Description
9 B,.e ispiele.Process for the preparation of akridine derivatives. It has been found that the hitherto unknown compounds, which are obtained by reacting ammonia or primary amines, which are not arylamines, with 9-haloacridines, are valuable for medicinal purposes because of their quinine-like action. After their formation, the new compounds correspond to the general ones Constitutional formula in which the acridine residue is replaced by atomic groups, e.g. B. alkyls or alkyloxy groups, can be substituted. Aus ihm wird- durch, Ammoniak das in Wasser unlösliche gelbe 2-Äthoxy-9-äthanolaminoakridin abgeschieden, welches, aus verdünntem Alkohol kristallisiert, bei 146' schmilzt.1. 14 parts: 2-ethoxy-q-ch "lorakriidin - are heated to 100 for several hours with 6 parts of ethanolanline and 20 parts of absolute alcohol. The precipitated yellow crystal mass, which is the hydrochloric acid salt of the new base, is Sucked off and recrystallized from dilute alcohol; it is easily soluble in water, but less soluble in alcohol. From it, ammonia precipitates the water-insoluble yellow 2-ethoxy-9-ethanolaminoacridine, which, crystallized from dilute alcohol, melts at 146 ' .
2. 6.5 parts of 2-ethoxy-g-dilorakridine and 20 parts of a 3: 2 percent alcoholic ethylamine solution are heated to 1401 for a few hours, with or without the addition of a very small amount of a copper salt. A solution is created, from which the acidification. the hydrochloric acid salt of the new base is separated with dilute hydrochloric acid; it is easily soluble in water, but less soluble in alcohol. The yellow free base 2-ethoxy-9-ethylaminoakri-d-yne, which, crystallized from alcohol, melts at IV to 132- ', is boiled off from the chlorohydrate by means of ammonia.
3. 13 parts of 2-ethoxy-q-chlorakri # din, 7 parts of p-oxyphenylethylamine and 6o parts of alcohol are 8 to. Heated to ioo 'for 9 hours. The resulting solution separates on the addition of. Ammon # ia # the new yellow base, idas 2-ethoxy-q - 4! -Oxyphenylethylaminoakridi-n, - from. To remove any chloracridine that is still present, it is boiled with benzene and then crystallized from alcohol; it melts at 233 '. With hydrochloric acid and sulfuric acid it forms sparingly soluble salts, with glycolic acid it forms an easily soluble salt which melts at 2o8 '.
4-Aminoantipyrine are refluxed with #oo parts of alcohol. The reaction is over in a short time and the 2-ethoxy-q-antipyrylaminoakri-din is precipitated when it cools down. It is separated from the mother liquor and recrystallized from alcohol; it is easily soluble in water. Ammonia separates the yellow-red free base from it, which is insoluble in water and most common organic solvents. is crystallized from pyridine, it melts at: 2570-5. i-iTeileg-C # hhloral,: ri-din, g parts of ethanotamine and 25 parts of absolute alcohol are heated to 100% for about 6 hours. Ammonia is then added and the product which has separated out is filtered off; In addition to Akridon, it contains the new base: 9-Athanolaminoakridin. This is dissolved in dilute hydrochloric acid, separated by ammonia and crystallized from dilute alcohol; it forms long yellow needles which melt at 2o6 '.
6. 7 parts of 9-chloroacridine are heated to 1400 for 5 hours with 50 parts of i2% alcoholic ammonia solution and a little copper acetate. The akridon formed is then filtered off and the solution is evaporated on a water bath.
The residue is taken up in dilute hydrochloric acid, and the 9-aminoacridine is precipitated from the solution with sodium bicarbonate. It crystallized from diluted alcohol in Al yellow needles, the smacking at 23:20, in dilute hydrochloric acid, it is easily soluble, wirrt by excess hydrochloric acid. (read chlorohydrate excreted.
