Aus der Patentschrift 926 248 ist ein Verfahren zur Herstellung von tuberkulostatisch
wirksamen Abkömmlingen des Isonicotinsäurehydrazids bekannt, das darin besteht,
daß p-Methylmercaptopropionaldehyd nach an sich bekannten Methoden mit Isonicotinsäurehydrazid
kondensiert und das entstandene jB-Methylmercaptopropylidenhydrazid gegebenenfalls
durch Behandlung mit einem Oxydationsmittei in das entsprechende Sulfoxyd übergeführt

pharmakologischen Eigenschaften aufweist. Die neue Verbindung stellt ein Sulfoxyd
dar, das sich von dem aus der Patentschrift 926 248 bekannten Sulfoxyd durch
eine
längere Seitenkette unterscheidet und die folgende Strukturformel besitzt:
Sowohl gegenüber dem einfachen Isonicotinsäurehydrazid als auch gegenüber den nach
Patentschrift 926 248 erhältlichen schwefelhaltigen Derivaten zeichnet sich die

Gegenstand der vorliegenden Erfindung ist dementsprechend ein Verfahren
zur Herstellung eines schwefelhaltigen Isonicotinoylhydrazons, das darin besteht,
daß man nach an sich bekannten Methoden Isonicotinsäurehydrazid mit ß-Äthylthio-äthylthioacetaldehyd
zum entsprechenden Hydrazon kondensiert und die erhaltene, zwei Thioäthergruppen
enthaltende Verbindung durch Behandlung mit einem Oxydationsmittel in das entsprechende
Disulfoxyd überführt oder daß man B-Äthylthio -äthylthioacetaldehyd durch Behandlung
mit einem Oxydationsmittel in das entsprechende Disulfoxyd überführt und dieses
mit Isonicotinsäurehydrazid zum Isonicotinsäure- (iß-äthylsulfin- äthylsulfinäthyliden)
-hydrazid kondensiert. Der als Ausgangsmaterial benötigte ß-Äthylthio-äthylthioacetaldehyd
wird z. B. durch Umsetzung von Bromacetal mit dem entsprechenden Mercaptan hergestellt.
Der so erhaltene schwefelhaltige Aldehyd kann direkt mit Isonicotinsäurehydrazid
kondensiert und das gebildete Hydrazon durch Behandlung mit einem geeigneten Oxydationsmittel
in das zugehörige Disulfoxyd übergeführt werden. Es ist jedoch auch möglich, die
Oxydation zum Disulfoxyd vor der Hydrazonbildung vorzunehmen, wonach ß-Äthylsulfin-äthylsulfinacetaldehyd
mit Isonicotinsäurehydrazid kondensiert wird. Die Um-

in 1250 ccm Alkohol werden 188 g 30%iges Hydroperoxyd eingetragen, wobei die Temperatur
des Gemisches nicht über 50°C ansteigen soll. Nach 3stündigem Rühren bei Raumtemperatur
und Stehen über Nacht wird im Vakuum eingedampft. Durch Umkristallisieren aus absolutem
Alkohol erhält man das Isonicotinsäure-(p-äthylsulfin-äthylsulfinäthyliden)-hydrazid

wird unter Rühren eine Lösung von 84,5 g Isonicotinsäurehydrazid in 425 ccm Wasser
zugefügt. Es wird noch eine weitere halbe Stunde gerührt und dann im Vakuum eingedampft.
Der Rückstand liefert beim Umkristallisieren aus 850 ccm
absolutem Alkohol 135 g
Isonicotinsäure-(ß-äthylsulfin-äthylsulfin-äthyliden)-hydrazid in farblosen Kristallen
vom Schmelzpunkt 157 bis 158°C. Example 2 In a solution of 101 g of fl-ethylthio-ethylthioacetaldehyde
