Verfahren zur Herstellung von Moschusriechstoffen und von Zwischenprodukten
dafür, Aus der niederländischen Patentschrift 94 49o ist bekannte dass man. durch
Reaktion von gegebenenfalls alkylsubstituierten Styrolen mit bestimmten, durch mindestens
zwei Alkylgruppen substituierten Aethylenen in Gegenwart eines Kondensation,..
mittels und Einführung einer Acylgruppe mit 1 bis 3 Kohlen-Stoffatomen in den Benzolring
der so erhaltenen Verbindung wertvolle Moschusriechstoffe erhalten kann, In dieser
Patentschrift ist auch die Herstellung der bei der besagten Kondensation gebildeten,
noch nicht acylierten Zwischenprodukte er-
wähnt,, Diese werden als
Polyalkylindane bezeichnet, aber in der Beschreibung wird erwähnt, dass
diese Struktur nicht feststeht
und dass in einigen Fällen auch Isonere
dieser Polyalkylindane entstehen können,
Die Erfindung
bezieht sich auf ein neuen Verfahren zur Herstellung obiger Moschusriechstoffe
und Zwischenprodukte dafürg wobei man von denselben substituierten Styrolen
ausgeht. Dieses
Verfahren besteht darin., dann man ein Styrol
der allgemeinen For-
mel:
# in der eine der Gruppen R und R1 eine Alkylgruppe mit 1 bis
3
Kohlenstoffatomen und die andere Wasserstoff oder eine solche
Alkylgruppe darstellt, in Gegenwart eines Kondensationsmittels
mit einem 4 bis 7 C-Atome etthaltenden tert.Alkohol oder mit
einem
4 bis 7 C-Atome enthaltenden sekundären Karbinol mit verzweigter
Kohlenstoffkette kondensiert* Die dabei erhaltenen Verbindungen
entsprechen den in der niederländischen Patentschrift 94 49o
be-
schriebenen Zwischenprodukten; sie lassen sich in der dort
beschrie-
benen Weise durch Einführung einer Acylgruppe mit 1
bis 3 C-Atomen
in. die entsprechenden Moschusriechstoffe umsetzen,
Die Kondensation verläuft am besten mit den tert,Alkoiƒolen.
Bei
Anwendung sekundärer Karbinole ist die Ausbeute oft
niedriger
während die primären Alkohole im allgemeinen anders reagieren
und
dann nicht die gewünschten Verbindungen ergeben,
Bei der Kondensation der Styrolkomponente mit der Alkoholkomponente
erhält man meistens Polyalkylindane, jedoch entstehen in einigen
Fällen Polyalkyltetraline, Die als Endprodukte erhaltenen Moschus-
riechstoffe sind dementsprechend entweder oder Acyltetralin-
derivate,
Für den vorliegenden Zweck geeignete Alkohole sind tert.Butanol,
tert.
Anylalkoholg Dimethylisopropylkarblnolg Methyläthylisopropylkarbinol
und Pinacolylalkohol,
Als Styrole kommen insbesondere ac@Methylstyrolli p--ap#Dimethylstyrol
und p-Aethyl-a-methylstyrol in Betracht.
Auch andere als die vorgenannten Alkohole und Styrole sind
brauch-
bar, aber meistens werden sie durch ihren hohen Preis oder
wegen
schwerer Zugänglichkeit für die praktische Anwendung
weniger in
Betracht kommen.
Die geeignetsten Kondensationsmittel sind Gemische aus
Eisessig
und Schwefelsäure, vorzugsweise im Verhältnis 1:19 8o bis 95%ige
Schwefelsäure und SulfosKuren des Typus Methan-, Aethan- oder
Propansulfonsäure oder Gemische derselben.
Die Kondensation kann sowohl bei normaler wie erhöhter Temperatur
durchgeführt werden. Im allgemeinen kann man bei Temperaturen
von
1o bis 6o00 arbeiten, aber die besten Ausbeuten werden meistens
bei 35 bei 4o°0 erhalten. Bei Anwendung einer Alkansulfonsäure,wie
Bethansulfonsäure, ist es von Vorteil, die Temperatur etwas
höher
zu wählen, z.B. 500C.
Die Acylgruppe kann in das Polyalkylindan bzw. -tetralin nach
an
sich bekannten Methoden eingeführt werden, die in der niederlän-
dischen Patentschrift 94 49o näher beschrieben werden. Zweckmässig
stellt man die Acetylverbindungen her, und zwar dadurch, dass
das»
Polyalkylindan oder das Tetralin mit einem Acetylhalogenid
oder
mit Essigsäureanhydrid in Gegenwart von Aluminiumchlorid oder
einem
anderen Friedel-Crafts-Katalysator umgesetzt wird*
Die gemäss der Erfindung erhaltenen Verbindungen sind im allgemeinen
dieselben wie die, welche nach obigem älteren Verfahren entstehen
und für Einzelheiten über diese Verbindungen sei deshalb auf
die
Tabellen in der niederländischen Patentschrift 94 49o verwiesen*
Die Windung wird anhand naohstebjnder Beispiele erläutert,
Diese wurden in Laboratoriunsaasstab in einem Rmdbsienkolben
mit
Rührer9 Therso=eter, Kühler und Tropftrichter durchgeführt.

Eine Mischung von Zoo gEisessig und Zoo g konzentrierter
Schwefel»
säure wird in den Ruadisiwakolben eingebracht. Unter Rühren
und
Aufrechterhalten der Temperatur auf 4o100 wird innerhalb
1 Stunde
eine Mischung aus 132 g (1 1o1) p-«m.Dimethylstyrol
und 88 g (1 1o1)
tert.MWlalkohol zufliessen gelassen, Nachdem alles zugegeben
ist$
wird noch weitere 2o Minuten gerührt, Bein Stehen bildet
das Reak-
tionsgezisch spei Schichten, die in einem Scheidetrichter
voneinänder
getrennt werden, Die obere Schicht wird mit verdünnter Natronlauge
und Nasser bis zur neutralen Reaktion gewaschen. Sodann
wird unter
verminderten Druck destilliert, wobei zunächst ein Vorlauf
übergeht
und ansohliessand eine Mischung aus oa,?o % 1 #19293l3,5-Hezamethylw#
indan und aas 3o % 1 .1 .39"stranethyl-3-äthylindan als
farblose
Vl$ssigkeit tesi äp ?5,bis 8210/2«, Die Kohlenwasserstoffmischung
hat einen Erechungsindez von ap # 195o8 bis 19511,
In den Destillationkolben bleiben p-a-Dinethyletyrolpolymmere
zurück.
Beide obigen Indanderivate sind mit der siebten bzw, der
dritten
Verbindung der Tabelle II der niederländischen Patentschrift
94 49o
identisch,.
Zur Herstellung der lcetyl«farbindungen obiger Indanmischung
kann
man nach Beispiel IV der niederländischen Patentschrift
94 49o wie
folgt vertahren:
In einem mit Rührer, Tropftrichter und Gasabfuhrröhre versehenen
Kolben werden 4oö g (3,25 Mol) reines Nitrobenzol
mit 14o g (1,o5 Mol)
wasserfreiem Aluminiumchlorid verrührt, bis diese sich völlig
ge-
löst hat, Dabei wird durch äussere Kühlung mit Wasser gesorgt#
dass
die Temperatur der Mischung nicht wesentlich über 3o'0 anateigte
Durch den Tropftrichter lässt man in einem Zeitverlauf von
3 Stunden
2o4 g (1 Mol) obigen Indangemisches, 85 g (1,o8 Mol) Acetylchlorid
und Zoo g (1,63 Mol) Nitrobensol zufliessen' Während der
Reaktion
wird die Temperatur durch Kühlung mit Wasser auf oae2oe0
gehalten.,
Nachdem die Gasentwicklung aufgehört hat, rührt man eine
weitere
halbe Stunde. Die dickflüssige Masse wird nun unter kräftigem
Rühren
auf eine Mischung aus Eis und konzentrierter Salzsäure gegossen,
bis die komplexe Verbindung des Aluminiumchlorids völlig
zersetzt
ist, Das schwere 0e1 wird mit Wasser und Natriumaoetatlösung
bis
zur neutralen Reaktion gewaschen. Nach Abdestillieren des
Nitro-
benzols, von dem 385 g zurückgewonnen werden, erhält man
ca.2oo g
einer Fliäasigkeit, die bei 116 bis 126'%,7 mmNg
und aus
einer Mischung von 6-Acetyl-1,1,2,3,3,5-hexamethylindan
und 6-Acetyl-
1,1,3,5-tetramethyl-3-äthylindan besteht, Beim Stehen wird
dieses
Produkt grösstenteils fest.
Diese beiden Acetylverbindungen entsprechen der siebten
bzw: der drit-
ten Verbindung der Tabelle I der niederländischen Patentschre94
49o.
Beispiel 2 't
Gemäss Beispiel 1 stellt man., ausgehend von 132 g (1 Mol)
p-cu-Dime-
thylotyrol und 1o2 g (1 Mol) Dimet#ylisopropylkarbinol oder

colylalkohol, 1,1,3 ,4,4,6-Hexamethyltetralin dar' Der rohe
Kohlen-
wasserstoff bei 92 bis 1oo°C/2 mm und wird bei längerem
Stehen grösstenteils:bste Näch Abnutachen und Uakristallisieren
aus Aethanol kann man daraus das Reinprodukt in Form von
bei 65-
bis 660C schmelzenden Kristallen erhalten, Das Dimethylisopropyi#
karbinol ergibt eine höhere Ausbeute als Pinacolylalkohol&
Die erhaltene Verbindung ist identisch mit der elften Verbindung
der Tabelle II der niederländischen Patentschrift 94 49o!
wo dieser
Stoff allerdings irrtümlich als ein Indanderivat bezeichnet
wird,
Die Acetjrlverbindung dieses Tetralinderivats kann man in
der im
Beispiel 1 beschriebenen Weise herstellen, Das so erhaltene
1,1,3,
4,4,6..hexamethyltetralin ist eine feste Substanz vom Kp
1190C/o,2em
und vom Schmelzpunkt 55,5 bis 56'C0
Die Formylverbindung dieses Tetralinderivates kann nach
Beispiel YII
der niederländischen Patentschrift 94 49o wie dolgt hergestellt
werden:
In einem mit Rührer, Gaseinfuäärohr, Thermometer und Liebig-Kühler
versehenen Kolben suspendiert man 18,6 g (o,14 Mol) wasserfrei6a
Zinkchlorid und 22,4 g (o975 Mol) Paraformaldehyd in 25o
g (1,15 Mol)
1,1,3j4,4,6-Hexamethyltetralin, Nach Erwärmen der Masse
auf 9o00
leitet man unter Rühren einen kräftigen Staom trockenes
HC1-Gas ein,
wobei die Temperatur zwischen 88 und 9200
gehalten wird.
Die erste Stunde wird das HCl-Gas rasch absorbiert, Um die
Reaktion
vollständig zu Ende zu führen, muss noch weitere 2 Stunden
Gas ein-
gileitetaerden; die Einleitegeschwindigkeit kann dabei wesentlich
herabgesetzt werden, Nach Kühlung werden die Schichten in
einem
Scheidetrichter voneinander getrennte Nachdem die obere
Schicht mit
Wasser und verdünnter Natronlauge bis zur neutralen Reaktion
ge-
wasohen ist, destilliert man unter vermindertem Druck. Dabei
geht
zunächst ein Vorlaufaus unveränderten Hexamethyltetralin
ikber,
nodann das Ohlormethyl-1,1,3,4,4j6-Hexamethyltetralin. Diese
Sub-
stanz siedet bei 121 bis 13o'%,5 an und schmilzt, nach Unkri-
,stallisieren aus Aethanol, bei 91 bis 93#0.
42 g (o,16 Mol) der so erhaltenen Verbindung werden
2 Stunden mit
44 g (o,31 Mol) Hexamethylentttramin, 65 ca3 Eiseseig und
65 033
Wasser am Rüokflusskühler gekocht. Man kühlt danach ein
wenig ab,
setzt 25 cm 3 36%ige Salzsäure zu undkocht wieder während
2o Minuten.
Nach Abkühlen und Verdünnen mit Wasser extrahiert man mit
Benzol
Die Benzollösung wird danach mit Wasser und Bicarbonatlösung
bis zur
neutralen ]Reaktion gewaschen. Nach Abdestillieren des Benzols
frak-
tioniert Man in Vakuum, wobei man 29
g Aldehyd vom Siedepunkt 125
bis 14o°0/1 um erhält, Durch Umkristallisieren aus einen
Methanol-
Aethanol-Gesisch wird das Formyl-1,1,3,4,4,6-hexamethyltetralin
weiter gereinigt. Es fällt in Form von farblosen Kristallen
von
Schmelzpunkt 91,5 bis 93°C-ane
Der in dieser Weise erhaltene Moschusriechetoff ist mit
der zwölften
Verbindung der Tabelle I der niederländischen Patentschrift
94 49o
identisch; dort wurde jedoch irrtümlichwrweise die Indanstruktur
angegeben,
Beispiel -
Mm verfährt wie in Beispiel 1, jedoch unter Ersatz des tertelmyl#.
alkohols durch ?4 g (1 Mol) tert.Butanole Man erhält in
dieser Weise
1 ..1,3,3,5-Pentamethyliodan in Form einer farblosen Flüssigkeit,
die
bei 62 bis 72'C/2 au und dieach Fraktionieren das reine
Indan
von Siedepunkt 67°0/2 mm und den Brechungsindex aß o
= 1,5o3 bis
19505 ergibt.
Die erhaltene Substanz ist die zweite Verbindung aus der
Tabelle II
der n2derländischen Patentschrift 94 49o.
Beis Acetylieren dieser Verbindung in der in Beispiel 1
beschriebenen
Weise erhält man 6-Acetyl-1,1,3,3,5-pentamethylindan vom
Siedepunkt
112 bis 116'G/1 mm. Nach Unristallisieren aus Aethanol ist
der
Schmelzpunkt 6o bis 61'C. Hie Substanz entspricht der zweiten
Ver#.
bindung der Tabelle I der niederländischen Patentschrift
94 49o,
Beispiel 4
Man verfährt wie in Beispiel 1, jedoch unter Ersatz des
p-«,»Dimethyl.
st'rols durch 146 g (1 Mol) p-Aethyl##«-methylstyrol,
Man erhält 1,1,
2,393#Fentasethyl-5-äthylindan als farblose Flüssigkeit,
die bei
85 bis 97'G/2 mm kocht. Bei weiterer Reinigung erhält
man ein Produkt
von Siedepunkt 92'G/2 ms und den Brechungsindex aDo
= 1,5o6. Diese
Substanz ist mit der neunten Verbindung aus Tabelle II der
Patent.
Schrift 94 49o identisch,
Das erhaltene Indanderivat kann in der iri Beispiel 1 beschriebenen
Weise acetyliert w.&rden. Man erhält dann das 6-Acetyl##1,1,2,3,3»penta-
methyl#»5-äthylindan, das in reines Zustand bei 141°G/2
mm und
bei 74 bis 75'G schmilzt und der zahnten Verbindung der
Tabelle I
in der niederländischen Patentschrift 94 49o entspricht,
Beispiel 5
146 g(1 Mol) p-Aethyl-a-methylstyrol werden in der im Beispiel
1 be-
schriebenen Weise mit 'log g (1 Mol) Dimethylisopropylkarbinol
oder
Pinacolylalkohol umgesetzt. Man erhält den Kohlenwasserstoff,
der
nach Fraktionieren bei 1o6 bis 1o8#0/2 mm und den Brechungs-
index n# o # 1i515 bis 1,517 besitzt und sich als
das 1,1*3,4,4»
P»tamethyl-6-äthyltetralin erweist, Diese Substanz ist mit

Die Verbindung wir«in der in Beispiel I angegebenen Weise
acetylierti
wobei 7-Acetyl-1 i1 93.4,4-pentamethyl-6##äthyltetralin
erhalten wird.
Dieses bei 128 bis 133°0/095 mm und schmilzt nach Umkristalli-
sieren aus Aethanol bei 6o bis 61'0.
Die Verbindung entspricht der fünfzehnten Verbindung der
Tabelle I
der niederländischen Patentschrift 94 49o, wo. jedoch wieder
irrw.
tümlicherweise die Indanstruktur angegeben wird.
Beispiel i
Man verfährt wie in Beispiel 19 jedoch unter Ersatz des
p-CO.Dimethyl»
styrols durch 118 g(1 Kol) a4-Het»lstyrol. Es zeigt sich,
dass das
Reaktionsprodukt im wesentlichen aus 1i1 9293,3-Pentamethylindan
be-
steht. Die Flüssigkeit bei 63 bis 72°0/2 mm. Durch eine
zweite
fraktionierte Destillation erhält man die reine Verbindung

Die erhaltene Substanz entspricht der fünfzehnten Verbindung

Durch Acetlyieren dieses Indans in der in Beispiel 1 beschriebenen

se erhält man 5-kcetyl-1919293#3-pentamethylindan von Siedepunkt

Man kann auch in der in Beispiel 2 beschriebenen Weise das

und entspricht der zwanzigsten Verbindung der Tabelle I

In der in Beispiel 1 beschriebenen Weise werden 118 g (1
Mol) oc.-Methyl-
styrol mit 74 g (1 Mol) tert.Butanol umgesetzt, Man erhält

393-Tetramethylindan in Form einer farblosen Flüssigkeit,

Beispiel 8
In der in Beispiel 1 beschriebenen Weise werden 118 g (1
Mol)
a-Methylstyrol mit 1o2 g (1 Mol); Dimethylisopropylkarbinol
oder
ginocolylalkohol umgesetzt. Man erhält ein Gemimch aus Kohlenwasser-
stoffen, das im wesentlichen aus 1,1,2,4,4-Pentamethyltetralin
besteht.
Es ist eine Flüssigkeit, die bei 81 bis 91'C/2 am kochiiGad
einen
Erechungeindex nDo # 1,514 bis 1,517 hat*
Diese Verbindung ist die
sechzehnte Verbindung der Tabelle II der niederländischen
Patent-
schrift 94 49o, die dort zu Unrecht als ein Indanderivat
bezeichnet
worden ist.
Durch koetylierung in der im Beispiel 1 beschriebenen Weise
erhält
man aus diesem Tetralin das Aoetyl-1,1,2,4,4-pentamethyltetralin
von
Siedepunkt 140'C/2 am und Brechungsindez aDo #
1,534 bis 1,538. Die
Substanz besteht aus eines Gemisch von zwei Isomeren und
entspricht
der einundzwanzigsten Verbindung der Tabelle I
der niederländischen
Patentschrift 94 49o., die dort wieder als Aoetylindan angegeben
worden ist,
In. Beispiel 1 und allen weiteren, sich darauf beziehenden
Beispielen
wurde die Kondensation des Styrols mit dem tertiären oder
sekundämren
Alkohol unter Anwendung eines Gemisches aus Zoo g Eisessig
und Zoo g
konzentrierter Schwefelsäure als Kondensationsmittel ausgeführt.
In e11 diesen Beispielen kann man statt dieses Gemisches
auch 4oo g
85%ige Schwefelsäure, 4oo g 9o%ige gethansulfonsäure oder
4oo g
eines 4 bis 8 9I6 Wasser enthaltenden Gemisches aus Methan-,
Aethan-
und 1'ropansulfonsäure verwenden. In den letzten zwei fällen
hat es
Vorteile, eine etwas höhere Reaktionstemperatur, vorzugsweise

50'C, anzuwenden.
