Description
Aus der DE-OS 32 23 949 sind insektizid wirksame Thiophosphorsäureester
der Formel (I)
bekannt. Die Herstellung dieser
Verbindungen geschieht nach den üblichen Verfahren durch Veresterung
eines Phenols oder Phenolats der Formel (VI) (M = H, Na, K,
1/2 Ca) mit einem O,S-Dialkylthiophosphorsäurechlorid der
Formel (VII). Dieses Verfahren hat den Nachteil, daß die an
sich bekannten Thiophosphorsäurechloride (VII) nur schwer in
genügender Reinheit herstellbar sind. Bei der Synthese von
(VII) nach bekannten Verfahren fallen Nebenprodukte an, die
schlecht abtrennbar sind. Die zur Reinigung von (VII) erforderliche
Destillation führt wegen der geringen thermischen Stabilität
von (VII) zur teilweisen Zersetzung.
Des weiteren ist bekannt, O-Alkyl-O-aryl-S-alkyl-thiophosphorsäureester
durch S-Alkylierung von Alkalisalzen (JP 74 80 247,
GB 13 11 773) oder Erdalkalisalzen (JP 76 144 728) von O-Alkyl-O-aryl-thiophosphaten
herzustellen.
In der DE-OS 33 16 180 schließlich ist ein Verfahren beschrieben,
in welchem zunächst ein O,O-Dialkyl-O-aryl-thiophosphorsäureester
unter Abspaltung einer O-Alkylgruppe dealkyliert wird. Danach
wird durch Umsetzung mit einem Alkalimetallhydroxid zu einem
Alkalisalz der O-Alkyl-O-aryl-thiophosphorsäure umgesalzt und
mit einem Alkylierungsmittel zum gewünschten O,S-Dialkyl-O-arylthiophosphorsäureester
S-alkyliert.
Bei den zuvor genannten Synthesen wird im letzten Verfahrensschritt
ein Alkalisalz der O-Alkyl-O-aryl-thiophosphorsäure
in wäßrigem Medium alkyliert. Es besteht dabei die Gefahr einer
hydrolytischen Zersetzung des Edukts. Ausbeute und Qualität
des Produkts werden so vermindert.
Überraschenderweise wurde nun gefunden, daß O,S-Dialkyl-O-aryl-thiophosphorsäureester
der Formel
in der
R1 und R2 die gleich oder verschieden sein können C1-C3-Alkyl,
R3 für C1-C8-Alkyl
X für S oder O
Y unabhängig voneinander für Chlor, Brom oder Fluor
und
n für 1 oder 2 steht,
in guten Ausbeuten und ohne Hydrolyse des Zwischenprodukts hergestellt werden können indem man einem Thiophosphorsäureester der Formel mit einem Amin der Formel
R1 und R2 die gleich oder verschieden sein können C1-C3-Alkyl,
R3 für C1-C8-Alkyl
X für S oder O
Y unabhängig voneinander für Chlor, Brom oder Fluor
und
n für 1 oder 2 steht,
in guten Ausbeuten und ohne Hydrolyse des Zwischenprodukts hergestellt werden können indem man einem Thiophosphorsäureester der Formel mit einem Amin der Formel
in der R4 und R5 die gleich oder verschieden sein können und
für C1-C8-Alkyl stehen,
zu einem Salz der Formel in wäßriger Lösung umgesetzt und anschließend, gegebenenfalls nach Einengen mit einem Alkylierungsmittel der Formel
zu einem Salz der Formel in wäßriger Lösung umgesetzt und anschließend, gegebenenfalls nach Einengen mit einem Alkylierungsmittel der Formel
in der Z für Halogen steht, gewünschtenfalls unter Phaspentransferbedingungen
zu dem Produkt der Formel (I) alkyliert.
Unmittelbarer Erfindungsgegenstand ist also ein Verfahren zur
Herstellung von O,S-Dialkyl-O-aryl-thiophosphorsäureestern
(I) bei dem die sek.-Aminsalze der Formel (IV) nicht zu den
entsprechenden Alkalisalzen umgesalzt werden müssen, sondern
die Alkylierungsreaktion mit dem Alkylhalogenid (V) bereits
auf der Stufe des Aminsalzes (IV) in vorteilhafter Weise im
Hinblick auf Ausbeute und Reinheit des Produkts (I) durchgeführt
wird.
Des weiteren betrifft die Erfindung die neuen Aminsalze der
Formel (IV)
in der die Reste R1 bis R5, X, Y und n wie zuvor definiert
sind.
Die im ersten Verfahrensschritt aus einem O,O-Dialkyl-thiophosphorylhalogenid
(VIII) und Phenolen bzw. Phenolaten (VI, M
M = H, Na, K, 1/2 Ca) hergestellten O,O-Dialkyl-O-aryl-thiophosphate
(II) sind teilweise neu.
Die Erfindung betrifft somit auch O,O-Dialkyl-O-aryl-thiophosphate
der Formel
in der R1, Y, n und R3 wie zuvor definiert sind und X für
Sauerstoff steht.
Im Rahmen der verwendeten Definition sind mit Halogen, Fluor,
Chlor, Brom und Jod vorzugsweise Fluor, Chlor und Brom gemeint.
C1-C8-Alkyl steht für Methyl, Ethyl, Propyl, Isopropyl und
die isomeren Butyl-, Pentyl-, Hexyl-, Heptyl- und Octylradikale.
Bevorzugt ist C1-C4-Alkyl, also Methyl, Ethyl, Propyl, Isopropyl,
n-Butyl, iso-Butyl, sek-Butyl und tert.-Butyl.
Als Alkylierungsmittel der Formel (V) werden vorzugsweise die
Chlor, Brom und Jodalkane verwendet, wie Brompropan, Ethylbromid,
Ethylchlorid, Methyliodid, Methylbromid, Isopropylbromid, Isopropylchlorid
etc.
Die Entalkylierung sowie die nachfolgende Alkylierung kann
bei Temperaturen zwischen Erstarrungstemperatur und der Siedetemperatur
des Reaktionsgemisches durchgeführt werden. Bevorzugt
ist der Temperaturbereich von 40-90°C besonders bevorzugt ist
50-80°C, wobei im Falle leichtflüchtiger Alkylierungsmittel
gegebenfalls unter leichtem Überdruck in einer geschlossenen
Apparatur gearbeitet wird.
Das Alkylierungsmittel (V) wird in leichtem Überschuß vorzugsweise
in 1,1 bis 1,7facher und besonders bevorzugt in 1,2 bis 1,5
facher Menge eingesetzt.
Für das Verfahren der Entalkylierung der O,O-Dialkyl-O-arylthiophosphate
(II) sind die reinen sekundären aliphatischen Amine,
sowie deren handelsübliche wäßrige Lösungen geeignet. Zu nennen
sind Dimethylamin, Diethylamin, Di-propylamin, Di-isopropylamin,
Di-butylamin sowie auch die gemischt substituierten Amine,
wie Methyl-ethyl-amin, etc. Da es nur auf die Bildung des
Salzes (II) ankommt, ist es erforderlich, daß die Aminkomponente
eine einheitliche Struktur hat.
Die Entalkylierung des O,O-Diethyl-O-arylthiophosphats (II)
zum Salz (IV) sowie die Alkylierung des Salzes zum Produkt
(I) wird in wäßrigem Medium durchgeführt, wobei aber zur Verbesserung
der Löslichkeit der einzelnen Komponenten ein Löslichkeitsvermittler,
wie etwa Ethanol, Methanol, Propanol, Dimethylformamid
etc. zugesetzt werden kann.
Die nachstehenden Beispiele sollen die Erfindung näher erläutern:
O,O-Diethyl-O-(4-chlor-2-methoxymethylphenyl)-thiophosphat.
Zu einer Lösung von 4-Chlor-2-methoxymethylphenol (172,5 g)
und O,O-Diethylthiophosphorylchlorid (200 g) in Toluol
(500 ml) wird bei Raumtemperatur unter Rühren eine äquimolare
Menge einer wäßrigen NaOH-Lösung (31%) getropft.
Anschließend wird für etwa 5 Stunden auf 70 bis 80°C erwärmt.
Nach dem Erkalten wird die Reaktionslösung alkalisch gestellt
und bei Raumtemperatur kurz ausgerührt.
Die organische Phase wird abgetrennt, getrocknet und am Rotationsverdampfer
eingeengt. Man isoliert als Rohausbeute 318 g
(98,2%) der Titelverbindung. Das Rohprodukt wird durch
Destillation gereinigt. Man isoliert 310 g (95,5%) der Titelverbindung
als Öl mit Kp (0,1 Torr) : 140-145°C.
Um die Reaktion zu beschleunigen, kann dem zweiphasigen Reaktionsgemisch
ein üblicher Phasentransferkatalysator zugesetzt werden.
O-Ethyl-S-propyl-O-(4-chlor-2-methoxymethylphenyl)-thiophosphat.
O,O-Diethyl-O-(4-chlor-2-methoxymethylphenyl)-thiophosphat
(325 g) werden zusammen mit 500 ml einer wäßrigen Dimethylaminlösung
(40%ig) für mindestens 7 Stunden bei 60-65°C gerührt.
Anschließend
wird am Rotationsverdampfer unter vermindertem Druck (20 Torr)
eingeengt und die so erhaltene wäßrige Lösung des Thiophosphorsäure-O-Ethyl-O-(4-chlor-2-methoxymethylphenyl)N-Ethyl--N,N-
dimethylammoniumsalzes
wird mit Wasser (600 ml) verdünnt, mit
1-Brompropan (184,5 g) und etwas Dimethylformamid (1 ml) versetzt
und für mehrere Stunden bei 70°C gerührt.
Die erkaltete Lösung wird mit Ethylenchlorid (500 ml) und wäßriger
NaOH-Lösung (200 ml 1-M-NaOH) geschüttelt, die organische Phase
wird abgetrennt, getrocknet und eingeengt.
Man isoliert 294 g der Titelverbindung als Öl vom Kp. (0,05 Torr)
145-155°C. Die Reinheit des so erhaltenen Produkts wird
gaschromatographisch mit 95,1% bestimmt. Entsprechend dem
in den Beispielen 1a und 1b beschriebenen Verfahrensschritten
erhält man:
O-Ethyl-S-propyl-O-(2-chlor-4-methylthiomethylphenyl)-thiophosphat
(Kp 0,1 = 105-8°C)
ausgehend von O,O-Diethyl-thiophosphorylchlorid und 2-Chlor-4-methylthiomethylphenol
erhält man O,O-Diethyl-O-(2-Chlor-4-methylthiomethylphenyl)-thiophosphat,
welches analog zu Beispiel 1b
mit Dimethylamin zum Thiophosphorsäure-O-ethyl-O-(2-chlor-4-methylthiomethylphenyl)
N-Ethyl-N,N-dimethylammoniumsalz
reagiert und anschließend mit Brompropan zur Titelverbindung
alkyliert wird.
O-Ethyl-S-propyl-O-(4-chlor-2-methylthiomethylphenyl)-thiophosphat,
ausgehend von O,O-Diethyl-thiophosphorylchlorid und 4-Chlor-2-methylthiophenol
erhält man O,O-Diethyl-O-(4-chlor-2-methylthiomethylphenyl)-thiophosphat,
welches mit Dimethylamin zum
Thiophosphorsäure-O-ethyl-O-(4-Chlor-2-methylthiomethylphenyl)
N-Ethyl-N,N-dimethylammoniumsalz reagiert und anschließend
mit Brompropan zur Titelverbindung alkyliert wird.
O-Ethyl-S-propyl-O-(3-chlor-4-methoxymethylphenyl)-thiophosphat,
ausgehend von O,O-Diethyl-thiophosphorylchlorid und 3-Chlor-4-methoxymethylphenol
erhält man O,O-Diethyl-O-(3-chlor-4-methoxymethylphenyl)-thiophosphat,
welches mit Diethylamin zum Thiophosphorsäure-O-ethyl-O-(3-chlor-4-methoxymethylphenyl)
Triethylammoniumsalz
reagiert und anschließend mit Brompropan zur Titelverbindung
alkyliert wird.
O-Ethyl-S-propyl-O-(4-chlor-3-methyl-thiomethylphenyl)-thiophosphat,-
ausgehend von O,O-Diethylthiophosphorylchlorid und 4-Chlor-3-methylthiomethylphenol
erhält man O,O-Diethyl-O-(4-Chlor-3-methylthiomethylphenyl)-thiophosphat,
welches mit Dimethylamin zum
Thiophosphorsäure-O-ethyl-O-(4-chlor-3-methylthiophenyl)
N-Ethyl-N,N-dimethylammoniumsalz reagiert und anschließend mit
Brompropan zur Titelverbindung alkyliert wird.
O-Ethyl-S-propyl-O-(4-chlor-2-ethoxymethylphenyl)-thiophosphat
(n D 25 : 1,5149),
ausgehend von O,O-Diethyl-thiophosphorylchlorid und 4-Chlor-2-ethoxymethl-phenol
erhält man O,O-Diethyl-O-(4-chlor-2-ethoxymethylphenyl)-thiophosphat,
welches mit Dimethylamin zum
Thiophosphorsäure-O-ethyl-O-(2-ethoxymethyl-4-chlorphenyl)N-Ethyl-
N,N-dimethylammoniumsalz reagiert und anschließend mit Brompropan
zur Titelverbindung alkyliert wird.
O-Ethyl-S-propyl-O-(4-Chlor-2-isopropyloxymethylphenyl)-thiophosphat-
(n D 25 : 1,5100)
ausgehend von O,O-Diethyl-thiophosphorylchlorid und 4-chlor-2-isopropyloxymethylphenol
erhält man O,O-Diethyl-O-(4-chlor-2-isopropyloxymethylphenyl)-thiophosphat,
welches mit Dimethylamin
zum Thiophosphorsäure-O-ethyl-O-(4-chlor-2-isopropyloxymethylphenyl)
N-Ethyl-N,N-methylammoniumsalz reagiert und
anschließend mit Brompropan zur Titelverbindung alkyliert wird.
O-Ethyl-S-propyl-O-(2-Chlor-5-methoxymethylphenyl)-thiophosphat,
ausgehend von O,O-Diethylthiophosphorylchlorid und 2-chlor-5-methoxymethylphenol
erhält man O,O-Diethyl-O-(2-chlor-5-methoxymethylphenyl)-thiophosphat,
welches mit Di-isopropylamin zum
Thiophosphorsäure-O-ethyl-O-(2-chlor-5-methoxymethylphenyl)-
N-Ethyl-N,N-diisopropylammoniumsalz reagiert und anschließend
mit Brompropan zur Titelverbindung alkyliert wird.
O-Ethyl-S-propyl-O-(4-brom-3-propylthiomethylphenyl)-thiophosphat,
ausgehend von O,O-Diethylthiophosphorylchlorid und 4-Brom-3-propylthiomethylphenol
erhält man O,O-Diethyl-O-(4-brom-3-propylthiomethylphenyl)-thiophosphat,
welches mit Dimethylamin zum
Thiophosphorsäure-O-ethyl-O-(4-brom-3-propylthiomethylphenyl)
N-Ethyl-N,N-dimethylammoniumsalz reagiert und anschließend
mit Brompropan zur Titelverbindung alkyliert wird.
O-Ethyl-S-propyl-O-(2,5-dichlor-4-methoxymethylphenyl)-thiophosphat,-
ausgehend von O,O-Diethyl-thiophosphorylchlorid und 2,5-Dichlor-4-methoxymethylphenol
erhält man O,O-Diethyl-O-(2,5-dichlor-4-methoxymethylphenyl)-thiophosphat,
welches mit Dimethylamin
zum Thiophosphorsäure-O-ethyl-O-(2,5-dichlor-4-methoxymethylphenyl)-
N-Ethyl-N,N-dimethylammoniumsalz reagiert und anschließend
mit Brompropan zur Titelverbindung alkyliert wird.
Claims (3)
1) Verfahren zur Herstellung von Thiolphosphorsäureester der
Formel
in der
R1 und R2 die gleich oder verschieden sein können für C1-C3-Alkyl,
R3 für C1-C8-Alkyl
X für S oder O
Y unabhängig voneinander für Chlor, Brom oder Fluor
und
n für 1 oder 2 steht,
dadurch gekennzeichnet, daß man ein Thiophosphorsäureester der Formel mit einem Amin der Formel die gleich oder verschieden sein können und für C1-C8-Alkyl stehen, zu einem Salz der Formel in wäßriger Lösung umsetzt und anschließend, gegebenenfalls nach Einengen mit einem Alkylierungsmittel der Formel in der Z für Halogen steht, gewünschtenfalls unter Phaspentransferbedingungen zu dem Produkt der Formel (I) alkyliert.
R1 und R2 die gleich oder verschieden sein können für C1-C3-Alkyl,
R3 für C1-C8-Alkyl
X für S oder O
Y unabhängig voneinander für Chlor, Brom oder Fluor
und
n für 1 oder 2 steht,
dadurch gekennzeichnet, daß man ein Thiophosphorsäureester der Formel mit einem Amin der Formel die gleich oder verschieden sein können und für C1-C8-Alkyl stehen, zu einem Salz der Formel in wäßriger Lösung umsetzt und anschließend, gegebenenfalls nach Einengen mit einem Alkylierungsmittel der Formel in der Z für Halogen steht, gewünschtenfalls unter Phaspentransferbedingungen zu dem Produkt der Formel (I) alkyliert.
