Abstract
Vektor zur Verwendung in einem Verfahren zur selektiven Entfernung von Antibiotikaresistenz aus einem antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistenten Bakterienstämmen in einem gesunden Probanden, der einen antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistente Bakterienstämme trägt, oder in einem Patienten, der einen antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistente Bakterienstämme trägt,wobei der Vektor für ein DNA-modifizierendes Enzym kodiert, das das Antibiotikaresistenzgen bzw. die Antibiotikaresistenzgene, die für die Antibiotikaresistenz des Bakterienstamms bzw. der Bakterienstämme verantwortlich ist/sind, inaktiviert,wodurch die Antibiotikaresistenz aus dem/den antibiotikaresistenten Bakterienstamm/-stämmen in der Mikrobiota des Probanden oder Patienten selektiv entfernt wird.
Claims (15)
- Vektor zur Verwendung in einem Verfahren zur selektiven Entfernung von Antibiotikaresistenz aus einem antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistenten Bakterienstämmen in einem gesunden Probanden, der einen antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistente Bakterienstämme trägt, oder in einem Patienten, der einen antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistente Bakterienstämme trägt, wobei der Vektor für ein DNA-modifizierendes Enzym kodiert, das das Antibiotikaresistenzgen bzw. die Antibiotikaresistenzgene, die für die Antibiotikaresistenz des Bakterienstamms bzw. der Bakterienstämme verantwortlich ist/sind, inaktiviert, wodurch die Antibiotikaresistenz aus dem/den antibiotikaresistenten Bakterienstamm/-stämmen in der Mikrobiota des Probanden oder Patienten selektiv entfernt wird.
- Vektor zur Verwendung in einem Verfahren zur Verhinderung einer mit Antibiotikaresistenz zusammenhängenden Infektion bei einem gesunden Probanden, der einen antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistente Bakterienstämme trägt, oder bei einem Patienten, der einen antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistente Bakterienstämme trägt, wobei der Vektor für ein DNA-modifizierendes Enzym kodiert, welches das/die Antibiotikaresistenzgen(e) inaktiviert, das/die für die Antibiotikaresistenz des Bakterienstammes bzw. der Bakterienstämme verantwortlich ist/sind, wodurch die Antibiotikaresistenz von einem antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistenten Bakterienstämmen in der Mikrobiota des Probanden oder Patienten selektiv entfernt wird und eine mit Antibiotikaresistenz zusammenhängende Infektion in dem Probanden oder Patienten verhindert wird.
- Rekombinanter Phage zur Verwendung in einem Verfahren zur selektiven Entfernung von Antibiotikaresistenz aus einem antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistenten Bakterienstämmen in einem gesunden Probanden, der einen antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistente Bakterienstämme trägt, oder in einem Patienten, der einen antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistente Bakterienstämme trägt, wobei der rekombinante Phage eine Nukleinsäure umfasst, die für ein DNA-modifizierendes Enzym kodiert, das das Antibiotikaresistenzgen bzw. die Antibiotikaresistenzgene, die für die Antibiotikaresistenz des Bakterienstamms bzw. der Bakterienstämme verantwortlich ist/sind, inaktiviert, wodurch die Antibiotikaresistenz aus dem/den antibiotikaresistenten Bakterienstamm/-stämmen in der Mikrobiota des Probanden oder Patienten selektiv entfernt wird.
- Rekombinanter Phage zur Verwendung in einem Verfahren zur Verhinderung einer mit Antibiotikaresistenz zusammenhängenden Infektion bei einem gesunden Probanden, der einen antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistente Bakterienstämme trägt, oder bei einem Patienten, der einen antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistente Bakterienstämme trägt, wobei der rekombinante Phage eine Nukleinsäure umfasst, die für ein DNA-modifizierendes Enzym kodiert, das das Antibiotikaresistenzgen bzw. die Antibiotikaresistenzgene, die für die Antibiotikaresistenz des Bakterienstamms bzw. der Bakterienstämme verantwortlich ist/sind, inaktiviert, wodurch die Antibiotikaresistenz von einem antibiotikaresistenten Bakterienstamm bzw. antibiotikaresistenten Bakterienstämmen in der Mikrobiota des Probanden oder Patienten selektiv entfernt wird und eine mit Antibiotikaresistenz zusammenhängende Infektion in dem Probanden oder Patienten verhindert wird.
- Vektor zur Verwendung nach
Anspruch 1 oder2 oder rekombinanter Phage zur Verwendung nachAnspruch 3 oder4 , wobei das DNA-modifizierende Enzym eine Nuklease, wie eine CRISPR-assoziierte Nuklease, oder ein Baseneditor ist. - Vektor oder rekombinanter Phage zur Verwendung nach einem der
Ansprüche 1 bis5 , wobei der Vektor oder rekombinante Phage weiter einen konditionalen Replikationsursprung umfasst, der in dem anvisierten antibiotikaresistenten Bakterienstamm bzw. in den anvisierten antibiotikaresistenten Bakterienstämmen inaktiv, in einer Spenderbakterienzelle jedoch aktiv ist. - Vektor oder rekombinanter Phage zur Verwendung nach einem der
Ansprüche 1 bis6 , wobei antibiotikaresistente Bakterien aus dem Darm des gesunden Probanden oder Patienten entfernt werden. - Vektor oder rekombinanter Phage zur Verwendung nach einem der
Ansprüche 1 bis7 , wobei die selektive Dekolonisierung erfolgt, bevor der Proband oder der Patient sich einer Operation unterzieht und/oder sich einer Krebsbehandlung und/oder einer immunsuppressiven Behandlung unterzieht. - Vektor oder rekombinanter Phage zur Verwendung nach
Anspruch 8 , wobei es sich bei der Operation um eine Operation zur Transplantation eines festen Organs der Niere oder der Leber handelt. - Vektor oder rekombinanter Phage zur Verwendung nach einem der
Ansprüche 1 bis9 , wobei die Mikrobiota eine Darmmikrobiota ist. - Vektor oder rekombinanter Phage zur Verwendung nach einem der
Ansprüche 1 bis10 , wobei das Antibiotikum Methicillin, Vancomycin oder ein Antibiotikum der Carbapenem-Klasse oder der Beta-Lactam-Klasse ist. - Vektor oder rekombinanter Phage zur Verwendung nach einem der
Ansprüche 1 bis11 , wobei das Verfahren weiter das Erhalten einer Mikrobiota-Probe von dem Probanden oder dem Patienten und die Bestätigung des Vorhandenseins des antibiotikaresistenten Bakterienstammes bzw. der antibiotikaresistenten Bakterienstämme in der Probe umfasst. - Vektor oder rekombinanter Phage zur Verwendung nach einem der
Ansprüche 1 bis12 , wobei mindestens zwei verschiedene Vektoren oder rekombinante Phagen verwendet werden, bevorzugt vier verschiedene Vektoren oder rekombinante Phagen, wobei jeder Vektor oder rekombinante Phage ein anderes Bakterium, insbesondere ein anderes E. coli-Bakterium, anvisiert und/oder für ein DNA-modifizierendes Enzym kodiert, das eine andere Nukleinsäuresequenz innerhalb des anvisierten Bakteriums anvisiert. - Vektor oder rekombinanter Phage zur Verwendung gemäß
Anspruch 13 , wobei der Patient an einer hämatologischen Malignität wie hämatologischem Krebs leidet und/oder wobei der Patient einer Krebsbehandlung und/oder immunsuppressiven Behandlung unterzogen wird. - Zusammensetzung oder Kombination, umfassend mindestens 4 rekombinante Phagen, wie in einem der
Ansprüche 3 bis13 definiert, zur Verwendung bei einem Verfahren zur Verhinderung einer mit antibiotikaresistenten Escherichia coli in Zusammenhang stehenden Infektion bei einem Patienten, der einen antibiotikaresistenten Stamm bzw. antibiotikaresistente Stämme von Escherichia coli trägt, wobei der rekombinante Phage eine Nukleinsäure umfasst, die für ein DNA-modifizierendes Enzym kodiert, das das Antibiotikaresistenzgen bzw. die Antibiotikaresistenzgene, die für die Antibiotikaresistenz des Stamms bzw. der Stämme von E. coli verantwortlich ist/sind, inaktiviert, wodurch die Antibiotikaresistenz von antibiotikaresistenten E. coli-Stämmen in der Mikrobiota des Patienten selektiv entfernt und eine mit antibiotikaresistenten E. coli in Zusammenhang stehende Infektion des Patienten verhindert wird, wobei der Patient an einer hämatologischen Malignität wie hämatologischem Krebs leidet und sich optional einer Krebsbehandlung und/oder einer immunsuppressiven Behandlung unterzieht.
