Description
Process for the preparation of mercury compounds which are colloidally soluble in water of the oxybenzenesulfocarboxylic acids and their homologues. The so-called mercury salicylate (Hydrargyrum salicylicum basicum of the German Pharmacopoeia), which therapeutic Is used, has the disadvantage of insolubility in water, whereby the Absorption in the body is extremely slowed. It was therefore natural to try by introducing a sulfo group into the molecule of mercury salicylate to produce sulphonic acid of the mercurated compound which is readily soluble in water. the Attempts so far made in this direction have been unsuccessful; so there the patent specification 216267 of class 12 q indicates that the mercury compounds of the Oxybenzenesulfocarboxylic acids, which can be obtained, for example, by boiling with mercury oxide and water can be obtained from these acids, are also sparingly soluble in water.
However, this information is only partially correct. It has been found that, under suitable test conditions, it is possible to obtain mercury compounds of oxybenzenesulfocarboxylic acids which are readily soluble in water. Thus the sulphonic acid of mercury salicylate, which is obtained in a well-crystallized state, shows the surprising property, which opens up new possibilities for use, of providing colloidal solutions with water. The compound is intended to find therapeutic use as such or in the form of its colloidal solutions. The new compounds contain the mercury in masked form (see Der. D. Deutsch. Chem. Gesellsch. 35 [190--1 pp. 2877 ff); they are soluble in alkalis without precipitation of mercury oxide and are not precipitated in aqueous solution by hydrogen sulfide, but give brown hydrosols of mercury sulfide.
The formation of such mercury compounds or the colloidal ones Solutions take place when the "mercurying" takes place in the presence of free acid, z. B. if you are on the oxybenzenesulfocarboxylic acid mercury sulfate, nitrate or acetate solution or mercury oxide or carbonate in one to saturate the three acidic groups of sulfooxybenzoic acid can act in insufficient amount, so Less than 1/2 molecule of mercury oxide to one molecule of sulfonic acid during exposure of 1l / 2 or more molecules of mercury oxide according to the specifications of the patent 21C267 leads to water-insoluble compounds. The opportunity to form the The products mentioned are generally given when there is somehow bound mercury meets with oxybenzenesulfocarboxylic acids in acidic solution. E.g. arise initially insoluble in the mercuration according to the specifications of patent specification 216267, salty mercury compounds of oxybenzenesulfocarboxylic acids that can still be precipitated by alkalis, which then turn into insoluble, but no longer precipitable with alkali, when heated Convert connections. By heating with acids can now both the former as well as the latter. converted into the water-soluble colloidal compounds will.
C6 H3 (OH) CO - 0 - Hg -E- 2 H2 S04 = C6 H3 (OH) (S03 H) COOH -1- Hg S04+ H2 0. z. C6 H3 (OH) (S03 H) COOH -E- Hg S04 -f- H2 0 - C6 H2 (OH) (S03H) (COOH) Hg OH -i- H2 SO4. Beispiel I.A simple embodiment of the mercuration in sulfuric acid solution is finally that z. B. Mercury salicylate with concentrated sulfuric acid heated, diluted with water and further heated. The formation of the mercury sulfosalic acid takes place here in two phases: x. C6 H3 (OH) CO - 0 - Hg -E- 2 H2 S04 = C6 H3 (OH) (S03 H) COOH -1- Hg S04 + H2 0 e.g. C6 H3 (OH) (S03 H) COOH -E- Hg S04 -f- H2 0 - C6 H2 (OH) (S03H) (COOH) Hg OH -i- H2 SO4. Example I.
Die Lösung wird durch Ammoniumsulfat flockig gefällt, durch Schwefelanimonium wird selbst beim Kochen nur langsam Quecksilbersulfid abgeschieden, Schwefelwasserstoff erzeugt eine kolloidale Lösung des letzteren.6 g of crude sulfosalicylic acid (5-sulfo-2-oxybenzene-x-carboxylic acid), dissolved in 10 cc of water, with a solution of 3.7 g of mercury oxide in the required Amount of dilute sulfuric acid regardless of any precipitating salt restricted to the water bath. At first a whitish jelly forms, which is soon turned into crystals. These are genetically sucked and washed with alcohol. The sulphuration and mercuration of salicylic acid can also be carried out in one operation done by warming salicylic acid with sulfuric acid, diluting with water and after stirring in the calculated amount of mercury oxide it is concentrated. The new mercury sulfosalicylic acid forms white cup-shaped crystals, which are very difficult to dissolve in common organic solvents are. In contrast, it dissolves easily and clearly in water to a colloidal, extremely viscous liquid. The solution is through Ammonium sulphate is precipitated flaky, due to sulphurous ammonium, even when cooked Mercury sulphide is only slowly deposited, hydrogen sulphide produces a colloidal one Solution of the latter.
BeispielII.Alkaline solutions, ammonia, and saline solution dissolve the mercuric sulfosalicylic acid with salt formation. The solution leaves a whitish skin on evaporation, which with water supplies the colloidal solution again. ExampleII.
The mercury oxide obtained from 5.5 g of mercury chloride is with a solution of i i g of sulfosalicylic acid in about zoo cc of water and mixed some Minutes. The result is a thick solution, which in the cold becomes a translucent jelly freezes. Evaporation on a water bath gives solid mercurisulfosalicylic acid; add excess before evaporation dilute sulfuric acid (I: .4) added, see above. separates from the remaining one acid concentrated the new compound in crystals.
Beispie11II. Example11II. The obtainable according to Example II of the patent specification 216267 water-insoluble mercury compound of sulfosalicylic acid is diluted with Sulfuric acid is heated on the water bath until a sample of the crystalline Clearly soluble in water. On part of the dried mercury compound 5 parts sulfuric acid (i: x) are to be used.
Beispiel IV.Example IV.
io g Quecksilbersalicylat (Mercurisalicylsäure} werden mit i 5 ccm konzentrierter Schwefelsäure verrieben und so lange auf dein Wasserbade erwärmt, bis eine Probe wasserlöslich ist.10 g of mercury salicylate (mercuric salicylic acid) are mixed with 5 ccm concentrated sulfuric acid and warmed up on your water bath for so long, until a sample is water soluble.
So much water is now added until a clear solution is obtained is, and further heated. The mercuric sulfosalicylic acid separates after a short time Time in crystals.
Example V. m-cresotinic acid is sulphurised on a water bath using concentrated sulfuric acid. 6.4 g of the dried crude sulphonic acid are dissolved in 2o g of sulfuric acid and 30 g of warm water and the liquid is heated on the water bath with a solution of 3.6 g of mercury oxide in the required amount of dilute sulfuric acid until no sodium hydroxide solution is found in a sample Mercury oxide falls more.
The mercuration product separates during the heating process, Mercurisulfozn-cresotinic acid, in crystal warts consisting of needles.
Die neue Verbindung ähnelt in ihren Eigenschaften, insbesondere der Bildung einer kolloidalen Lösung mit Wasser, der Mercurisulfosalicyls@äure:The new compound is similar in its properties, especially the Formation of a colloidal solution with water, the mercurisulfosalicylic acid:
