Process for increasing the solubility of halogen-substituted oxypyridines
in water The invention aims to provide compounds of the pyridine series that are sparingly soluble in water,
the hydroxyl groups and also halogen atoms, such as iodine, bromine, or even more
contain various of these elements substituted in the core in the agent mentioned
easier to dissolve and their properties in other directions
to improve therapeutic aspects, e.g. B. such that irritant effects in their
Can be eliminated or reduced.
Stellungen substituiert enthalten. Als schwer lösliche Ausgangsstoffe kommen vorzugsweise
in Betracht 2-Oxy-5-jodpyridin, Dijodverbindungen usw.The pyridine compounds in question contain the OH group
in the 2-position and the halogen atoms can be in the 3- or 5-position or in both
Positions contain substituted. The sparingly soluble starting materials are preferred
consider 2-oxy-5-iodopyridine, diiodine compounds, etc.
According to the present process, the solubility of such starting materials
increased by treating them with alkylating agents, among those
Agents also contain acidic groups, in particular the carboxyl group on the alkyl radical,
are to be understood.

geht aus folgenden vergleichenden Zahlen her-
spiel q.) in Wasser etwa 25 : ioo löslich. Auch
die freie 2-Oxo-5-jodpyridin-N-essigsäure ist
in Wasser ungefähr zehnmal stärker löslich
For this purpose, the compounds whose solubility is to be improved are treated with alkyl halides, halogenated fatty acids or other alkylating agents, advantageously in the presence of acid-binding substances. The products of the reaction include. For example, compounds according to the following formulas can be considered: Example i 25 g of 2-oxo-5-iodopyridine are heated to boiling on a reflux condenser with 6.5 g of finely powdered potassium hydroxide dissolved in 100 cc of 96% alcohol on a water bath. A portion of 6.5 g of methyl iodide is added through the cooler in the course of 10 minutes. After two hours of boiling, the mixture is allowed to cool, the crystals which have separated out are filtered off with suction and the mother liquor is concentrated in vacuo. The dried crystals are boiled several times with benzene, and the N-methyl-2-oxo-5-iodopyridine obtained by evaporating the filtered benzene solution obtained in this way is purified by distillation in vacuo. It forms yellow crystals with a melting point of 73 to 74 ° and a boiling point of 182 to 185 ° under 12 mm pressure. Example e

before. While the 2-oxy-5-iodopyridine as

is the free 2-oxo-5-iodopyridine-N-acetic acid
about ten times more soluble in water
like 2-oxy-5-iodopyridine. Quite similar
are the conditions with the other two

an increase in water solubility by at least ten times that of the starting material is achieved.
