Description
The invention relates to a method and a device to determine the adhesive behavior of pressure-sensitive adhesive surfaces such as medical adhesive tape and in particular pressure-sensitive adhesive application systems such as transdermal thera human systems (TTS) on the skin of humans or sow animal.
The temporary connection by means of pressure sensitive adhesive comprises one Period from a few hours to 14 days. The term Skin also covers the mucous membrane of the upper airways, the Eye and the genital organs.
The mentioned products are medical African adhesive tapes for fixation purposes or for wound dressing care ("adhesive plaster").
Applied on the skin are also included on systems, which are to be understood as dosage forms, the one or more active substances or test substances primarily an adhesive layer on the skin or through this on the body submit by. Alternatively, such systems can also be used Removal of chemical substances from the body through the skin be used. Such an application has to be diagnosed purposes or for detoxification. In particular refers to the method and the device according to the Er invention on transdermal therapeutic systems (TTS) for controlled, long-term delivery of one or more Active ingredients on the skin. The distribution of these active substances can remain localized or affect the entire The organism of the wearer.
TTS are known in the pharmaceutical industry and in large number on the market.
The invention is particularly suitable as a means for Er research of the pressure-sensitive adhesive of such products on the skin, especially with longer wear.
One problem with the development of TTS is security a contiguous connection with the skin Wear for periods from a few hours to 7 days or more. In this context, contour-fit means that edge separation of the TTS from the skin is avoided becomes.
Influences on wearing behavior depend on the size of the TTS and from the application site on the human body. Out These two parameters are largely the result of a TTS mechanical forces, e.g. B. stretching, Compression, torsion and shear forces, which are required to separate from the Skin.
To the adhesion on the skin for comparatively longer Ensuring periods is an optimization of the skin side adhesive layer required. Depending on the structure of the TTS can this layer contain active ingredients and also phar contain pharmaceutical excipients, the skin permeation favor the active ingredient. In such cases Adhesion behavior of the layer by type and amount of the included acted or auxiliary substances more or less affected flows. Any additional layers of the system can Condition of the adhesive layer on the skin due to absorption change or dispensing of substances also.
The absorption of skin probes plays a special role into the TTS. These include the separation of gas shaped water (transpiration), of liquid water (Sweat), salts (sweat) and fatty substances (sebum). The absorption of water in particular leads to many TTS a reduced cohesion and shear strength of the inner Structure, which retains the adhesive force until detachment can hen.
Furthermore, the internal structure of a TTS can change as a result of Delivery of individual components to the application site strong in terms of cohesion, shear strength and adhesive strength and changed unexpectedly.
In summary, the setting of optimal adhesive properties mainly formulate the formulation of the glue layer concerned and also strong from the inside and external structure of the TTS dependent task. The one with it associated variabilities make it generalization Successful formulations largely impossible. For each new TTS is a more or less complex optimization the adhesive properties required.
Testing TTS under development follows advantageously under real conditions on humans or Animal. Such studies require, in particular, humans However, due to the often pharmaceutical law Clinical trial status as well as statistical for clear statements required a very high number of individuals financial expense. Optimizing adhesive properties Experience has shown that in the course of the product range process to be repeated several times.
For these reasons, according to current practice, testing is carried out Exercise on humans only in a late stage of development. The adhesive strength of TTS is preferably used as a parameter in clinical studies on therapeutic efficacy and safety added thoughtfulness.
In the early stages of development, subjective play so far Criteria of the people involved in the development major role. This also includes manual tests of the adhesives layer with hands as well as brief attempts at wearing People.
A large part of the objective test methods come from from the development of technical adhesives or pressure sensitive adhesives (compare official test methods of AFERA; Asso ciation des Fabricants Europeens de Rubans Auto-adhesifs, Vitry sur Seine, France). With simple mechanical bela The pressure sensitive adhesive formulation to be tested is tested next applied to a layered carrier, with to which it forms a firm bond. This tape-like Test product is then glued to test surfaces and then For example, pulling forces become different Trigger angles and speeds determined. For that can e.g. B. serve an arrangement as in DE 41 27 018 C2 is described with which the adhesive force of an adhesive film Steel by means of one interacting with a pressure roller Pull-off and deflection roller with connected force measurement direction is determined.
With a suitable structure, the tendency of an adhesive can also occur mass to flow under mechanical load (cold River) due to lack of cohesion.
The tests can be carried out by varying parameters such as Be quality of test surfaces as well as temperature and of humidity in a wide range the. However, the individual tests always reflect physically only a fraction of the dynamic load process on the Skin reflected. In particular, an exchange of substances between the system and the carrier can be difficult or even impossible cannot be adjusted. The mechanical loads are one-sided and practically always aim at one extreme strain on the adhesive that is not common on the skin is.
In summary, these methods are primarily suitable as Means of quality control in ensuring the Uniformity of a product and less for optimization its properties in the development phase.
For a differentiated assessment of the adhesive properties there are other, technically essential on the shelf more elaborate procedures available. For example can by measuring wetting angles between drops different test liquids and the surface of one Pressure sensitive adhesive Data on its surface properties be won. It can be the energetic relationship nisse and in particular the distribution on chemically polar and non-polar interaction on the outside of the adhesive determine. Ideally, these relationships are the same few on the surface to be glued, in the case of TTS the skin.
With the help of this method, the physico-chemical Affinity of an adhesive to the application area mechanical properties, such as that Ability to adapt quickly to a rough surface, remain completely ignored.
The mechanical, internal properties of the adhesive the methods of rheology and dynamic mechanical analysis (DMA).
With rheometers, examinations on thin glue can be done layers carried out under the action of torsional forces become. The type of torsion load as well as the temperature and humidity during the test can vary widely Limits can be varied. At the DMA, torsions forces out z. B. also bending, stretching and compression forces be allowed to act on a test material.
Both methods are very suitable for the investigation of Cohesion and shear strength especially when changing temperature. Statements about the cold flow as well as elastic and plastic parts in the Deformation of an adhesive mass can be taken. The procedure Ren are powerful, but technically complex. However, they only consider the internal properties of an adhesive while the properties of the outer surfaces as the actual stickiness can not be detected.
An exchange of material with the environment, as is the case with TTS at the Skin takes place, cannot react with today's equipment be lized.
In summary, there is a wide arsenal of experiments methods that, however, are always only statements that are relatively narrow Allow partial aspects of skin adhesion. Erhe The data are practiced under highly abstract conditions gene based on the extraction of individual physico-chemical Characteristics are aligned in the purest form.
Realistic investigation methods and mo are missing dents that wear TTS on the skin in less abstracted form and as holistic as possible to simulate the associated problems possible. Hence the demand, such Model without high technical effort and in a kind and Way of realizing a wide use for routi nine examinations allowed.
The invention has for its object a method and to specify a test apparatus which makes it possible to Pressure sensitive behavior of fabrics on human or animal skin under realistic conditions to test and for this for each active ingredient composition to determine optimal pressure sensitive adhesive formulations.
According to the invention, this object is achieved by a test method according to the features of claim 1 and a device solved according to the features of claim 15. An in vitro Test methods can also be found in DE 41 18 200 C1 men, however, this is an assessment of the un desired bonding of wound dressings to the wound an agar / collagen matrix with fibrin that forms network, while according to the invention a standardized duration test for the assessment of adhesive properties, in particular of TTS is provided.
The results obtained according to the invention give good results Consistent with that when wearing TTS on the skin replacement problems that occur again. Combined with The test procedure demonstrates objective evaluation procedures the invention a surprisingly large differentiation gene.
For the test method according to the invention, either uses all or part of TTS. As part becomes a flat section separated from the TTS understood, the layer structure is identical to the TTS. However, a part can only have one in its layer structure fabric that partially matches the TTS be congruent in its external form with the TTS or is different from him.
In any case, the TTS and, if applicable, the te part of it an identically formulated pressure sensitive adhesive layer and at least one more possibly from each other deviating layer. The term TTS includes the following which are also parts of a TTS as defined above.
The TTS to be tested is first put on a piece of a foil glued, elastic carrier. The TTS points in any case a smaller area than the carrier on and is stuck on so that there is a gap on all sides the edge of which is present. The carrier is together with the TTS in the form of a separating membrane between two half-shelled Brought elements. The two half-shell elements are congruent along their edge and enclose together a defined interior, in principle similar the shells of a walnut. The halves of each enclosed volumes can be the same size or of one other deviate.
The two shells are made using suitable devices held firmly and contiguously against each other. The carrier with the TTS is wrinkle-free, planar and at most fixed slightly pre-stretched between the halves. This The device is referred to below as a "test cell". The TTS lies completely inside the test cell and holds distance from their walls on all sides.
As part of the test procedure, there is a recurrence mechanical load and relief of the wearer together the TTS on it. This burden is recorded via the time the entire area of the carrier and takes place before moves from the surface of the carrier facing away from the TTS forth. This corresponds to the conditions when wearing on the Skin covered by the mechanical stress from the Area to which TTS is transferred.
The mechanical load on the wearer can be affected piston-like reciprocating stamp made of through a corresponding opening in the test cell leads. However, the load is preferably caused by a circle at low speed the solid body moving away from the TTS realized. This body is completely within the Test cell, is pressed into the elastic support and causes its stretching. This stretch does not go over the elastic deformability of the carrier, so that practically no plastic deformation occurs. The Verfor The wearer's measurements are mainly recorded on the TTS as Transfer shear, stretch, compression, bending and bending forces.
The rotation of the above body by one in it or axis lying outside of it and its pressure force against the TTS carrier can be effected in different ways become. In one variant, the body can roll freely and lies only through the surface of the TTS carrier Gravity on. The rolling of the body is then through Tilting the test cell against gravity and reaching the Pressure force results from weight and contact surface of the body.
In a preferred arrangement, pressure forces and Movement of the rolling body through an external, variable magnetic field determined.
Finally, the body can be made by suitable mechanical Connecting elements (e.g. rod-shaped axis or push rod ge) are moved from outside the test cell.
In total there is a recurring mechani stress on the TTS that lasts for hours and days can be stretched.
The special performance of the model consists in the Linking the mechanical load with a material exchange between the TTS and its surroundings.
This mass transfer is made possible by the two halves the test cell, which in the simplest case contains air, too wholly or partly with liquid, semi-solid or solid Fabrics can be filled.
The transpiration of the skin can be simulated, for example the half of the cell opposite the TTS le wholly or partly with water or an aqueous solution solution is filled. The water can be what if you choose one sera impermeable but water vapor permeable carrier penetrate into the TTS in the form of water vapor. Kind and order Catch this "transpiration" can be over wide the choice of the carrier film, the temperature and osmotic effective additives to the water phase can be controlled. Becomes a microporous carrier film is used, so is under the conditions even contacting the TTS with rivers water and any substances dissolved in it possible. This corresponds to sweating through the pores in of the skin.
In addition to the mechanical load, egg ne continuous release of substances from the TTS achieved become. This is made possible when facing the TTS half of the test cell with the medium completely or is partially filled, the low molecular weight ingredient fe of the TTS is able to absorb by diffusion. Type and off The diffusion can also be determined by the choice of intermediate carrier film can be influenced. To such Way z. B. the release of a low molecular weight substance, a permeation-enhancing auxiliary or a soft makers can be simulated from a TTS. In practice it can the adhesive behavior of TTS on the skin Change processes strongly and unpredictably. In the invent The inventive model can do such operations in simpler Can be linked with mechanical influences. Finally, half of the TTS Test cell filled with liquid. Then this comes in direct contact with the TTS. When using what water or dilute aqueous solutions Influences when washing, showering, bathing or swimming simulate the adhesive behavior of a TTS on the skin.
The evaluation of the experiments with the Mo shown above dell can be qualitative and quantitative. With the qua the period of time is recorded after the liturgical evaluation which the TTS has completely detached from the carrier. This In the simplest case, evaluation can be done at intervals visual observation can be realized, possibly with the exact time is not recorded.
For the exact determination of the time of the complete The model will be replaced by an optical control device tion attached or introduced. It is preferably a signal-modulated light barrier in the visible or in red area.
The test cell is aligned with the light barrier so that a completely detached TTS in the direction of gravity falls through the light beam and interrupts it.
As an alternative to an optical control device, the Test cell equipped so that it is in the direction of the gravity force falling TTS under an electrical circuit breaks or closes. To close a circuit it may be necessary to connect the TTS with at least one equip current-conducting layer made of metals or conductive polymers can be formed.
The complete detachment of a TTS from the carrier can finally also by a conductivity measurement between these two are done. The electrical resistance increases steadily increases during the detachment of the TTS in the event of complete detachment.
For a more differentiated evaluation of the tests, the quantitative recording of the detached area of the TTS before moves. This takes place at one or more times before there was a complete detachment.
The conductivity measurement mentioned above can also be used for this Purpose can be used because the conductivity between Trä ger and TTS are proportional to their contact area behaves. In this way, the degree of detachment can be continuous be grasped.
In the case of a batch process, the attempt canceled after a defined period and the TTS together with the carrier is removed from the test cell. Those parties s of the TTS that have detached from the carrier material, can be stained with a dye solution or suspension become. Where there is still a network between TTS and The dye solution, on the other hand, cannot adhere to the carrier reach the surface of the TTS or the support and this do not stain. In this way, only those can be replaced stained lots that are colored with the edge of the TTS di be in direct contact. Everywhere from remaining contact Detachments surrounded by surfaces are not detected.

In the simplest case, the TTS ( 5 ) consists of a pressure-sensitive layer ( 6 ) which sticks to the carrier film ( 4 ), as well as a backing layer ( 7 ) covering the adhesive layer ( 6 ). In the lower half-shell ( 2 ) of the test cell ( 21 ) there is a cylindrical support body ( 8 ) in a central arrangement. The free surface of this body ( 8 ) is preferably ground flat and lies in one plane with the flat ground edges ( 3 ) of the lower half shell ( 2 ).

The distance between the two contact surfaces of the Rührele element ( 9 ) on the carrier film ( 4 ) preferably corresponds to the average of the outer and inner diameter of the sheet in the form of the TTS ( 5 ). If the test cell ( 21 ) is placed on a magnetic stirring unit with a support plate, the stirring element ( 9 ) in the test cell ( 21 ) can be rotated in a circle around its center.
This is preferably done at a speed of 1 to 30 revolutions per minute. In such an arrangement, the pressing force of the stirring element ( 9 ) on the carrier film ( 4 ) from the weight of the stirring element ( 9 ) and the attractive force of the magnetic stirring unit acting on it adds up. Both the magnetic properties of the stirring element ( 9 ) and of the magnetic stirring unit thus have an influence on the test result and must be standardized.
The two contact surfaces of the stirring element ( 9 ) describe a circular path on the carrier film ( 4 ) during the course of the experiment. The center of this circular path preferably corresponds to that of the TTS ( 5 ).
In the area of the contact surfaces there is a temporary elastic expansion of the carrier film ( 4 ), which is transferred to the TTS which is pressure-sensitive adhesive on the counter surface. This load leads in the course of the experiment to a more or less extensive until the TTS ( 5 ) is completely detached from the carrier film ( 4 ).
In an alternative construction of the test arrangement, magnetic power transmission is dispensed with in favor of a mechanical one ( FIG. 3). The body loading the TTS is once again rotated by a shaft ( 10 ) fed in from the outside in the form of a dumbbell. The two end pieces ( 11 ) of the stirring element ( 9 ) are preferably freely rotatably mounted on its transverse axis ( 12 ) so that they can roll over the carrier film ( 4 ) when the stirring element ( 9 ) rotates. The penetration depth of the stirring element ( 9 ) into the carrier film ( 4 ) can be adjusted via the shaft ( 10 ).
The choice of a circular TTS ( 5 ) in connection with the central support body ( 8 ) in the lower half of the test cell results in a particularly favorable loading geometry.
In conjunction with the dumbbell-shaped stirring element ( 9 ), the TTS ( 5 ) can be loaded until it completely detaches from the carrier film ( 4 ). This detachment process is shown successively by FIGS . 6a to 6c.
In the case of a circular, full-surface TTS without a central contact surface, the load with the dumbbell-shaped magnetic stirrer only leads to a detachment of the TTS ( FIG. 7). However, this version represents a comparatively ineffective arrangement.
The circular shape of the TTS makes it full compared to one flat design continues the ratio of edge zone enlarged to inner surface. The relationship between yourself detaching and the still adhering surface also larger, so that the ability to differentiate the Model increases. Finally, the circular ring shape reduces that Probability of centered, completely within the Area of the TTS lying detachments, which is a preferred Quantification technology are not accessible.
The is of particular importance for the test result Material quality of the carrier film. Preferred materials are polymers that have a pronounced elasticity and a have good water vapor permeability and at the same time are practically insoluble in water. These properties show in particular polyurethanes and ethyl vinyl acetate Copolymers and silicone rubbers. Polyethylene and poly propylene and the block polymers styrene-isobutylene-styrene (SIS) and styrene-butadiene-styrene (SBS) are sufficient the elasticities available. Its water vapor permeable However, speed is low and not very suitable for transp ration of the skin.
Human carriers also come as carrier materials for the TTS or animal skin in question. This can be as full skin or in the form of isolated layers of skin. Such difficult to standardize biogenic carriers, however not preferred according to the invention because they are for one standardizable routine check in a conventional La boron not suitable.
When using a water-vapor-permeable carrier film ( 4 ), the transpiration of the skin can be included in the test implementation by completely or partially filling the upper part of the test cell ( 21 ) with water or an aqueous solution. This is done by first filling the lower half-shell ( 2 ) with liquid and assembling the test cell ( 21 ) upside down. Then it is turned over so that the liquid comes into contact with the carrier film ( 4 ) and completely or partially covers it. Such a structure is shown in Fig. 4. The liquid speed ( 13 ), hereinafter referred to as medium A, covers the carrier film ( 4 ) here completely. Preferred liquids are water or isotonic saline.
With an arrangement according to FIG. 4 can continue the delivery of low molecular weight Stoffan (molecular weight up to 1000 Da) are simulated from the TTS in or through the skin during wear if a carrier film (4) with suffi chender diffusibility of these substances is used. Substances are to be understood here in particular as active ingredients and permeation enhancers and plasticizers. The latter in the groups are available in large numbers for the formulation of TTS and are known to the person skilled in the art.
If these substances dissolve poorly in the purely aqueous medium A. other liquids can also be used dress. In particular, these are mixtures of water with monohydric or polyhydric alcohols, polyethylene glycols or Polyvinyl pyrrolidones. As solubilizers, anioni cal, cationic, amphoteric or nonionic surfactants be included. Also proteins, preferably serum albumin from Human or animal, can be used for this purpose.
In a further experimental arrangement, a water-insoluble film which is water-permeable due to micropores (pore diameter below 100 μm) is used as the carrier. As an example, microporous polyethylene or polypropylene films are mentioned. The pores of this film are preferably treated with a hydrophilizing agent in order to facilitate the passage of liquid water. In a structure of FIG. 3 A, the medium passes through the pores into contact directly with the TTS. This corresponds to sweating on human skin, in which liquid water together with salts from the glands of the skin are released onto the skin surface via pores. In particular, this form of water contact leads to a deterioration in the adhesive properties or detachment from the skin with the formation of a moisture film that occurs between the adhesive layer and the skin in many pressure-sensitive adhesives.
In the experimental arrangement shown in FIG. 5 can finally, the TTS during the experiment from the outside are contacted with liquid (medium B) in direct contact. Medium B ( 14 ) is preferably water or a solution of surfactants in water to simulate the effect of washing, showering, bathing or swimming on the TTS.
The test is evaluated by quantifying the detached area of the TTS in relation to the total area of the TTS. The procedure described below is shown in FIGS. 8a to 8c.
The carrier film ( 15 ) and the still adhering TTS ( 16 ) who removed the test cell at the end of the test.
The edges of the TTS are drizzled with a dye solution ( 17 ) which is able to penetrate into the cavities between the TTS and the carrier film ( 18 ), which are caused by detachment. The dye must be able to stain either the carrier film or the adhesive layer of the TTS. The back layer of the TTS (the layer of the TTS furthest away from the carrier film), on the other hand, must practically not be able to be colored with the dye in a short time (5 to 10 minutes). After a short exposure time (5 to 10 minutes), the excess dye solution is wiped off with a cloth. The back layer of the TTS is optionally cleaned briefly with the solvent of the dye solution.
The stained area beyond the area of the TTS the carrier film is removed. This is done by using die-cut carrier film is punched away. As a punching tool serves the identical tool with which the TTS originally was brought into its shape.
At the end of the procedure, an annular TTS ( 19 ) is obtained, some of which adheres to a carrier film of the same geometry. Only the detached surface parts ( 20 ) are stained by the dye and thus accessible for further evaluation.
In the simplest case, this evaluation is carried out by Fotoko the colored TTS on a scale of 1: 1 or another defined scale on a paper with uniform Basis weight. The colored areas are from the photo cut out copy and by weighing your share in of the total area of the TTS.
Compared to this imprecise and labor-intensive method is the evaluation by computer-assisted image processing preferable. The image of the stained TTS is also included by means of a commercially available scanner or by a Digital camera (CCD camera) generated and in computer readable Formed.
The ratio of the colored Area to the total area and thus the detached portion of the TTS are computer-aided. This stands for the specialist to a larger number of computer programs Available (e.g. LECO 2003 from LECO Corporation, Image Ana lysis group).
The Re obtained with experimental setup and implementation results show the high value of the test procedure and give the Ab in a surprisingly reproducible way solution phenomena of TTS on the skin again.
The backing layer preferably consisted of a transparent film (Hostaphan® RN from Hoechst). A 51 µm thick EVA film with 9% vinyl acetate (MSP 98792 from 3M) served as the carrier film. The test duration was 24 hours. The saturated solution of Sudan red 7 B in ethanol was used as the coloring solution. The evaluation was carried out using a Sony CCD color camera on a tripod with connected image processing system LECO 2003 on a conventional PC.
Im einzelnen ergaben die Versuche:
The individual tests showed:
The uncrosslinked Durotak 1051 acrylic adhesive shows one significantly larger release area than the cross-linked adhesive same type (Durotak 1052).
When n = 2, uncured adhesive Durotak 1051 and above graded crosslinker additives tested. No addition of 0.001% Al give an identical 6% detached area, with 0.01% Al the detachment is 4.5% and 0.05% Al finally at 3%. This is in line with that for the Sen adhesive known optimal crosslinking additives of 0.01 up to 0.05% Al.
Adding 1% Eudragit E100 to Durotak 1051 leads to a physical cross-linking that is already unique can be distinguished by reduced detachment.
Hotmelt formulations based on ethyl cellulose already showed extreme water absorption and almost complete detachment after 1 hour using microporous PE film as a carrier (according to FIG. 4, the upper half of the test cell was filled with water). The moisture sensitivity of these systems was already known from wearing tests. Three of these formulations were still tested on an EVA film and again under the influence of water (now only water vapor). The degree of detachment matched the experience from attempts to wear. One of the systems turned out to be particularly bad and had a special effect in the test: the initially cloudy matrix was finally transparent to glass, which demonstrated the water absorption during the test procedure.
Moisture-sensitive TTS were tested on a micro-porous PE film with and without the action of water (according to FIG. 4). Partial detachment only occurred under the influence of water and the TTS could then be easily removed from the support. The dry-fermented system, on the other hand, showed practically no detachment and was very difficult to remove manually from the carrier.
