The main component of opium, the most famous hypnotic, sedative and Anodynums, forms the morphine. -But the effect of opium is different from that a corresponding amount of morphine different. This difference is due to the Presence of other alkaloids in the opium, the so-called minor alkaloids. It was not previously known, the total amount of minor alkaloids contained in opium in the proportions found in opium, in solid form free from morphine to manufacture. This is the purpose of the present invention. It has been shown that the secondary alkaloids freed from morphine in the proportions as they are in the Opium are an extremely good sleep aid. They make you Sleep, which is particularly compared to the absence of any side effects the sleep produced by morphine. The patients are awakening fresh, without any drowsiness in the head and without tiredness in the limbs.
However, this procedure was extremely cumbersome and ultimately yielded the alkaloids not in solid form, but in solution. As the starting material of the The present process can be opium or, with advantage, that according to the patent specification 229905, class 30 h, manufactured product that contains the total alkaloids of opium in the form of their contains hydrochloric acid salts. To manufacture the new preparation can for example, be proceeded as follows.
This leaching is repeated until the opium alkaloids are residues, which after three 4 ^ is usually to four times leaching the case. The free sulfuric acid is then precipitated in the acid extract with barium hydroxide, whereupon the liquid is made alkaline with alkali carbonate, preferably sodium bicarbonate and a little soda, about 10 percent, and shaken out repeatedly with chloroform.
Auf diese Weise erhält man ein violettbraunes Pulver, das sich klar und leicht in WasserThe free sulfuric acid that is present can also be neutralized with an alkali, which is then made alkaline with alkali carbonate, preferably with sodium bicarbonate and a little soda. In order to remove traces of morphine dissolved in chloroform, this solution is shaken out once with the calculated amount of 1 Z 10 normal sodium hydroxide solution. The chloroform solution contains the alkaloids of opium with the exception of morphine; it is evaporated, whereupon the residue is taken up in alcohol, the solution is neutralized with alcoholic hydrochloric acid and evaporated in vacuo. In this way you get a purple-brown powder that is clear and light in water
übergeht.solves; it reacts sourly to litmus paper, but neutrally to congo paper. The preparation shows the following behavior against some common alkaloid reagents:
Fröhde's reagent immediately gives a blue-green color, while a morphine-containing product first turns an intense purple, whereupon the color only later changes to moss green.
ίο Mayer's reagent produced in equally concentrated Solutions of morphine-free and morphine-containing products show a thicker white precipitate in the former than in the latter. Concentrated sodium acetate solution, mainly narcotine, behaves in the same way and papaverine falls. Here, too, a thicker precipitate arises when there is no morphine. . Bismuth iodide-potassium iodide gives an orange to red-brown precipitate, which in comparison is reddish tinge to morphine-containing preparations. Bourchardat's reagent (iodine-iodine potassium) behaves similar to bismuth iodide-potassium iodide.
Statt Salzsäure kann natürlich auch jede andere Säure, die wasserlösliche Salze bildet, verwendet werden.100 g of the product prepared according to patent specification 229905, class 30 h, are dissolved in distilled water. This solution is first mixed with chloroform and then made alkaline with sodium bicarbonate. The excreted secondary alkaloids are absorbed by the chloroform, while the morphine remains in the liquid as a flaky precipitate. The chloroform solution is separated from the water, which is then extracted again with chloroform. The two extraction liquids are combined and, in order to remove small traces of dissolved morphine, extracted once with the calculated amount of 1 Z 10 normal sodium hydroxide solution, then evaporated in vacuo, whereupon the residue is taken up with alcohol and with a small excess of alcoholic Hydrochloric acid is added. By evaporating this solution in a vacuum, all of the secondary alkaloids of opium are obtained in solid form. The product obtained according to this example corresponds to that according to example 1. Instead of hydrochloric acid, any other acid that forms water-soluble salts can of course also be used.
The possibility, with the present process, of the products of the process of the patent specification 229905, cl. 30h, or an appropriately prepared extract from opium, completely withdrawing the morphine without anything else To change something in the ratio of the secondary alkaloids must be described as surprising will.
According to Hager (Handbook of Pharmaceutical Practice, 1903, Volume II, page 396), as is also expressly stated, only an impure morphine salt is obtained; So it works according to Hager's method, it is not possible to obtain morphine free from secondary alkaloids, but this affects the ratio of the remaining minor alkaloids. Even from the known solubility of pure morphine and individual pure secondary alkaloids in the solvents used was an indication of the success of the separation not to pull; because mixtures of organic compounds often behave perfectly different from the pure substances. It is a known fact that individuals are organic Substances influence each other in their solubility, a fact that theirs Separation is usually extremely difficult. Then it should also be emphasized that the differences in solubility in the individual alkaloids are not such that one, so extensive separation was to be predicted.
