethyl- oder Methoxygruppen; undX und Y, die gleich oder verschieden sein können, einStickstoffatom, eine .C-H-Gruppe oder eine .C-F-Gruppe ;sowie die pharmazeutisch akzeptablen Basenaddi^ionssalze derselben.2. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß in der Formel (I) bedeuten:Y eine^C-H-Gruppe,X eine_^rc-F-Gruppe und 5 R Ethyl.3. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß in der Formel (I) bedeuten:Y eine ^C-H-Gruppe,10 X eine^C-F-Gruppe und R Cyclopropyl.4. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß in der Formel (I) sowohl X als auch Y ein Stickstoff-15 atom bedeuten und R für Ethyl steht.5. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß in der Formel (I) bedeuten:X eine^C-F -Gruppe, 20 Y ein Stickstoffatom und R Ethyl.6. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß sowohl X als auch Y^C-F bedeuten und R für Ethyl25 steht.7. Verbindung nach Anspruch 1, gekennzeichnet durch die Formel 1-Ethyl-6-fluoro-1,4-dihydro-4-oxo-7-(4-(2-(2-chlorophenyl) ethen-1 -yl) -1 -piperazinyl) -3-ch inolincarbo'nsäure.8. Verbindung nach Anspruch 1, gekennzeichnet durch die Formel 1-Ethyl-6-fluoro-1,4-dihydro-4-oxo-7-(4-styryl-1-piperazinyl)-3-chinolincarbonsäure.9. Verbindung nach Anspruch 1, gekennzeichnet durch die Formel 8-Ethyl-5,8-dihydro-5-oxo-2-(4-styryl-1-piperazinyl) -pyrido/"2 ,3-d/pyrimidin-6-carbonsäure.-ΒΙΟ. Verfahren zur Herstellung der Verbindungen der allgemeinen Formel (I) nach einem der Ansprüche 1 bis 9, dadurch gekennzeichnet, daß Verbindungen der allgemeinen FormelCOOH(II)worin R, X und Y die in den Ansprüchen 1 bis 9 angegebenen Bedeutungen haben,
umgesetzt werden mit Phenylacetaldehyden der allgemeinen15 FormelR1(Λ "^V-CH -CHO(III)worin R1 und R" die in den Ansprüchen 1 bis 9 angegebenen Bedeutungen haben.11. Verfahren nach Anspruch 10, dadurch gekennzeichnet, daß die Umsetzung in Toluol bei Rückflußtemperatur in Gegenwart katalytischer Mengen von Säuren durchgeführt wird,12. Pharmazeutische Zusammensetzungen mit einer antibakteriellen Aktivität, dadurch gekennzeichnet, daß sie eine wirksame Menge einer oder mehrerer Verbindungen der Formel (I) nach einem der Ansprüche 1 bis 9 im Gemisch mit pharmazeutisch verträglichen Hilfsstoffen oder Verdünnungsmitteln enthalten.
